Acute Pulmonary Injury In Hematology Patients Supported With Pathogen-Reduced And Conventional Platelet Components, Allison P Wheeler, Ronald Jackups, Et Al.
2020-Current year OA Pubs
Patients treated with antineoplastic therapy often develop thrombocytopenia requiring platelet transfusion, which has potential to exacerbate pulmonary injury. This study tested the hypothesis that amotosalen-UVA pathogen-reduced platelet components (PRPCs) do not potentiate pulmonary dysfunction compared with conventional platelet components (CPCs). A prospective, multicenter, open-label, sequential cohort study evaluated the incidence of treatment-emergent assisted mechanical ventilation initiated for pulmonary dysfunction (TEAMV-PD). The first cohort received CPC. After the CPC cohort, each site enrolled a second cohort transfused with PRPC. Other outcomes included clinically significant pulmonary adverse events (CSPAE) and the incidence of treatment-emergent acute respiratory distress syndrome (TEARDS) diagnosed by blinded …

Antibiotics are a strong risk factor for Clostridioides difficile infection (CDI), and CDI incidence is often measured as an important outcome metric for antimicrobial stewardship interventions aiming to reduce antibiotic use. However, risk of CDI from antibiotics varies by agent and dependent on the intensity (i.e., spectrum and duration) of antibiotic therapy. Thus, the impact of stewardship interventions on CDI incidence is variable, and understanding this risk requires a more granular measure of intensity of therapy than traditionally used measures like days of therapy (DOT).

The duck gastrointestinal tract (GIT) harbors an abundance of microorganisms that play an important role in duck health and production. Here, we constructed the first relatively comprehensive duck gut microbial gene catalog (24 million genes) and 4437 metagenome-assembled genomes using 375 GIT metagenomic samples from four different duck breeds across five intestinal segments under two distinct rearing conditions. We further characterized the intestinal region-specific microbial taxonomy and their assigned functions, as well as the temporal development and maturation of the duck gut microbiome. Our metagenomic analysis revealed the similarity within the microbiota of the foregut and hindgut compartments, but distinctive …

MV140 is an inactivated whole-cell bacterial mucosal vaccine with proven clinical efficacy against recurrent urinary tract infections (UTIs). These infections are primarily caused by uropathogenic E. coli (UPEC) strains, which are unique in their virulence factors and remarkably diverse. MV140 contains a non-UPEC strain, suggesting that it may induce an immune response against different UPEC-induced UTIs in patients. To verify this, we experimentally evaluated the cellular and humoral responses to UTI89, a prototypical UPEC strain, in mice vaccinated with MV140, as well as the degree of protection achieved in a UPEC UTI89 model of acute cystitis. The results show that …

Application Of Finite-State Wake Models To Rotors Re-Entering Their Own Wake, Michael Seay Morrow
Application Of Finite-State Wake Models To Rotors Re-Entering Their Own Wake, Michael Seay Morrow
McKelvey School of Engineering Graduate Student Theses & Dissertations
This paper details the development of a finite-state, dynamic wake model for the purpose of modeling a helicopter rotor reentering its own wake. This model stems directly from both the Peters-Morillo wake model to determine the velocity field on and above the helicopter rotor and the Peters-Fei model to compute the velocity field below the rotor. The specific case of nonzero net thrust was evaluated under assumptions of axial flow and potential flow theory. Results consist of computations of the inflow velocity for this case, along with comparison to closed-form, potential flow solutions for validation. The finite-state model is shown …

Improving And Modeling Heterogeneous Streaming Computation, Clayton Faber
Improving And Modeling Heterogeneous Streaming Computation, Clayton Faber
McKelvey School of Engineering Graduate Student Theses & Dissertations
Data streaming algorithms are a class of problems that deal with moving data through a system while being processed. When implementing these types of algorithms a developer will spend time tuning an implementation for deployment, but if they are using heterogeneous architectures or when nodes of computation are physically separate from one another performance may be lost to unforeseen data movement complications. In this work we aim to alleviate some of these pain points through a combination of programming advice and mathematical models. One of the pain points often unseen and underappreciated by developers is a type of data streaming …

