Irrespective of the reason for hypoperfusion, hypocoagulable and/or hyperfibrinolytic hemostatic aberrancies afflict up to one-quarter of critically ill patients in shock. Intensivists and traumatologists have embraced the concept of SHock-INduced Endotheliopathy (SHINE) as a foundational derangement in progressive shock wherein sympatho-adrenal activation may cause systemic endothelial injury. The pro-thrombotic endothelium lends to micro-thrombosis, enacting a cycle of worsening perfusion and increasing catecholamines, endothelial injury, de-endothelialization, and multiple organ failure. The hypocoagulable/hyperfibrinolytic hemostatic phenotype is thought to be driven by endothelial release of anti-thrombogenic mediators to the bloodstream and perivascular sympathetic nerve release of tissue plasminogen activator directly into the microvasculature. …

Transcription factors (TFs) are transported from the cytoplasm to the nucleus and disappear from the nucleus after they regulate gene expression. Here, we discover an unconventional nuclear export of the TF, orthodenticle homeobox 2 (OTX2), in nuclear budding vesicles, which transport OTX2 to the lysosome. We further find that torsin1a (Tor1a) is responsible for scission of the inner nuclear vesicle, which captures OTX2 using the LINC complex. Consistent with this, in cells expressing an ATPase-inactive Tor1aΔE mutant and the LINC (linker of nucleoskeleton and cytoskeleton) breaker KASH2, OTX2 accumulated and formed aggregates in the nucleus. Consequently, in the mice expressing …
Uncovering Novel Regulators Of Memory Using C Elegans Genetic And Genomic Analysis
Uncovering Novel Regulators Of Memory Using C Elegans Genetic And Genomic Analysis
Faculty, Staff and Students Publications
How organisms learn and encode memory is an outstanding question in neuroscience research. Specifically, how memories are acquired and consolidated at the level of molecular and gene pathways remains unclear. In addition, memory is disrupted in a wide variety of neurological disorders; therefore, discovering molecular regulators of memory may reveal therapeutic targets for these disorders. C. elegans are an excellent model to uncover molecular and genetic regulators of memory. Indeed, the nematode's invariant neuronal lineage, fully mapped genome, and conserved associative behaviors have allowed the development of a breadth of genetic and genomic tools to examine learning and memory. In …

Impact Of Penicillin Allergy Labels On Children Treated For Outpatient Respiratory Infections, Debra L Palazzi, Jeffrey S Gerber
Faculty, Staff and Students Publications
BACKGROUND: Penicillin allergy is the most common antibiotic allergy, yet most children labeled as allergic tolerate penicillin. The impact of inaccurate penicillin allergy labels (PALs) on pediatric outpatients is unknown. The objective of this study was to compare outcomes between children with and without a PAL after treatment for outpatient respiratory tract infections (RTI).
METHODS: A retrospective, longitudinal birth cohort study was performed in children who received care in 90 pediatric primary care practices in Philadelphia and Houston metropolitan areas. Prescribing and clinical outcomes of children with a PAL at the time of an RTI were compared to non-allergic children, …

BACKGROUND: The absence of consensus for outcomes in pediatric antibiotic trials is a major barrier to research harmonization and clinical translation. We sought to develop expert consensus on study outcomes for clinical trials of children with mild community-acquired pneumonia (CAP).
METHODS: Applying the Delphi method, a multispecialty expert panel ranked the importance of various components of clinical response and treatment failure outcomes in children with mild CAP for use in research. During Round 1, panelists suggested additional outcomes in open-ended responses that were added to subsequent rounds of consensus building. For Rounds 2 and 3, panelists were provided their own …

The asymmetric increase in average nighttime temperatures relative to increase in average daytime temperatures due to climate change is decreasing grain yield and quality in rice. Therefore, a better genome-level understanding of the impact of higher night temperature stress on the weight of individual grains is essential for future development of more resilient rice. We investigated the utility of metabolites obtained from grains to classify high night temperature (HNT) conditions of genotypes, and metabolites and single-nucleotide polymorphisms (SNPs) to predict grain length, width, and perimeter phenotypes using a rice diversity panel. We found that the metabolic profiles of rice genotypes …

Detection of Volatile Organic Compounds (VOCs) is One of the Most Challenging Tasks in Modelling Breath Analyzers Because of their Low Concentrations (Parts-Per-Billion (Ppb) to Parts-Per-Million (Ppm)) in Breath and the High Humidity Levels in Exhaled Breaths. the Refractive Index is One of the Crucial Optical Properties of Metal-Organic Frameworks (MOFs), Which is Changeable Via the Variation of Gas Species and Concentrations that Can Be Utilized as Gas Detectors. Herein, for the First Time, We Used Lorentz–Lorentz, Maxwell–Ga, and Bruggeman Effective Medium Approximation (EMA) Equations to Compute the Percentage Change in the Index of Refraction (∆n%) of ZIF-7, ZIF-8, ZIF-90, …

