Background and Significance: Ewing Sarcoma (EWS) is the second most common malignant bone tumor of children and adolescents. Patients with metastatic or recurrent disease have very poor outcomes. The receptor tyrosine kinase(RTK) insulin-like-growth-factor-1-receptor (IGF-1R) is upregulated in 93% of EWS patients with anti-IGF-1R antibodies and kinase inhibitors in clinical studies. However, with only ~10% of patients achieving objective responses, delineation of novel pathways that facilitate IGF-1R-driven oncogenesis in EWS could provide avenues for more effective therapy. The RTK levels and compartmentalization at the cell surface determine their access to growth factors, thus dictating the downstream oncogenic signaling. Our lab has …

Child Health Research Institute Pediatric Research Forum

Background: Access to medically indicated caesarean sections is an essential strategy for reducing maternal and infant mortality rates worldwide. However, overuse of medically unnecessary caesarean sections is associated with excess maternal-child morbidity. Previous studies suggest higher rates of caesarean section among women who identify as racial/ethnic minorities.

Significance of Problem: Despite national efforts to prioritize the reduction of medically unnecessary caesarean sections, caesareans rates in the United States have remained stable over the last decade. Women who identify as racial or ethnic minorities experience disproportionally higher rates of caesarean, even when controlling for demographic, behavioral, medical, and institutional level factors. …

Exploring Challenges And Triumphs Within The Transgender Community Through Comics-Based Research, Carly Caldwell May 2022

Exploring Challenges And Triumphs Within The Transgender Community Through Comics-Based Research, Carly Caldwell

Art Therapy Counseling Final Research Projects

Transgender people are marginalized and pathologized within our society, which leaves transgender individuals vulnerable to numerous negative outcomes. The minority stress model describes the factors experienced by marginalized communities contribute to adverse outcomes (Meyer, 2015). Health care professionals are often not properly equipped to serve the needs of transgender individuals due to marginalization and pathologization. Used to challenge harmful stereotypes, as a form of cultural resistance, and to incite social change comic books and zines have been used as forms of expression for individuals with marginalized identities. Through the creation of a comic book, I aimed to educate others, encourage …

Conductive thread is a common material in e-textile toolkits that allows practitioners to create connections between electronic components sewn on fabric. When powered, conductive threads are used as resistive heaters to activate thermochromic dyes or pigments on textiles to create interactive, aesthetic, and ambient textile displays. In this work, we introduce Embr, a creative framework for supporting hand-embroidered liquid crystal textile displays (LCTDs). This framework includes a characterization of conductive embroidery stitches, an expanded repertoire of thermal formgiving techniques, and a thread modeling tool used to simulate mechanical, thermal, and electrical behaviors of LCTDs. Through exemplar artifacts, we annotate a …

Resource Use And Water Implications Of Material Consumption In Consumer Electronics, Hema Madaka May 2022

Resource Use And Water Implications Of Material Consumption In Consumer Electronics, Hema Madaka

Theses

Rapid technological innovation has introduced a broad spectrum of materials in the consumer electronics sector. Consumption of these materials increases the demand for water and potentially discharge contaminants into the water resources across their life cycle, exacerbating freshwater scarcity and pollution. These water impacts have not yet been fully studied, as most literature on consumer electronics focuses on supply chain energy, carbon footprint, or end of life management. Evaluating water impacts requires data on material content, life cycle water consumption and emissions at spatial level, and availability of impact assessment models that connects life cycle data to water impacts. Data …

Personality Type Posters, Sabrina Awdi May 2022

Personality Type Posters, Sabrina Awdi

Theses

Humans are highly visual creatures and have been responding to visual cues from the beginning of time for survival and information. Visual design is an important tool to use for to convey ideas through imagery instead of words or typed out information. Using design to help the person visually decipher and interpret information can serve as a solution in many aspects of life. I am using personality types to show introversion, extroversion, ambiversion, and empathy visually through the use of digital art (illustration) and graphic design. The goal is to evoke an emotional response from the viewer to be able …

Between, Abigail Benkovich May 2022

Between, Abigail Benkovich

Theses

I’m finally finding safety through learning to set boundaries in my life, which has carried into my artwork as physical barriers, whether worn or observed. Through the protection of volume, space, netting, and sharp objects, I can reclaim control over my immediate surroundings and my body. My sculptures were constructed to protect the body. As I dove deeper into why that brings me comfort, I realized it’s a reaction to the current culture that objectifies women’s bodies. This is specifically significant to me as I grew up subjugated to purity culture within the Evangelistic Christian church, which often leaves young …

Probing Convergence In Orthogonal Conjugated Catalysis By T4 Lysozyme Utilizing Biophysical Characterization, William Turner May 2022

Probing Convergence In Orthogonal Conjugated Catalysis By T4 Lysozyme Utilizing Biophysical Characterization, William Turner

