Trophish: Building A Global Database Of Freshwater Trophic Interactions, Jacob M. Ridgway May 2022

Trophish: Building A Global Database Of Freshwater Trophic Interactions, Jacob M. Ridgway

Honors Thesis

Freshwater management and research frequently use the trophic data of freshwater fishes. Despite this fact, it is difficult to perform a simple search of dietary information for any one fish species. FishBase represents, to our knowledge, the largest compilation of freshwater dietary information to date. However, it excludes a large portion of the ecological literature due to its development taking place prior to the creation of most modern scientific search engines. Our project (TroPhish) is building upon FishBase by digitizing approximately 130 years of data from the fish predation literature. Data from the primary and grey (e.g. theses, dissertations, reports) …

Mechanical Characterization Of 3d Printed Hydrogel Lattices, Annabella Mascot May 2022

Mechanical Characterization Of 3d Printed Hydrogel Lattices, Annabella Mascot

Mechanical Engineering and Materials Science Independent Study

White matter brain tissue is largely inaccessible and is therefore difficult to mechanically characterize although this would be useful in understanding injuries and injury prevention. Thus, soft gels and 3D bioprinted materials allow for the estimation of the mechanical properties of brain tissue through non-invasive means. Through previous studies, it is determined that brain tissue is inherently anisotropic. To properly model it, the use of anisotropic cubic, diamond, and vintile type lattice 10 x 10 x 10 cm cube structures were used in compression testing to determine the elastic modulus of each lattice type in each of its orientations. Each …

Gut Commensals Modulate Siv/Shiv Pathogenesis And Therapeutics, Samuel Johnson May 2022

Gut Commensals Modulate Siv/Shiv Pathogenesis And Therapeutics, Samuel Johnson

Theses & Dissertations

Despite significant advancements in combination antiretroviral therapy (cART), ongoing inflammation in the brain and gut remain two of the most significant hurdles in the health of people living with human immunodeficiency virus (HIV). Additionally, a viral reservoir in each compartment inhibits cure efforts by allowing rapid viral rebound following cART interruption. Emerging understanding of the gut-brain axis (GBA) implicates each compartment in the modulation of the other in a complex bi-directional interaction mediated by vagus innervation, circulating lymphocytes, and microbiome composition and biproducts. Using multiple models of the simian (and simian-human) immunodeficiency virus (SIV/SHIV) and therapeutic intervention, I present how …

Pendulums Of Personhood? Exploring The Multitudes Of Immigrant Womanhood In Spanish-Maghrebi Literature, Kaitlyn C. Sisco

Honors Theses

Often considered articulations of in-between-ness and bearers of fraught selfhoods, the work of Spanish-Maghrebi authors has been widely debated in literary fields, with academics arguing that it constitutes a largely homogenous set of texts about the standard immigrant experience. However, by placing these texts in a single category, such arguments end up erasing the immensely varied identities expressed and represented by Spanish-Maghrebi authors. This thesis seeks to address this issue by paying particular attention to how Spanish-Maghrebi authors negotiate different types of immigrant subjectivities in their writing. Specifically, I analyze the works of three contemporary Spanish-Maghrebi writers, Najat El Hachmi, …

Visceral Adipose Tissue Remodeling In Pancreatic Ductal Adenocarcinoma Cachexia: The Role Of Activin A Signaling, Pauline Xu May 2022

Visceral Adipose Tissue Remodeling In Pancreatic Ductal Adenocarcinoma Cachexia: The Role Of Activin A Signaling, Pauline Xu

Theses & Dissertations

Pancreatic ductal adenocarcinoma (PDAC) is currently the third leading cause of cancer death in the United States and is projected to become the second leading cause by the year 2030. Prognosis for patients with metastatic disease remains dismal, with cachexia as a main contributor to the low survival rate. Emerging reports indicate that PDAC patients display distinct phenotypes of cachexia development, with either adipose tissue loss preceding skeletal muscle wasting or loss of only adipose tissue. While muscle wasting has been the most frequently studied mechanism in cachexia research, changes in adipose tissue are increasingly understood as important components of …

