Protein kinase-mediated phosphorylation plays a critical role in many biological processes. However, the identification of key regulatory kinases is still a great challenge. Here, we develop a trans-omics-based method, central kinase inference, to predict potentially key kinases by integrating quantitative transcriptomic and phosphoproteomic data. Using known kinases associated with anti-cancer drug resistance, the accuracy of our method denoted by the area under the curve is 5.2% to 29.5% higher than Kinase-Substrate Enrichment Analysis. We further use this method to analyze trans-omic data in hepatocyte maturation and hepatic reprogramming of human dermal fibroblasts, uncovering 5 kinases as regulators in the two …

Background: Near the beginning of the COVID-19 pandemic in the United States, medical students were pulled out of all in-person patient care activities. This resulted in massive disruption to the required clinical rotations (clerkships), necessitating creative curricular solutions to ensure continued education for medical students. Approach: In response to the lockout, our school adopted a “flipped” clinical rotations model that assigned students to remote learning activities prior to in-person patient care activities. This approach allowed students to continue their clinical education virtually with a focus on knowledge for practice while awaiting return to the shortened in-person portions of their rotation. …

We explored water use management and trends in irrigated agriculture in the U.S. West using operator-level USDA-NASS Farm and Ranch Irrigation Survey (FRIS)/Irrigation and Water Management Survey (IWMS) data aggregated for the first time to the county instead of state scale. Our hope is that this study will guide water managers in developing regional conservation programs, inform extension and research initiatives related to irrigation, promote grower-informed water conservation education, and provide researchers with key information regarding if, how, and why growers consider water conservation in irrigation decision-making.

Genetic and preclinical studies have implicated adenylyl cyclase 1 (AC1) as a potential target for the treatment of chronic inflammatory pain. AC1 activity is increased following inflammatory pain stimuli and AC1 knockout mice show a marked reduction in responses to inflammatory pain. Previous drug discovery efforts have centered around the inhibition of AC1 activity in cell-based assays. In the present study, we used an in vitro approach focused on inhibition of the protein-protein interaction (PPI) between Ca2+/calmodulin (CaM) and AC1, an interaction that is required for activation of AC1. A protein’s structure is determined by its amino acid sequence and post-translational modifications, and provides the basis for its physiological functions. Roughly a third of the proteome in mammalian species comprises proteins that contain highly unstructured or intrinsically disordered regions. Proteins with extensive unstructured regions are referred to as intrinsically disordered proteins (IDPs). Recent advances in experimental and computational analyses predicted multiple interacting partners for the disordered regions of proteins, implying critical roles in signal transduction and regulation of biological processes. In response to increasing threats from sea-level rise and storm surge, the City of Charleston, South Carolina, and the US Army Corps of Engineers (USACE) propose constructing a seawall around the Charleston peninsula. The proposed seawall will terminate close to lower wealth, predominantly minority communities. These communities are identified as environmental justice (EJ) communities due to their history of inequitable burdens of industrial and urban pollution and proximity to highways and US Environmental Protection Agency (EPA) designated Superfund sites. Ferroelectric domain walls provide a fertile environment for novel materials physics. If a polarization discontinuity arises, it can drive a redistribution of electronic carriers and changes in band structure, which often result in emergent 2D conductivity. If such a discontinuity is not tolerated, then its amelioration usually involves the formation of complex topological patterns, such as flux-closure domains, dipolar vortices, skyrmions, merons, or Hopfions. The degrees of freedom required for the development of such patterns, in which dipolar rotation is a hallmark, are readily found in multiaxial ferroelectrics. BACKGROUND: Every year, an estimated one million children and young adolescents become ill with tuberculosis, and around 226,000 of those children die. Xpert MTB/RIF Ultra (Xpert Ultra) is a molecular World Health Organization (WHO)-recommended rapid diagnostic test that simultaneously detects Mycobacterium tuberculosis complex and rifampicin resistance. We previously published a Cochrane Review 'Xpert MTB/RIF and Xpert MTB/RIF Ultra assays for tuberculosis disease and rifampicin resistance in children'. The current review updates evidence on the diagnostic accuracy of Xpert Ultra in children presumed to have tuberculosis disease. The social consciousness of residents has been changing since China's reform and opening up. The grassroots governance of the society has also become more and more difficult. In the last decade, the Chinese government has been improving the level of social community governance, and since COVID-19 pandemic, the government has been strengthening democratic social governance. It has become very possible to cultivate social consciousness and democratic consciousness among the residents in the community. I conducted two rounds of questionnaires and three rounds of interviews in several communities in Shandong province.

Ceramic technology was adopted by hunter-gatherers of the Paleo-Inuit Norton tradition in the Western Arctic between 2800 and 2500 years B.P., corresponding with an increase in the use of aquatic resources. Pottery production and use continued until approximately 1,500 BP, and resumed during the Neo-Inuit Birnirk and Thule periods, approximately 1,350 years BP. The technical characteristics of Norton and Thule ceramics suggest they performed differently when used for cooking, with Norton ceramics best suited for cooking using direct or suspended heat, and Thule ceramics best suited for indirect heat. Accuracy in photovoltaic (PV) module temperature modeling is crucial to achieving precision in energy performance yield calculations and subsequent economic evaluations of PV projects. While there have been numerous approaches to PV temperature modeling based on both the steady-state and transient thermal assumptions, there have been few attempts to account for changing convective cooling on PV module surfaces resulting from changes in the PV system layout. Spaceflight poses risks to the central nervous system (CNS), and understanding neurological responses is important for future missions. We report CNS changes in Drosophila aboard the International Space Station in response to spaceflight microgravity (SFμg) and artificially simulated Earth gravity (SF1g) via inflight centrifugation as a countermeasure. While inflight behavioral analyses of SFμg exhibit increased activity, postflight analysis displays significant climbing defects, highlighting the sensitivity of behavior to altered gravity. Multi-omics analysis shows alterations in metabolic, oxidative stress and synaptic transmission pathways in both SFμg and SF1g; however, neurological changes immediately postflight, including neuronal loss, glial cell count alterations, oxidative …

Nurses working in emergency departments (EDs) of trauma centers are integral members of the trauma team during the initial resuscitation of severely injured patients. Decisions regarding these resuscitations are made in high stress environments and are inevitably rife with ethical issues due to the high frequency and severity of injury and its exorbitant costs to society. These decisions are made with little background knowledge about the patients during a complex process of rapid assessment of physiological status overlaid by ethical principles, societal norms and expectations, and legal mandates.
