Ubiquitin-specific protease-7 (USP7) is an important drug target as it regulates multiple proteins and genes (such as MDM2 and p53) with roles in cancer progression. Its inhibition can hinder the function of oncogenes, increase tumor suppression, and enhance immune response. The current study was designed to express USP7 in a prokaryotic system, followed by screening of small molecules against it using biophysical methods, primarily STD-NMR technique. Among them, 12 compounds showed interaction with USP7 as inferred from NMR-based screening. These compounds further caused destabilization of USP7 by reducing its melting temperature (Tm) up to 6 °C in …

Machine learning traditionally emphasizes developing models for given datasets, but real-world data is often messy, making model improvement insufficient for enhancing performance. Data-Centric AI (DCAI) is an emerging field that systematically improves datasets, leading to significant practical ML advancements. While experienced data scientists have manually refined datasets through trial-and-error and intuition, DCAI approaches data enhancement as a systematic engineering discipline. DCAI represents a shift from focusing on models to the underlying data used for training and evaluation. Niclosamide, a drug originally used to treat parasitic infections, has shown potential as a possible cancer treatment, but poor absorptive and solubility properties are believed to limit its effectiveness. Researchers are exploring modified structures of the drug along with advanced delivery methods, particularly nanotechnology, to improve niclosamide’s absorption profile and improve its specificity in regard to targeting cancerous cells. These efforts intend to produce more effective tumor-targeting compounds, which can be employed as pharmacological cancer treatment options when appropriate. Introduction
The Vestibular/Ocular Motor Screening (VOMS) was created as a brief clinical screening tool for identifying vestibular and ocular motor symptoms and impairments post-concussion. It was found to have predictive validity in correctly identifying concussed athletes from healthy controls. In 2018, the Military Acute Concussion Evaluation 2 (MACE2) replaced the original Military Acute Concussion Evaluation (MACE); the most prominent change between the MACE and MACE2 was the addition of the VOMS. Despite its adoption into military medicine, it is not known if the addition of the VOMS to the MACE2 is acutely helpful, and if it provides additional information for …

Haydn spent his formative musical years in Vienna, a city with a strong Marian cult that was expressed musically through such works in German as Heinrich Rademin’s Mater dolorum (set by Werner, Reutter the Younger, and Wagenseil), and by innumerable settings of the Latin sequence Stabat mater. In Vienna, musical settings of the Stabat mater were traditionally performed not only on the feast of Septem Dolorum B. V. M., but also on Saturdays in Lent and during Holy Week. A “Rosary” Symphony? The Impact Of Haydn’S Religious Faith On His Symphonies, Henry Stratmann

Despite expressing a strong religious faith throughout his life, Haydn’s secular music far outnumbers his sacred works. Certain symphonies, however, include either allusions to religious themes or project what can be interpreted as a devotional mood. Specifically, some unusually long symphonic slow movements during Haydn’s “Sturm und Drang” period may be orchestral depictions of a meditative, contemplative, prayerful state of mind. They likely reflect the particular practices of Roman Catholic belief and worship that were a pervasive part of the culture in which Haydn lived his entire life and also part of his personal belief system. Mater Dolorum Settings By Georg Reutter The Younger And Gregor Werner, As Reflected In Music By Joseph Haydn, Janet K. Page

Mater dolorum, a text by Heinrich Rademin (1674–1731), is a German-language oratorio intended for performance before a model of the Holy Sepulcher on Good Friday. Such works, amalgamating court sepolcro and popular traditions, were performed in churches throughout Central Europe during Holy Week. Mater dolorum, one of the most popular texts, was set by three of Haydn’s older contemporaries: Georg Reutter Jr. (1726), Gregor Werner (ca. 1733), and Georg Christoph Wagenseil (ca. 1739). In this essay, I consider how these settings, and the attitudes displayed in them, intersected with Haydn’s world and could have influenced him, especially in his …

Millions of United States (U.S.) troops deployed to the Middle East and Southwest Asia were exposed to toxic airborne hazards and/or open-air burn pits. Burn pit emissions contain particulate matter combined with toxic gasses and heavy metals. Ongoing research has demonstrated that exposures to the airborne hazards from military burn pits have profound and lasting health and wellness consequences. Research on the long-term health consequences of exposure to open burn pits has been limited. This Note analyzes a 2023 Fifth Circuit opinion concerning whether a contractual pre-dispute jury waiver is enforceable utilizing the knowing and voluntary factors and when challenged under general fraud allegations. The Fifth Circuit incorrectly weighed two of the voluntary and knowing factors—conspicuousness and unequal bargaining power—to be in favor of enforcing the jury waiver when the facts of the case and prior case law prove otherwise. The Fifth Circuit should have first applied a historical analysis in analyzing a constitutional right, but the court was reasonable in applying the arbitration rule to jury waiver provisions because waiving a jury right …

The Biden administration has conducted the government’s first ever negotiations with pharmaceutical companies on the cost of life saving drugs. They have been able to negotiate prices down by nearly 80%. The drugs at the center of negotiations are ones that people are prescribed for years at a time in order to manage illnesses such as heart disease, Crohn’s disease, and more. In order to conduct these negotiations, the Biden administration used march-in rights built into the Bayh-Dole Act.