Master of Science in Chemical Sciences Theses

Conjugated polymers have become highly attractive as they afford unique material properties that make them promising for a wide range of applications, such as photovoltaics and drug delivery systems. However, these conjugated polymers require extensive synthetic steps involving hazardous organic solvents or metal-based catalysts yielding toxic waste streams. To remedy this, enzymes have emerged as a highly valuable alternative to synthesizing these polymers as they are able to be produced in environmentally benign conditions and have played a pivotal role in various biosynthetic strategies in recent years. Serving as model systems, lysozymes have been shown to polymerize 2-ethynylpyridine (2-EP) via …

Determining The Structure And Activity Of A Pseudomonas Aeruginosa Protein, Ivyp2, Katherine Letsinger

Master of Science in Chemical Sciences Theses

Various biophysical methods were employed to structurally characterize and assess the activity of an important resistance factor (Ivyp2) from the multi-drug resistant Gram-negative bacterium Pseudomonas aeruginosa. This opportunistic pathogen accounts for approximately 10% of all hospital-acquired infections in the United States and contains a number of virulence factors that aid in its ability to infect and colonize immunocompromised individuals and those with cystic fibrosis. One of these factors – inhibitor of vertebrate lysozyme, or Ivy – neutralizes the lytic activity of lysozyme, an antimicrobial enzyme part of the innate immune system that hydrolyzes the linkages between bacterial cell wall subunits. …

Making The Old New: The Recontextualization And Traditionalization Of Tree Spirits In Video Games, Alexandria Ziegler

All Graduate Plan B and other Reports, Spring 1920 to Spring 2023

Folklorists study the active rituals between humans and deities, as well as the inactive participation between them in narrative. However, they do not study the active participation that comes in the form of video games between them, though with shifts in society, this new way of engaging through digital forms is widespread and accessible. In my research, I studied Russian and Japanese tree spirits in a variety of video games to understand this new form of engagement with ancient deities. These video games are Okami, The Legend of Zelda: Breath of the Wild, Black Book, and The Witcher 3: The …

As a mainstay of treatment for acute lymphoblastic leukemia (ALL), vincristine’s side effect profile is well known. Parallel administration of the antifungal fluconazole has been shown to interfere with the metabolism of vincristine, potentially resulting in increased side effects. We conducted a retrospective chart review to determine whether concomitant administration of vincristine and fluconazole during pediatric ALL therapy impacted side effects of vincristine, namely, hyponatremia and peripheral neuropathy. We also evaluated whether the incidence of opportunistic fungal infections was impacted by fluconazole prophylaxis. Medical charts of all pediatric ALL patients treated with induction chemotherapy at Children’s Hospital and Medical Center …

Oral Antibiotics And Early Discharge For Febrile Neutropenia In Pediatric Oncology, Alex Ciurej, Melissa Acquazzino May 2022

Oral Antibiotics And Early Discharge For Febrile Neutropenia In Pediatric Oncology, Alex Ciurej, Melissa Acquazzino

Child Health Research Institute Pediatric Research Forum

Background

Febrile neutropenia is defined as a temperature of ≥38.0°C with an absolute neutrophil count (ANC) <1500. This is a common reason for hospital admission in pediatric cancer patients given the myelosuppressive effects of many chemotherapeutic agents. In one study, nearly 20% of emergency department visits in pediatric cancer patients were due to fever, and periods of severe neutropenia (ANC < 500) present the highest risk of morbidity and mortality due to infection. For this reason, pediatric oncology patients with fever and severe neutropenia are admitted to the hospital for management. These hospital stays lead to risks of nosocomial infection, mounting healthcare costs, and lessened quality of life. We aim to observe the safety and impact on length of stay for early discharge (prior to count recovery) on oral antibiotics in the treatment of febrile neutropenia in pediatric oncology patients.

Methods

This retrospective case-control study compares 1) unplanned readmission rates within 96 hours and 2) length of stay in pediatric oncology patients with febrile neutropenia who were discharged home on oral levofloxacin before count recovery (cases) or stayed until evidence of count recovery (controls). Study patients had an oncologic diagnosis admitted with or developed febrile neutropenia at Children’s Hospital Omaha, without history of Trisomy 21, acute myeloid leukemia, bone marrow transplant, and without discharge home on IV antibiotics.

Results

Cases (n=32) were an average …

Two Patients With Klinefelter Syndrome Fortuitously Diagnosed By Non-Invasive Prenatal Test, Sarah Craven, Terence Zach May 2022

Two Patients With Klinefelter Syndrome Fortuitously Diagnosed By Non-Invasive Prenatal Test, Sarah Craven, Terence Zach

Child Health Research Institute Pediatric Research Forum

BACKGROUND

Non-invasive prenatal testing (NIPT) uses cell-free DNA (cfDNA) in maternal blood to screen for fetal aneuploidy. Maternal and fetal cfDNA are both present in maternal blood; however, fragmentation patterns are different. This allows the fetal fraction to be identified and screened for autosomal chromosome aneuploidy, specifically Trisomy 13, 18, and 21. Although not currently recommended for routine use, NIPT can also be used to screen for sex chromosome abnormalities. This practice is increasingly common, as allows parents to determine the baby’s sex earlier than ultrasound. Klinefelter syndrome (XXY karyotype) is one sex chromosome aneuploidy that NIPT may recognize. Currently, …
