The cellular microenvironment varies significantly across tissues, and it is constituted by both resident cells and the macromolecules they are exposed to. Cues that the cells receive from the microenvironment, as well as the signaling transmitted to it, affect their physiology and behavior. This notion is valid in the context of stem cells, which are susceptible to biochemical and biomechanical signaling exchanged with the microenvironment, and which plays a fundamental role in establishing fate determination and cell differentiation events. Solid tumors are comprised of multiple cell types which communicate and work together to promote tumor progression. Advances in the treatment of solid tumors have armed clinicians with more efficacious pharmacologic agents and combinations. However, a focus on drug adjuvants in the treatment of solid cancer has left gaps in knowledge regarding non-pharmacologic treatment adjuvants, like aerobic exercise. The current dissertation investigates pharmacologic and non-pharmacologic based approaches to remodel the solid tumor microenvironment landscape and alter therapeutic efficacy. Metabolic disease and obesity affect millions of individuals worldwide and the prevalence of obesity is expected to continue to rise. Mechanisms underlying diet-induced metabolic disease and obesity are complex and are largely unknown. In this dissertation research, we aimed to make advances toward understanding how body temperature and high-fat diet consumption contribute to the development of obesity. Herein, we describe the progress that was made toward three research projects. Pancreatic ductal adenocarcinoma (PDAC) is a devastating disease with dismal prognosis. The only curative option for patients is surgery, but over 80% of patients are not surgical candidates. Unfortunately, PDAC is resistant to the three remaining options. PDAC is characterized by a profoundly hypoxic and immunosuppressive stroma, which contributes to its therapeutic recalcitrance. Alpha-smooth muscle actin+ (αSMA+) cancer-associated fibroblasts (CAFs) are the most abundant stromal component, as well as mediators of stromal deposition. Atlantic killifish (Fundulus heteroclitus) are euryhaline teleosts, which means they can thrive in environments with varying salinities, ranging from concentrated seawater (SW) to dilute fresh water (FW) and thus survive in estuaries. To be able to maintain ionic and osmotic homeostasis in these diverse salinities, osmoregulatory organs like gill and opercular membrane must be able to adjust the permeability and transport of water and ions. INTRODUCTION: Breathwork, or the specific act of controlling breathing to elicit a desired outcome, is not only a potential way to improve one’s mental well-being but also to alter adaptative responses through gas exchange at the capillary level. Increasing tolerance to carbon dioxide (CO2) has been positively correlated with improvement in athletic performance and reduction of stress and anxiety. Advanced age at the time of spinal cord injury (SCI) exacerbates damage from reactive oxygen species (ROS). Mechanisms underlying this age-dependent response are not well understood and may arise from decreased antioxidant defense. We investigated how spinal cord levels of the antioxidant glutathione (GSH), and its regulation, change with age and SCI. GSH is used by GSH peroxidase to sequester ROS and is recycled by GSH reductase. Male and female, 4- and 14-month-old (MO) mice received a 60 kDyn contusion SCI, and the levels of GSH and its regulatory enzymes were evaluated at one and three days post-injury (dpi). The …

The two are highly related; however, the relative association of physical activity and mindfulness on anxiety has yet to be examined. The present study aimed to evaluate the unique variance accounted for by physical activity and mindfulness on anxiety. International Journal of Exercise Science 15(3): 1085-1104, 2022. Individuals with spinal cord injury (SCI) may experience cardiovascular, musculoskeletal and organ function dysregulation. Sequelae include reduced catecholamine secretion and attenuated immune responses which may impact exercise-induced leukocytosis. The purpose of this study was to characterize major leukocyte subtypes following 30 minutes of acute, submaximal aerobic exercise, in line with updated international SCI exercise guidelines for adults. The current study examined the impact of wearing a face mask (FM) at rest and while exercising on cardiorespiratory variables and aerobic performance between males and females. Nine males (21.4 ± 0.5 yr) and 9 females (21.8 ± 3.6 yr) performed a pulmonary function test and a graded maximal exercise test (GXT) on a treadmill with an FM and without an FM in random order. The purpose of this study was to examine Australian Football athletes’ responses to a grade one muscular injury from a psychophysiological perspective to understand the strength of the association between stress, optimism, and cortisol. Forty-five players listed with one professional Australian Football club volunteered for this study. The aim of this study was to assess the effect of repeated-sprint training (RST) on vertical jump ability and aerobic power in college volleyball players. Nineteen male volleyball players, aged between 18-24 years, were randomized into the RST group (RST; n = 10) and control group (CG; n = 9). The RST included 2-3 sets of 6x30m all-out sprints, twice per week, in addition to the regular training routine. PURPOSE: 1) Compare the effects of whole-body heat stress (HS) and resistance exercise (RE) on thermoregulatory responses and skeletal muscle heat shock and hypertrophy related signaling. 2) Examine the effects of acute heat stress (HEAT) on myotube growth and fusion compared to controls (CON), hypertrophy (HYPER), and atrophy (RAPA) treatments in-vitro. METHODS: 1) Eight healthy, physically active and resistance trained individuals (18-45 years) completed RE and HS. 2) 48 hours following treatments, C2C12 myotubes were assessed for myotube area and nuclear fusion index. However, few studies have assessed the prolonged impact of participation in different sports on health and fitness. The main purpose of the current study was to provide an in-depth description of individual player’s intra-game physiological responses during an ice hockey game. A secondary aim was to compare these responses between forwards and defensemen. Six elite junior ice hockey players, three forwards and three defensemen, median (interquartile range) 17 (17–17) years, 182 (180–185) cm, and 78 (74– 80) kg were recruited to participate in the study. Objectives: The purpose of this study was to compare the effects of two acute doses of Capsiate (CAP; 6 vs. 12 mg) on upper body resistance exercise performance in trained men. Methods: Using a randomized, crossover and double-blind design, 20 resistance-trained males were supplemented with low-dose CAP (6 mg), high-dose CAP (12 mg) or placebo 45 minutes before exercise. Anthropogenically driven climate changes are altering marine habitats globally. Rising sea surface temperatures and coastal eutrophication, arising from global warming and coastal nutrient loading, have resulted in progressive ocean deoxygenation. This may restrict available habitat of marine organisms as studies suggest that the balance between metabolic oxygen demand and environmental supply plays an important role in limiting viable habitat and species fitness. As ectothermic predators, with temperature-dependent metabolism and high metabolic demands, coastal shark species may be susceptible to shifts in ocean temperature and oxygen. Ectopic pelvic kidney is a known congenital anomaly; however, the presence of renal cell carcinoma (RCC) in an ectopic kidney is rare with the evidence available in the form of a few case reports only. In this case report, we present a case of metastatic RCC in the pelvic kidney which became a diagnostic challenge because of atypical contrast-enhanced computed tomographic (CT) characteristics and unusual pattern of lymph node involvement including cervical lymph node in the absence of visceral metastasis.
