Introduction: There is generally a concerning likelihood of burnout in healthcare workers. Given the impact of COVID-19 on healthcare workers, our institution identified the need for wellness interventions to foster adaptive functioning and mitigate burnout. The purpose of this pilot project was to assess the feasibility of virtual holistic interventions like meditation, art, laughter therapy and dance and their impact on overall well-being of physicians and advanced practice providers (APPs).

Methods: A series of 12 virtual sessions (art therapy, dance therapy, mindfulness-based practices/meditation and laughter therapy) were offered to providers over a 6-month period. Participants completed an online survey reporting …

Introduction. Accumulating evidence indicates that exposure to electronic cigarettes (e-cigs) promotes sympathetic dominance and electrophysiologic instability in the heart, potentially culminating in arrhythmogenesis. E-liquids contain various formulations of nicotine and flavorings, but the effects of specific e-liquid constituents and their concentrations on e-cig-induced autonomic imbalance and electrical dysfunction are unresolved. To that end, we tested the hypothesis that e-cigs modify cardiac autonomic balance and ventricular arrhythmogenesis in mice dependent on e-liquid constituent type and concentration. Methods. Background: Recent technological advances allow for the repeated sampling of real-time data in natural settings using electronic ecological momentary assessment (eEMA). These advances are particularly meaningful for investigating physical activity, sedentary behavior, and sleep in young adults who are in a critical life stage for the development of healthy lifestyle behaviors.

Objective: This study aims to describe the use of eEMA methodologies in physical activity, sedentary behavior, and sleep research in young adults.

Werner syndrome (WS) is an autosomal recessive disorder that results in premature aging and occurs in 1 in 1,000,0000 to 1 in 10,000,000 people. In humans, WS is the result of mutations that render the WRN gene, that contains a helicase and an exonuclease domain, non-functional. Currently, there is no cure for WS in humans, making dietary and lifestyle interventions attractive for increasing the quality and longevity of lives. Introduction: Similar to prior reviews, the Journal of Wellness editors searched the literature from the second half of 2022 for an interesting and impactful selection of publications on wellness in healthcare professionals.

Methods: Editors conducted a standard keyword search in Pubmed, focusing chiefly on large journals, interventional trials, and other prospective research. We included papers published between July 1, 2022 and December 31, 2022.

Introduction: Burnout is a major risk in healthcare professions and is a significant contributor to the current nursing shortage. Strategies to combat burnout of healthcare professionals are in desperate need. The purpose of this project is to introduce the clinical peer supervision model as a method to alleviate burnout in nursing professionals.

Approach: Eight nurses from in-patient settings participated in a peer-supervision support group, modeled after existing European nursing and mental health provider-support protocols. Exercise increases cardiac workload, escalating the demand for oxygen. The myocardial vasculature responds to the greater demand for oxygen by increasing blood flow to match the needs of the heart during augmented work. Increasing blood flow requires the coronary arteries dilate (a.k.a. vasodilation); this is mediated predominantly by vascular smooth muscle cell relaxation. Vasodilation is driven through inhibition of calcium influx into vascular smooth muscle cells. The prevention of calcium influx is largely mediated by efflux of potassium via potassium channels causing membrane hyperpolarization, which in turn closes voltage-dependent (or gated) calcium channels (VDCC). Regular aerobic exercise promotes physiological cardiac growth, which is an adaptive response thought to enable the heart to meet higher physical demands. Cardiac growth involves coordination of catabolic and anabolic activities to support ATP generation, macromolecule biosynthesis, and myocyte hypertrophy. Although previous studies suggest that exercise-induced reductions in cardiac glycolysis are critical for physiological myocyte hypertrophy, it remains unclear how exercise influences the many interlinked pathways of metabolism that support adaptive remodeling of the heart. The most commonly used platforms for drug testing are in vivo animal models and in vitro cell culture models. While both have been of paramount importance for the discovery and detection of many cardiotoxicities and mechanisms of action, they lack the ability to model an intact human myocardium. Microvascular disease is hallmarked by pathophysiological conditions such as endothelial senescence, intimal thickening which impairs vasodilation, and regression of the capillary beds causing tissue ischemia in the myocardium or in peripheral vascular networks. Adipose-derived stromal vascular fraction (SVF) has previously demonstrated the ability to revascularize tissue. Increasing evidence shows that regenerative cells elicit their therapeutic benefit by paracrine mechanisms, leaving open extracellular vesicles (EVs) as a potential crux of the cell therapy paradigm. Introduction: Prevalence of stress among Indian medical undergraduate students is currently higher than the global stress level. Notably, although India is considered home to yogic practices, attempts at incorporating them in the daily routines of medical students have been lacking. The maintenance of myocardial oxygen supply during stress is essential for sustaining cardiac health. Enhancement of coronary blood flow upon increases in myocardial oxygen demand (i.e., hyperemia) relies on regulation of voltage-gated K+ (Kv) channels by their intracellular β subunits (i.e., Kvβ proteins). Considering that, Kvβ proteins are aldo-keto reductases (AKRs) and regulate Kv channel gating, we tested the hypothesis that elevation of myocardial oxygen demand modifies intracellular NAD(H) in arterial myocytes. Introduction: Burnout in emergency medicine and in residency training has been well-described. The impact of demographic, individual, and programmatic factors on burnout have not previously been determined in a national survey of emergency medicine residents. This study aimed to identify personal and environmental factors impacting resident burnout in a national sample of emergency medicine residents.

