Insects are crucial for ecosystem functions, such as pollination and food web maintenance. This study aims to assess insect biodiversity in La Salle Botanical Gardens (LSBG) in Porac, Pampanga, focusing on its grassland and forestland areas. Using rapid biodiversity assessment, this study provides baseline data on insect diversity, abundance, dominance, and evenness for conservation. Sampling employed transect line intercept and quadrat methods. Specimens were collected using different collection and preservation techniques. The purpose-bred dog breeding industry for research is little known. Each year, thousands of dogs are bred for scientific and medical testing, supplied to research facilities by a handful of major commercial breeders. But gaps in enforcement and regulation have allowed abuses to persist, even as the industry increasingly turns to alternatives to animal testing. Animal rights advocates have been decrying the treatment of these dogs. A federal Inspector General report found lax oversight of the industry, raising further concerns about how dogs are treated. Integrin beta 1 (ITGB1) is an adhesion receptor that link cells to the extracellular matrix and regulates pathways involved in survival, proliferation, and migration. In order to understand its role in Ewing Sarcoma, we applied CRISPR-Cas9 with two guide RNAs to knock down ITGB1 in ES8 cells. We then assessed changes in cell growth, death, movement, and gene expression using Incucyte live-cell imaging, colony formation assays, wound healing assays, and qRT-PCR. qRT-PCR showed partial ITGB1 reduction, with ITGB1_gRNA2 producing the strongest decrease. ITGB1_gRNA2 also led to a small increase in caspase activity and a 20-30% increase in proliferation at the …

Buffer Vs. Swabs: Efficacy Of Carrier Nucleic Acids In Robotic Extractions, Shawn Cecil

This study compared the efficiency of two carrier nucleic acid methods, specifically using salmon sperm DNA (spDNA), in robotic solid-phase extractions of cotton swab samples. Robotic extractions reduce processing time and workload but result in substantially lower DNA recovery than manual organic extraction. This research tested 10 µg of spDNA being added to the lysis buffer or pre-treating the swabs with it before DNA was added. Both methods showed a significant increase in recovery compared to extractions with no carrier. In the buffer extractions, recovery ranged from 69.4% to 117.5% and the swabs being pre-treated ranged from 84.2% to 125.8% …

Droplet-based microfluidics has rapidly advanced numerous fields, including chemistry, biology, materials science, medicine, food science, and cosmetics. In these systems, fluorocarbon oil combined with fluorinated surfactants is the preferred medium for fluid encapsulation, offering exceptional stability and biocompatibility essential for sensitive biological and chemical processes. Knowledge of novel and revised bacterial taxonomy facilitates routine operations in the medical microbiology laboratory and communication with important stakeholders. Notable new gram-positive taxa accepted in 2024 include the clinically significant Staphylococcus brunensis sp. nov., as well as Streptococcus suis subsp. hashimotonensis subsp. nov., which can be delineated from other Streptococcus suis subsp. by possession of the Lancefield group A antigen. Four novel Enterobacterales species are discussed, plus a novel eighth family Gallaecimonadaceae. One of these, Providencia huashanensis sp. nov. is clinically significant and harbors multiple genetic determinants that encode antimicrobial resistance mechanisms. Clinical transplantation demands have increased immensely in recent years and, along with increasing supply shortages, many barriers to successful organ donation arise from a lack of dedicated testing to rule out microbial or bloodborne infections. Investigating The Effects Of Cbd And Caffeine Interaction On Embryonic Development In Xenopus Laevis, Jeanesia M. Pierre

This study examined the interactive effects of cannabidiol (CBD) and caffeine on Xenopus laevis embryos over a 96-hour exposure period. Developmental toxicity was assessed using toxic unit isobole analysis, with LC₅₀, EC₅₀, and teratogenic index (TI) values calculated for CBD, caffeine, and mixtures at 3:1, 1:1, and 1:3 ratios.
Results showed that mixtures produced concentration-dependent interactions. When caffeine levels were higher, the mixtures displayed synergistic toxicity; as the proportion of CBD increased, the interaction shifted toward a concentration-addition model. Mortality was elevated in mixtures relative to single-compound exposures, while malformation rates decreased, indicating reduced teratogenicity. Overall, findings suggest that CBD …

