Drug delivery is the process of transporting a drug to its site of action. It includes controlled delivery, where the drug is released at the tissue in a regulated manner. Additionally, it includes targeted delivery, where the drug is aimed at a specific tissue or region to avoid unwanted side effects (that would occur if the drug were distributed throughout the body). A cellulose acetate phthalate coated chitosan bead was developed as a formulation for controlled and targeted release of dexamethasone to the ilium and colon. Manganese-enhanced magnetic resonance imaging (MEMRI) rose to prominence in the 1990s as a sensitive approach to high contrast imaging. Following the discovery of manganese conductance through calcium-permeable channels, MEMRI applications expanded to include functional imaging in the central nervous system (CNS) and other body systems. MEMRI has since been employed in the investigation of physiology in many animal models and in humans. Here, we review historical perspectives that follow the evolution of applied MRI research into MEMRI with particular focus on its potential toxicity. Lionfish (Pterois spp.), an invasive species native to the Indo-Pacific, have permanently established themselves throughout the greater Caribbean, Gulf of Mexico, and regions of the Western Atlantic ranging from as far north as North Carolina to central Brazil. As their fundamental range expands, lionfish threaten to migrate into estuarine environments as they have been found to tolerate low salinities and an eclectic range of temperatures. The physiological capacity of invasion was assessed by quantifying the visual ecology of lionfish utilizing corneal electroretinography (ERG) as well as their metabolic scope and hypoxia tolerances under various temperature-oxygen-regimes utilizing intermittent-flow respirometry. According to the World Health Organization, drowning fatalities are a neglected public health issue ranked as the third most frequent cause of unintentional death by injury. Data used to record drowning incidents are acknowledged as inaccurate. Recommendations on drowning prevention based on these data are currently not informed by accurate, detailed and reliable evidence. This dissertation aimed to explore the usefulness of using force derivatives for onset detection in the isometric mid-thigh pull test. First, we examined applications of three differential calculus principles, first and second derivative, and curvature using visual detection as a reference under different baseline conditions. Second, we compared the best derivative method to a threshold-based method using visual detection as a reference. Results of our first investigation showed trivial differences between many differential calculus methods and visual detection. Bacterial populations grow clonal populations; however, individual cells have a variety of phenotypes. The physiological heterogeneity observed in populations has been attributed to variations in the processes of gene expression. For example, promoter expression has been shown to be heterogeneous within a population and contribute to increased stress tolerance in a subpopulation of cells. In comparison to transcription, the influence of translation on single cells is unclear. In this study, my collaborators and I have developed a dual-fluorescence reporter that allows us to measure the mistranslation rate in single cells in vivo. Postsurgical and neuropathic pain are each clinically common, and often associated with ongoing pain. Ongoing pain has been linked to ongoing activity (OA) in human C-fiber nociceptors. Preclinical studies using rodent neuropathic models have concentrated on allodynia driven by OA generated in non-nociceptive Aβ fibers, but little attention has been paid to postsurgical pain in sham controls or to C-fiber nociceptor OA promoting ongoing pain.

Gastropods are one of the most imperiled groups of animals in North America, yet there are major gaps in the literature pertaining to pollutants and climate change, and especially sublethal impacts. This dissertation assesses the effects of climate warming and unionized ammonia (NH3), one of the most abundant water pollutants, on the behavior and physiology of two caenogastropod snails: fine-ridged elimia (Elimia semicarinata) and Shawnee rocksnails (Lithasia obovata) (Gastropoda: Neotaenioglossa: Pleuroceridae). Methanogens, some of Earth’s most primitive prokaryotic organisms, are candidates for possible life forms capable of inhabiting Mars. Specifically, four different species (Methanobacterium formicicum, Methanococcus maripaludis, Methanosarcina barkeri, Methanothermobacter wolfeii) were analyzed for their tolerance to the presence of illite clay. Illite is a crystalline mineral that has been identified from regions of Mars’s surface. Results indicated that all four species grew with some success in the illite at different concentrations. With temperatures rising globally, assessing the possible impacts of the changing climate becomes more and more urgent. Ectotherms are excellent indicators of potential climatic ramifications on biodiversity because of their heavy reliance on the environment for their thermoregulation. Studies have historically looked at thermal tolerance values to establish predictive models for population and species extinctions.

