The gut–brain axis represents a dynamic two-way signaling network whose dysregulation has been implicated in a wide range of neurogastrointestinal disorders. In this context, extracellular vesicles (EVs) have emerged as critical mediators of intercellular signaling and as promising non-invasive biomarkers. Derived from host and microbial cells, EVs carry bioactive cargo—including proteins, lipids, nucleic acids, and metabolites—that reflect the physiological or pathological state of their cells of origin. Their ability to cross biological barriers, such as the blood–brain barrier, underscores their potential for diagnosing and monitoring gut–brain axis dysfunctions. In July and August 2025, China’s People’s Liberation Army (PLA) employed a unique type of civilian deck cargo ship known as a Landing Craft Tank (LCT) to transport military vehicles directly to shore as part of an amphibious landing exercise. While civilian deck cargo ships of this type have been observed in previous PLA over-the-shore logistics exercises, they had only been used to transport vehicles and equipment from port-to-port—simulating the transport of forces into a captured port—not directly to an unimproved beachhead. Despite the harm of substances, coworkers in hospitals, healthcare facilities, and urgent care centers found themselves among healthcare professionals who misused substances such as alcohol and prescription medication. Grounded in social cognitive theory, the purpose of this qualitative descriptive phenomenological study was to explore the lived experiences of coworkers who have been exposed to healthcare professionals’ substance misuse. The participants consisted of 12 healthcare professionals working in various hospitals, healthcare facilities, and urgent care centers in the United States. The experiences of parenting a child with autism spectrum disorder (ASD) have been widely studied in Western contexts. Still, limited research has explored how sociocultural factors shape these experiences in India. This qualitative study aims to address this gap by exploring and comparing the lived experiences of mothers and fathers raising a child with ASD in Kerala, India. The purpose of this study was to investigate how gender roles, cultural expectations, and social structures impact parental adaptation, coping, and caregiving practices. The COVID-19 pandemic caused a sudden transition from face-to-face to online instruction. The problem that was addressed through this study is that little is known about the experiences of community college faculty members when teaching online in the post-pandemic era. Grounded in Hall’s and Hord’s concerns-based adoption model, the purpose of this qualitative study was to explore the experiences of community college faculty members’ perceptions of their teaching experiences working online post-pandemic. Recent developments in spatially resolved -omics have enabled the joint study of gene expression, metabolite levels and tissue morphology, offering greater insights into biological pathways. Integrating these modalities from matched tissue sections to probe spatially-coordinated processes, however, remains challenging. Here we introduce MAGPIE, a framework for co-registering spatially resolved transcriptomics, metabolomics, and tissue morphology from the same or consecutive sections. We show MAGPIE's generalisability and scalability on spatial multi-omics data from multiple tissues, combining Visium with MALDI and DESI mass spectrometry imaging. Physical inactivity among older adults remains a major national and global public health concern linked to preventable morbidity, mortality, and rising healthcare costs. Physical therapists are uniquely positioned to promote physical activity (PA) through health education, yet their engagement in this role is not well defined. This quantitative cross-sectional study examined associations between clinical practice factors—years of experience, board certification status, and practice setting—and three domains of the theoretical domains framework (knowledge, skills, and behavioral regulation) related to PA promotion among older adults. Depression exists as a common yet often undetected mental health issue which affects many elderly residents who live in long-term care facilities. The combination of multiple depression screening tools with inadequate staff training leads to delayed resident diagnosis and produces variable treatment methods which raises the possibility of negative results for residents. This Doctor of Nursing Practice (DNP) project addressed a practice gap related to inconsistent implementation of the Patient Health Questionnaire–2 (PHQ-2) and Patient Health Questionnaire–9 (PHQ-9) through the development and implementation of an evidence-based staff education intervention in a long-term care setting. A Policy Reversal As Structural Break: A Lesson From China’S Land Lease Financing For Infrastructure, Yilin Hou, Lin Li, Lei Shao

Land-based revenues as a fiscal instrument to finance infrastructure tie public investment to real estate markets, where demand fluctuations cause fiscal consequences. This paper takes policy reversals as structural breaks and examines their impacts on infrastructure investment. We dissect China’s 2016 policy reversal that disallowed developers to finance land purchase with debt, causing sharp declines in local government land revenues, slashing their fiscal base for infrastructure investment. Using prefecture-level panel data and continuous difference-in-differences identification, we estimate the reversal reduced land-lease revenues by 55%. Two-stage least squares estimates suggest a 10% land-revenue decline translates into 6.6% investment reduction. Decomposing the …