Description of new and revised taxa originating from non-domestic animal species continues to be pursued. The majority are bacteria isolated from the alimentary system of a variety of healthy wildlife, adding to our understanding of microbial flora present in the normal state. A few new and revised taxa are associated with disease, including a novel Erysipelothrix species associated with sepsis in albatross and a revised species, Allocoenonia anatina comb. nov., associated with Riemerella anatipestifera-like disease in ducks and geese. Music influences emotion, physiology, and cognition, yet little is known about how the frequency it is tuned to affects these influences. Prior research has shown that music tuned to 432 Hz can reduce stress, lower blood pressure, and improve sleep. My colleagues and I conducted two studies to investigate this. Our research found that music tuned to 432 Hz promotes a significant increase in memory retention and induces a state of focus and relaxation. These results suggest that shifting from the standard tuning frequency of 440 Hz to 432 Hz could have a profoundly positive impact on our daily lives. …

Recognizing and updating bacterial names is key to communication between the veterinary clinical microbiology laboratory, veterinarians, and clients. Moraxella oculi sp. nov., distinct from Moraxella bovis and Moraxella bovoculi, was isolated from a cow with infectious bovine keratoconjunctivitis. Several respiratory pathogens were recognized, including Neisseria leonii sp. nov. described from rabbits, Mannheimia indoligenes sp. nov. described from cattle in Europe, and Moraxella haemolytica sp. nov. described from a goat in China. Stenotrophomonas forensis sp. nov. is a novel designation within the Stenotrophomonas maltophilia complex associated with isolates derived from horses. Aim: In sub-Saharan Africa, approximately 40 million pregnant women are exposed to parasitic diseases such as malaria caused by Plasmodium falciparum, Schistosome parasites, and soil-transmitted helminths (STHs). When parasitic diseases share the same habitat and overlap in distribution, then high co-infection rates occur. The co-infection can lead to consequences for the child, such as intrauterine growth retardation, low birth weight, pre-term delivery, and neonatal mortality.

The bay scallop (Argopecten irradians), a commercially and ecologically significant bivalve along the Eastern coast of the United States, has faced severe population declines due to various anthropogenic impacts. At the same time, the proliferation of plastics, particularly in maritime industries, has introduced new threats to marine environments through both physical pollution and chemical leaching. This study aimed to assess the acute toxicological effects of leachates from two commonly used plastic products, polyethylene Netron and polypropylene rope, on A. irradians larvae. Rising ocean temperatures and increasingly frequent marine heatwaves threaten the physiological performance and survival of marine larvae. This study quantifies the thermal tolerance, respiration, and swimming behavior of Chthamalus fissus cyprid larvae to establish a baseline for understanding their vulnerability under future climate scenarios. Larvae were collected from the Scripps Pier in La Jolla, CA and exposed to a range of temperatures (12–50°C, dependent on the experiment) to assess oxygen consumption, mortality, and swimming speeds. Oxygen consumption rates increased significantly with temperature, peaking at 30°C, with cyprids exposed to temperatures above 26°C showing significantly higher respiration rates than those below …

Many preoperative and postoperative cataract patients struggle with comprehending the use of prescription medication as directed. Language barriers and low health literacy levels are major factors contributing to improper use of prescriptions. To increase patients comprehension, the Fort Lauderdale Eye Institute employed an educational intervention consisting of a live presentation and an instructional video. Results found that 44% of patients were hesitant to ask questions to clinical staff, 32% felt overwhelmed, and nearly 70% lacked confidence in using their prescribed eye drops. Following the intervention, 91% of patients reported increased confidence in their medications, and most indicated that the video …

The competencies identified in the development of the “Fundamental Characteristics of the Translational Scientist” may not consider and fully integrate the concepts of knowledge integration and engagement, key components of the translational science spectrum. This gap may be widening and deepening the “valley of death” and exacerbating ongoing challenges in the field of translational science. This dissertation explored the association of knowledge integration, stakeholder engagement, and community engagement principles compared with translational science competencies.

