Dysregulated fatty acid metabolism is an attractive therapeutic target for colorectal cancer (CRC). We previously reported that fatty acid synthase (FASN), a key enzyme of de novo synthesis, promotes the initiation and progression of CRC. However, the mechanisms of how upregulation of FASN promotes the initiation and progression of CRC are not completely understood. Here, using Apc/VillinCre and ApcMin mouse models, we show that upregulation of FASN is associated with an increase in activity of β-catenin and expression of multiple stem cell markers, including Notum. Genetic and pharmacological downregulation of FASN in mouse adenoma organoids decreases the activation of β-catenin …

Toxoplasma gondii forms tissue cysts in neurons and astrocytes in the brain to establish chronic infection, and astrocytes express the CXCL10 chemokine in chronically infected mice. Since chemokines mediate the migration of T cells to attack their targets, and since CXCL10 plays key roles in T cell-mediated control of the proliferation of tachyzoites (the acute stage form) of T. gondii during the acute stage of infection, we examined whether CXCL10 is involved in recruiting anti-cyst CD8+ cytotoxic T cells to eliminate the cysts in their brains. The Impact Of Pdcd4, A Translation Inhibitor, On Drug Resistance, Qing Wang, Hsin-Sheng Yang

Programmed cell death 4 (Pdcd4) is a tumor suppressor, which has been demonstrated to efficiently suppress tumorigenesis. Biochemically, Pdcd4 binds with translation initiation factor 4A and represses protein translation. Beyond its role in tumor suppression, growing evidence suggests that Pdcd4 enhances the chemosensitivity of several anticancer drugs. To date, numerous translational targets of Pdcd4 have been identified. These targets govern important signal transduction pathways, and their attenuation may improve chemosensitivity or overcome drug resistance. This review will discuss the signal transduction pathways regulated by Pdcd4 and the potential mechanisms through which Pdcd4 enhances chemosensitivity or counteracts drug resistance.

In GMM estimation, it is well known that if the moment dimension grows with the sample size, the asymptotics of GMM differ from the standard finite dimensional case. The present work examines the asymptotic properties of infinite dimensional GMM estimation when the weight matrix is formed by inverting Brownian motion or Brownian bridge covariance kernels. These kernels arise in econometric work such as minimum Cramer-von Mises distance estimation when testing distributional specification. The properties of GMM estimation are studied under different environments where the moment conditions converge to a smooth Gaussian or non-differentiable Gaussian process. This paper develops a welfare accounting decomposition that identifies and quantifies the origins of welfare gains and losses in general economies with heterogeneous individuals and disaggregated production. The decomposition — exclusively based on preferences and technologies — first separates efficiency from redistribution considerations. Efficiency comprises i) exchange efficiency, which captures allocative efficiency gains due to reallocating consumption and factor supply across individuals, and ii) production efficiency, which captures allocative efficiency gains due to adjusting intermediate inputs and factors, as well as technical efficiency gains from primitive changes in technologies, good endowments, and factor endowments. Amyloid-β protein (Aβ) is widely studied due to its key role in Alzheimer’s disease (AD) pathology. One characteristic of patients suffering from AD is the deposition of amyloid plaques comprised of Aβ aggregates, which are associated with synapse damage. Thus, studying Aβ aggregation and aggregation inhibition may provide pathological and therapeutic insights. Aggregation of Aβ is a nucleation dependent process that may be influenced by the presence of pre-formed Aβ aggregate seeds that can facilitate formation of protein aggregates. This research evaluates the dynamic behavior and volume change of sand with gravel subjected to cyclic loading. Historically, research on the dynamic behavior of cohesionless soils has primarily focused on the study of liquefaction due to its known implications on soil shear strength loss. Gravelly soil is generally assumed to be less susceptible to liquefaction since its large particle size facilitates drainage during cyclic loading. Gravel’s particle size also makes it difficult to test using standard cyclic shearing laboratory equipment. During the 2023-2024 academic year, this action research study was conducted at local, mid-sized high school in South Carolina using a mixed-methods approach to evaluate the efficacy and impact of a mandatory athletic study hall (ASH) program on high school athletes’ academic performance. Through this program, during each academic quarter, any in-season athletes with any grades below 69% to either attend after-school study hall sessions or complete tutoring. The study both reviewed the academic performance of athletes and nonathletes and collected qualitative data from stakeholder surveys completed by nonathletes, athletes, teachers, coaches, and parents. This work examines two applications of pooling: group testing and pooled biomonitoring. Group testing, introduced by Dorfman in the early 1940s, was initially developed to screen for syphilis among U.S. inductees during World War II. Since then, the approach has demonstrated cost-saving benefits in diverse fields, including drug discovery, genetics, and infectious disease testing. While various regression methods—parametric, nonparametric, and semiparametric—have been proposed to analyze group testing data, they fall short in addressing age-related disparities in disease presence if such variations exist. Through teaching experiences in the math classroom, students voiced that solving mathematical problems did not benefit them in the real-world. This study discussed the use of a performance task in a high school geometry class as a way to engage students, increase their confidence when solving math problems, and determine the impact they have on engagement compared to supportive assignments. Constance Mellon formally identified library anxiety in the mid-1980s. Library anxiety can result in students avoiding the library, its resources, and its staff. Students with library anxiety perform worse on academic tasks than their more comfortable peers. Library anxiety causes procrastination on assignments and undermines academic success. Technology anxiety has evolved with library anxiety changing as new information technology becomes available. This study examined the relationship between a state high-stakes summative mathematics assessment and an interim third-party math diagnostic assessment and data literacy among teachers in a Southeastern school district. Despite ongoing federal and state accountability efforts, the gap between policy intentions and student achievement continues to grow. In this quantitative correlational study, the relationship between the Spring 2023 state summative and interim assessment data for fourth and eighth-grade students was examined.
