Brugler, Angelo Fraga Bernardino, Thaís Lima De Brito, Zhenjian Lin, Francisca Andréa Da Silva Oliveira, Paulo Sumida, Craig R. Smith, Amaro Trindade-Silva, Kenneth M. Halanych, Daniel L. Distel

School of Biological and Marine Sciences

The bivalve families Teredinidae and Xylophagaidae include voracious consumers of wood in shallow-water and deep-water marine environments, respectively. The taxa are sister clades whose members consume wood as food with the aid of intracellular cellulolytic endosymbionts housed in their gills. This combination of adaptations is found in no other group of animals and was likely present in the common ancestor of both families. Despite these commonalities, the two families have followed dramatically different evolutionary paths with respect to anatomy, life history, and distribution. Here, we present 42 new mitochondrial genome sequences from Teredinidae and Xylophagaidae and show that distinct trajectories …

Inflammatory Breast Cancer: The Cytokinome Of Post-Mastectomy Wound Fluid Augments Proliferation, Invasion, And Stem Cell Markers, Alshaimaa Tarek, Shrouk Khalaf El-Sayed, Wendy A Woodward, Mohamed El-Shinawi, Jon Mark Hirshon, Mona Mostafa Mohamed

Faculty, Staff and Student Publications

Inflammatory breast cancer (IBC) is an aggressive phenotype with a high recurrence and low survival rate. Approximately 90% of local breast cancer recurrences occur adjacent to the same quadrant as the initial cancer, implying that tumor recurrence may be caused by residual cancer cells and/or quiescent cancer stem cells (CSCs) in the tumor. We hypothesized that wound fluid (WF) collected after modified radical mastectomy (MRM) may activate cancer cells and CSCs, promoting epithelial mesenchymal transition (EMT) and invasion. Therefore, we characterized the cytokinome of WF drained from post-MRM cavities of non-IBC and IBC patients. The WF of IBC patients showed …

Development And Evaluation Of A Remote Training Strategy For The Implementation Of Mental Health Evidence-Based Practices In Rural Schools: Pilot Study Protocol., Ricardo Eiraldi, Barry Mccurdy, Muniya S Khanna, Jessica Goldstein, Rachel Comly, Jennifer Francisco, Laura E Rutherford, Tara Wilson, Kathryn Henson, Thomas Farmer, Abbas F Jawad

PCOM Scholarly Works

BACKGROUND: An increasing number of schools in rural settings are implementing multi-tier positive behavioral interventions and supports (PBIS) to address school-climate problems. PBIS can be used to provide the framework for the implementation of evidence-based practices (EBPs) to address children's mental health concerns. Given the large service disparities for children in rural areas, offering EBPs through PBIS can improve access and lead to better long-term outcomes. A key challenge is that school personnel need technical assistance in order to implement EBPs with fidelity and clinical effectiveness. Providing ongoing on-site support is not feasible or sustainable in the majority of rural …

Chemistry Faculty Research & Creative Works

Enantiomers of a series of hydantoin derivatives were prepared from d- and l-amino acids with p-tolyl isocyanate and 3,5-bis(trifluoromethyl)phenyl isocyanate as guests for chiral recognition by 1H NMR spectroscopy. Meanwhile, several tetraaza macrocyclic compounds were synthesized as chiral solvating agents from d-phenylalanine and (1S,2S)-(+)-1,2-diaminocyclohexane. An uncommon enantiomeric discrimination has been successfully established for hydantoin derivatives, representatives of five-membered N,N-heterocycles, in the presence of tetraaza macrocyclic chiral solvating agents (TAMCSAs) 1a-1c by means of 1H NMR spectroscopy. Several unprecedented nonequivalent chemical shifts (up to 1.309 ppm) were observed in the split 1H NMR spectra. To evaluate practical applications …

Mixed Method Analysis Of Undergraduate Student Interpretation Of Different Phylogenetic Trees, Faith Frings

USF Tampa Graduate Theses and Dissertations

Phylogenetic trees are common tools used to visualize evolutionary concepts such as historical patterns of ancestry, divergence of species, and descent of species. However, students have misconceptions when reading these abstract diagrams. The purpose of this study was to compare student performance and evolutionary thinking when using two styles of phylogenetic trees: cladograms and phylograms. The study also assessed the validity of a hierarchal theoretical framework evaluating student phylogenetic tree interpretation. Introductory biology students from two research universities were assigned to two groups, one solely given assessments with phylograms, and one solely given assessments with cladograms. One-on-one student interviews were …

