Temporal Localization Of Insulin-Like Growth Factor 1 (Igf1) Positive Cells In Developing Hypothyroid Brain, Shelby Anne Kline
Temporal Localization Of Insulin-Like Growth Factor 1 (Igf1) Positive Cells In Developing Hypothyroid Brain, Shelby Anne Kline
All Graduate Theses, Dissertations, and Other Capstone Projects
Hypothyroidism has been known to result in numerous neuroanatomical defects in the developing brain. To investigate further into the mechanism in which these defects are obtained, local brain insulin-like growth factor 1 (Igf1) was investigated in relation to the hypothyroid brain. There are many similarities between hypothyroidism and the Igf1 deficient brain. Additionally, TH (thyroid hormone) is known to interact with Igf1. Therefore, the purpose of this study was to investigate the effect of induced congenital hypothyroidism on the temporal expression of Igf1-positive cells in the brain. To investigate the relationship between TH and Igf1 in the brain, we inhibited …
Female Social Rank And Steroid Production In Semi-Free Ranging Lemur Catta On St. Catherines Island, Georgia, Tiffany Burgess
Female Social Rank And Steroid Production In Semi-Free Ranging Lemur Catta On St. Catherines Island, Georgia, Tiffany Burgess
College of Graduate Studies: Theses & Dissertations
Many mammalian species live in complex social groups that profoundly affect the behavior and ecology of their members. In ring-tailed lemurs (Lemur catta) these groups are characterized by female dominance, where females have priority over males for food, grooming, and space. Among females there is also a linear dominance hierarchy. L. catta have a strong matrilineal social core due to females staying within their natal group and males dispersing at sexual maturity. Daughters “inherit” their mother’s social rank. However, it is not known if the matrilineal lines are maintained through learned dominant/aggressive behavior or inherited via genetically determined hormonal …
The Relationship Between Vagal Tone, A Marker Of Parasympathetic Activity, And Pro-Social Behavior, Emily A. Goodlin
The Relationship Between Vagal Tone, A Marker Of Parasympathetic Activity, And Pro-Social Behavior, Emily A. Goodlin
Scripps Senior Theses
Vagal tone, a measure of parasympathetic activity via the vagus nerve, is known to be associated with positive emotion because it promotes social engagement and self-soothing behavior. Heart rate variability (HRV), especially high frequency oscillation, is a direct measure of vagal tone, and has been used in previous studies to test the correlation between vagal tone and positive emotion. This study aims to determine if the two major oscillations of heart rate variability, high frequency (HF-HRV) and low frequency (LF-HRV) can predict pro-social behavior, which is classified as giving donations to charities. Baseline LF- and HF-HRV levels were recorded, and …

Open Access Theses & Dissertations
The impact of restraint stress on changes in cytokine expression and oxidative stress markers in hippocampal (Hipp) and hypothalamic (Hyp) regions of the brain, were assessed in a rat model. In addition, immune and inflammatory variables were assessed in peripheral blood. Male Wistar rats were acutely (30 min) or repeatedly (30 min/d for 14 consecutive days) exposed to a restraint stress, or maintained as non-stressed controls. At the time of sacrifice, whole blood and brain tissues enriched in Hyp or Hipp regions were collected; blood was collected with EDTA as an anticoagulant and centrifuged to yield plasma. All tissue samples …
L30a Mutation Of Phospholemman Mimics Effects Of Cardiac Glycosides On Isolated Cardiomyocytes, Ryan David Himes
L30a Mutation Of Phospholemman Mimics Effects Of Cardiac Glycosides On Isolated Cardiomyocytes, Ryan David Himes
Dissertations
In order to determine if mutations made to phospholemman (PLM) could increase PLM binding to the Na/K-ATPase (NKA) and cause positive inotropy we performed scanning mutagenesis of the transmembrane domain of PLM and measured FRET between each mutant and NKA. We observed increased binding to NKA for several PLM mutants compared to WT, including L27A, L30A, and I32A. In isolated cardiomyocytes, overexpression of WT PLM increased the amplitude of the Ca2+ transient compared to GFP control. Ca2+ transient amplitude was further increased by L30A PLM overexpression. L30A mutation also delayed Ca2+ extrusion and increased the duration of the cardiomyocyte contraction. …

As the central pacemaker, the suprachiasmatic nucleus (SCN) has long been considered the primary regulator of blood pressure circadian rhythm; however, this dogma has been challenged by the discovery that each of the clock genes present in the SCN is also expressed and functions in peripheral tissues. The involvement and contribution of these peripheral clock genes in the circadian rhythm of blood pressure remains uncertain. Here, we demonstrate that selective deletion of the circadian clock transcriptional activator aryl hydrocarbon receptor nuclear translocator-like (Bmal1) from smooth muscle, but not from cardiomyocytes, compromised blood pressure circadian rhythm and decreased blood …
Sensorimotor Analysis Of Oxaliplatin Treated Rats, Krystyna Blanka Wieczerzak
Sensorimotor Analysis Of Oxaliplatin Treated Rats, Krystyna Blanka Wieczerzak
Browse all Theses and Dissertations
There is currently no direct evidence that chemotherapy induced peripheral neuropathy (CIPN) necessarily explains the sensorimotor deficits seen in 90% patients treated with oxaliplatin (OX). Some patients develop sensory symptoms without CIPN. Our laboratory reported abnormal signaling from IA afferents in OX treated rats with no evidence of neuropathy. We hypothesized that in the absence of CIPN, the behavioral disability is associated with impaired sensory encoding in OX treated rats. The purpose of this study was to investigate the sensorimotor abilities of OX treated rats. The battery of behavioral tests was designed to address proprioception and sensorimotor integration. In the …

