Supporting An Infrastructure For Experienced Nurses: A Fellowship Foundation, Melissa Oberle

Nursing

A Nursing Professional Development (NPD) Specialist identified 90% of first-year turnover occurred with experienced nurses in the acute care setting, which accrues a cost of $60,000 per nurse. Experienced nurses departed within the first year of hire due to the work environment and personal reasons. This high-risk population had minimal targeted training, specifically nurses with greater than six months of experience. A survey of newly hired experienced nurses revealed interest in a program to support their transition. An experienced nurse fellowship program was created to promote a healthy work environment, professional growth, and retention by allowing organizational assimilation. The program …

Targeted Deletion Of Nr2f2 And Vcam1 In Theca Cells Impacts Ovarian Follicular Development: Insights Into Polycystic Ovary Syndrome?†, Nicholes R Candelaria, Joanne S Richards

Faculty, Staff and Students Publications

Defining features of polycystic ovary syndrome (PCOS) include elevated expression of steroidogenic genes, theca cell androgen biosynthesis, and peripheral levels of androgens. In previous studies, we identified vascular cell adhesion molecule 1 (VCAM1) as a selective androgen target gene in specific NR2F2/SF1 (+/+) theca cells. By deleting NR2F2 and VCAM1 selectively in CYP17A1 theca cells in mice, we documented that NR2F2 and VCAM1 impact distinct and sometimes opposing theca cell functions that alter ovarian follicular development in vivo: including major changes in ovarian morphology, steroidogenesis, gene expression profiles, immunolocalization images (NR5A1, CYP11A1, NOTCH1, CYP17A1, INSL3, VCAM1, NR2F2) as well as …

Patterns, Drivers, And Implications Of Postfire Delayed Tree Mortality In Temperate Conifer Forests Of The Western United States, Sebastian Busby, Cody Evers, Andrés Holz

Geography Faculty Publications and Presentations

Conifer forest resilience may be threatened by increasing wildfire activity and compound disturbances in western North America. Fire refugia enhance forest resilience, yet may decline over time due to delayed mortality—a process that remains poorly understood at landscape and regional scales. To address this uncertainty, we used high-resolution satellite imagery (5-m pixel) to map and quantify delayed mortality of conifer tree cover between 1 and 5 years postfire, across 30 large wildfires that burned within three montane ecoregions in the western United States. We used statistical models to explore the influence of burn severity, topography, soils, and climate moisture deficit …

Steroid receptor coactivator 3 (SRC-3) is a critical mediator of many intracellular signaling pathways that are crucial for cancer proliferation and metastasis. In this study, we performed structure-activity relationship (SAR) exploration and drug-like optimization of the hit compound SI-2, guided by in vitro/in vivo metabolism studies and cytotoxicity assays. Our efforts led to the discovery of two lead compounds, SI-10 and SI-12. Both compounds exhibit potent cytotoxicity against a panel of human cancer cell lines and demonstrate acceptable pharmacokinetic properties. A biotinylated estrogen response element (ERE) pull-down assay demonstrated that SI-12 could disrupt the recruitment of SRC-3 and p300 in …

Latinx Science Students’ Sense Of Belonging In Rural Wisconsin, Patrick Robinson, Cynthia Baeza, Luis Gonzalez-Quizhpe, Abigail Robinson, Diego Román, Katherine Ward

Journal of Extension

This study explores the sense of belonging of rural middle and high school Latinx science students in Wisconsin. Between 2000 and 2015, the Latinx population was the fastest growing ethnic group in Wisconsin and increased by 95%. Yet, little is known about the experiences of these students in rural schools or their engagement in science learning. Our research contributes to addressing this knowledge gap and provides insights into Latinx student experiences relative to the science classroom and community. Our findings indicate that a primary determinant of Latinx science students’ positive sense of belonging is a positive relationship with their teachers.