Molecular Research In Pancreatic Cancer: Small Molecule Inhibitors, Their Mechanistic Pathways And Beyond, Shaila A. Shetu, Nneoma James, Gildardo Rivera-Sanchez, Debasish Bandyopadhyay
School of Integrative Biological & Chemical Sciences (Formerly Dept. of Chemistry)
Pancreatic enzymes assist metabolic digestion, and hormones like insulin and glucagon play a critical role in maintaining our blood sugar levels. A malignant pancreas is incapable of doing its regular functions, which results in a health catastrophe. To date, there is no effective biomarker to detect early-stage pancreatic cancer, which makes pancreatic cancer the cancer with the highest mortality rate of all cancer types. Primarily, mutations of the KRAS, CDKN2A, TP53, and SMAD4 genes are responsible for pancreatic cancer, of which mutations of the KRAS gene are present in more than 80% of pancreatic cancer cases. Accordingly, there is a …

Among college students, evidence of relatively lower levels of concern of COVID-19 infection, rapid rates of transmission, and lagging vaccination rates substantiates a need to combat college student COVID-19 vaccine hesitancy. This two-phase study comprised formative research, development, and pilot testing of a campaign to combat COVID-19 vaccine hesitancy among college students. In phase one, focus group discussions with college students (N = 48) uncovered their specific attitudes, subjective norms, and perceived behavioral control toward the COVID-19 vaccine. Thematic analysis revealed concerns about vaccine safety, efficacy, cost, and politicization, as well as perceived barriers to accessing information and the …

On Refinements To Qmfd Based Chirp Parameter Estimation, Balu Santhanam, Thalanayar Santhanam
Electrical & Computer Engineering Technical Reports
Commuting matrix methods furnish a full basis of orthog- onal eigenvectors for the discrete Fourier transform or its centered version needed for computing the discrete fractional Fourier transform and multicomponent chirp signal analysis. However, these approaches suffer from ill-conditioning issues at higher matrix sizes, and require a computationally expensive eigenvalue decomposition.
In this paper, ill-conditioning issues associated with the QMFD approach developed previously by the authors are addressed via diagonal modification. Further symmetries of the eigenvectors are used to reduce the size of the underlying eigenvalue problem. These modifications are then incorporated into the real-arithmetic implementation of the QMFD approach …

Home – No Home reflects through expanded photographic artworks and elucidate – through heuristic introspection, auto-ethnography, and critical text analysis – notions of expatriation and social integration, such as home, (Blunt & Dowling 2006; Blunt et al., 2007) home displacement and attachment, nostalgia (Vidler, 1992; Koerner, 2017) and homesickness (Fink et al., 2007). Attached to these concepts are deeper questions rooted in societal discourses about today’s global trends and discourses of expatriation, (Fechter, 2007; Leonard 2010; Walsh, 2010, 2012, 2014; Mathur, 2011; and Kunz, 2016) and the politics of border assertion and control. Is the ability to live …

The northeast of Madagascar is as diverse as it is threatened. The area bordering the Analanjirofo and SAVA regions contains six protected areas and at least 22 lemur species. Many applied research and conservation programs have been established in the region with the aim of ensuring both wildlife and people thrive in the long term. While most of the remaining humid evergreen forest of northeast Madagascar is formally protected, the local human population depends heavily on the land, and unsustainable natural resource use threatens this biodiversity hotspot. Drawing from our collective experiences managing conservation activities and research programs in northeast …

Split And Join: An Efficient Approach For Simulating Stapled Intestinal Anastomosis In Virtual Reality, Di Qi, Suvranu De
Engineering Faculty Articles and Research
Colorectal cancer is a life-threatening disease. It is the second leading cause of cancer-related deaths in the United States. Stapled anastomosis is a rapid treatment for colorectal cancer and other intestinal diseases and has become an integral part of routine surgical practice. However, to the best of our knowledge, there is no existing work simulating intestinal anastomosis that often involves sophisticated soft tissue manipulations such as cutting and stitching. In this paper, for the first time, we propose a novel split and join approach to simulate a side-to-side stapled intestinal anastomosis in virtual reality. We mimic the intestine model using …

This paper unpacks the assumptions underpinning England’s new Core Content Framework (CCF) in respect of the educational research required for teacher expertise, with particular attention to the sources referenced in the final part of the CCF and claims that these constitute the ‘best available educational research’. Drawing on sociological studies of educational knowledge, and assessments of the quality of educational research in England, in addition to the philosophy of expertise as related to teaching, it is argued that the CCF is currently orientated towards a scientism that (i) marginalises longstanding traditions of educational thought, and (ii) technicises and instrumentalises teaching …
