The last 10 years of scientific research analyzing asylum-seekers’ mental health has established high rates of trauma exposure throughout the migratory trajectory. However, limited studies have identified gender-based violence among Central American asylum-seeking women. The purpose of this study was to identify the frequencies of gendered-base violence among asylum-seeking women from Central and South America at a humanitarian respite center (USA) and a tent encampment (Mexico) on both sides of the United States–Mexico Border using data from three independent studies in 2016, 2019, and 2023, respectively. Video games are a popular form of interactive media and, since their inception, have become one of the largest forms of media consumed. As such, they have evolved greatly from their humble beginnings into much more complex experiences, and adjusting the difficulty level to suit the needs of the player has become common practice. While there are simple ways to do so, the best games often feature a dynamic difficulty-scaling system that adapts to the player. Especially during and following the ‘lockdown era’ of the COVID-19 pandemic, college writing instructors have sought ways to mitigate the harms of neoliberal and capitalist practices in the academy and writing classroom. Incorporating elements of Slow Pedagogy, Disability Studies, Anti-Capitalist Composition, and Participatory Action Research methodology, this dissertation argues for practicing a Pedagogy of Slow Resistance, centering co-creation and relationship-building, to resist neoliberal and capitalist values, and promote student belonging and well-being. This portfolio consists of the artifacts of the creative, artistic and research work realized during my doctoral studies. In its core, all recitals had Brazilian music, especially that of H. Villa-Lobos, as a unifying element, although personal taste and reasoning were also a driving force behind choosing repertoire and building a theme for each recital. This research addresses inefficiencies in Manufacturing Execution System (MES)-driven workflows, where excessive user interactions and fragmented access to work instructions introduce transactional and cognitive waste. To address these limitations, an integrated architecture was developed using Ignition as a SCADA-based operator interface connected to the MES through API-based communication. The system replaces direct MES interaction with a unified interface that embeds work instructions and automates transaction execution within the production workflow. The approach was implemented and evaluated through an industrial case study on a 15 station electronics manufacturing line. Results show a 51% reduction in total cycle time, decreasing from 2,996 …

Radiotherapy can lead to late-onset toxicity, to varying extents between individuals due to differences in radiosensitivity. Predicting which patients are most at risk is key to augmenting the therapeutic window. However, the underlying biological mechanisms remain poorly understood, and current experimental methods often lack clinical applicability. This study employs Raman spectroscopy to analyse biochemical profiles in peripheral lymphocytes and plasma, aiming to monitor radiotherapeutic response and predict intrinsic radiosensitivity in high-risk localised prostate cancer patients treated with stereotactic radiotherapy. Agricultural productivity in East Africa is increasingly threatened by climate-sensitive insect pests. This study investigates the occurrence of key vegetable pests: Brevicoryne brassicae, Plutella xylostella, Maruca vitrata, and Liriomyza mik. A comparative species distribution modelling evaluated pest risk projections under climate change. Four variable selection strategies were tested: case 1 (correlation and variance inflation factor [VIF] filtering across all predictors), case 2 (correlation and VIF filtering of bioclimatic variables merged with other predictors), case 3 (filtering based on environmental values at species occurrence point to approximate realized niches), and case 4 (dimensionality reduction using principal component analysis). Alveolar capillary dysplasia with misalignment of pulmonary veins (ACDMPV) is a rare lethal lung developmental disorder caused by haploinsufficiency of FOXF1. While larger-sized coding and noncoding copy-number variant (CNV) deletions involving the FOXF1 locus are detected in approximately half of histopathologically-diagnosed ACDMPV patients, small CNVs remain diagnostically challenging. Here, we revisited an unsolved case of familial ACDMPV with two affected siblings. Whole genome sequencing (WGS) of 30-year-old archival lung autopsy tissue analyzed using AI powered platform revealed a 151 bp CNV deletion involving a highly GC-rich portion of exon 1 of FOXF1 that was not detected using Sanger sequencing and …

