Purpose: Potassium-Chloride Cotransporter 2 (KCC2) is a neuronal membrane protein specific to the central nervous system. It is responsible for removing Cl- ions from the intracellular space, maintaining a normal Cl- gradient essential for proper function at inhibitory synapses. Dysregulation causes an upward shift in the Cl- reversal potential resulting in a hyperexcitable state of the postsynaptic neuron. Existing literature indicates that KCC2 may be involved in the addiction pathway of a variety of drugs of abuse, including opioids and alcohol. Ticks are the most important arthropod disease vectors in both North America and Europe. Field collection of wild ticks is vital for research on the ecology of vector-borne diseases. Dragging and trapping are the two most common methods for collecting wild ticks. Dragging involves a researcher pulling a canvas through a field to collect ticks searching for a host, while trapping exploits ticks’ natural attraction to carbon dioxide to lure them onto a canvas where they get caught on tape. One of the most important tests in the clinical laboratory is the Complete Blood Count, which involves identifying the white blood cells in a patient’s blood. The respective counts of the different white blood cell types correlate with various states of health and disease and are critical to diagnosing diseases such as leukemia. Leukemic cells (blasts) are considered especially difficult to distinguish, and it is of the upmost importance that these cells are identified correctly. Anaplastic thyroid cancer is a rare, fatal cancer with a five-year survival of 4%. Universally diagnosed at stage IV, anaplastic thyroid cancer is characterized by its lack of differentiation, rapid proliferative rate, highly inflammatory tumor microenvironment, and metabolic dysregulation. Refractory to all established therapies, anaplastic thyroid cancer requires a novel therapeutic approach that targets all of these drivers of anaplastic thyroid cancer carcinogenesis. We propose natural alkaloid berberine as a therapeutic with multitarget efficacy to alter mitochondrial metabolism and reprogram anaplastic thyroid cancer’s aggressive phenotype. Background: Mycoplasmoides genitalium remains a difficult sexually-transmitted infection (STI) to manage due to its potential for antimicrobial resistance and post-infection sequelae. University students are especially vulnerable, as this demographic has the highest rate of STI in the United States. As a result, investigating prevalence rates and therapeutic outcomes in this population is essential to minimize future impact of M. genitalium. The purpose of this study was to investigate a university student population for M. genitalium distribution and treatment outcome.
Design: Retrospective chart-review of university health clinic attendees, augmented by laboratory detection of M. genitalium following therapeutic intervention.

The endangered Blanding’s turtle (Emydoidea blandingii) is a species with a generally diverse diet in their freshwater wetland habitats; they consume prey such as crayfish and snails. In captivity, these options often become more limited due to logistical reasons. As ex situ conservation efforts such as captive rearing and head-starting are becoming more popular for this species, it will be important to gain more insight on how these more limited diets might affect these turtles. Purpose: To determine strength changes and hypertrophy differences at the biceps brachii between an isokinetic bicep curl machine created by C&M Machines and preacher curl exercise with a standard curl bar. The study also aimed to determine the effectiveness of the aforementioned machine and to give insight into this unique method of training.

It is so important to explore the potential side effects of antidepressants especially if these side effects could lead to a future detriment to an individual’s financial and social well-being. A better understanding of these side effects in regards to human developmental issues is imperative considering issues related to mental health and depression affect a large percentage of the population. In this study, we tested if Caenorhabditis (C.) elegans could be a potential model organism to predict possible side effects associated with select antidepressants. Their developmental problems potentially encountered upon drug treatment may be encountered in humans.

Alcohol is associated with increased mortality and morbidity globally. Pulmonary infections with opportunistic pathogens can occur in healthy humans; however, binge alcohol intoxication (≥ 0.08% BAC) is a major risk factor. We have previously shown that a single dose of alcohol comparable to binge alcohol intoxication increases infection by reducing alveolar macrophage function in vivo. The aim of this study was to 1) test the therapeutic potential of the phytonutrient sulforaphane (SFN) given as a pre-treatment, and 2) test the alcohol-induced effects on phagocytic function in murine and human macrophages in vitro.

The purpose of this experiment is to see if there is a correlation between Ozempic (semaglutide) treatment for weight loss and alcohol consumption. Ozempic is a glucagon-like peptide-1 (GLP-1) agonist that lowers blood sugar and appetite as it mimics the effects of GLP-1. In previous studies involving rodents, it has been shown that GLP-1 agonists can also be used to reduce addictive substance intake. We hypothesize that subjects who receive Ozempic treatment will exhibit reduced consumption of alcohol over the span of a month compared to subjects who do not receive Ozempic treatment.

