Assessment Of The Impact Of Attention Deficit Hyperactivity Disorder On Type 1 Diabetes, Kellee Miller
Assessment Of The Impact Of Attention Deficit Hyperactivity Disorder On Type 1 Diabetes, Kellee Miller
USF Tampa Graduate Theses and Dissertations
Individual day-to-day management and effective control of type 1 diabetes (T1D) is ultimately driven by decisions made by the individual. Individuals with attention deficit hyperactivity disorder (ADHD) have a higher tendency to be inattentive, impulsive, and hyperactive. Attention deficits and impulsivity among adolescents and adults with T1D could result in poor diabetes management through infrequent self-monitoring of blood glucose and inadequate insulin dosing – key components of achieving optimal glycemic control.
This study included 7,380 adolescents and young adults, ages 13-25 years, participating in the T1D Exchange clinic registry (median age 17.4 years, duration 7.7 yrs, 50% female, 81% white). …
The Acute Effects Of Multiple Resisted Sled-Pull Loads On Subsequent Sprint-Running Performances, Carl Stephen Crouse
The Acute Effects Of Multiple Resisted Sled-Pull Loads On Subsequent Sprint-Running Performances, Carl Stephen Crouse
Dissertations and Theses @ UNI
Sled-pulling is a commonly implemented form of training for various sports. However, few studies have used sled-pulling as a means of acutely enhancing sprintrunning performances. The purpose of this study was to investigate how various sledpulling resistance loads (95% and 110% of body mass) effect subsequent, unloaded, sprint-running performances, possibly with aid from the effect known as Post-Activation Potentiation (PAP). PAP is a physiological phenomenon which increases the rate of force development of skeletal muscle which may result in the enhancement of power (speed-strength) dominant activities such as sprint-running and jumping.
Participants were a mix of males (n = 11; …

Mutations in the Plasmodium falciparum chloroquine resistance transporter, PfCRT, are the major determinant of chloroquine resistance in this lethal human malaria parasite. Here, we describe P. falciparum lines subjected to selection by amantadine or blasticidin that carry PfCRT mutations (C101F or L272F), causing the development of enlarged food vacuoles. These parasites also have increased sensitivity to chloroquine and some other quinoline antimalarials, but exhibit no or minimal change in sensitivity to artemisinins, when compared with parental strains. A transgenic parasite line expressing the L272F variant of PfCRT confirmed this increased chloroquine sensitivity and enlarged food vacuole phenotype. Furthermore, the introduction …

Sigma Receptor Activation Mitigates Toxicity Evoked By The Convergence Of Ischemia, Acidosis And Amyloid-Beta, Adam Alexander Behensky
USF Tampa Graduate Theses and Dissertations
Stroke is the fifth leading cause of death in the United States and a major cause of long-term disability in industrialized countries. The core region of an ischemic stroke dies within minutes due to activation of necrotic pathways. Outside of this core region is the penumbral zone, where some perfusion is maintained via collateral arteries. Delayed cell death occurs in this area due to the triggering of apoptotic mechanisms, which expands the ischemic injury over time. The cellular and molecular events that produce the expansion of the ischemic core continue to be poorly understood. The increases in the amyloid precursor …
Minocycline Treatment And The Necessity To Develop A Novel Outcome Measure For Children With Angelman Syndrome, Joseph Christopher Grieco
Minocycline Treatment And The Necessity To Develop A Novel Outcome Measure For Children With Angelman Syndrome, Joseph Christopher Grieco
USF Tampa Graduate Theses and Dissertations
Angelman syndrome (AS) is a rare genetic disorder affecting 1/10,000 to 1/20,000 births. This disorder arises through the genetic disruption of the maternal UBE3A allele, which when coupled with epigenetic silencing of the paternal allele UBE3A allele, gives rise to an absence of UBE3A protein in the central nervous system. Clinical manifestations of the syndrome vary in severity and include poor motor function, deficits in language and severe intellectual impairments. Previous research in the Angelman syndrome mouse model revealed abnormalities in dendritic spine density and morphology of hippocampal pyramidal cells. As seen in humans with AS, mice show abnormal behavioral …

