Triple-negative breast cancer (TNBC) is an aggressive breast cancer subtype associated with limited targeted treatment options, heterogeneous treatment response, and high risk of early recurrence. Artificial intelligence (AI) has rapidly emerged as a powerful tool to address key clinical challenges in TNBC across diagnosis, treatment response assessment, and prognosis. Diagnostic and staging challenges persist due to variable imaging features in TNBC and limitations in conventional modalities, increasing the risk of delayed detection. Predicting response to neoadjuvant systemic therapy remains difficult, as patient responses are heterogeneous, and existing clinical markers provide limited early predictive value. An ideal cancer therapy enhances anti-tumor effects while minimizing side effects. iRGD, a non-cytotoxic peptide that activates a tumor-specific molecular transport machinery, promotes the penetration of co-injected drugs into tumor tissues. Clinical trials have demonstrated its potential as a tumor-specific delivery scaffold and potentiator of anti-cancer agents. In this study, we synthesized an iRGD conjugate containing monomethyl auristatin F (MMAF), a highly toxic antimitotic agent, and characterized its dual function as a tumor-specific cytotoxic agent and co-injected drug delivery scaffold. The iRGD-MMAF conjugate internalized and killed cultured tumor cells in an αv integrin-dependent manner. When injected systemically, iRGD-MMAF homed selectively …

This study investigated the role of cytokines in Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (ME/CFS), a debilitating multi-system disease with unknown etiology. Using data from a case-control study (60 cases, 61 controls) previously collected by Dr. Roxana Moslehi’s lab, serum levels of 48 cytokines were analyzed. Statistical analyses, including T-tests, Wilcoxon Rank Sum tests, and logistic regression (unadjusted and adjusted for medication use), were performed to identify differences between cases and controls.
Results revealed a statistically significant difference in mean Fractalkine levels between cases and controls (p=0.031). Borderline significant differences were also observed for IL-6 (p=0.046) and MIP-1α (p=0.046). Unadjusted logistic regression …

To better advance our knowledge of cellular behavior, development of an organism, and disease mechanisms understanding cellular heterogeneity in the functional proteome is essential. Bulk measurements across a cell population often fail to capture the variations which exist between individual cells, however single cell proteomic technology has increasingly demonstrated the characterization of cellular heterogeneity. A single cell functional proteomics assay needs to be both high-throughput and multiplexed to effectively identify biomarkers and provide a quantitative correlation to better understand the interrelationships within complex protein networks. Among these, the metabolism of homocysteine (HCY), a sulfur-containing amino acid derived from dietary methionine, has attracted growing attention due to its involvement in one-carbon metabolism and its potential link to cancer. Members of the gram-positive bacterial genus Staphylococcus have historically been classified into coagulase-positive Staphylococcus (CoPS) and coagulase-negative Staphylococcus (CoNS). This classification is based on the ability of clotting of plasma by the coagulase enzyme, which converts fibrinogen to fibrin. CoNS species are ubiquitous commensals that frequently colonize the skin and mucosal surfaces. They are also opportunistic pathogens implicated in many human and animal infections. The evolutionary history of CoNS has generally been overlooked because of the historical lack of recognition for their clinical importance and poor taxonomic sampling. Salmonella enterica is a globally distributed bacterial pathogen that is commonly acquired through consumption of contaminated food and water. The enterica subspecies includes over 1500 unique subvariants, or serovars, that are classified by antigen presentation. Due to its global environmental prevalence, broad host range, and increasing pathogenicity, serovar Typhimurium is of particular concern for human and animal health, as well as food safety. Once in the gastrointestinal tract, Salmonella Typhimurium (STm) exploits host immune mechanisms, implements an arsenal of virulence factors, and outcompetes the microbiota to invade the intestinal epithelium. The p53 family of transcription factors regulates diverse gene regulatory networks such as those involved in DNA damage repair, epidermal commitment, and neuronal development. As a result, dysfunction in these pathways lead to numerous human health conditions, including multiple types of cancer and craniofacial developmental disorders. Background: This dissertation addresses the urgent issue of child trafficking in high-incidence areas of Ghana, where sociocultural, economic, and structural factors sustain and exacerbate this complex social challenge. The rationale for the study is grounded in the need for a holistic understanding of child trafficking beyond traditional justice-based approaches. The research emphasizes the necessity of community-led interventions informed by local cultural norms and protective values that have been largely unexplored in the Ghanaian context. Specifically, this dissertation applies an integrative approach, rooted in the Ubuntu philosophy, to explore community interdependence as a central component of sustainable anti-trafficking interventions.

