Protein-protein interactions (PPIs) through the phosphotyrosine-Src homology 2 (SH2) interaction play critical roles in many signal transduction pathways. CT10 regulator of kinase (Crk) and Crk-like (CrkL) are structurally and functionally similar adaptor proteins that mediate PPIs through their SH2 domains. Crk and CrkL are essential in tumor cell migration and invasion and have been proposed as therapeutic targets. However, there are no Crk and CrkL inhibitors available. Previously, we designed a conserved heptapeptide that binds to Crk by analyzing the 15 YXXP motifs in p130Cas. The peptide bound to the SH2 domain of Crk (CrkSH2) and inhibited the CrkSH2-p130Cas interaction. …

BACKGROUND: Black women diagnosed with ductal carcinoma in situ (DCIS) experience disproportionately higher risk of second breast cancer events (SBEs), including both ipsilateral and contralateral recurrences, yet remain underrepresented in molecular biomarker studies compared to non-Hispanic White women.

METHODS: We performed RNA sequencing on 200 archival DCIS samples from the Resource of Archival Breast Tissue cohort, including 33% self-reported Black women. We correlated transcriptomic and clinical data to identify predictors of SBEs. Analysis Of Approximations In Flash Thermal Diffusivity Measurements Using High‑Fidelity Simulations, Tage T. Burnett, Jakob G. Bates, Matthew R. Jones, Christopher R. Dillon, John Tencer

Thermal diffusivity is an important material property for understanding and characterizing transient behavior in many heat transfer applications. This study investigates the accuracy and approximations of inverse mathematical models for measuring thermal diffusivity of materials via the widely used Flash Method. High-fidelity simulations of the Flash Method in copper, silicon carbide, silicon, and glass were performed as numerical experiments and included physics such as in-depth absorption, radial conduction, and surface convection. Data from those numerical experiments were used to estimate material thermal diffusivity using seven traditional and new inverse models. Parker’s original model had relative errors 𝜖< 5% when the approximations it makes were enforced in numerical experiments. Newer models performed well even when experimental restrictions were relaxed. Models that include radial heat conduction were capable of accurately measuring thermal diffusivity (𝜖< 1%) when a Gaussian energy source was used. Models with radial conduction and in-depth material absorption of the laser source could calculate thermal diffusivity for semi-transparent materials such as silicon (𝜖< 1%) and even transparent materials like glass (𝜖< 10%). Convective losses from the material’s front surface had a negligible impact on measurements except for very low thermal diffusivity materials. Using temperatures from many locations of the test material’s surface increased resilience to noise, reducing the distribution of thermal diffusivity measurements by more than an order of magnitude. The models developed in this study could enable a more relaxed Flash Method experimental setup that maintains thermal diffusivity accuracy and extend the utility of the Flash Method to semitransparent materials.

Movement integration has been shown to positively influence elementary students’ cognitive performance, engagement, and on-task behavior; however, its use in classrooms remains inconsistent due to various teacher-perceived barriers. This qualitative study investigates the challenges teachers face when implementing movement integration in elementary classrooms and examines whether access to structured, movement-integrated lesson plans influences instructional practices. The study was conducted with one kindergarten teacher at Lafayette Elementary School. Data were collected through two semi-structured interviews—one conducted prior to implementation and one following a two-week intervention period during which the teacher implemented pre-developed, movement-integrated lesson plans aligned with required academic standards. Background: Not getting enough sleep is common among college students and is often linked to habits such as screen time, caffeine intake, stress, and irregular routines. This study examined how these lifestyle factors are associated with insufficient sleep among college students ages 18-22 and how poor sleep affects their overall wellbeing. Methods: This study used a cross-sectional survey design and collected data through an anonymous 25 question online Qualtrics questionnaire. This was completed by 150 participants. T-cell acute lymphoblastic leukemia (T-ALL) is an aggressive pediatric hematologic malignancy that frequently presents with mediastinal involvement. Delayed recognition can result in rapid clinical deterioration due to airway compression, metabolic derangements, and treatment-related complications. We report the case of a previously healthy 12-year-old male who developed acute hypoxemic respiratory failure secondary to a large mediastinal mass initially treated as pneumonia, in whom imaging demonstrated a large anterior mediastinal mass with airway compression and a malignant right-sided pleural effusion with the diagnosis of T-ALL. The hospital course was complicated by severe tumor lysis syndrome resulting in oligo-anuric acute kidney injury, recurrent …