UNF Graduate Theses and Dissertations
Shark fisheries are a multimillion dollar industry in the United States and have significant contributions both recreationally and commercially. In order to maintain this industry, fisheries must be properly managed. An understanding of life history and reproduction is crucial in order to adequately manage shark fisheries. The finetooth shark, Carcharhinus isodon, is a member of the small coastal shark (SCS) fishery complex. It is found in Atlantic waters from South Carolina to Florida and throughout the Gulf of Mexico. This species has recently come under increased fishing pressure and has previously been overfished. New data is needed in order …
Molecular Identification And Functional Characteristics Of Peptide Transporter 1 (Pept1) In The Bonnethead Shark (Sphyrna Tiburo), Hannah Hart
UNF Graduate Theses and Dissertations
Many elasmobranchs are considered top predators with worldwide distribution, and in general these fish play an important role in the transfer of energy from the lower to the upper trophic levels within the marine ecosystem. Despite this, little research has been done regarding the rates of prey ingestion, digestion, and the processes of energy and nutrient absorption. Specifically understudied is enzymatic digestion within the intestinal brush border, which functions to break down macromolecules into smaller subunits for luminal absorption across the gastrointestinal epithelium. Given their carnivorous diet, the present study sought to expand knowledge on nutrient intake in elasmobranchs by …

Gender Differences In Social Support During Injury Rehabilitation, Chelsey Ann Bruns
Gender Differences In Social Support During Injury Rehabilitation, Chelsey Ann Bruns
Dissertations and Theses @ UNI
Athletic injury can be a stress-inducing event that could have negative consequences on the psychological health of the athlete (Anderson & Williams, 1988; Gould, Udry, Bridges, & Beck, 1997). Injuries not only challenge the athlete physically, but also mentally challenge their ability to cope and overcome. A growing amount of research has indicated social support as a key factor in the rehabilitation process for an injured athlete (Yang, Peek, Lowe, Heiden, & Foster, 2010). Athletic injury causes an emotional disruption, and social support could influence the athlete’s reaction to the injury and his or her ability to cope with rehabilitation …
The Effects Of Hibernation On The Hemostatic Properties Of The American Bullfrog, Rana Catesbeiana, Kiana Cullinan
The Effects Of Hibernation On The Hemostatic Properties Of The American Bullfrog, Rana Catesbeiana, Kiana Cullinan
Honors Program Theses
It is known that hibernating animals display a drastic reduction in metabolic rate, accompanied by reduced heart rate and rate of blood flow through vessels. Sluggish blood flow increases the risk for developing clots, yet it has been observed that hibernators conclusively display elongated clotting times. This observed impairment may be caused by the decrease in body temperature, due to a negative impact on the functioning of the enzymes involved in coagulation. Some hibernating endotherms, or “warm-blooded” animals, employ biological mechanisms as preventative means to prevent clotting during hibernation, such as the down-regulation of certain clotting factors or the production …

Understanding The Physiological Effects Of Suspended Material On Rainbow Trout (Oncorhynchus Mykiss), Tyler J. Weinhardt
Understanding The Physiological Effects Of Suspended Material On Rainbow Trout (Oncorhynchus Mykiss), Tyler J. Weinhardt
Theses and Dissertations (Comprehensive)
The effect of global warming on northern environments is becoming increasingly evident. Melting of underlying permafrost is associated with widespread impacts in these environments. The loss of permafrost results in a destabilizing of underlying sedimentary layers resulting in thermokarst slumping. When this occurs on a large scale (mega-slumping) soil material becomes mobilized and is carried into local streams and rivers. The purpose of this study is to examine the sub-lethal physiological effects that suspended material has on rainbow trout (Oncorhynchus mykiss) in the context of the Peel River Plateau. Juvenile rainbow trout were exposed (following Environment Canada exposure …

The Effects Of Lampricide 3-Trifluoromethyl-4-Nitrophenol Toxicity On The Gills Of Larval Sea Lamprey And Non-Target Rainbow Trout And Lake Sturgeon, Lisa A. Sorensen
Theses and Dissertations (Comprehensive)
The pesticide, 3-trifluoromethyl-4-nitrophenol (TFM), is widely used in the Great Lakes to control invasive sea lampreys (Petromyzon marinus) populations, but much about its sub-lethal effects remains unknown. A better understanding of its toxicity is needed to improve TFM effectiveness and to protect non-target organisms from its potential adverse effects. The objectives of this thesis were to test the following two hypotheses: (1) impairment of mitochondrial ATP production by TFM interferes with ATP-dependent ion-uptake by fish, leading to altered electrolyte balance, and (2) perturbations of gill function by TFM are exacerbated in soft, ion poor water (SW; 40 mg …
Estimates Of Persistent Inward Current In Human Motor Neurons During Postural Sway, Ryan Foley, Jayne M. Kalmar Dr.
Estimates Of Persistent Inward Current In Human Motor Neurons During Postural Sway, Ryan Foley, Jayne M. Kalmar Dr.
Theses and Dissertations (Comprehensive)
Persistent inward current (PIC) is a membrane property critical for increasing gain of motor neuron output. In humans, most estimates of PIC are made from plantarflexor or dorsiflexor motor units with the participant in a seated position with the knee flexed. This seated and static posture neglects the task-dependent nature of the monoaminergic drive that modulates PIC activation. Seated estimates may drastically underestimate the amount of PIC that occurs in human motor neurons during functional movement. The current study estimated PIC using the conventional paired motor unit technique which uses the difference between reference unit firing frequency at test unit …