Several reports, including our previous studies, indicate that hyperglycemia and diabetes mellitus exert differential effects on vascular function in males and females. This study examines sex differences in the vascular effects of type 2 diabetes (T2D) in an established monogenic model of obesity-induced T2D, Zucker Diabetic Fatty (ZDF) rats. Acetylcholine (ACh) responses were assessed in phenylephrine pre-contracted rings before and after apocynin, a NADPH oxidase (NOX) inhibitor. The mRNA expressions of aortic endothelial NOS (eNOS), and key NOX isoforms were also measured. We demonstrated the following: (1) diabetes had contrasting effects on aortic vasorelaxation in ZDF rats, impairing relaxation to …

Mass Exodus? Police Officer Turnover Intention And Organizational Commitment In New York City, Kenneth M. Quick, Kevin T. Wolff

Purpose – This study assesses the relationship between job satisfaction, perceived organizational support, and workplace factors on officer turnover intention within an urban, municipal police organization.
Design/methodology/approach – Using data from an online survey of New York City Police Officers (n=1,823), both bivariate analysis and logistic regression models were utilized to assess the salience of police officer job satisfaction, perceived organizational support, and perception of six workplace domains, including financial compensation, environmental factors, professional fulfillment, work/quality of life balance, treatment from management, and occupational risk on predicting turnover intention.
Findings – The cross-sectional study finds that job satisfaction, financial factors …

Cultivating library collections includes not just thoughtful acquisition of materials, but also careful upkeep of physical materials and digital records. When the time has come to make space in the library, many different factors must be considered and dealt with to successfully complete a bound journal removal project. This poster highlights a project in the Science Library at the College of the Holy Cross. Critical Comparison Of Potential Machine Learning Methods For Lightning Thermal Damage Assessment Of Composite Laminates, Juhyeong Lee, Scott L.J. Millen, Xiaodong Xu

The present study assesses the potential of using machine learning (ML) methods to predict the extent of lightning thermal damage in fiber-reinforced composite laminates using three supervised machine learning (SML) algorithms: (1) linear regression (LR), (2) decision tree (DT)-based, and (3) MLP models. These models were based on the 10 most significant factors that influence the severity of lightning damage, including three current waveform parameters, four material configurations, and three orthogonal electrical conductivities of each composite. All models demonstrated good performance with coefficient of determination (R2) values between 0.84 ~ 0.96. The multilayer perceptron (MLP) regression model trained …

Sepsis remains an important healthcare challenge. The lungs are often affected in sepsis, resulting in acute lung injury characterized by inflammation. Mechanisms involving lipopolysaccharide (LPS) stimulation of toll-like receptor (TLR) signaling with induction of proinflammatory pathways have been implicated in this process. To date, however, studies targeting these pathways have failed to improve outcomes. We have found that LPS may also promote lung injury through the activation of α4 nicotinic acetylcholine receptors (α4 nAChRs) in immune cells. We observed increased expression of α4 nAChRs in human THP-1 monocytic cells exposed to LPS (100 ng/mL, 24 h). We also observed that …

Misregistration between CT and PET in PET/CT is mainly caused by respiratory motion or irregular respiration during the CT scan in PET/CT. Other than repeat CT, repeat PET/CT, or data-driven gated (DDG) CT, there is no practical approach to mitigate the misregistration artifacts and subsequent CT attenuation correction (CTAC) of the PET data. DDG PET derives a respiratory motion model based on the multiple phases of PET images without hardware gating and it allows for a potential correction of the misregistration artifacts based on the respiratory motion model. Dishonest behaviours such as tax evasion impose significant societal costs. Ex ante honesty oaths-commitments to honesty before action-have been proposed as interventions to counteract dishonest behaviour, but the heterogeneity in findings across operationalizations calls their effectiveness into question. We tested 21 honesty oaths (including a baseline oath)-proposed, evaluated and selected by 44 expert researchers-and a no-oath condition in a megastudy involving 21,506 UK and US participants from Prolific.com who played an incentivized tax evasion game online. Of the 21 interventions, 10 significantly improved tax compliance by 4.5 to 8.5 percentage points, with the most successful nearly halving tax evasion. Limited …

Starch is one of the widely required excipients in pharmaceutical dosage forms manufacturing. Indonesia’s rich biodiversity, encompassing tubers like arrowroot, taro beneng, porang, ganyong and yams, harbours the potential for novel starch excipient sources. These tubers provide starch variations with distinct functional properties influenced by the natural starch granules’ properties and amylose-amylopectin ratio. Previous research utilizing natural and modified starches from these tubers has demonstrated their potential in pharmaceutical formulation. These starch tubers have been studied and provided promising results in tablet formulation and film development. The COVID-19 pandemic significantly disrupted public transit systems across the US, resulting in substantial declines in ridership and revenue. At the same time, ridership has increased to different levels in cities. This thesis explores the determinants of transit recovery in American cities post-pandemic. It analyzes data from the National Transit Database (NTD) and employs a two-stage least squares (2SLS) regression model first laid out by Taylor et al. (2008) for data from 2000.