Pseudomonas aeruginosa is an opportunistic pathogen that is highly antibiotic resistant and causes tens of thousands of infections each year. It persists in hospitals by forming biofilms on medical equipment like ventilators and catheters, making it difficult to eradicate. Biofilm formation, among other virulence factors of P. aeruginosa, is regulated through quorum sensing, a mechanism of bacterial communication. Quorum sensing is primarily coordinated through LuxI-R type systems comprised of an autoinducer synthase and receptor, respectively. Canonically, the first system to be expressed is LasI-R, and LasR upregulates the second arm of the quorum sensing network, the RhlI-R system. Research …

Many participants in clinical trials of psychedelic-assisted therapy (PAT) report mystical experiences. Researchers often use the 30-item Mystical Experience Questionnaire (MEQ-30) to measure mystical experiences. Several studies have validated a four-factor structure of the MEQ-30, but only as a subset of items within broader versions of the scale (e.g., MEQ-43). As such, the factor structure of the standalone MEQ-30 remains unvalidated in English. Additionally, some data suggest that certain factors might be more strongly associated with psychedelic-associated outcomes than others, which could provide insight to enhance the therapeutic potential of PAT. Studies indicate that a grave mental health crisis is disproportionately impacting female-identified youth. While school districts have made attempts to address this problem, those most impacted - young people - have frequently found their voices overlooked by adults seeking solutions. Upon the introduction of the Metropolis et. al (1953) [6] algorithm, the question of how many steps in the Markov chain were needed to achieve convergence to stationarity became apparent. The purpose of this qualitative case study was to explore how homeschooling parents (HPs) conceptualize the role of motivation in early reading development and how these conceptualizations inform how they design and implement their reading instruction. Over 50,000 Canadians have died of unregulated drug poisoning since 2016. And while the toxic drug crisis is the leading cause of death in British Columbia for people between the ages of 10 and 59, some British Columbians are at a higher risk than others. In 2023, First Nations people—who comprise 3.4% of BC’s population—accounted for 17.8% of its toxic drug deaths. Other marginalized communities are also at heightened risk: racialized people, young people, and people living in poverty or without shelter are all disproportionately impacted. This review paper identifies key stability and control screening parameters needed to design low-risk, general-purpose high-speed aircraft. These derive from MIL-STD-8785C, MIL-STD-1797, and older AGARD reports, and are suitable for assessing conceptual high-speed vehicles. We demonstrate their applicability using published ground test, computation, and flight test data from the Bell X-2, North American X-15, Martin X-24A, Northrop HL-10, Lockheed Blackbird (YF-12/SR-71), and North American XB-70 as well as the Rockwell Space Shuttle Orbiter. The relative success of the X-15 and Blackbird and the performance limitations of the others indicate the need to scrutinize lateral-directional stability at the preliminary design phase. …

Objective: This study presents a mathematical model and finite element simulations to investigate interstitial fluid flow and nanodrug transport in a solid tumor, incorporating transvascular exchange, convection–diffusion–reaction dynamics, and intratumoral injection mechanisms. Impact Statement: Optimizing nanodrug distribution remains a critical challenge in cancer therapy. The proposed model advances nanomedicine by enhancing the mechanistic understanding of nanodrug transport in a solid tumor. Objective: To test the hypothesis that the association of smoking with long-term colorectal cancer incidence may be stronger for tumours with higher mutational and neoantigen loads.
Methods and analysis: In the Nurses' Health Study (1980-2012) and the Health Professionals Follow-up Study (1986-2012), our novel prospective cohort incident-tumour biobank method (PCIBM) used 3053 incident colorectal carcinoma cases including 752 cases with whole-exome sequencing data. Using the multivariable duplication-method Cox regression model with the inverse probability weighting to adjust for the selection bias due to tissue availability, we assessed a differential association of cigarette smoking with colorectal carcinoma incidence by an exome-wide …