Many key factors – including but not limited to – sleep, nutrition, travel, stress, and practice influence the optimization of athletic performance. Although previous studies have investigated the use of wearable technology in sport to track several such factors, peer-reviewed research specific to WHOOP technology is limited at best. The purpose of this study was to examine the impact of WHOOP technology on sleep, recovery, and batting performance in healthy NAIA baseball players. PURPOSE: To examine the influence of three distinct training phases on isometric mid-thigh pull (IMTP) measures in well-trained weightlifters. METHODS: Pre- and post-block IMTP data from 11 collegiate weightlifters was used for analysis. The mean of the best two attempts from each athlete for measures of PF and RFD from 0-50ms, 0-100ms, 0-150ms, 0-200ms, and 0-250ms were used for comparison. In total, results from five timepoints for each of the 11 athletes were examined in order to compare the effects of the three training phases. Seventeen strength and power athletes (n = 11 males, 6 females; height: 177.5 ± 7.0 cm, 165.8 ± 11.4 cm; body mass: 90.0 ± 14.1 kg, 66.4 ± 13.9 kg; age: 30.6 ± 10.4 years, 30.8 ± 8.7 years), who regularly performed weightlifting movements during their resistance training programs, were recruited to examine the effect of altering body posture and barbell position on the between-session reliability of force-time characteristics generated in the isometric mid-thigh pull (IMTP). High-density lipoprotein (HDL) have long been studied for their protective role against cardiovascular diseases, however recently relationship between HDL and cancer came into focus. Several epidemiological studies have shown an inverse correlation between HDL-cholesterol (HDL-C) and cancer risk, and some have even implied that HDL-C can be used as a predictive measure for survival prognosis in for specific sub-population of certain types of cancer. Abdominal aortic aneurysms (AAA) are characterized by extensive extracellular matrix (ECM) fragmentation and inflammation. However, the mechanisms by which these events are coupled thereby fueling focal vascular damage are undefined. Here we report through single-cell RNA-sequencing of diseased aorta that the neuronal guidance cue netrin-1 can act at the interface of macrophage-driven injury and ECM degradation. Netrin-1 expression peaks in human and murine aneurysmal macrophages. Targeted deletion of netrin-1 in macrophages protects mice from developing AAA. Through its receptor neogenin-1, netrin-1 induces a robust intracellular calcium flux necessary for the transcriptional regulation and persistent catalytic activation of matrix metalloproteinase-3 (MMP3) …

The last decade has seen a sharp increase in the number of scientific publications describing physiological and pathological functions of extracellular vesicles (EVs), a collective term covering various subtypes of cell-released, membranous structures, called exosomes, microvesicles, microparticles, ectosomes, oncosomes, apoptotic bodies, and many other names. However, specific issues arise when working with these entities, whose size and amount often make them difficult to obtain as relatively pure preparations, and to characterize properly. The International Society for Extracellular Vesicles (ISEV) proposed Minimal Information for Studies of Extracellular Vesicles (“MISEV”) guidelines for the field in 2014. Subthalamic (STN) deep brain stimulation (DBS) is an established treatment to alleviate the appendicular motor symptoms of Parkinson's Disease (PD). Current steering during DBS allows the unequal fractionation of current between two electrodes on the lead, resulting in a non-spherical electrical field. It is hypothesized that the way the electrical field is shaped will affect a patient’s upper limb symptom alleviation. Seven PD patients who underwent bilateral STN-DBS were tested over four weeks post-operation. 16 current fractionation settings were tested each week at an amplitude that increased weekly. Patients with end stage renal disease (ESRD) suffer high mortality from arrhythmias linked to fibrosis, but are contraindicated to late gadolinium enhancement magnetic resonance imaging (MRI). We present a quantitative method for gadolinium-free cardiac fibrosis imaging using magnetization transfer (MT) weighted MRI, and probe correlations with widely used surrogate markers including cardiac structure and contractile function in patients with ESRD. In a sub-group of patients who returned for follow-up imaging after one year, we examine the correlation between changes in fibrosis and ventricular structure/function. Introduction: The alignment of a prosthesis is clinically determined based on observations by clinicians and the subjective perception of amputees during gait. However, this process has been reported to be unreliable. Socket reaction moment has been reported to be significantly impacted by alignment changes, but the impact of these alignment changes on other gait parameters is unclear. The aim of this study was to investigate the effects of coronal alignment changes of a transtibial prosthesis on socket reaction moment, spatiotemporal parameters, and perceptions of alignment during gait in amputees.

A Diversity, Epidemiology and social justice unit was incorporated into the Physiology and Disease curriculum in Fall 2016. Students discussed topics such as social and cultural influences on diversity thinking, and selective treatment in hospitals based on race. It was very noteworthy that students were united in their thinking regarding diversity and inclusion, despite the fact that they were from different backgrounds and diverse cultures. In 2017, additional discussions were held on equity in education. Students who were otherwise reserved were encouraged enough to express their views, and everyone appreciated the fact that they had been included.

