This study investigates the respiratory physiology of bioluminescent larvae of Pyrearinus termitilluminans in relation to their tolerance to hypoxia and hyperoxia and to the supply of oxygen for bioluminescence. The partial pressure of oxygen (P(O2)) was measured within the bioluminescent prothorax by in vivo electron paramagnetic resonance (EPR) oximetry following acclimation of larvae to hypoxic, normoxic and hyperoxic (normobaric) atmospheres and during periods of bioluminescence (during normoxia). The P(O2) in the prothorax during exposure to an external P(O2) of 15.2, 160 and 760 mmHg was 10.3+/-2.6, 134+/-0.9 and 725+/-73 mmHg respectively (mean +/- s.d., N=5; 1 mmHg=0.1333 kPa). The theoretical mechanisms underlying the symptoms of Parkinson’s disease are based on dysfunctional output from the basal ganglia projected to the cortex via the thalamus. However, the subthalamic nucleus (STN) and substantia nigra pars reticularis (SNr) as part of the basal ganglia, also project to the brainstem, in particular to the pedunculopontine nucleus (PPN). Both the STN and SNr have altered neuronal activity in the 6-hydroxydopamine (6-OHDA) Parkinson’s model rat. Thus, it is hypothesized that the PPN is altered in this model of Parkinson’s disease.

Gap junctions play a crucial role in embryogenesis, especially of the heart where connexin43 is thought to be important for its formation as well as synchronized contraction. Several reports have shown that connexin43 protein is phosphorylated on serine residues in vitro and in vivo.

Studies performed in our laboratory have demonstrated that cell-to-cell communication in cells expressing connexin43 can be controlled rapidly and reversibly by microinjection of active protein kinases or phosphatases that target serine or threonine residues. Previously it has been shown that persistent activation of the stimulatory adenylyl cyclase pathway with cholera toxin (CT) downregulates the Gs alpha polypeptide (80%) in a cAMP-independent manner in C6 glioma cells (Shah, 1997). This study was conducted to examine the short and long term effects of CT on the regulation of pertussis toxin-sensitive and -insensitive G proteins and their transcripts in C6 glioma cells. Treatment of C6 cells with CT (100 ng/ml) up to 16 h had no effect on either Gi or Gq/11 alpha proteins. However, prolonged exposure (24-48 h) caused increased expression of Gi (20-30%) and Gq/11 …

To our knowledge, this is the first series of in vitro studies to investigate the effects of long-term, high-altitude hypoxemia on the contractility of fetal coronary vessels, namely the left circumflex, left anterior descending, and right coronary arteries. We have found that in response to chronic hypoxia, isolated fetal ovine coronary arteries exhibit attenuated contractile responsiveness to potassium and the thromboxane A2 mimetic. U46619. Furthermore, long-term, high-altitude hypoxia produced no significant alteration in the maximum relaxation response and sensitivity to adenosine in all vessel groups examined. Although the neonatal immune system is functionally immature, immune response capabilities are not the same in all newborns. The objective of this thesis was to determine development of immunocompetence and test the hypothesis that activation of maternal inflammatory immunity during pregnancy influences maturation of the neonatal immune system.

Lymphocytes in blood and spleen were immunophenotyped by flow cytometric analysis to determine differentiation characteristics, and splenocytes were assessed for cytokine production capabilities. Macrophage-derived factors have been implicated in the etiology of preterm labor but little is known about trafficking of this immune cell into the uterus or its regulation of uterine contractility in mice. Enhanced number and activation of macrophages was hypothesized to precede parturition. Studies characterized contractile activity and the distribution of macrophages in the uterus of C3/HeN mice before and after term (=day 19). Contractile activity by uterine strips on days 15 and 18 of pregnancy, the day of delivery, and 1 day postpartum indicate that an endogenous oscillator sustains high frequency contractions. Our previous studies have shown that subthreshold concentrations of two platelet agonists exert synergistic effects on platelet aggregation. Here we studied the mechanism of synergistic interaction of 5-hydroxytryptamine (5-HT) and epinephrine mediated platelet aggregation. We show that 5-HT had no or little effect on aggregation but it did potentiate the aggregation response of epinephrine. The synergistic interaction of 5-HT (1-5 microM) and epinephrine (0.5-2 microM) was inhibited by alpha2-adrenoceptor blocker (yohimbine; IC50= 0.4 microM), calcium channel blockers (verapamil and diltiazem with IC50 of 10 and 48 mM, respectively), PLC inhibitor (U73122; IC50=6 microM) and nitric oxide (NO) donor, SNAP (IC50=1.6 …

Peptidylglycine alpha-amidating monooxygenase (PAM) is an essential enzyme that catalyzes the COOH-terminal amidation of many neuroendocrine peptides. The bifunctional PAM protein contains an NH2-terminal monooxygenase (PHM) domain followed by a lyase (PAL) domain and a transmembrane domain. The cytosolic tail of PAM interacts with proteins that can affect cytoskeletal organization. A reverse tetracycline-regulated inducible expression system was used to construct an AtT-20 corticotrope cell line capable of inducible PAM-1 expression. Upon induction, cells displayed a time- and dose-dependent increase in enzyme activity, PAM mRNA, and protein. Induction of increased PAM-1 expression produced graded changes in PAM-1 metabolism. An optical Fourier transform represents the interference of wavefronts produced as light passes through multiple slits. Theoretically, any image containing periodic structure causes diffraction. The cellular arrangement of a plant leaf and the regularity in venation are ideal conditions for diffracting light. As light passes through columns of cells, its path is altered according to the distance and orientation present.

