Aging of the blood system impacts systemic health and can be traced to hematopoietic stem cells (HSCs). Despite multiple reports on human HSC aging, a unified map detailing their molecular age-related changes is lacking. We developed a consensus map of gene expression in HSCs by integrating seven single-cell datasets. This map reveals previously unappreciated heterogeneity within the HSC population. It also links inflammatory pathway activation (TNF/NFκB, AP-1) and quiescence within a single gene expression program. This program dominates an inflammatory HSC subpopulation that increases with age, highlighting a potential target for further experimental studies and anti-aging interventions.

In vitro fertilization is a relatively new medical practice that helps people who face infertility be able to reproduce. Reproductive medicine providers face ethical dilemmas often in their field, only some of which have standards of care. There are questions in the medical landscape pertaining to physician autonomy versus patient autonomy, which heavily apply to reproductive medicine providers. Patient autonomy is highly valued in medical ethics; however, physician autonomy should also be considered. The co-occurrence of per- and polyfluoroalkyl substances (PFAS) and nanoplastics (NPs) poses a synergistic threat to environmental and human health, yet the molecular mechanisms governing PFAS–NP complexation and membrane interactions remain unclear. Using atomistic molecular dynamics simulations, we investigated the adsorption of neutral polytetrafluoroethylene (PTFE) and anionic perfluorinated compounds (perfluorooctanoic acid, PFOA, and perfluorooctanesulfonic acid, PFOS) on polystyrene NPs (3.1 and 6.7 nm) and their interactions with 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) membranes. Lipid nanoparticle (LNP)-encapsulated nucleoside-modified mRNA vaccines elicit robust CD4+ T cell responses, yet the mechanisms underlying this T cell priming remain unknown. Antigens presented to CD4+ T cells on major histocompatibility complex class II (MHC II) are traditionally acquired by antigen presenting cells (APCs) from extracellular sources. Here we show that vaccine specific CD4+ T cell responses instead rely on antigen directly expressed within APCs, without extracellular transit. Murine APCs treated with mRNA-LNP vaccines activate T cells more efficiently when presenting antigen produced internally, rather than acquired externally. The LEGEND Collaboration is searching for neutrinoless double-beta (0νββ) decay by operating high-purity germanium detectors enriched in 76Ge in a low-background liquid argon environment. Building on key technological innovations from the GERmanium Detector Array (GERDA) experiment and the MAJORANA DEMONSTRATOR experiment, LEGEND-200 has performed a first 0νββ decay search based on 61.0 kg yr of data. Over half of this exposure comes from our highest performing detectors, including newly developed inverted-coaxial detectors, and is characterized by an estimated background level of 0.5+0.3−0.2 cts/(keV ton yr) in the 0νββ decay signal region. Background: Antibiotic overuse in Neonatal Intensive Care Units (NICUs) is a major contributor to antimicrobial resistance and adverse neonatal outcomes. This study aims to evaluate baseline antibiotic utilization (AU), identify factors influencing variability, and assess the impact of neonatal characteristics and sepsis incidence. Methods: A multicenter retrospective analysis examined AU in seven NICUs from 2019 to 2023, involving 25,532 neonatal admissions during national antibiotic stewardship program implementation. Data encompassed neonatal clinical parameters, sepsis incidence, and AU metrics, including days of therapy (DOT) per 1000 patient-days. Statistical analyses included correlation assessments and multivariate regression to identify determinants of antibiotic use. Results: …

Purpose: RAPGEF2 encodes a guanine nucleotide exchange factor (GEF) that activates small GTPases and has not been linked to a Mendelian disorder. RAPGEF2 is highly intolerant to loss-of-function variants. We report 5 de novo heterozygous variants in RAPGEF2 in unrelated individuals with developmental delay, attention deficit hyperactivity disorder, epilepsy, dysmorphic features, or other manifestations. We used a Drosophila model to assess the functional impact of the identified human variants.

Methods: We generated a Kozak-GAL4 null allele of the Drosophila ortholog of RAPGEF2, PDZ-GEF, and used the allele to determine the gene expression pattern as well as the loss-of-function phenotypes. We …

Purpose The rapid evolution of antibiotic-resistant bacteria poses an urgent global health crisis. A key gap in current antibiotic discovery approaches is the absence of automated chemical synthesis methods designed to systematically generate and evaluate compounds within specific antibiotic classes. We address this gap through fragment-based computational experiments that systematically explore antibiotic chemical spaces.

Methods Our computational methodology consists of three steps: fragmentation of known compounds (eMolFrag), generating new molecular structures by recombining fragments (eSynth), and filtering of candidates based on desired properties (eFilter). eFilter combines structural analysis, pathway information, and protein targets …

Student Disruptions And Teacher Turnover, Kyuhan Choi, Ying Shi, Maria Zhu

This paper examines how exposure to disruptive students affects teacher retention using linked teacher–student administrative records from North Carolina. To address non-random classroom assignment, we instrument for classroom exposure using the school-by-grade share of disruptive students based on prior-year disciplinary infractions. A 10 percentage point increase in the share of disruptive students raises the probability of an early-career teacher leaving the school in the following year by 1.0 percentage points. Notably, working in a school environment with supportive leadership and greater teacher autonomy, particularly around student conduct policies, mitigates the impact of student disruptions.

This article analyzes the FDA's proposed policy plan issued in May 2025 regarding the approval of Covid-19 boosters for different groups of persons. It offers two constructive recommendations that will help prevent constitutionality challenge on impermissible intrusion of the freedom of health.

The first is to avoid using the confusing term "benefit" but instead focus on the statutory parameters of "safety, purity, and potency." The affected group can make a strong case that their free choice of vaccination should not be deprived, as long as the vaccine qualifies for the statutory thresholds of being safe, pure, and potent. This paper reports results from a discrete choice experiment conducted with Oregon residents regarding possible policy changes in spatial management of nearshore habitat. We evaluate public preferences across several policy scenarios, each characterized by varying levels of marine reserve size (bounded areas where extractive activities are prohibited), coastal jobs generated or lost, and carbon sequestration by seagrass beds, tidal marshes and kelp forests(blue carbon habitat expansion), with models estimated in both utility and willingness to pay (WTP) space. Efficient environmental search is an important and adaptive everyday skill. A particular feature of theoretical interest is whether large-scale search is informed by the spatial statistics of the environment - probability cueing is a robust effect in two-dimensional visual search tasks, but studies of large-scale search have generated equivocal findings. Here, we examined whether sensitivity to a statistical cue specified within an allocentric reference frame is modulated by the presence and location of environmental landmarks. Now that artificial intelligence tools for lawyers are widely available, we decided to integrate them for a semester in our Entrepreneurship Clinic. We have some important takeaways for legal education in general and the transactional practice of law in particular.

First, employers and educators need to account for law students who already are using AI tools in their legal work and guide new lawyers about how to use such tools appropriately.