Malignant transformation is the process by which cells develop cancer properties. While many causes for malignant transformation are known (i.e. common genetic mutations and/or exposure to toxins or viruses), the basic requirements that allow a cell to stay alive with altered nutrient and energy requirements are just now being studied. In some tumor types malignant cells undergo changes that result in metabolic differences compared to normal cells. These can include defects in mitophagy resulting in an accumulation of dysfunctional mitochondria and/or a metabolic switch resulting in increased glycolysis, termed the Warburg effect. Increased tumor growth and metastasis have also been …

A high prevalence of sleep-disordered breathing (SDB) after spinal cord injury (SCI) has been reported in the literature; however, the underlying mechanisms are not well understood. My studies had 2 aims: 1) to determine the effect of the withdrawal of the wakefulness drive to breathe on the degree of hypoventilation in SCI patients and able-bodied controls and 2) to determine the response of …
Crebh, A Novel Liver Clock Keeper For Energy Metabolism, Ze Zheng
Crebh, A Novel Liver Clock Keeper For Energy Metabolism, Ze Zheng
Wayne State University Dissertations
Circadian rhythms play crucial roles in orchestrating diverse physiological processes that are critical for health and disease. Cyclic AMP responsive element binding protein 3-like 3 (CREB3L3, also known as CREBH) is a liver-enriched, endoplasmic reticulum (ER)-tethered transcription factor known to regulate hepatic acute-phase response and energy homeostasis under stress conditions. Here, we demonstrate that CREBH is regulated by the circadian clock and functions as a diurnal regulator of hepatic lipid and glucose metabolism. CREBH is required to maintain circadian profiles of blood triglycerides, fatty acids, and glucose as well as hepatic glycogen storage. CREBH rhythmically regulates expression levels and amplitudes …
Trophoblast Retrieval And Isolation From The Cervix (Tric) For Non-Invasive Prenatal Genetic Diagnosis And Prediction Of Abnormal Pregnancy Outcome, Rani Fritz
Wayne State University Dissertations
The placenta is vital for the short- and long-term health of the fetus, and significantly impacts the health of the mother. During the first and second trimesters of pregnancy, extravillous trophoblast (EVT) cells invade the uterus and remodel the maternal spiral arteries, which, if inadequate, leads to pregnancy complications, including early pregnancy loss (EPL), preeclampsia (PE), and intra-uterine growth restriction (IUGR). EVT migration into the uterine wall is dependent on growth factors and cytokines that signal between maternal and fetal tissues. The epidermal growth factor (EGF) signaling system plays a significant role in trophoblast function. Using immunocytochemistry (ICC), we evaluated …
The Acute And Chronic Effects Of Resveratrol On Renal Function And Blood Pressure, Kevin L. Gordish
The Acute And Chronic Effects Of Resveratrol On Renal Function And Blood Pressure, Kevin L. Gordish
Wayne State University Dissertations
We investigated the acute and chronic effects of resveratrol on renal function and blood pressure. We hypothesized that resveratrol would act as a renal vasodilator through a nitric oxide-dependent mechanism. In our acute studies, we found an intravenous bolus of resveratrol influenced changes in renal hemodynamics by increasing renal blood flow and decreasing renal vascular resistance. The mechanism of renal vasodilation was nitric oxide dependent and through a reduction of endogenous reactive oxygen species. Resveratrol-induced renal vasodilation was not influenced by COX metabolism and vasodilatory prostanoids. We found with continuous intravenous resveratrol infusion induced significant renal vasodilation while not altering …

Behavior, Energetics, And Swimming Performance Of The American Paddlefish, Polyodon Spathula, In A Lower Mississippi River Oxbow, Bryan Andrew Cage
Electronic Theses and Dissertations
Paddlefish often frequent oxbows and backwater habitats between periods of migration. Speculation as to the use and energetic savings associated with this seasonal residence has not been substantiated. This study used radio telemetry to track the seasonal movements and swim tunnel respirometry to estimate metabolic rates and swimming efficiency of paddlefish within an oxbow located just north of Clarksdale, Mississippi (Moon Lake). Paddlefish overwintered in the deepest portions of Moon Lake with little movement into shallower areas. Paddlefish swimming performance was measured at four swimming speeds (76.04, 91.27, 106.50, and 121.72 cm/s) to determine the cost of locomotion over season …