An Instrument To Evaluate 4-H Cloverbud Stem Programming, Alisha M. Hardman, Donna J. Peterson, Mariah S. Morgan, H. Elizabeth Solace

Journal of Extension

Evaluation data is needed to demonstrate the impact of 4-H science, technology, engineering, and mathematics (STEM) programming on children and youth. However, collecting evaluation data from cloverbuds (ages 5-7) is particularly challenging given their developmental age. We adapted an observational Cloverbud evaluation tool to measure basic life skills, essential elements of positive youth development experiences, and, unique to this tool, STEM specific experiences. We developed the STEM items using educational science standards allowing Extension to document STEM in addition to youth development outcomes. Doing so, may increase the adoption of 4-H STEM programming by teachers in the school setting.

Modulating brain oscillations has strong therapeutic potential. Interventions that both non-invasively modulate deep brain structures and are practical for chronic daily home use are desirable for a variety of therapeutic applications. Repetitive audio-visual stimulation, or sensory flicker, is an accessible approach that modulates hippocampus in mice, but its effects in humans are poorly defined. We therefore quantified the neurophysiological effects of flicker with high spatiotemporal resolution in patients with focal epilepsy who underwent intracranial seizure monitoring. In this interventional trial (NCT04188834) with a cross-over design, subjects underwent different frequencies of flicker stimulation in the same recording session with the effect …

Two decades into its tenure as a field, dissemination and implementation (D&I) scientists have begun a process of self-reflection, illuminating a missed opportunity to bridge the gap between research and practice-one of the field's foundational objectives. In this paper, we, the authors, assert the research-to-practice gap has persisted, in part due to an inadequate characterization of roles, functions, and processes within D&I. We aim to address this issue, and the rising tension between D&I researchers and practitioners, by proposing a community-centered path forward that is grounded in equity. We identify key players within the field and characterize their unique roles using …

2020-Current year OA Pubs

Hydroxyproline-rich glycoproteins (HRGPs) are a ubiquitous class of protein in the extracellular matrices and cell walls of plants and algae, yet little is known of their native structures or interactions. Here, we used electron cryomicroscopy (cryo-EM) to determine the structure of the hydroxyproline-rich mastigoneme, an extracellular filament isolated from the cilia of the alga Chlamydomonas reinhardtii. The structure demonstrates that mastigonemes are formed from two HRGPs (a filament of MST1 wrapped around a single copy of MST3) that both have hyperglycosylated poly(hydroxyproline) helices. Within the helices, O-linked glycosylation of the hydroxyproline residues and O-galactosylation of interspersed serine residues create a …

2020-Current year OA Pubs

Nucleoli are multicomponent condensates defined by coexisting sub-phases. We identified distinct intrinsically disordered regions (IDRs), including acidic (D/E) tracts and K-blocks interspersed by E-rich regions, as defining features of nucleolar proteins. We show that the localization preferences of nucleolar proteins are determined by their IDRs and the types of RNA or DNA binding domains they encompass. In vitro reconstitutions and studies in cells showed how condensation, which combines binding and complex coacervation of nucleolar components, contributes to nucleolar organization. D/E tracts of nucleolar proteins contribute to lowering the pH of co-condensates formed with nucleolar RNAs in vitro. In cells, this …

2020-Current year OA Pubs

Chemotherapy is a widely used treatment for a variety of solid and hematological malignancies. Despite its success in improving the survival rate of cancer patients, chemotherapy causes significant toxicity to multiple organs, including the skeleton, but the underlying mechanisms have yet to be elucidated. Using tumor-free mouse models, which are commonly used to assess direct off-target effects of anti-neoplastic therapies, we found that doxorubicin caused massive bone loss in wild-type mice, a phenotype associated with increased number of osteoclasts, leukopenia, elevated serum levels of danger-associated molecular patterns (DAMPs; e.g. cell-free DNA and ATP) and cytokines (e.g. IL-1β and IL-18). Accordingly, …