BACKGROUND: Black women diagnosed with ductal carcinoma in situ (DCIS) experience disproportionately higher risk of second breast cancer events (SBEs), including both ipsilateral and contralateral recurrences, yet remain underrepresented in molecular biomarker studies compared to non-Hispanic White women.
METHODS: We performed RNA sequencing on 200 archival DCIS samples from the Resource of Archival Breast Tissue cohort, including 33% self-reported Black women. We correlated transcriptomic and clinical data to identify predictors of SBEs. Apathy, or loss of motivation, is a prominent syndrome accompanying both Alzheimer's and Parkinson's diseases, in addition to other disorders. One approach to understanding motivational loss is to examine the processes underlying goal-directed behaviour. Weighing up rewarding outcomes against the effort costs required to obtain them (effort-based decision-making) is a core computation when deciding to act for outcomes. Although a growing body of evidence points to disruption of this computation in people with apathy, which underlying decision parameters drive this disruption, their neural associations and whether these would generalize or differ across brain disorders have not been examined. Despite effective lipid-lowering therapies, atherosclerosis continues to be a leading cause of death, with considerable residual cardiovascular risk. Atherosclerotic lesions develop preferentially at arterial regions exposed to disturbed flow (d-flow), which induces genomic stress, endothelial injury, and barrier dysfunction. Hemodynamic forces are known to reprogram endothelial metabolism, but the role of de novo purine synthesis (DNPS), which supplies nucleotides for genome maintenance and whose terminal steps are catalyzed by the bifunctional enzyme ATIC, remains undefined in atherosclerosis. Background: Soluble recombinant expression of Aedes aegypti mosquito midgut proteases in Escherichia coli prove to be difficult. These enzymes depend on disulfide bond formation for structural stability. Initial attempts in BL21(DE3) were unsuccessful due to a reducing cytoplasm. The use of T7 SHuffle cells (with a more oxidizing cytoplasm) led to soluble expression. However, other factors had to be altered (use of richer media and lower (< 25 °C) growth temperature). Not all mosquito proteases were equally soluble. Therefore, given the importance of IPTG in initiating transcription and translation, we set out to determine if low IPTG concentrations (< 0.1 mM) at 10 °C would increase soluble production of midgut proteases. Additionally, we investigated the effect of the small molecule osmolyte betaine on the soluble expression of midgut proteases. Results: For this study, the focus was on Aedes aegypti Late Trypsin (AaLT), Early Trypsin (AaET), Serine Protease I (AaSPI), Serine protease V (AaSPV), and Juvenile Hormone Associated 15 (JHA15). The colder bacterial growth, along with low IPTG, slows the rate of transcription/translation of T7 RNA polymerase. Lower expression of T7 RNA polymerase, along with slower transcription activity at 10 °C, prevents rapid simultaneous translation of midgut proteases thereby promoting soluble expression. In addition, we found that different growth periods also varied among the proteases. Soluble expression for AaLT and AaET was maximal at 52 h post-induction, 72 h for JHA15, and 168 h for AaSPI and AaSPV. Surprisingly, for AaET, temperature was the only important factor. The addition of betaine to the growths had a more pronounced effect at higher (> 0.05 mM) IPTG. Conclusions: Low IPTG at 10 °C slows the rate of transcription/translation of recombinantly expressed mosquito proteases in bacteria. Contamination of aquatic environments with per- and polyfluoroalkyl substances (PFAS) may disrupt wildlife reproduction by altering hormonal-behavioural pathways. In birds, nest attendance is a key incubation behaviour, mediating embryonic development and hatchability. Yet, the potential associations between PFAS and nest attendance remains underinvestigated. In 2022 and 2023, we quantified plasma PFAS concentrations in breeding females from two Finnish waterfowl species: common goldeneyes (Bucephala clangula), an income breeding cavity nester, and common eiders (Somateria mollissima), a predominantly capital breeding ground nester. This study aims to analyze the structure, content, and integration of a play-based learning implementation guide as a pedagogical instrument to support reflective learning in early childhood education at TK Alam Jingga. A descriptive qualitative approach with document analysis was employed. The primary data source was the Play-Based Learning Implementation Guide, which integrates operational guidelines, technical procedures, instructional planning tools, and a monitoring and evaluation system. Data were collected through intensive and iterative document review and analyzed using qualitative content analysis. Mixed neuroendocrine-non-neuroendocrine neoplasms (MiNENs) of the rectum are rare tumors that typically demonstrate aggressive behavior, largely driven by high-grade neuroendocrine carcinoma components. Evidence guiding management is limited, particularly for early-stage lesions and for patients unable to tolerate standard oncologic surgery. We report the case of an elderly man with extensive comorbidities who was found to have a 1.3 cm rectal MiNEN following transanal endoscopic microsurgery (TEMS) performed for rectal bleeding. Histopathology revealed a moderately differentiated adenocarcinoma component and a well-differentiated grade 1 neuroendocrine tumor (NET) component, with submucosal invasion greater than 2 mm, negative margins, low tumor budding, small-vessel lymphovascular …