The Physiological Stress Response Caused By Hypoxia And Reperfusion Injury In Striped Bass (Morone Saxatilis) And Nile Tilapia (Oreochromis Niloticus), Amanda C. Reynolds
College of Graduate Studies: Theses & Dissertations
Approximately five million people in the United States are affected by cardiovascular related diseases yearly contributing to 300,000 annual deaths, making CVD the leading cause of mortality worldwide. It has been suggested that apoptosis (programmed cell death) contributes to the pathogenesis of cardiovascular diseases. When blood flow is reduced or cut off from the heart, usually by a thrombus, this results in oxygen deprivation (hypoxia) to the cardiomyocytes (heart cells). In response to this hypoxic stress, cardiomyocytes will undergo apoptosis. Since many species of fish can survive levels of hypoxia that would be fatal to mammals, fish are an ideal …

Reproductive information, including seasonality in behavior and physiology, is key to our ability to breed species for maintaining assurance colonies or for future introductions. Limited scientific data is available about the reproduction of Helodermatid lizards. Several species of helodermatid lizard are uncommon, rare, or simply exist at low population densities and in need of the development of conservation programs and management. Most helodermatid reproductive data collected to date has been done on deceased animals using histological techniques. Until this study, no hormones have been analyzed, and individual animals have not been followed through a complete cycle. The purpose of this …

Ontogenetic Scaling Patterns And Functional Anatomy Of The Pelvic Limb Musculature In Emus (Dromaius Novaehollandiae), Luis P. Lamas, Russell P. Main, John R. Hutchinson
Department of Basic Medical Sciences Faculty Publications
Emus (Dromaius novaehollandiae) are exclusively terrestrial, bipedal and cursorial ratites with some similar biomechanical characteristics to humans. Their growth rates are impressive, as their body mass increases eighty-fold from hatching to adulthood whilst maintaining the same mode of locomotion throughout life. These ontogenetic characteristics stimulate biomechanical questions about the strategies that allow emus to cope with their rapid growth and locomotion, which can be partly addressed via scaling (allometric) analysis of morphology. In this study we have collected pelvic limb anatomical data (muscle architecture, tendon length, tendon mass and bone lengths) and calculated muscle physiological cross sectional area …

When a static stimulus appears successively at two distant locations, we perceive illusory motion of the stimulus across them–long-range apparent motion (AM). Previous studies have shown that when the apparent motion stimuli differ in shape, interpolation between the two shapes is perceived across the AM path. In contrast, the perceived color during AM has been shown to abruptly change from the color of the first stimulus into that of the second, suggesting interpolation does not occur for color during AM. Here, we report the first evidence to our knowledge, that an interpolated color, distinct from the colors of either apparent …

Aims: Cancer cachexia is a syndrome which results in severe loss of muscle mass and marked fatigue. Conditioned media from cachexia-inducing cancer cells triggers metabolic dysfunction in skeletal muscle, including decreased mitochondrial respiration, which may contribute to fatigue. We hypothesized that Lewis lung carcinoma conditioned medium (LCM) would impair the mitochondrial electron transport chain (ETC) and increase production of reactive oxygen species, ultimately leading to decreased mitochondrial respiration. We incubated C2C12 myotubes with LCM for 30 min, 2, 4, 24 or 48 h. We measured protein content by western blot; oxidant production by 2′,7′-dichlorofluorescin diacetate (DCF), 4-amino-5-methylamino-2′,7′-difluorofluorescein diacetate (DAF), and …