Targeted In Vivo Gene Transfer To B Lymphocytes Using Adenoviral Vectors, Paul Boucher
McKelvey School of Engineering Graduate Student Theses & Dissertations
In recent years, engineered cells have been successfully developed for a variety of therapies including cancer and genetic disorders using cells such as T lymphocytes and hematopoietic stem cells. A novel approach that has been enabled by the advent of CRISPR/Cas gene editors is the engineering of B lymphocytes. B lymphocytes are the antibody producing cells of the adaptive immune system and are pivotal in control of pathogens through the binding of membrane bound antibody, or the B cell receptor, to cognate antigen. In conjunction with appropriate co-stimulatory signals, this binding results in activation of the cells and eventual secretion …
Air Quality Assessment On Local And Hyperlocal Scales, Zhiyao Li
Air Quality Assessment On Local And Hyperlocal Scales, Zhiyao Li
McKelvey School of Engineering Graduate Student Theses & Dissertations
Air quality monitoring across local and hyperlocal scales has attracted increased attention from both the public and research community. There are several approaches such as passive samplers and low-cost air quality sensors, depending on the pollutant and measurement objectives. Local and hyperlocal air pollutant measurements help to characterize local emission sources such as traffic and industry, including separating these signals from large scale influences (e.g., urban and regional scales), and better understand how land use types and meteorology influence pollutant spatiotemporal patterns. Low-cost sensors (LCS) are now a popular approach to monitor several criteria pollutants (e.g., particulate matter (PM), ozone, …

Focused ultrasound (FUS) has emerged as a promising noninvasive technique for noninvasive and transient opening of the blood-brain barrier (BBB). FUS-induced BBB opening enables "two-way trafficking" between the brain and bloodstream to benefit the development of effective therapeutic approaches and advance the diagnosis of brain diseases. This technology not only enables targeted drug delivery to the brain for the treatment of various neurological disorders but also can enhance the release of brain-specific biomarkers from the FUS-targeted brain location into the blood circulation for noninvasive and localized molecular diagnosis of brain diseases, a technique called sonobiopsy. However, the lack of precise …

Novel Mass Spectral Data Acquisition And Data Analysis Tools For The Study Of Organic Gases And Particles In Indoor And Atmospheric Environments, Sohyeon Jeon
McKelvey School of Engineering Graduate Student Theses & Dissertations
Particulate matter (PM) has a significant impact on visibility, hydrological cycles, and climate change through direct and indirect radiative properties. PM is also ubiquitous in both indoor and outdoor air, presenting adverse effects on human health. Organic matter (OM) constitutes 20-50% of these particulate matter, known as organic aerosols (OA). The chemical composition of OA is linked to its sources and processes, making it crucial to understand its characteristics for developing aerosol-related control policies, air quality regulations, and climate models. Semi-volatile organic compounds (SVOCs) compose a substantial portion of OA, existing in both gas and particle phases. The distribution of …
Deep Learning-Based Motion Compensation For Four-Dimensional Cone-Beam Computed Tomography Reconstruction In Radiation Therapy, Zhehao Zhang
Deep Learning-Based Motion Compensation For Four-Dimensional Cone-Beam Computed Tomography Reconstruction In Radiation Therapy, Zhehao Zhang
McKelvey School of Engineering Graduate Student Theses & Dissertations
The fundamental objective of radiation therapy is to deliver high doses of ionizing radiation to the target while minimizing the dose to surrounding normal tissues. Cone-beam computed tomography (CBCT) plays an essential role in ensuring the treatment precision by providing on-board volumetric images of the patient’s anatomy, facilitating target localization and patient positioning correction prior to treatment sessions. However, conventional CBCT imaging is susceptible to motion artifacts, potentially leading to inaccuracies in patient setup. To address this challenge, four-dimensional (4D) CBCT has been developed to capture time-resolved positional information of the patient, thereby reducing setup errors in the presence of …
Improved Models Of Elastic Scheduling, Marion Baumli Sudvarg
Improved Models Of Elastic Scheduling, Marion Baumli Sudvarg
McKelvey School of Engineering Graduate Student Theses & Dissertations
In real-time computing systems, \textit{timely} execution is a requirement of \textit{correct} execution. Such systems are widely found in robotics and autonomous vehicle applications, mobile spectrometry of atmospheric aerosols, real-time hybrid simulation for natural hazards engineering, and in prompt localization of transients such as gamma-ray bursts for time-domain and multi-messenger astrophysics. \textit{Elastic scheduling} provides a framework to adjust computational rates and workloads in systems for which timeliness cannot otherwise be guaranteed. While originally proposed for periodic tasks executing on a single processor, elastic scheduling has since been extended to sequential and parallel execution on multiple processors and to earliest deadline first …