Background: Magnetophosphenes are among the most reliably reported effects resulting from magnetic induction. The frequency dependence of the perception threshold is crucial, as guideline agencies use this information to set exposure limits whose purpose is to protect public and workers.

Objective:Establish the magnetophosphene perception thresholds throughout the extremely low frequency range (0-300 Hz) and evaluate the use of EEG as a biomarker.

Hypothesis:Perception thresholds will be lowest at ~30 Hz. EEG occipital alpha power will decrease upon perception.

Physiology and Disease is a Biology elective at IMSA that has been developed to be mostly student-centered. Some examples of student projects include modeling heart structure to reflect function and creating LED arduino monitors to measure heart rate. Students also measure their lung capacity and blood pressure to demonstrate correlation of these values with heart rate, and trace the correlation back to neuronal controls. Projects such as these integrate other disciplines such as engineering and conform to NGSS Science and Engineering standards and NGSS Cross cutting Concepts standards. Students take responsibility for their own learning and articulate better on tests.

Menopause is associated with adverse changes in cardiovascular disease risk factors. A reduction in estrogens is most commonly associated with changing cardiovascular disease risk; however, recent observations suggest that the increase in follicle stimulating hormone that accompanies menopause may also influence risk, potentially through its influence on lipid levels. The changes in cardiovascular disease risk factors may adversely affect endothelial cell function, a pre-clinical marker for cardiovascular disease. Whether cardiorespiratory fitness is protective of endothelial health in this population, thereby mitigating the changes in risk that accompany menopause, is unclear. Monitoring the resistance training volume load (VL) (sets × reps × load) is essential to managing resistance training and the recovery–adaptation process. Subjects: Eight trained weightlifters, seven of which were at national level, participated in the study. Methods: VL was measured both with (VLwD) and without (VL) the inclusion of barbell displacement, across twenty weeks of training, in order to allow for comparisons to be made of these VL calculating methods. Whilst the phenomenon of an electrical resting membrane potential (RMP) is a central tenet of biology, it is nearly always discussed as a phenomenon that facilitates the propagation of action potentials in excitable tissue, muscle, and nerve. However, as ion channel research shifts beyond these tissues, it became clear that the RMP is a feature of virtually all cells studied. The RMP is maintained by the cell's compliment of ion channels. Transcriptome sequencing is increasingly revealing that equally rich compliments of ion channels exist in both excitable and non-excitable tissue. The giant acorn barnacle, Balanus nubilus, is a resident of the subtidal and low intertidal rocky shoreline on the Pacific Coast of North America (Alaska to Baja California). B. nubilusis notable for having the largest muscle fibers in the animal kingdom; fiber diameters that can exceed 3mm in adults! At such extreme sizes these muscle cells may be at risk for insufficient oxygen delivery to mitochondria owing to low SA:V ratios and long intracellular diffusion distances. Behind only Alzheimer’s disease, vascular contributions to cognitive impairment and dementia (VCID) is the second most common cause of dementia, affecting roughly 10–40% of dementia patients. While there is no cure for VCID, several risk factors for VCID, such as diabetes, hypertension, and stroke, have been identified. Elevated plasma levels of homocysteine, termed hyperhomocysteinemia (HHcy), are a major, yet underrecognized, risk factor for VCID. B vitamin deficiency, which is the most common cause of HHcy, is common in the elderly. The purpose of this analysis was to construct a preliminary scale of reference values for reactive strength index-modified (RSImod). Countermovement jump data from 151 National Collegiate Athletic Association (NCAA) Division I collegiate athletes (male n = 76; female n = 75) were analyzed. Using percentiles, scales for both male and female samples were constructed. For further analysis, athletes were separated into four performance groups based on RSImod and comparisons of jump height (JH), and time to takeoff (TTT) were performed. RSImod values ranged from 0.208 to 0.704 and 0.135 to 0.553 in males and females, respectively. Males had greater RSImod …

Sepsis is a life-threatening inflammatory condition with high morbidity and mortality rates. Though improvements to diagnosis and management of sepsis have been made, the complexity of the disease process and an incomplete understanding of its endpoint mechanisms have prevented major breakthroughs for early diagnosis and treatment. Representing a common endpoint in a number of inflammatory injuries including sepsis, endothelial barrier dysfunction causes fluid leakage and leukocyte infiltration that leads to tissue damage and multiple organ failure. Therefore, elucidating mechanisms of endothelial barrier regulation is needed to further develop targeted therapies in inflammatory disease.