In order to grow, crustaceans must shed their exoskeleton in a process known as "moulting'. Although this process is short and intermittent, it remains dominant over the life of a crustacean. Physiological changes in the period between moults (intermoult) are comparatively quiescent as opposed to the periods directly before and after the actual moult, known as premoult, and postmoult respectively (West, 1997). Moulting is associated with distinct physiological changes including the breakdown of muscle (Musgrove and Geddes, 1985). Consistent with previous studies, we demonstrated that dexamethasone (Dex) caused cytolysis/apoptosis in log phase Nb2 lymphoma cells, while prolactin (Prl) inhibited this effect. The Nb2 model was used to investigate the mechanisms of apoptosis control with the aid of immunocytochemistry (ICC). We established with absorption the specificity of staining due to Fas, Fas Ligand, Bcl-2, and Bax antibodies (the specificity of anti-p53 could not be verified). Dex-induced cytolysis/apoptosis was detected in synchronized (G⁰/G¹) cells after 6 and 8 hours of Dex exposure. The purpose of this study was to compare factors that may influence menstrual irregularities of female adolescent high school cross-country runners, ranging from 13-18 years of age. A survey was utilized to determine the subjects age, year in school, exercise history, menstrual history, and training regimens and where the subjects sought nutritional information. The groups were separated according to their current menstrual status. One group (normal) consisted of subjects who currently had a normal menstrual cycle for at least one year (n=17), while the other group (abnormal) (n=18) experienced an irregular menstrual cycle.

The Nb2 lymphoma cell line is susceptible to glucocorticoid (GC)-induced apoptosis in the absence of mitogen and this effect is inhibited by the mitogen prolactin (PRL). This process is believed to be mediated by the glucocorticoid receptor (GR) and various downstream signals. The purpose of this study was to use the limiting dilution method to isolate subsets of Nb2 clones which expressed either highly enriched amounts of GR or an absence of GR and to relate the amount of GR to the magnitude of GC induced cytolysis. The acousticolateralis sensory system is characterized by a specific receptor cell type called the sensory hair cell and is found in all vertebrates. There are two types of hair cell sensory epithelia based on location: those of the inner ear, such as the organs for hearing and balance, and the lateral line system located within the epidermis. Background. Gap junctions are thought to have a crucial role in the synchronized contraction of the heart and in embryonic development. Connexin43, the major protein of gap junctions in the heart, is targeted by several protein kinases that regulate myocardial cell-cell coupling. We hypothesized that mutations altering sites critical to this regulation would lead to functional or developmental abnormalities of the heart.

Methods. Connexin43 DNA from 25 normal subjects and 30 children with a variety of congenital heart diseases was amplified by the polymerase chain reaction and sequenced. Mutant DNA was expressed in cell culture and examined for its effect …

Many lower extremity anomalies are linked to an increased incidence of knee pain. A relationship between increased sports participation and knee pain has also been hypothesized. This study examined the relationship between lower extremity anomalies and the level of sports participation to knee pain. Fifty middle and high school female students completed a questionnaire to determine level of sports participation and the presence of knee pain/pathology. Measurements were made of Q angle, iliotibial band flexibility, knee hyperextension, hamstring flexibility, patella alta and calcaneal position. The Truckee River in California and Nevada is subject to diverse water regimes and a corresponding variety of flow rates. Original riparian vegetation has been altered by these variable flow rates and by a variety of human uses resulting in loss of native riparian vegetation from its historic extent. The purpose of this study was to examine the effects of dehydration and rehydration on the one repetition maximum (1-RM) bench press of weight trained males. The ten subjects displayed the following characteristics: 25.3 ± 3.2 years old, 85.5 ± 16.4 kilograms body mass and a 117.9 ± 24.0 kilogram base-line 1-RM. Ten subjects were tested on two separate days, with one week between each test day. The subjects were randomly assigned to begin their testing in either the A protocol or the B protocol. A previous study found that the patellofemoral joint compressive force (PFJRF) during backward running was less than during forward running at free speed. Therefore, the purpose of this study was to compare the patellofemoral joint compressive forces during backward and forward running at the same speed. Ten runners (4 females, 6 males) between the age of 20-25 (X=22, SD=2) ran in backward and forward directions at the slow speed of 2.3 mph (1.0 m.sec-1). Macroinvertebrate communities were examined on 4 winter dates over a 4-yr period in 2 high-altitude Rocky Mountain streams to document overwintering assemblages potentially experiencing spring acid pulses. Taxa richness values were comparable to other published lists for alpine and montane stream systems despite the fact that most literature reflected summer collections. Mean benthic density ranged from 1406 to 19,734 organisms/m2, and drift rates ranged from 0 to 1740 organisms/100 m3. Benthic collections showed higher taxa richness than drift collections while the Ephemeroptera and Plecoptera occurred in greater proportions in drift than in benthos. Leukemia inhibitory factor (LIF) is a pleiotropic cytokine which demonstrates perplexing physiological effects. It has been demonstrated that LIF is essential for implantation in mice. Little is known relating to the manner by which LIF effects pre-implantation and post-implantation development. The objectives of this project were to determine the effects LIF on pre-implantation development, to determine the effects that it may have on implantation rates, successful pregnancy rates, and resorption rates, and to determine the effects that LIF has on the skeletal development of mice.
