The proinflammatory cytokine interferon-gamma (IFNγ) is upregulated in a variety of infections and contributes to bone marrow failure through hematopoietic stem cell (HSC) activation and subsequent exhaustion. The cell surface protein, bone marrow stromal antigen 2 (BST2), is a key mediator of this process, as it is induced upon interferon stimulation and required for interferon-dependent HSC activation. To identify the mechanism by which BST2 promotes interferon-dependent HSC activation, we evaluated its role in niche localization, immune cell function, lipid raft formation, and intracellular signaling. Our studies indicated that knock out (KO) of BST2 in a murine model does not disrupt …

The mobile nature of migratory marine animals across jurisdictional boundaries can challenge the management of biodiversity, particularly under global environmental change. While projections of climate-driven habitat change can reveal whether marine species are predicted to gain or lose habitat in the future, geopolitical boundaries and differing governance regimes may influence animals’ abilities to thrive in new areas. Broad geographic movements and diverse governance approaches elicit the need for strong international collaboration to holistically manage and conserve these shared migratory species. In this study, we use data from the Tagging of Pacific Predators program to demonstrate the feasibility of using climate-driven …

Background: Night shift workers face increased risk of cardiovascular disease (CVD) compared to non-shift workers. Evidence supports on-shift napping and regular non-invasive monitoring of endothelial function for risk mitigation, yet neither strategy is widely used. Methods: We evaluated the feasibility of non-invasive assessment of peripheral arterial tone (PAT) to assess the effect of napping during simulated night shift work on endothelial function. We used a single-site, randomized crossover trial of simulated night shift work with a 45-min nap condition versus a control, no-nap condition (ClinicalTrials.gov NCT05436951). Results: The primary outcome was the number of participants with ≥ 70% of endothelial …

This paper introduces a novel computational framework for evaluating above-knee prostheses, addressing a major challenge in gait deviation studies: distinguishing between prosthesis-specific and patient-specific contributions to gait deviations. This innovative approach utilizes three separate computational models to quantify the changes in gait dynamics necessary to achieve a set of ideal gait kinematics across different prosthesis designs. The pilot study presented here employs a simple two-dimensional swing-phase model to conceptually demonstrate how the outcomes of this three-model framework can assess the extent to which prosthesis design impacts a user's ability to replicate the dynamics of able-bodied gait. Furthermore, this framework offers …

Evaluation Of 3d Beach Profiles: A New Method For Monitoring Changes In Beach Morphology And Grain Size, Kalinda Ellen Roberts
Evaluation Of 3d Beach Profiles: A New Method For Monitoring Changes In Beach Morphology And Grain Size, Kalinda Ellen Roberts
Graduate Masters Theses
Beach profiles are a simple and affordable approach to monitoring changes in beach morphology. However, traditional methods offer limited resolution and dimension. Recent advancements in technologies, particularly structure-from-motion (SfM) and grain size detection models (GSDMs), can be integrated into conventional beach profiling techniques, enabling more detailed spatial and temporal analysis of slope and grain sizes. This research introduces a new technique for coastal monitoring, termed “3D Beach Profiles,” which integrates SfM technology to create high-resolution three-dimensional digital surface models (DSMs) and orthomosaics of beach surfaces. These digital surfaces enable the extraction of beach slopes with orders of magnitude more data …
Frequency Distribution Of Graphemic-Phonemic Correspondences Of Vowels In English Children's Literature, Steven Michael Paquin
Frequency Distribution Of Graphemic-Phonemic Correspondences Of Vowels In English Children's Literature, Steven Michael Paquin
Graduate Masters Theses
Phonics instruction is widely regarded as the most efficacious method for teaching children to read English. In order to acquire reading, young-learners must connect orthography to phonology. An obstacle to English reading acquisition is the language’s opaque orthography. Its grapheme to phoneme correspondence is not 1:1. Instead, a range of multi-letter strings, or graphotactics, correspond to various phonemic sequences. This network of graphophonemic correspondences renders English reading acquisition an arduous undertaking for young-learners. Phonics curricula like Letters and Sounds (Department for Education and Skills, 2007) in the U.K. and UFLI Foundations (Lane et. al., 2022) in the U.S. aid young-learners …