Mechanisms Of Sorting And Fission At The Endosomes, Kanika Dhawan May 2022

Mechanisms Of Sorting And Fission At The Endosomes, Kanika Dhawan

Theses & Dissertations

Endocytic trafficking is a fundamental cellular process that regulates the transport of lipids and proteins. Our lab focuses on the intracellular trafficking of receptors involved in cellular processes such as cell division, migration, and proliferation. Accordingly, the regulation of these trafficking pathways is tightly controlled, involving a complex series of events, of which a key step is the endosomal fission. Perturbations in the endosomal network can eventually lead to impaired receptor recycling to the plasma membrane (PM) and, therefore, have pathological consequences like Alzheimer’s disease and multiple cancers. Upon internalization, cargo-laden vesicles released from the PM fuse with the sorting …

Extracellular Mechanotransduction In Marfan Syndrome: An Equivalence Principle, Stephen Haller May 2022

Extracellular Mechanotransduction In Marfan Syndrome: An Equivalence Principle, Stephen Haller

Theses & Dissertations

Biological tissues continuously experience mechanical stress and have evolved sophisticated mechanisms to sense mechanical stimuli. While traditional viewpoints regard cells as the ultimate sensors and processors of mechanical information, compounding evidence demonstrates that extracellular matrix, the structural component of tissues, also exhibits evolved molecular responses to force. This led us to propose a new paradigm termed extracellular mechanotransduction, in which matrix orchestrates a complementary form of force integration distinct from traditional cellular and extracellular viewpoints. We thus propose that force-sensitive signaling mechanisms evolved within the extracellular space to help cells maintain mechanical homeostasis in tissues. In this dissertation, we apply …

Genome-Wide Distribution Of Acape1 And Its Implications In Dna Repair And Transcription, Mason Tarpley May 2022

Genome-Wide Distribution Of Acape1 And Its Implications In Dna Repair And Transcription, Mason Tarpley

Theses & Dissertations

Endogenous DNA damage can occur throughout the genome and is resolved through the Base Excision Repair (BER) pathway. The apurinic/apyrimidinic sites that result from endogenous DNA damage, called AP-sites, are created by DNA glycosylases removing oxidized or modified bases. These AP-sites are primarily repaired by AP endonuclease 1 (APE1) through the BER pathway. Modification of APE1 by acetylation of specific lysine residues in the proteins’ unstructured, N-terminal tail, have been shown to increase the residence time of APE1 (AcAPE1) at the AP-sites. It is unknown where AcAPE1 is binding to chromatin in the genome. Here, we are testing the hypothesis …

Phos-Tag-Based Screens Identify Novel Therapeutic Targets In Ovarian Cancer And Pancreatic Cancer, Renya Zeng May 2022

Phos-Tag-Based Screens Identify Novel Therapeutic Targets In Ovarian Cancer And Pancreatic Cancer, Renya Zeng

Theses & Dissertations

Multiple Phos-tag-based screens were conducted in anti-tubulin drug (paclitaxel and nocodazole)-treated cells to explore novel targets implicated in cell cycle regulation, resistance against paclitaxel, and yes-associated protein (YAP) nucleocytoplasmic transport. The Phos-tag-based screen of protein kinases identified that several proteins were degraded or phosphorylated in response to anti-tubulin drug treatment. A tyrosine kinase, fibroblast growth factor receptor 4 (FGFR4), was significantly degraded upon anti-tubulin drug treatment, suggesting its potential role in cell response to paclitaxel. Further functional investigations identified that FGFR4 mediated paclitaxel resistance in ovarian cancer, and specific inhibitors targeting FGFR4 could sensitize ovarian cancer cells to paclitaxel. Mechanistically, …