Previous studies have indicated that specific CpG sites may be linked to the risk of prostate cancer (PCa) by regulating the expression of PCa target genes. However, most existing studies aim to identify DNA methylation (DNAm) biomarkers through blood tissue genetic instruments, which impedes the identification of relevant biomarkers in prostate tissue. To identify PCa risk-associated CpG sites in prostate tissue, we established genetic prediction models of DNAm levels using data from normal prostate samples in the GTEx (N = 108) and assessed associations between genetically predicted DNAm in prostate and PCa risk by studying 122,188 cases and 604,640 controls. …

Transcriptome-wide association studies (TWAS) have been widely used to identify thousands of likely causal genes for diseases and complex traits using predicted expression models. However, most existing TWAS methods rely on gene expression alone and overlook other regulatory mechanisms of gene expression, including DNA methylation and splicing, that contribute to the genetic basis of these complex traits and diseases. Here we introduce a multi-omics method that integrates gene expression, DNA methylation, and splicing data to improve the identification of associated genes with our traits of interest. The accumulation of amyloid plaques and neurofibrillary tangles are pathological hallmarks of Alzheimer's disease (AD). While amyloid-beta propagation through prion-like mechanisms has been extensively studied in both central and peripheral pathways, the potential spreading of tau aggregates in the periphery remains largely unexplored. Emerging evidence suggests that hyperphosphorylated tau (ptau) aggregates may propagate beyond the central nervous system, as they have been detected in peripheral tissues and biological fluids from humans and mouse models of tauopathies. Background: Magnetic resonance (MR) guided radiation therapy combines high-resolution image capabilities of MRI with the precise targeting of radiation therapy. However, MRI does not provide the essential electron density information for accurate dose calculation, which limit the application of MRI. In this presented work, we evaluated the potential for Deep Learning (DL) based synthetic CT (sCT) generation using 3D MRI setup scans acquired during real-time adaptive MRI-guided radiation therapy.

Background: Inflammatory breast cancer (IBC) is rare but aggressive, characterized by the rapid onset of diffuse skin erythema, edema, tenderness, induration, and fast metastasis. Both hormone receptor positive (HR+) and human epidermal receptor 2 positive (HER2+) are associated with inferior response after neoadjuvant systemic therapy (NAST) compared to non-IBC. Despite multimodal treatment approaches, the 3-year overall survival rate for stage III IBC remains low at approximately 74%, around 10% lower than the 83% for non-IBC.

Despite tremendous advancements in multiple myeloma (MM) therapeutics, outcomes remain heterogeneous, heavily influenced by clinical and cytogenetic factors. Among these, deletion of the short arm of chromosome 17 (del(17p)) is a strong predictor of poor prognosis. The aim of this study was to evaluate real-world outcomes in patients with newly diagnosed MM (NDMM) with del(17p) undergoing upfront autologous hematopoietic stem cell transplantation (auto-HCT). How performance informs budgeting and budgets is contentious and unclear. Applying punctuated equilibrium theory, this research seeks to understand if performance information once placed on the media and macropolitical agendas can lead to long term budgetary shifts. This research highlights how the accumulation of performance information punctuated the funding scheme and created a new equilibrium over time.

The “memory wars” resurgence has renewed debates over the validity of repressed memories. This revival appears linked to factors such as changing statutes of limitations, confusion about repression, and unchallenged social media content. In a nationally representative online survey of American adults (N = 1581), we examined (a) beliefs in repression and the body keeps the score, (b) the prevalence of recovered memory claims, and (c) the impact of question phrasing on recovered memory reporting. This exploratory study compares the output of the free versions of six generative artificial intelligence (AI) chatbots—ChatGPT, Gemini, Copilot, Claude, Perplexity and HuggingChat. Five questions were submitted to all of the chatbots and examined according to the length of the response, tone of the response, and any additional features provided by the chatbots such as citations, web links, and helpful follow-up questions.