The research examines plant resource management and food processing practices of the Manteño occupation of the Río Blanco, Manabí, Ecuador, through the analysis of starch grain residues recovered from archaeological artifacts. Twenty-six samples from four archeological sites (N4C4-044, N4C4-064, N4C4-068, N4C4-090) associated with domestic artifacts such as fragments of comales, vessels, and metates were analyzed, from which a total of 546 starch grains were identified, corresponding to several plant taxa including maize (Zea mays), beans (Phaseolus spp.), sweet potato (Ipomoea batatas), manioc (Manihot esculenta), chili pepper (Capsicum spp.), and squash (Cucurbita …

Pavlovian biases reflect how evolutionarily hard-wired tendencies—automatic approach toward reward cues and withdrawal from threat cues—can interfere with flexible, goal-directed action. Such biases arise through three mechanisms: (a) anticipated rewards energize action while anticipated punishments suppress it (response bias), (b) agents learn differently from actions than from inactions (learning bias), and (c) reward/punishment cues themselves drive repetitive behavior, independent of outcomes (perseveration bias). The neural origin of these biases is unclear. Past evidence suggests dorsal anterior cingulate cortex (dACC) and anterior insula (aIns) as part of a “reset network” that rapidly responds to salient information and might contribute to these …

Mitochondrial electron transport flavoprotein (ETF) insufficiency causes metabolic diseases known as a multiple acyl-CoA dehydrogenase deficiency (MADD). In contrast to muscle, ETFDH is a non-essential gene in acute lymphoblastic leukemia NALM6 cells, and its expression is reduced across human cancers. In various human cancer cell lines and mouse models, ETF insufficiency caused by decreased ETFDH expression limits flexibility of OXPHOS fuel utilisation but paradoxically increases bioenergetics and accelerates neoplastic growth via activation of the mTORC1/BCL-6/4E-BP1 axis. Collectively, these findings reveal that while ETF insufficiency is rare and has detrimental effects in non-malignant tissues, it is common in neoplasia, where ETFDH …

Spatial transcriptomics (ST) unlocks potential for studying gene functions in processes that depend on orchestration of transcription across space. However, analysis tools for ST remain aimed at data exploration, with few resources for hypothesis testing. What's missing is a way to test whether a factor of interest affects functionally relevant parameters of a gene's spatial distribution. We present a tool to fill this gap, which we call a warped sigmoidal Poisson-process mixed-effects (WSP, pronounced "wisp") model. WSP models are the first ST tool allowing researchers to test critical questions without bespoke preprocessing pipelines for identifying key spatial parameters. By aligning …

Elucidating the genetic contributions to Parkinson's disease aetiology across diverse ancestries is a critical priority for the development of targeted therapies in a global context. We conducted the largest sequencing characterization of potentially disease-causing, protein-altering and splicing mutations in 710 cases and 11 827 controls from genetically predicted African or African admixed ancestries. We explored copy number variants (CNVs) and runs of homozygosity in prioritized early onset and familial cases. Our study identified rare GBA1 coding variants to be the most frequent mutations among patients with Parkinson's disease, with a frequency of 4% in our case cohort. Of the 18 …

This paper explores the transformative potential of contemplative environmental education in addressing the growing disconnection between humans and the natural world, particularly within K-12 education systems shaped by anthropocentric and capitalist paradigms. Modern schooling often frames nature as an object of study, rather than a sacred, living presence, contributing to ecological alienation amid intensifying environmental crises. We propose a radical shift toward contemplative, holistic education that draws on spiritual, Indigenous, and ecological knowledge systems. Grounded in Daoist philosophy, Islamic teachings on stewardship, Hindu ethics, and Indigenous worldviews, we reimagine education as a relational and sacred engagement with the Earth. We …