Size Matters: The Characterization Of Telomere Function In Germ Cell Development In Danio Rerio, Jessica Macneil
Graduate Doctoral Dissertations
Infertility is a growing problem worldwide. According to the WHO, 48 million couples across the globe suffer from infertility and many couples turn to Assisted Reproductive Technology (ART) to become pregnant. One readout of gamete quality and successful ART is telomere integrity because telomeres are necessary for chromosome movements during meiosis so that the segregation of chromosomes happens correctly. Telomeres are well known for their role in aging; however, they play an important and understudied role during meiosis. In meiosis, telomeres attach to the nuclear envelope (NE) via a protein chain spanning the NE that interacts with cytoplasmic motor proteins, …

High-resolution mass spectrometry (HRMS), as its name suggests, possesses high resolving power that enables the separation of ions with very close mass values. This is particularly beneficial for analyzing complex samples where numerous compounds may have similar masses, as it helps prevent signal overlaps and ensures an accurate spectral analysis. The current work explores the diverse applications of high-resolution mass spectrometry in oligonucleotide research, organic synthesis and medicinal chemistry. Oligonucleotide research: Chapters 2-5 present the research results from studies that employed ultra-high-performance liquid chromatography coupled with high-resolution mass spectrometry (UHPLC-HRMS) to understand the pharmacokinetics of RNA interference (RNAi) therapeutics. Removing Methane Via Soil Amendments: The Legal Framework, Korey Silverman-Roati, Romany M. Webb
Sabin Center for Climate Change Law
Rapid and sustained reductions in human-driven methane emissions are critical to limiting warming in line with the 2015 Paris Agreement. Reductions in certain methane emissions, particularly from the fossil fuel sector, can be implemented cheaply and efficiently. Other forms of methane emissions, like those from agricultural sources, are more difficult to control. This fact, coupled with concerns that climate change could unlock significant additional natural emissions from tropical wetlands and melting permafrost, has increased interest in atmospheric methane removal (“AMR”).
AMR refers to human interventions to accelerate the conversion of methane in the atmosphere to a form that causes less …

Sabin Center for Climate Change Law
This paper provides a detailed examination of the regulatory landscape surrounding Ocean Alkalinity Enhancement (“OAE”), a form of marine carbon dioxide removal (“CDR”) that seeks to mitigate climate change. The paper explores the domestic (U.S.) regulation of two of the most commonly discussed approaches to OAE: a mineral based approach and an electrolytic approach.
While no federal laws specifically mention OAE, the practice might be subject to various pollution control and other federal statutes and regulations. One key question is the extent to which federal law preempts state regulation of OAE. This paper aims to answer that question by reviewing …

Construction Management
This paper details the preconstruction process involving the replacement and redesign of the north wall at the CAED Support Shop at Cal Poly, San Luis Obispo. Accomplished by an interdisciplinary team of Architectural Engineering and Construction Management students, this project explores the requirements and procedures of generating a project proposal to submit to Cal Poly Facilities Management. The project, initiated in the fall of 2024, aimed to redesign the wall aligned with the adjacent Engineering West building and replace it with a CMU low lift grouted wall. The CAED Support Shop is offered as a resource to all CAED students …

Objective
Extracting DNA is essential in wildlife genetic studies, and numerous methods are available. However, the process is costly and time-consuming for non-model organisms, including most wildlife species. Therefore, we optimized a cost-efficient protocol to extract DNA from the muscle tissue of White-tailed Deer using the DNAdvance kit (Beckman Coulter), a magnetic-bead-based approach. We devised a 3 × 3 factorial design using combinations of tissue mass (10 mg, 50 mg, or 100 mg) and reaction volume (25%, 33%, and 50% of the manufacturer's recommended volumes). DNA was extracted for N = 81 tissue sub-samples (9 replicates/treatment).

Libyan Moringa oleifera leaves were extracted using methanol, and the bioactive components' antibacterial, antioxidant, anti-inflammatory, and anticancer characteristics were assessed using gas chromatography mass spectrometry analysis (GC-MS). Moringa oleifera exhibited antioxidant activity, with an IC50 of 11.53 µg/mL. Wi38 cell line's viability decreased with increasing M. oleifera extract concentration, while its cytotoxicity increased with an IC50 of 393.41±7.77 μg/mL. Alloxan-induced diabetes model in Swiss albino mice was implemented. Three different doses (100, 200, or 300 mg/kg of mice body weight) of M. oleifera’s extract were evaluated for their effects on blood sugar concentration and the histology of the pancreatic islets. …