Contaminated sediments in marine environments have been shown to be good indicators of ecological risk and a means to assess anthropogenic impacts on marine habitats and the animals that inhabit them (Long et al. 1995, Rattner 2009). Estuarine sediments are especially complex media with regard to physical, chemical, and biological characteristics that trap, store, modify and sometimes release contaminants to the biota (Long et al. 1995). Especially vulnerable are animals that are in constant contact with the sediments, such as flatfishes that partially bury themselves for camouflage (Costa et al. 2011). Impacts can be assessed in a number of ways, …

DAX-1 is known to play a key role in the maintenance of pluripotency in mouse embryonic stem cells (mESC), such that elimination of DAX-1 (through methods such as RNAi) leads to the differentiation of ESC (Khalfallah et al. 2009). Earlier research by the Tzagarakis-Foster lab identified target genes of DAX-1 in mESC through PCR array methodology (Torres 2013). The Dickkopf 1 gene (Dkk1) was identified as a potential target of DAX-1 by these studies and has since been confirmed to be a DAX-1 target in mESC. However, to date very little is understood of the mechanism of DAX-1 repression of …

Glutamyl-tRNA (Glu-tRNAGlu) is the common substrate for both protein translation and heme biosynthesis via the C5 pathway. Under normal conditions, an adequate supply of this aminoacyl-tRNA is available to both pathways. However, under certain circumstances, Glu-tRNAGlu can become scarce, resulting in competition between the two pathways for this aminoacyl-tRNA. In Acidithiobacillus ferrooxidans, glutamyl-tRNA synthetase 1 (GluRS1) is the main enzyme that synthesizes Glu-tRNAGlu. Previous studies have shown that GluRS1 is inactivated in vitro by hydrogen peroxide (H2O2). This raises the question as to whether H2O2 negatively affects …

Prostate Field Cancerization -- Thinking Outside The Tumor, Dor Shoshan, Marco Bisoffi
Student Scholar Symposium Abstracts and Posters
Prostate field cancerization (or field effect) is characterized by the presence of molecular alterations in histologically normal tissues adjacent to adenocarcinomas. Accordingly, our research indicates deregulated expression of several proteins that define this type of molecular pathology. The scope of the present study was to determine the expression of the key transcription factor and potential marker of field cancerization early growth response 1 (EGR-1) in human prostate tissues derived from prostatectomies and biopsy cores.
EGR-1 was detected by immunofluorescence using a polyclonal anti-human EGR-1 and Alexa Fluor 488-conjugated secondary antibodies. EGR-1 expression was quantitated by determining the pixel count per …

The brain senses circulating levels of angiotensin II (AngII) via circumventricular organs, such as the subfornical organ (SFO), and is thought to adjust sympathetic nervous system output accordingly via this neuro-hormonal communication. However, the cellular signaling mechanisms involved in these communications remain to be fully understood. Previous lesion studies of either the SFO, or the downstream median preoptic nucleus (MnPO) have shown a diminution of the hypertensive effects of chronic AngII, without providing a clear explanation as to the intracellular signaling pathway(s) involved. Additional studies have reported that over-expressing copper/zinc superoxide dismutase (CuZnSOD), an intracellular superoxide (O2·-) scavenging enzyme, in …

Movement is as natural to humans as breathing is, and, yet, passivity starts early in schools. We are taught to sit still and in silence for long periods of time. By the time we reach adulthood and enter the workforce, we have almost forgotten our sense of embodiment. This lack of movement is counter-productive, not only to learning but to the development of creativity as well. For this project, I designed a tool to recapture the joy and playfulness of movement. Furthermore, the …
The Role Of Ceramides In Mediating Endotoxin-Induced Mitochondrial Disruption, Melissa Ellen Hansen
The Role Of Ceramides In Mediating Endotoxin-Induced Mitochondrial Disruption, Melissa Ellen Hansen
Theses and Dissertations
Ceramides are sphingolipids that serve as important second messengers in an increasing number of stress-induced pathways. Ceramide has long been known to affect the mitochondria, altering both morphology and physiology. Lipopolysaccharide (LPS) is a prevalent circulating inflammatory agent in obesity, potentially mediating some of the pathologies associated with weight gain. Given previous findings of TLR4-mediated ceramide accrual and ceramide-mediated mitochondrial disruption, we questioned whether ceramide is necessary for LPS-induced mitochondrial disruption. We found that LPS treatment increased gene transcript levels of ceramide synthesis enzymes and mitochondrial fission proteins and increased ceramide content in cultured myotubes and in mouse tissue. Mitochondrial …
