Animal models of retinal degeneration are critical for understanding disease and testing potential therapies. Inducing degeneration commonly involves the administration of chemicals that kill photoreceptors by disrupting metabolic pathways, signaling pathways, or protein synthesis. While chemically induced degeneration has been demonstrated in a variety of animals (mice, rats, rabbits, felines, 13-lined ground squirrels (13-LGS), pigs, chicks), few studies have used noninvasive high-resolution retinal imaging to monitor the in vivo cellular effects. Here, we used longitudinal scanning light ophthalmoscopy (SLO), optical coherence tomography, and adaptive optics SLO imaging in the euthermic, cone-dominant 13-LGS (46 animals, 52 eyes) to examine retinal structure …

Previously, we found that Mycobacterium tuberculosis (Mtb) infection in type 2 diabetes mellitus (T2DM) mice enhances inflammatory cytokine production which drives pathological immune responses and mortality. In the current study, using a T2DM Mtb infection mice model, we determined the mechanisms that make T2DM mice alveolar macrophages (AMs) more inflammatory upon Mtb infection. Among various cell death pathways, necroptosis is a major pathway involved in inflammatory cytokine production by T2DM mice AMs. AntiTNFR1 antibody treatment of Mtb-infected AMs from T2DM mice significantly reduced expression of receptor interacting protein kinase 3 (RIPK3) and mixed lineage kinase domain-like (MLKL) (necroptosis markers) and …

A Correlational Study Of The Relationship Of Nurses' Pain Assessment And Patients' History Of Substance Abuse, Valerie A. Esposito Kubanick

Dissertations

Patients experience physical pain for a myriad of reasons. The patients' expression of pain is often difficult to quantify and treat based on individuality of the patients and the viewpoint of the nurses. Therefore, the treatment of pain can become discounted or marginalized, especially in patients who use alcohol or drugs. The use of opioids is the gold standard for pain management, but current administering trends are to use less, or to use non-opioid treatments for fear of patient dependency. There is also an opioid epidemic and patients are sometimes categorized into this unfortunate situation as a precaution. Nevertheless, pain …

Development Of A Rapid Bacterial Quantification Method Based On Droplet Microfluidics, Shawn Bliss

Electronic Theses and Dissertations

The capacity to identify bacteria quickly and accurately is critical for applications such as medical diagnosis, environmental monitoring, and food safety. For instance, sepsis diagnosis requires blood cultures that take as long as 1-5 days, preventing timely intervention and increasing mortality rate. Here, we propose the joint use of microfluidics and a machine learning algorithm for rapid bacterial cell capture and quantification. The combination of high-throughput droplet microfluidics and a support vector machine (SVM) enables analysis and quantification of bacterial samples within as short as 4 hours. We have performed successful encapsulation of pathogenic bacteria such as E. coli, …

A Novel Method For Multiple Phenotype Association Studies Based On Genotype And Phenotype Network, Xuewei Cao, Shuanglin Zhang, Qiuying Sha

Michigan Tech Publications

Joint analysis of multiple correlated phenotypes for genome-wide association studies (GWAS) can identify and interpret pleiotropic loci which are essential to understand pleiotropy in diseases and complex traits. Meanwhile, constructing a network based on associations between phenotypes and genotypes provides a new insight to analyze multiple phenotypes, which can explore whether phenotypes and genotypes might be related to each other at a higher level of cellular and organismal organization. In this paper, we first develop a bipartite signed network by linking phenotypes and genotypes into a Genotype and Phenotype Network (GPN). The GPN can be constructed by a mixture of …

BACKGROUND: The immune microenvironment impacts tumor growth, invasion, metastasis, and patient survival and may provide opportunities for therapeutic intervention in pancreatic ductal adenocarcinoma (PDAC). Although never studied as a potential modulator of the immune response in most cancers, Keratin 17 (K17), a biomarker of the most aggressive (basal) molecular subtype of PDAC, is intimately involved in the histogenesis of the immune response in psoriasis, basal cell carcinoma, and cervical squamous cell carcinoma. Thus, we hypothesized that K17 expression could also impact the immune cell response in PDAC, and that uncovering this relationship could provide insight to guide the development of …