Cuff-style peripheral nerve interfaces (PNIs) communicate with the central nervous system and offer interesting advantages when compared with other neural interfaces, including brain-computer interfaces (BCIs) and non-cuff PNI designs. PNI development must consider design factors such as volume of neural activity being driven or recorded, specificity of stimulus activation, and longevity of the device. This dissertation provides an analytical framework for PNI experiments, showcasing two experiments where PNIs were implanted in highly trained macaque monkeys. We applied foundational psychophysical procedures and other behavioral paradigms to examine the relationship between the input electrical stimulus delivered by a cuff electrode and the …
Develop And Optimize Clinical System Of Focused Ultrasound-Enabled Liquid Biopsy (Sonobiopsy) For Brain Cancer Diagnosis, Lu Xu
McKelvey School of Engineering Graduate Student Theses & Dissertations
Brain cancer is a devastating disease characterized by the abnormal growth of cells within the brain. It disrupts neurological function, carries a grim prognosis, and significantly decreases the quality of life. Glioblastoma (GBM), the most common malignant primary brain tumor in adults, is particularly aggressive, with less than 5% of patients surviving for five years. The conventional treatment portfolio, including radiation therapy, chemotherapy, and surgical resections, is less effective for GBM than other cancer diseases due to its high spatial heterogeneity and adaptability to treatment. This suggests that an accurate and comprehensive diagnosis of the molecular subtypes is critical for …

Tree Recovery By Dynamic Programming, Gustavo Alberto Gratacos
Tree Recovery By Dynamic Programming, Gustavo Alberto Gratacos
McKelvey School of Engineering Graduate Student Theses & Dissertations
Tree-like structures are common, naturally occurring objects that are of interest to many fields of study, such as plant science and biomedicine. Analysis of these structures is typically based on skeletons extracted from captured data, which often contain spurious segments or cycles that need to be removed. We propose a dynamic programming algorithm which seeks to recover directed trees from these noisy skeletons. Our method recovers trees by removing edges and duplicating nodes while adhering to edge-label constraints. Our algorithm proceeds by iteratively merging graph nodes, such that the solution on the original graph can be obtained from those on …

Structural Dynamics Of The Human Brain In Vivo From Tagged Mri And Mr Elastography, Jordan De Niro Escarcega
McKelvey School of Engineering Graduate Student Theses & Dissertations
The underlying mechanics surrounding vulnerability of human brains in response to impact is critical to our ability to diagnose and treat traumatic brain injury (TBI), but our understanding of brain mechanical behavior is incomplete. Tagged MRI (tMRI) and magnetic resonance elastography (MRE) are two imaging techniques that are used to visualize and quantify deformation fields in human brains in vivo. The objective of this thesis is to gain a better understanding of brain vulnerability, by quantitively analyzing deformation fields extracted from in vivo human imaging studies. This was achieved through the following aims: (1) Extract mode shapes and natural frequencies …

Engineering Macrophages For Self-Regulated, Localized Delivery Of Anti-Inflammatory Biologics, Molly Klimak
McKelvey School of Engineering Graduate Student Theses & Dissertations
Immunoengineering continues to revolutionize healthcare, generating new approaches for treating previously intractable diseases. In chronic inflammatory diseases such as rheumatoid arthritis (RA), a chronic autoimmune disease characterized by joint inflammation and destruction, anti-cytokine treatments have previously been at that forefront of therapeutic innovation. However, while many of the existing anti-cytokine treatments are successful for a subset of patients, these treatments can also pose severe adverse events and off-target effects from systemic administration of anti-inflammatory drugs at constant high dosages or a lack of disease-specific targets. These limitations have motivated the development of new treatment strategies that are well-controlled, precise, safe, …
Exploring The Frontier Of Amorphous Materials: Structure, Dynamics, Nucleation Behavior, And Glass Forming Ability, Fangzheng Chen
Exploring The Frontier Of Amorphous Materials: Structure, Dynamics, Nucleation Behavior, And Glass Forming Ability, Fangzheng Chen
McKelvey School of Engineering Graduate Student Theses & Dissertations
The transition from a liquid to a crystal or a glass is a fundamental process that determines many properties of the final product. Although numerous experiments have been conducted to investigate this transition, the atomistic scale of such behavior remains elusive. The properties of a high-temperature liquid could play an important role during solidification. For example, it has been argued that the ordering of the structure factor in a liquid reveals the glass-forming ability. In this dissertation, computational methods were used to study the structure, dynamics, nucleation properties, and glass-forming ability of metallic liquids and silica glasses. Several Embedded Atom …