However, currently, the only effective cure is to undergo surgery to augment or receive a healthy donor heart, which could be very difficult due to the long waiting list and the associated risks. Breast cancer remains one of the most prevalent diseases worldwide, yet therapeutic equity is still lacking. Standard treatments including surgery, radiotherapy, chemotherapy, and long-term hormone therapy. These approaches are often accompanied by significant side effects. While newer methods such as immunotherapy and targeted therapies have emerged, chemotherapy protocols and incorporation have remained largely unchanged in the past decades. Preliminary experimentation has suggested that a research-use-only real-time transcription-mediated amplification assay for Treponema pallidum (RUO T. pallidum TMA) yields instances of T. pallidum nucleic acid detection that coincide with non-treponemal serology in men who have sex with men (MSM) at increased risk for sexually transmitted infection. To further characterize the specificity of RUO T. pallidum TMA testing, 3,586 rectal swab specimens reported as “not detected” by the assay generated a mean endpoint FAM fluorescence of 637.5 units. Introduction of Treponema denticola, Treponema phagedenis, and Treponema refringens nucleic acid into matrices generated mean endpoint FAM fluorescence ranging from 620.7 …

Recent work has optimized parameters of a real-time reverse transcriptase PCR-based laboratory-developed test on the Panther Fusion system which detects Mycoplasma genitalium-specific macrolide resistance-associated mutations LDT (MRM-LDT) from primary swab and urine specimens. In this study, MRM-LDT was applied to a large multi-demographic study set to further characterize M. genitalium resistance in the United States. A total of 2,145 primary clinical specimens testing positive for M. genitalium 16S rRNA by transcription-mediated amplification (TMA) were initially titered by the same assay using serial 10-fold dilutions to determine relative target nucleic acid burden. DNA extraction methods are evolving, from several steps using organic solvents to recent technology using magnetic nanoparticles as adsorbents for the purification step. BACKGROUND: Acute myeloid leukemia (AML) is an aggressive hematologic cancer with a notoriously bleak prognosis; for non-M3 AML, the overall 5-year survival rate is ∼30%. While 60-70% of newly diagnosed AML patients will achieve complete remission (CR), half of these patients will experience relapse (secondary resistance) by three years from their diagnosis. Moreover, 30-40% of AML patients present with refractory disease (primary resistance) and cannot respond to frontline treatments. Leukemia stem cells (LSCs) are implicated in both primary and secondary resistance, and their eradication is necessary to maintain CR. Transfer RNA (tRNA) is essential for accurate translation of genetic information and plays a role in cellular processes such as lipid aminoacylation and bacterial conjugation. Base modifications help tRNA fold correctly and function properly. An important base modification, N6-threonylcarbamoyladenosine (t6A) occurs at position A37 in the anticodon-stem-loop of tRNA. This modification is crucial for decoding ANN codons. Interestingly, an Archeal organism, Methanocaldococcus jannaschii, can have either t6A and hydroxynorvalylcarbamoyl-6- adenosine (hn6A) modification at the same position on tRNAs. As Tsa proteins catalyze the formation of both t6A and hn6A modifications, this raises the question of whether discrimination between these …

Developing efficient and durable electrocatalysts for the oxygen evolution reaction (OER) and hydrogen evolution reaction (HER) is crucial for advancing sustainable energy conversion technologies. In this study, high-entropy (FeCoNiMnAl)3O4 and its hybrid forms supported on carbon nanotubes (CNTs) and reduced graphene oxide (rGO) were synthesized with varying carbon concentrations (5 mM, 10 mM, and 15 mM) and evaluated for their bifunctional electrocatalytic performance in alkaline media. The rGO-supported catalyst with 10 mM concentration exhibited the best HER performance, achieving a low overpotential of 162 mV at 10 mA/cm², followed by rGO (15 mM) and rGO (5 mM) …

In this thesis, we applied 1,4-butanediol, which is an emerging biobased and low-cost monomer, was synthesized with a series of aliphatic dicarboxylic acids, namely succinic acid, adipic acid, and sebacic acid using melt polycondensation to Biobased polyesters. The resulting polymers poly(butylene succinate) (PPeS), poly(butylene adipate) (PPeA), and poly(butylene sebacate) (PPeSe) chemical structures were analysis through Fourier transfer infrared spectroscopy(FT-IR), 1H nuclear magnetic resonance (NMR) revealed the elimination of side reactions in the obtained polyesters. The design and development of atomically dispersed M-N-C catalysts (metal (M) supported on an nitrogen-carbon (NC) matrix with high multifunctional electrocatalytic performance is desirable but proved to be very challenging. Herein, we synthesized M-N-C catalysts (M = Fe, Co, Mn, and Ni) using Zn-assisted high temperature treatment and characterized using various techniques. The prepared catalysts were tested for their electrocatalytic performance towards oxygen and hydrogen evolution reaction (OER and HER) as well as oxygen reduction reaction (ORR) in alkaline media.
