C-Met Initiates Epithelial Scattering Through Transient Calcium Influxes And Nfat-Dependent Gene Transcription, Peter R. Langford
Theses and Dissertations
Hepatocyte growth factor (HGF) signaling drives epithelial cells to scatter by breaking cell-cell adhesions and migrating as solitary cells, a process that parallels epithelial-mesenchymal transition. HGF binds and activates the c-Met receptor tyrosine kinase, but downstream signaling required for scattering remains poorly defined. This study addresses this shortcoming in a number of ways.A high-throughput in vitro drug screen was employed to identify proteins necessary in this HGF-induced signaling. Cells were tested for reactivity to HGF stimulation in a Boyden chamber assay. This tactic yielded several small molecules that block HGF-induced scattering, including a calcium channel blocker. Multimodal sensory input directs simple and complex behaviors in animals. Most research to date has been limited to studies of individual senses rather than multiple senses working together, leading to important advances in our comprehension of the sensory systems in isolation, but not their complementary and alternative roles in difficult behavioral tasks, such as feeding. In the marine environment, a prey item might emit an odor, create a hydrodynamic disturbance, such as from gill movements or swimming, be visible to the predator, produce a sound, and/or produce a weak electrical field. Vasodilation of lower leg arterioles is impaired in animal models of chronic peripheral ischemia. In addition to arterioles, feed arteries are a critical component of the vascular resistance network, accounting for as much as 50% of the pressure drop across the arterial circulation. Despite the critical importance of feed arteries in blood flow control, the impact of ischemia on feed artery vascular reactivity is unknown. At 14 days following unilateral resection of the femoral–saphenous artery–vein pair, functional vasodilation of the profunda femoris artery was severely impaired, 11 ± 9 versus 152 ± 22%. Peripheral Artery Disease is a very serious disease characterized by an arterial occlusion due to atherosclerotic plaques. In response to an arterial occlusion, arteriogenesis occurs, causing smooth muscle cells to transition from a contractile to synthetic state. Also following an arterial occlusion, functional impairment was seen in the collateral circuit. An immunofluorescence protocol was developed in order to assess the impact of collateral enlargement (arteriogenesis) on smooth muscle phenotype at various time points. Long, uninterrupted bouts of sedentary behavior are thought to negatively influence insulin sensitivity, and may impact metabolic function regardless of adherence to general physical activity guidelines. The purpose of this study was to determine the combined effect of physical activity (1 h continuous exercise v. intermittent exercise throughout the day) and meal consumption on glucose excursions, insulin secretion, and appetite markers in obese individuals with prediabetes. Because of their simple fruiting bodies consisting of one to a few spores atop a finely tapering stalk, protosteloid amoebae, previously called protostelids, were thought of as primitive members of the Eumycetozoa sensu Olive 1975. The studies presented here have precipitated a change in the way protosteloid amoebae are perceived in two ways: (1) by expanding their known habitat range and (2) by forcing us to think of them as amoebae that occasionally form fruiting bodies rather than as primitive fungus-like organisms. Proper autonomic regulation of mammalian cardiac function is dependent upon very complex and precise communication among the intracardiac ganglia and individual neurons within the ganglia. An array of neuromodulators is found within the ganglia that direct neuronal activity by modulating the movement of calcium. The current study determines that opioidergic agonists, which have been found to contribute to severe cardiac disease states and intracellular calcium mobilization, are also responsible for changes in the function of the intracardiac neuron via their effects on store-operated calcium channels (SOCs).

Introduction: Chronic ankle instability (CAI) is a common consequence of lateral ankle sprains and a leading cause of post-traumatic ankle osteoarthritis. Despite a history of a lateral ankle sprain, there are some individuals (copers) who appear to have a mechanism that allows them to avoid recurrent lateral ankle sprains and giving way episodes. Muscular and respiratory V’O2-kinetics are influenced by many factors. At cellular level muscular V’O2 (V’O2musc) is affected by O2-delivery and O2-consumption. Further, these factors can be divided into muscle perfusion, capillary density, enzyme activity, mitochondrial proliferation, muscle fiber composition and some more.
Focusing on the difference between V’O2resp and V’O2musc the venous blood volume, the venous O2-stores and the dynamics of cardiac output can be identified as influences. Introduction: The hypertrophic response to overload in fast-twitch skeletal muscle is impaired in aged humans and rats, and impaired protein synthesis pathway activation is hypothesized to be a contributing factor. Muscle growth occurs when protein synthesis exceeds protein degradation. Dietary supplementation of the essential amino acid leucine has been shown to enhance protein synthesis in both young and aged skeletal muscle. Leucine acts in part by activating mammalian target of rapamycin (mTOR; a key upstream regulator of protein synthesis pathways) as well as by attenuating the activation of 5'-AMP-activated protein kinase (AMPK; a negative regulator of mTOR and protein synthesis). …

Progressive, resistance exercise training (RT) induces skeletal muscle hypertrophy, increases strength, power, and quality of muscle, and is potentially the most promising method to regenerate and re-grow muscle in populations suffering from involuntary atrophy. Multivitamin use is increasingly prevalent in the US among most every population. Yet, previous research noted no performance benefits among individuals following a supplementation period. This study focused on the potential physiological benefits from a three week multivitamin supplementation period using 24 (20.9 ± 2.6yr) aerobically untrained college aged students. Subjects were divided equally into three groups (placebo = PL, multivitamin = MV, control = CL) and asked to perform separate eight minute bouts of exercise, consisting of six min of moderate (60%VO2max) intensity followed immediately by two min of high (85%VO2max) intensity exercise on a cycle ergometer. Osteoporosis is a disease that affects many older individuals, resulting in a decrease in bone density. Involvement in impact loading sports has been associated with greater stiffness index (SI) of the calcaneus when compared to non-impact sports and abstinence from sports. However, comparisons between the effectiveness of impact and non-impact exercise equipment on calcaneal SI is less clear. These programs are intended for teenage athletes in various sports who have the potential of becoming elite athletes. The sports academia programs make it easier for these student athletes to focus on training for their sport while attending high school, and thereby, enable them to get a good education and enhance their athletic potential. Sodium bicarbonate (NaHCO3) is a nutritional aid that is proposed to enhance performance by reducing the state of exercise induced acidosis, however research is currently equivocal. Despite this, most research still focuses on its effects upon performance, and not whether the reduced acidosis impacts upon any other factors related to exercise. Athlete’s muscle function during Dragon Boat practice requires activation of a complex kinetic chain (composed of a great number of muscles of all sizes, starting from dorsal muscles, abdominal muscles and muscles of the limbs). The Dragon Boat competition includes, during the sporting year, races with length spanning from about 40 seconds (200 meters) up to 10 minutes (2000 meters) and beyond, in specific distances (long distances).

Dragon Boat is an activity that requires a participation of a large amount of muscles. The muscle groups involved are in order of involvement: the dorsal and abdominal muscles, biceps and triceps in the upper limbs and femoral quadriceps and gastrocnemius in the lower limbs. The athlete’s muscle building is based on strengthening exercises with weight lifting that involves both upper and lower limbs, with a particular attention to the heart. Children’s approach to Kayak as a sport takes place at about the age of ten. The winter weather conditions of the Northern Italian valley do not usually permit training at the river for a long period of time. Additionally the weather increases the risk of the boat to turn over in case of imperfect control. For this reason we have elaborated a training scheme in which going out on the boat starts in April when the temperature (both of the water and the atmosphere) begins to raise.