Methods: A prospective Emergency Medicine Resident Wellness Survey was administered in 2017. Introduction: Second Victim Syndrome (SVS) describes the phenomenon in which a caregiver experiences a traumatic psychological and emotional response to an adverse patient event or medical error. Using quantitative survey analysis, we aim to better understand the personal factors that affect SVS development and recovery.

Introduction: In post-menopausal women, small coronary microvessel dysfunction is the predominant heart disease presentation.There is currently no single treatment to tackle the multifactorial etiology of micro vessel dysfunction including oxidative stress, hyper constricted state, endothelial dysfunction, and blunted beta-adrenergic function. In the heart, G protein-coupled receptors (GPCRs), such as the b-adrenergic receptor (bADR) investigated in this experiment, are active in mediating vessel contractility vasodilation to facilitate blood flow. The presence of GPCR kinase 2 (GRK2), which inhibit bADRs, increase in old age and may mediate blunted capacity to vasodilate. Colorectal adenocarcinoma is the fourth most common cancer diagnosed worldwide and is a significant cause of morbidity and mortality. This dissertation performed an exploratory RNA-sequencing analysis comparing gene expression between colon adenocarcinoma tissue and paired normal colon epithelium. After identification of a number of lncRNAs that were increased in expression in colon adenocarcinoma compared to normal colon epithelium, we aimed to validate the expression and investigate their function in vitro. Gastropods are one of the most imperiled groups of animals in North America, yet there are major gaps in the literature pertaining to pollutants and climate change, and especially sublethal impacts. This dissertation assesses the effects of climate warming and unionized ammonia (NH3), one of the most abundant water pollutants, on the behavior and physiology of two caenogastropod snails: fine-ridged elimia (Elimia semicarinata) and Shawnee rocksnails (Lithasia obovata) (Gastropoda: Neotaenioglossa: Pleuroceridae). This dissertation is an analysis of skeletal muscle atrophy and a molecular assessment of potential preventative treatments. Chapter I begins with a background of skeletal muscle atrophy along with an analysis of associated molecular pathways. Here, we discuss how skeletal muscle atrophy is the consequence of protein degradation exceeding protein synthesis and can occur when a muscle is abnormally disused. Mitochondrial Transcription Factor A forms a mitochondrial nucleoid anti-oxidant structure and reduces pathophysiological abnormalities to structural and contractile proteins. Overall, this functions to physically stabilize mtDNA. Significant molecular changes such as a loss of TFAM drive pathophysiological concentrations of cytoplasmic Ca2+ and reactive oxygen species. Cardiac cell therapy using cardiac mesenchymal cells (CMC) significantly reduces ventricular dysfunction in patients with ischemic cardiomyopathy. Despite the improvement in function, a modest number of CMCs survive in the heart post-transplantation. In this study, we sought to improve the survival and retention of transplanted CMCs to prolong the therapeutic benefits afforded by cardiac cell therapy. To do this, we targeted the enzyme telomerase (TERT), known to be active in some highly proliferating cells (e.g. germ, stem). Spinal Cord Injury (SCI) in humans is a heterogeneous diagnosis, resulting in variable paralysis and paresthesia based on the mechanism, rostro-caudal location, and severity of injury. Both neurophysiological and anatomical studies have suggested that subclinical residual supraspinal-spinal connectivity exists in a subset of individuals deemed to have motor and sensory complete injuries. Recent reports of volitional movement in chronic, motor complete individuals during epidural spinal stimulation have provided compelling evidence that these residual projections may be capable of mediating volitional movement when the functional state of spinal circuitry is electrically modulated.