While there has been some progress on discovering clinically validated biomarkers for early detection in pancreatic ductal adenocarcinoma (PDAC), several challenges remain. Most approaches rely on single-modality biomarkers with limited sensitivity and/or specificity. Using data from a multicenter study with an age-matched cohort (n = 203 with healthy controls n = 46, pancreatitis controls n = 36, and diagnosed cases n = 121), we developed a machine learning approach integrating 2,096 microRNAs, 125 metabolites, and CA19-9. The paper presents simulation and experimental results to demonstrate heat transfer performance during cooking that can be practical in pot-in-pot arrangements for indirect cooking/heating. The cases included single pots, pot-in-pot with oil, and pot-in-pot with no oil. The aim was to optimize the heat transfer through conduction and the cooking process for a recently developed small-scale sensible heat storage system integrated with a cooking unit. The purpose of the study was to determine the change in storage of two water sheds after the impact of two rain events, considering factors such as rainfall, discharge and infiltration, and evapotranspiration. The two sub-watersheds, located on the Edward’s Aquifer Authority Field Research Park, each had a weir. Global biomedical and health research is increasingly relying on genomic and computational approaches, largely driven by the increasing volumes of nucleic acid sequencing. Concurrently, epidemiological studies and clinical records are generating enormous amounts of data amenable to disease modeling, machine learning, and artificial intelligence techniques. Background: The development of relevant and robust large animal models of hepatocellular carcinoma is needed to test new therapeutic strategies for this disease. Transgenic approaches hold promise in addressing this complex problem. One such model, the Oncopig, has been reported to develop tumors of up to 4 cm in diameter within 7-14 days at sites of in situ vector inoculation. However, the resulting lesions reportedly contained an extensive inflammatory component that has not been evaluated in detail.

To understand how the tumor immune microenvironment (TIME) impacts clinical outcomes and treatment response, researchers have been leveraging single-cell protein multiplex imaging techniques. These technologies measure multiple protein markers simultaneously within a tissue sample, providing a more complete assessment of the TIME. However, statistical challenges arise from the over-dispersed and zero-inflated nature of the data and from relationships among different immune cell populations. Objectives Perioperative randomised controlled trials (RCTs) in liver transplantation are relatively infrequent. RCTs performed in this complex patient population need to be robustly conducted to maximise patient benefit and graft utility given the scarcity of donor organs. Recruitment challenges can compromise RCTs and studies in this population face unique challenges due to recipient illness severity, their comorbidities, demographics and the geographical constraints of specialist transplant centres. Emergency presentation and after-hours admission may further limit patients’ capacity or readiness to consider trial participation. We report direct measurements of changes in diving and movement behavior for 53 goose-beaked whales (Ziphius cavirostris) in relation to experimentally controlled mid-frequency (3–4 kHz) active sonar (MFAS) signals. These signals simulate powerful Navy sources that have been associated with multiple mortal stranding events for this species. We deployed a multi-scale combination of tags to monitor individual whales, including 50 long-duration (weeks), coarse-resolution satellite-transmitting tags and 3 short-duration (hours), high-resolution archival depth, orientation, and acoustic tags. ESR1 mutations are the leading cause of endocrine therapy resistance and progression in ER-positive metastatic breast cancer. ESR1 mutations are detected in ~50% of metastatic breast cancer patients, and identification of effective targeted therapeutics are critically needed. Here, we identified enrichment of dysregulated replication stress and DNA damage responses in multiple ESR1 mutant models. Targeting the replication stress response utilizing checkpoint inhibition in combination with PARP inhibition synergistically suppressed growth, induced cell cycle arrest, and attenuated DNA replication. Mutations in epigenetic regulators are common in bladder cancer, yet their impact on therapeutic responses remains unclear. Here, we identify that loss-of-function mutations in KDM6A, a histone demethylase altered in about 26% of advanced bladder cancers, are associated with poor survival after cisplatin chemotherapy, whereas they correlate with improved outcomes with anti-PD-1 therapy. Using CRISPR-Cas9-engineered murine and human bladder cancer models, we show that KDM6A deficiency increases formation of extrachromosomal circular DNA carrying chemoresistance loci, promoting cisplatin resistance. In parallel, KDM6A loss impairs DNA repair and rewires tumor metabolism, reducing glycolysis and lactate output. This metabolic shift diminishes histone lactylation …

Spatially resolved transcriptomics (SRT) is a promising new technology that enables simultaneous analysis of gene expression and spatial information for biomedical research. However, the existing statistical and deep learning algorithms used for analyzing SRT data rely solely on two-dimensional (2D) spatial coordinates, which limits their ability to accurately identify spatial domains, spatially variable genes (SVGs), cell-to-cell communications, and developmental trajectories in a three-dimensional (3D) spatial manner.