Effects Of Slurry Type On Drilled Shaft Strength, Cesar Quesada Garcia Jun 2022

Effects Of Slurry Type On Drilled Shaft Strength, Cesar Quesada Garcia

USF Tampa Graduate Theses and Dissertations

The main objective of this research is to identify how the strength of drilled shafts is affected by the use of various types of drill slurry. When concrete is placed in drilled shafts it flows out radially and a laitance interface forms creating a crease on the backside of the reinforcement cage due to the laitance trapped in concrete flowing around the rebar. This can cause concrete to not fully encapsulate the steel making the shafts prone to corrosion and the strength of the cover concrete is locally reduced. Over the past 9 years 58, 42-inch diameter shafts have been …

Isolation And Rna Sequencing Of Single Nuclei From Drosophila Tissues, Colleen N Mclaughlin, Yanyan Qi, Stephen R Quake, Liqun Luo, Hongjie Li

Center on Aging Staff Publications

Many insect cells are encapsulated within the exoskeleton and cannot be dissociated intact, making them inaccessible to single-cell transcriptomic profiling. We have used single-nucleus RNA sequencing to extract transcriptomic information from multiple Drosophila tissues. Here, we describe procedures for the (1) dissociation of single nuclei, (2) isolation of single nuclei using two popular cell sorters, and (3) preparation of libraries for Smart-seq2 and 10× Genomics. This protocol enables generation of high-quality transcriptomes from single nuclei and can be applied to other species.

Faculty, Staff and Students Publications

Metabolic switches play a critical role in the pathophysiology of cardiac diseases, including heart failure. Here, we describe an assay for long-chain fatty acid oxidation in neonatal mouse cardiomyocytes by using a SeaHorse Flux Analyzer (Agilent). This protocol is a simplified but robust adaptation of the standard protocol that enables metabolic measurements in cells isolated from transgenic mouse models, which can be timesaving and informative. Cell isolation and culture represent a critical point that may require bench optimization.

Protocol To Analyze And Quantify Protein-Methylated Rna Interactions In Mammalian Cells With A Combination Of Rna Immunoprecipitation And Nucleoside Mass Spectrometry, Ning Tsao, Jennifer M Soll, Nima Mosammaparast

2020-Current year OA Pubs

Cellular RNAs are modified by both physiological factors and exogenous agents, such as methyl methanesulfonate (MMS). However, techniques for analyzing how proteins may interact with these modified RNAs are limited. Here, we provide a protocol combining RNA immunoprecipitation (RIP) with mass spectrometry (MS) to analyze the methylation state of the RNAs bound by Flag-tagged proteins in mammalian cells. The approach is highly quantitative and can simultaneously detect several methylated nucleosides in a single experiment. For complete details on the use and execution of this protocol, please refer to Tsao et al. (2021).

Understanding peripheral nerve micro-anatomy can assist in the development of safe and effective neuromodulation devices. However, current approaches for imaging nerve morphology at the fiber level are either cumbersome, require substantial instrumentation, have a limited volume of view, or are limited in resolution/contrast. We present alternative methods based on MUSE (Microscopy with Ultraviolet Surface Excitation) imaging to investigate peripheral nerve morphology, both in 2D and 3D. For 2D imaging, fixed samples are imaged on a conventional MUSE system either label free (via auto-fluorescence) or after staining with fluorescent dyes. This method provides a simple and rapid technique to visualize myelinated …

Bloch Spectra For High Contrast Elastic Media, Jayasinghage Ruchira Nirmali Perera Jun 2022

Bloch Spectra For High Contrast Elastic Media, Jayasinghage Ruchira Nirmali Perera

LSU Doctoral Dissertations

The primary goal of this dissertation is to develop analytic representation formulas and power series to describe the band structure inside periodic elastic crystals made from high contrast inclusions. We use source free modes associated with structural spectra to represent the solution operator of the Lame' system inside phononic crystals. Then we obtain convergent power series for the Bloch wave spectrum using the representation formulas. An explicit bound on the convergence radius is given through the structural spectra of the inclusion array and the Dirichlet spectra of the inclusions. Sufficient conditions for the separation of spectral branches of the dispersion …