This qualitative, culturally sensitive study elevated the voices of Black female superintendents in California’s PreK–12 public schools—voices historically marginalized in educational leadership discourse. Centering their lived experiences, the research investigated the challenges these leaders face in advancing equity, the unique assets they bring to their roles, and the ways their intersectional identities shape both their leadership and their navigation of systemic barriers. Four Black female superintendents, identified through purposeful sampling from a professional network, participated in short-answer surveys and in-depth, semistructured interviews. Especially during and following the ‘lockdown era’ of the COVID-19 pandemic, college writing instructors have sought ways to mitigate the harms of neoliberal and capitalist practices in the academy and writing classroom. This portfolio consists of the artifacts of the creative, artistic and research work realized during my doctoral studies. In its core, all recitals had Brazilian music, especially that of H. Villa-Lobos, as a unifying element, although personal taste and reasoning were also a driving force behind choosing repertoire and building a theme for each recital. This research addresses inefficiencies in Manufacturing Execution System (MES)-driven workflows, where excessive user interactions and fragmented access to work instructions introduce transactional and cognitive waste. To address these limitations, an integrated architecture was developed using Ignition as a SCADA-based operator interface connected to the MES through API-based communication. The system replaces direct MES interaction with a unified interface that embeds work instructions and automates transaction execution within the production workflow. The approach was implemented and evaluated through an industrial case study on a 15 station electronics manufacturing line. Results show a 51% reduction in total cycle time, decreasing from 2,996 …

Elucidating the genetic contributions to Parkinson's disease aetiology across diverse ancestries is a critical priority for the development of targeted therapies in a global context. We conducted the largest sequencing characterization of potentially disease-causing, protein-altering and splicing mutations in 710 cases and 11 827 controls from genetically predicted African or African admixed ancestries. We explored copy number variants (CNVs) and runs of homozygosity in prioritized early onset and familial cases. Our study identified rare GBA1 coding variants to be the most frequent mutations among patients with Parkinson's disease, with a frequency of 4% in our case cohort. Of the 18 …

What’s in it for us? examines how value can be created, abstracted, and redefined through material practice, participation, and shared experience. Situated in the space between maker and mender, the artist works in the symbiotic relationship between creation and repair. Wood is the primary material of creation, often in its imperfect form. It becomes both medium and metaphor representing intrinsic value while helping to challenge the systems that’s equate value with market and currency. The transition to a circular economy requires entrepreneurs who can create value from waste, design regenerative business models, and navigate resource-constrained environments. Yet understanding what enables circular entrepreneurial success remains fragmented across the sustainability and entrepreneurship literatures. This dissertation examines how entrepreneurial support systems, capability configuration, and external enabling conditions shape circular venture development and performance through three complementary studies that together advance a unified framework of circular entrepreneurial effectiveness. Driven by escalating threats and regulatory scrutiny, information security risk management (ISRM) is now a strategic imperative for organizations across the industrial spectrum. Although prior literature reviews offer valuable insights into ISRM subdomains, they remain fragmented and overly reliant on manual methods, limiting their scope and scalability. Additionally, only limited reviews have been able to comprehensively synthesize ISRM literature across its three foundational pillars: processes, governance, and strategy. Radiotherapy can lead to late-onset toxicity, to varying extents between individuals due to differences in radiosensitivity. Predicting which patients are most at risk is key to augmenting the therapeutic window. However, the underlying biological mechanisms remain poorly understood, and current experimental methods often lack clinical applicability. Agricultural productivity in East Africa is increasingly threatened by climate-sensitive insect pests. This study investigates the occurrence of key vegetable pests: Brevicoryne brassicae, Plutella xylostella, Maruca vitrata, and Liriomyza mik. A comparative species distribution modelling evaluated pest risk projections under climate change. Four variable selection strategies were tested: case 1 (correlation and variance inflation factor [VIF] filtering across all predictors), case 2 (correlation and VIF filtering of bioclimatic variables merged with other predictors), case 3 (filtering based on environmental values at species occurrence point to approximate realized niches), and case 4 (dimensionality reduction using principal component analysis). Mitochondrial electron transport flavoprotein (ETF) insufficiency causes metabolic diseases known as a multiple acyl-CoA dehydrogenase deficiency (MADD). In contrast to muscle, ETFDH is a non-essential gene in acute lymphoblastic leukemia NALM6 cells, and its expression is reduced across human cancers. In various human cancer cell lines and mouse models, ETF insufficiency caused by decreased ETFDH expression limits flexibility of OXPHOS fuel utilisation but paradoxically increases bioenergetics and accelerates neoplastic growth via activation of the mTORC1/BCL-6/4E-BP1 axis. Spatial transcriptomics (ST) unlocks potential for studying gene functions in processes that depend on orchestration of transcription across space. However, analysis tools for ST remain aimed at data exploration, with few resources for hypothesis testing. What's missing is a way to test whether a factor of interest affects functionally relevant parameters of a gene's spatial distribution. We present a tool to fill this gap, which we call a warped sigmoidal Poisson-process mixed-effects (WSP, pronounced "wisp") model. Elucidating the genetic contributions to Parkinson's disease aetiology across diverse ancestries is a critical priority for the development of targeted therapies in a global context. We conducted the largest sequencing characterization of potentially disease-causing, protein-altering and splicing mutations in 710 cases and 11 827 controls from genetically predicted African or African admixed ancestries. We explored copy number variants (CNVs) and runs of homozygosity in prioritized early onset and familial cases. Our study identified rare GBA1 coding variants to be the most frequent mutations among patients with Parkinson's disease, with a frequency of 4% in our case cohort. Of the 18 …