Dogs admitted to animal shelters experience psychological stressors resulting in elevated plasma cortisol. We previously found 30 min of human interaction reduced this response. The present study further characterized this effect, with the aim of developing a practical means of reducing stress of shelter dogs. We found that a second day of 30 min of petting reduced cortisol levels as effectively as the first. Further, 15 min of this interaction was as effective as 30 min. During petting, signs of excitation (vocalizations) and anxiety (panting) as well as escaped attempts were reduced, and social solicitation (tail-wagging) increased. However, cortisol levels …

Transcranial Direct Current Stimulation (tDCS) has been used to treat various mental and neurological illnesses. Rodent models have been used to examine physiological changes in the brain after tDCS, as well as to develop safety standards. However, most animal tDCS studies implant an electrode on the brain, potentially altering the path of current during stimulation. Additionally, no studies have been completed specifically examining maximum safe anodal tDCS limits, and a pilot study conducted to determine an electrode montage to examine biological changes of learning and memory from anodal tDCS indicated brain lesion was occurring before a commonly cited lesion threshold …

Sleep deprivation causes many adverse effects on work performance. Many experiments in both human and rodent models reveal detriments that sleep deprivation has on learning and memory, including performance in a water radial arm maze (WRAM) task. This study utilizes the modified multiple platform method (MMPM) of sleep deprivation; rats were sleep deprived in order to study memory errors they may make during the WRAM task. The findings indicate that 6 hours of sleep deprivation for 2 five-day week periods did not affect performance in the WRAM task except on the initial day compared to the large platform group. The …

Available data suggest that snake hemoglobins (Hbs) are characterized by a combination of unusual structural and functional properties relative to the Hbs of other amniote vertebrates, including oxygenation-linked tetramer-dimer dissociation. However, standardized comparative data are lacking for snake Hbs, and the Hb isoform composition of snake red blood cells has not been systematically characterized. Here we present the results of an integrated analysis of snake Hbs and the underlying α- and β-type globin genes to characterize 1) Hb isoform composition of definitive erythrocytes, and 2) the oxygenation properties of isolated isoforms as well as composite hemolysates. We used species from …

Mitochondrial Therapeutics During Ischemia-Reperfusion; Modulation Of Complex I: Effect Of Metformin., Shawn Y. Sunu
Theses and Dissertations
The modulation of the electron transport during ischemia-reperfusion has been shown to be protective. We hypothesized that metformin, a Complex I inhibitor, may exhibit characteristics of a pharmacological agent that could achieve long-term therapeutic intervention against ischemia-reperfusion injury. Mitochondria were harvested from adult male mice and incubated with or without metformin at 30oC for 15 minutes, while being shaken at 300 rpm. Metformin decreased Complex I oxidative phosphorylation and Complex I activity. However, metformin also increased injury and decreased the maximum membrane potential. Even though there was a decrease in maximum membrane potential, the proton motive force (PMF) …

The Effect Of Training Volume And Intensity On Improvements In Muscular Strength And Size In Resistance-Trained Men, Gerald Mangine
Electronic Theses and Dissertations
The magnitude of improvements in muscular strength and size are influenced by the volume and intensity of a resistance training program. While it is clearly advantageous for resistance-trained individuals to utilize programming specific to these goals, it not clear which is more important. Therefore the purpose of the present investigation was to determine the effect of focusing on training volume versus intensity on changes in muscle size and strength. Changes in muscular strength and size were examined in 29 resistance-trained men following 8 weeks of resistance training. Participants were randomly assigned to either a high volume (VOL, n = 14, …
Albumin Levels In Hispanic Dialysis Patients With And Without Type Ii Diabetes, Hector Hernandez
Albumin Levels In Hispanic Dialysis Patients With And Without Type Ii Diabetes, Hector Hernandez
Walden Dissertations and Doctoral Studies
Albumin provides the vital scaffolding for growth and tissue repair and supports oncotic blood pressure and hemodynamics. In hemodialysis patients, albumin aids with fluid removal by drawing excess fluid from edematous tissues back into the blood where it can then be removed by a dialyzer. The hyperglycemia seen in dialysis patients with Type II diabetes progressively damages kidney glomeruli, which permits albumin seepage into the urine, thus lowering serum albumin. The conceptual framework underpinning this research is the van't Hoff theory of osmotic pressure. Under this framework, the solute-solvent relationship largely contributes to the osmotic movement of fluid. The purpose …

Extracellular matrix (ECM) degradation after myocardial infarction (MI) leaves the myocardium structurally weakened and, as a result, susceptible to early infarct zone dyskinesia and left ventricular (LV) remodeling. While various cellular and biomaterial preparations have been transplanted into the infarct zone in hopes of improving post-MI LV remodeling, an allogeneic cardiac muscle-derived ECM extract has yet to be developed and tested in the setting of reperfused MI. We sought to determine the effects of injecting a novel cardiac muscle-derived ECM into the infarct zone on early dyskinesia and LV remodeling in a mouse model of MI. Cardiac muscle ECM was …
