Research outputs 2022 to 2026
Background: Given the high level of product complexity and limited regulatory guidance, designing and implementing appropriate potency assays is often the most challenging part of establishing a quality control testing matrix for a cell-based medicinal product. Among the most elusive tasks are the selection of suitable read-out parameters, the development of assay designs that most closely model the pathophysiological conditions, and the validation of the methods. Here we describe these challenges and how they were addressed in developing an assay that measures the anti-inflammatory potency of mesenchymal stromal cells (MSCs) in an M1 macrophage-dominated inflammatory environment. Methods: An in vitro …

The Role Of Pennation Angle And Architectural Gearing To Rate Of Force Development In Dynamic And Isometric Muscle Contractions, Bas Van Hooren, Per Aagaard, Andrea Monte, Anthony J. Blazevich
Research outputs 2022 to 2026
Background: Associations between muscle architecture and rate of force development (RFD) have been largely studied during fixed-end (isometric) contractions. Fixed-end contractions may, however, limit muscle shape changes and thus alter the relationship between muscle architecture an RFD. Aim: We compared the correlation between muscle architecture and architectural gearing and knee extensor RFD when assessed during dynamic versus fixed-end contractions. Methods: Twenty-two recreationally active male runners performed dynamic knee extensions at constant acceleration (2000°s−2) and isometric contractions at a fixed knee joint angle (fixed-end contractions). Torque, RFD, vastus lateralis muscle thickness, and fascicle dynamics were compared during 0–75 and 75–150 ms …

The enormous potential of multifunctional bilayer wound dressings in various medical interventions for wound healing has led to decades of exploration into this field of medicine. However, it is usually difficult to synthesize a single hydrogel with all the required capabilities simultaneously. This paper proposes a bilayer model with an outer layer intended for hydrogel wound treatment. By adding gelatin methacrylate (GelMA) and tannic acid (TA) to the hydrogel composition and using polyvinyl alcohol-carboxymethyl chitosan (PVA-CMCs) foam layer as supports, a photocrosslinkable hydrogel with an optimal formulation was created. The hydrogels were then examined using a range of analytical procedures, …

A Computational Investigation Into Thermo-Kinetic Parameters Governing The Decomposition Of Bio-Oil Model Compounds, Ali Huseein Aljefri
A Computational Investigation Into Thermo-Kinetic Parameters Governing The Decomposition Of Bio-Oil Model Compounds, Ali Huseein Aljefri
Theses
Among notable products from thermal degradation of biomass are simple oxygen-containing aromatics. These compounds typically appear in high loads in collected bio-oil derived from biomass. As such, it is important to comprehend the decomposition chemistry of this category of compounds. This thesis takes on board a general approach to report thermo-kinetic parameters that govern decomposition of a selected set of bio-oil model compounds in two distinct systems: firstly radical-derived pathways in the gas phase and secondly thermal degradation in their condensed media. The first is attained through accurate density functional calculations while the second is accomplished through the use of …

Clostridioides difficile infection (CDI) remains a significant public health threat globally. New interventions to treat CDI rely on an understanding of the evolution and epidemiology of circulating strains. Here we provide longitudinal genomic data on strain diversity, transmission dynamics and antimicrobial resistance (AMR) of C. difficile ribotypes (RTs) 014/020 (n=169), 002 (n=77) and 056 (n=36), the three most prominent C. difficile strains causing CDI in Australia. Genome scrutiny showed that AMR was uncommon in these lineages, with resistance-conferring alleles present in only 15/169 RT014/020 strains (8.9 %), 1/36 RT056 strains (2.78 %) and none of 77 RT002 strains. Notably, ~90 …

Choosing Sides: On The Manipulation Of Civil Litigation, Yotam Kaplan, Ittai Paldor
Vanderbilt Law Review
Our litigation system is broken. Scholars have long warned that professional litigants, such as debt-collecting firms, insurance companies, and commercial landlords, enjoy immense and unfair advantages over private individuals. What has gone unnoticed is professional litigants’ ability to manipulate their litigatory position—that is, to choose whether they will litigate as plaintiffs or defendants. Extant literature assumes that the parties’ litigatory positions are determined by the substance of the dispute: the party seeking a remedy is the plaintiff, and the party objecting to the award of a remedy is the defendant. We show that, in reality, professional litigants have both the …

Anti-Transgender Constitutional Law, Katie Eyer
Vanderbilt Law Review
Over the course of the last three decades, gender identity anti-discrimination protections and other transgender-supportive government policies have increased, as government entities have sought to protect and support the transgender community. But constitutional litigation by opponents of transgender equality has also proliferated, seeking to limit or eliminate such trans-protective measures. Such litigation has attacked as unconstitutional everything from laws prohibiting anti-transgender employment discrimination to the efforts of individual public school teachers to support transgender teens.
This Article provides the first systematic account of the phenomenon of anti-transgender constitutional litigation. As described herein, such litigation is surprisingly novel: while trans-protective measures …

We report the results of lattice numerical studies of the Sp(4) gauge theory coupled to fermions (hyperquarks) transforming in the fundamental and two-index antisymmetric representations of the gauge group. This strongly coupled theory is the minimal candidate for the ultraviolet completion of composite Higgs models that facilitate the mechanism of partial compositeness for generating the top-quark mass. We measure the spectrum of the low-lying, half-integer spin, bound states composed of two fundamental and one antisymmetric hyperquarks, dubbed chimera baryons, in the quenched approximation. In this first systematic, nonperturbative study, we focus on the three lightest parity-even chimera-baryon states, in analogy …

Underground constructions often encounter water environments, where water–rock interaction can increase porosity, thereby weakening engineering rocks. Correspondingly, the failure criterion for chemically corroded rocks becomes essential in the stability analysis and design of such structures. This study enhances the applicability of the Hoek-Brown (H-B) criterion for engineering structures operating in chemically corrosive conditions by introducing a kinetic porosity-dependent instantaneous mi (KPIM). A multiscale experimental investigation, including nuclear magnetic resonance (NMR), X-ray diffraction (XRD), scanning electron microscopy (SEM), pH and ion chromatography analysis, and triaxial compression tests, is employed to quantify pore structural changes and their linkage with the strength responses …

Purpose:
To describe the clinical, electrophysiological and genetic spectrum of inherited retinal diseases associated with variants in the PRPH2 gene.
Methods.
A total of 241 patients from 168 families across 15 sites in 9 countries with pathogenic or likely pathogenic variants in PRPH2 were included. Records were reviewed for age at symptom onset, visual acuity, full-field ERG, fundus colour photography, fundus autofluorescence (FAF), and SD-OCT. Images were graded into six phenotypes. Statistical analyses were performed to determine genotype–phenotype correlations.
Results:
The median age at symptom onset was 40 years (range, 4–78 years). FAF phenotypes included normal (5%), butterfly pattern dystrophy, …

Non-invasive brain stimulation techniques, such as transcranial direct current stimulation (tDCS), are popular methods for inducing neuroplastic changes to alter cognition and behaviour. One challenge for the field is to optimise stimulation protocols to maximise benefits. For this to happen, we need a better understanding of how stimulation modulates cortical functioning/behaviour. To date, there is increasing evidence for a dose-response relationship between tDCS and brain excitability, however how this relates to behaviour is not well understood. Even less is known about the neurochemical mechanisms which may drive the dose-response relationship between stimulation intensities and behaviour. Here, we examine the effect …