Ksp1 is a casein II-like kinase whose activity prevents aberrant macroautophagy/autophagy induction in nutrient-rich conditions in yeast. Here, we describe a kinase-independent role of Ksp1 as a novel autophagic receptor protein for Ssn2/Med13, a known cargo of Snx4-assisted autophagy of transcription factors. In this pathway, a subset of conserved transcriptional regulators, Ssn2/Med13, Rim15, and Msn2, are selectively targeted for vacuolar proteolysis following nitrogen starvation, assisted by the sorting nexin heterodimer Snx4-Atg20. Here we show that phagophores also engulf Ksp1 alongside its cargo for vacuolar proteolysis. Methamphetamine (METH) abuse remains a global health concern, with emerging evidence highlighting its genotoxic potential. In the central nervous system METH enters dopaminergic cells primarily through the dopamine transporter (DAT), which controls the dynamics of dopamine (DA) neurotransmission by driving the reuptake of extracellular DA into the presynaptic neuronal cell. Additional effects of METH on the storage of DA in synaptic vesicles lead to the dysregulated cytosolic accumulation of DA. Previous studies have shown that after METH disrupts intracellular vesicular stores of DA, the excess DA in the cytosol is rapidly oxidized. This generates an abundance of reactive oxygen species …

Adfluvial Brook Trout Salvelinus fontinalis are known to typically grow larger than riverine Brook Trout. The Kabekona River in Hubbard County, Minnesota exhibits suitable habitat for both fluvial and adfluvial Brook Trout populations. The objective of this study was to analyze the relationships between depth, stream width, canopy cover, water temperature, sediment size, and distance from Kabekona Lake on Brook Trout size distribution in the Kabekona River. Aging is the major non-modifiable risk factor for cardiovascular disease (CVD), which is the leading cause of death in the United States. Many common CVD are diseases of the arteries. Two major hallmarks of arterial aging are 1) increased large artery stiffness and 2) endothelial dysfunction which is characterized by impaired endothelium dependent vasodilation and blunted nitric oxide (NO) bioavailability. Importantly, these adverse alterations in the arteries occur prior to, and are predictors of the onset of overt CVD. Aside from their classical role in maintaining hemostasis, platelets are considered first-line vascular guardians that influence both the innate and adaptive immune systems. This is mediated by either direct interactions with immune cells or through the release of pre-stored cytokines and chemokines upon interacting with pathogens such as viruses. Prostate cancer is the leading cause of cancer-related deaths among men worldwide, and the development of resistance to existing treatments necessitates the exploration of new therapeutic strategies, including combination therapies. This study investigates the role of Polo-like kinase 1 (PLK1), located on chromosome 16p12.2, as an oncogene in prostate cancer. We generated a transgenic mouse model with prostate-specific overexpression of PLK1 to closely mimic human prostate cancer progression. Quantitative PCR (qPCR) analysis was employed to characterize the molecular profile of PLK1, revealing its implication in multiple pathways. Myotonic dystrophy type 1 (DM1) is a slowly progressive, multisystem disorder caused by a CTG repeat expansion in the DMPK 3’UTR that leads to global dysregulation of alternative splicing. The resulting decline in physical function is slow, and no reliable biomarkers exist for predicting disease progression; however, an RNA mis-splicing biomarker associated with weakness may have utility in predicting functional outcomes. The purpose of this research is to improve understanding of the neurodevelopmental effects of embryonic exposure to elevated inflammation and oxidative stress induced by the antipyretic drug acetaminophen (APAP). Our study was the first to examine the interactive effects of APAP and inflammation in zebrafish embryos and how the treatments affect brain development and larval behavior. Experimental groups of zebrafish larvae were exposed to lipopolysaccharide (LPS) to induce inflammation, APAP, or LPS + APAP and larval behavior was analyzed using Ethovision automated behavioral tracking software. BACKGROUND: Muscle unloading such as bed rest or space flight decreases strength and cross-sectional area concomitant with a slow-to-fast fiber type shift, of anti-gravitational muscles such as the quadriceps. Diffusion tensor imaging (DTI) quantifies tissue anisotropic properties such as Fractional Anisotropy, Mean Diffusivity, Axial Diffusivity, and Radial Diffusivity, as well as muscle volume, fascicle length, and physiological cross-sectional area, and has recently been adapted to assess skeletal muscle structure. In skeletal muscle, anisotropic properties correlate with muscle fiber type and contractile function and may be responsive to unloading-induced atrophy.

Glioblastoma (GBM) is the deadliest and most common malignant CNS tumor. Despite advances in understanding its biology and the development of targeted therapies, effective treatment options remain limited. As a result, patient survival rates are dismal, with a 5-year survival rate of only 6.8% and an overall survival of less than two years. This poor outcome is partly due to our inability to treat GBM cells that infiltrate the whole brain. Focal treatments like surgery and radiotherapy alone are insufficient due to these invasive tumor cells. Obesity is a multifactorial disease with many comorbidities leading to multiorgan dysfunction. Current studies point towards dysregulation of platelet signaling as a driver for increased risk of cardiovascular disease and stroke. The exact mechanisms remain elusive due to the complex interplay of factors involved, warranting the need to further understand how platelets are affected. Our preliminary data evaluating changes in platelet and coagulation parameters from bariatric patients showed that obesity did not affect platelet count or clotting time as demonstrated in INR values. Bioengineering has made significant contributions to the treatment of orthopedic injury through interdisciplinary approaches. Central to these advancements are many orthopedic implants, including anchors and joint replacements, which have been designed to restore mobility. Despite their functional efficacy, these products have limited lifespan and are associated with many complications, such as implant-associated infection. The success of implant design and function, both in the short and long term, is paramount for enhancing patient outcomes and overall well-being. Female Genital Schistosomiasis (FGS) is caused by Schistosoma haematobium, which causes chronic gynecological conditions that lead to substantial morbidity and infertility. This study’s objective is to determine the prevalence and burden of FGS based on the presence of S. haematobium-specific DNA in females across age groups using our previously field-acquired filtered human urine samples from Zambia, Tanzania, and Ghana, collected over multiple years.
