Targeting Mertk On Tumour Cells And Macrophages: A Potential Intervention For Sporadic And Nf2-Related Meningioma And Schwannoma Tumours, Kevin Herrera Keane Jan 2025

Targeting Mertk On Tumour Cells And Macrophages: A Potential Intervention For Sporadic And Nf2-Related Meningioma And Schwannoma Tumours, Kevin Herrera Keane

Peninsula Medical School Theses

Although schwannomas and meningiomas are generally benign and slow-growing, the mass effect of either tumour within the nervous system causes life-altering symptoms. Individuals with NF2-related schwannomatosis may develop schwannomas and meningiomas, increasing tumour burden. The primary treatment for large or symptomatic schwannomas and meningiomas is surgical resection, which may not be suitable in cases involving tumours located near sensitive neuroanatomy or clinically vulnerable individuals. Not all schwannomas and meningiomas respond to alternative targeted treatments such as stereotactic radiosurgery or the vascular endothelial growth factor inhibitor bevacizumab. The main objective of this thesis was to investigate targeting of the receptor tyrosine …

Dred Scott, Military Enslavement, And The Case For Reparations, Hugh Barrett Mcclean Jan 2025

Dred Scott, Military Enslavement, And The Case For Reparations, Hugh Barrett Mcclean

All Faculty Scholarship

For more than a decade, Dred Scott was enslaved by a U.S. Army officer in a military system that perpetuated the institution of slavery in the free states and Northern territories of the antebellum period. Scott’s life, his lawsuit for freedom, and the U.S. Supreme Court case holding that Black people have “no rights which the white man was bound to respect” became infamous. But the history of the Dred Scott case and the military’s use of enslaved persons is largely unknown. Drawing on reparations advocacy in the Rosewood and Tulsa race massacre cases and other historical precedent for reparations, …

Civil & Environmental Engineering Faculty Publications

Desertification remains a critical global ecological and environmental challenge that threatens sustainable development. Although our understanding of desertification dynamics and their underlying drivers has improved, continued research is needed due to the region-specific nature of these processes. This study focuses on the Western Inner Mongolia Plateau in China as a case study to examine the evolution of desertification and its driving factors using a multifaceted approach, including the Mediterranean Desertification and Land Use (MEDALUS) model. Results show that the desertification sensitivity index (DSI) across the plateau ranged from 1.12 in prairie regions to 1.87 in desert areas, with a spatial …

The undersigned are scientists with expertise in per- and polyfluoroalkyl substances (PFASs) and/or the management of chemicals. We assert that the Organization for Economic Co-operation and Development (OECD) definition of PFASs is scientifically grounded, unambiguous, and well suited to identify these chemicals. We are concerned that some individuals and organizations are seeking a redefinition of PFASs endorsed by the International Union of Pure and Applied Chemistry (IUPAC) to exclude certain fluorinated chemical subgroups from the scope of the existing definition. We are concerned that this effort is politically and/or economically, rather than scientifically, motivated. An IUPAC-endorsed and potentially narrower PFAS …

Bioaccumulation Of Legacy And Novel Pfas In The Environment, Hesham Taher, Rainer Lohmann Jan 2025

Bioaccumulation Of Legacy And Novel Pfas In The Environment, Hesham Taher, Rainer Lohmann

Graduate School of Oceanography Faculty Publications

The bioaccumulation of per- and polyfluoroalkyl substances (PFAS), both legacy and novel, in the environment presents significant ecological and health risks. PFAS are a diverse group of synthetic chemicals known for their persistence and bioaccumulation, which can cause widespread environmental contamination and health risks. Strong carbon-fluorine bonds give these compounds unparalleled stability, preventing them from degrading and enabling them to endure in a range of environmental matrices, including water, soil, and biota. Legacy PFAS, including perfluorooctanoic acid (PFOA) and perfluorooctane sulfonic acid (PFOS), have been extensively studied and regulated, resulting in lower concentrations in some environmental media. However, novel PFAS, …

Furthering The Capabilities Of Diffusive-Gradient Passive Samplers For Per- And Polyfluoroalkyl Substances, Jarod Snook, Jitka Becanova, Simon Vojta, Rainer Lohmann

Graduate School of Oceanography Faculty Publications

Per- and polyfluoroalkyl substances (PFAS) are chemical pollutants of growing concern for many stakeholders. Due to their ubiquity, persistence in the environment, and potential for toxicity at low environmental concentrations, it is necessary to have convenient and reliable methods to measure PFAS in natural waters. Passive sampling methods (in situ preconcentration of PFAS) may be suitable for monitoring situations. One passive sampling design successfully employed for other, well studied contaminants (e.g., methylmercury) is the diffusive gradient in thin film sampler (DGT). However, the application of DGT for PFAS requires development and validation. Here, we iterate on previous PFAS-DGT studies by …

The concept of regulatory capture has been extensively studied in academic literature, primarily within the social sciences. This phenomenon has been increasingly discussed in the environmental sciences as the impacts of regulatory capture on human and ecosystem health have become increasingly apparent. Regulatory capture is just one tactic employed by vested interests in the strategy of delaying, weakening, or abolishing policies designed to protect the public interest. Here, we define capture strategies as ‘the act of influencing individuals, organizations, or governments to prioritize corporate interests over those of human and ecosystem health’. Similar to the evolution of terms like whitewashing …

Achievements And Challenges With Equilibrium And Kinetic Passive Sampling Of Hydrophobic And Hydrophilic Organic Compounds In Surface Waters, Kees Booij, Rachel Mackie, Branislav Vrana, Rainer Lohmann, Sarit L. Kaserzon Jan 2025

Achievements And Challenges With Equilibrium And Kinetic Passive Sampling Of Hydrophobic And Hydrophilic Organic Compounds In Surface Waters, Kees Booij, Rachel Mackie, Branislav Vrana, Rainer Lohmann, Sarit L. Kaserzon

Graduate School of Oceanography Faculty Publications

Passive sampling in surface waters is an important method in the monitoring and risk assessment of hydrophobic and hydrophilic organic chemicals. Sampler designs can be optimized for fast equilibrium attainment (equilibrium sampling) or improved time-integrative capability (kinetic sampling). We argue that both equilibrium and kinetic sampling can be applied when aqueous concentrations do not vary with time, whereas kinetic sampling also yields useful results for time-variable concentrations that are often observed with hydrophilic compounds in surface waters. We show that these methods have similar accuracy in principle but that their dominant error sources are different: sampler-water sorption coefficients for equilibrium …

Per- and polyfluoroalkyl substances (PFAS) are persistent contaminants with global environmental and health implications. This study evaluated PFAS concentrations in drinking water, wastewater, and surface water in Reykjavik, Iceland, focusing on seasonal variability and potential pollution sources. Thirty-three samples from groundwater, wastewater, and surface water were analysed for up to 54 PFAS. Results reveal that PFAS concentrations in Reykjavik's drinking water were minimal, which most compounds below detection limits, and the sum of 18 PFAS below 0.5 ng L−1. These levels were significantly below EU Drinking Water Directive and European Food Safety Authority health limits, indicating effective source …

Black carbon (BC), the most recalcitrant part of the pyrogenic carbon continuum, is formed by the incomplete combustion of biomass and fossil fuels. Methods for detecting BC include the chemical degradation of condensed aromatic compounds into benzenepolycarboxylic acids (BPCA), chemothermal oxidation of organic carbon at 375°C (CTO), 13C nuclear magnetic resonance combined with a molecular mixing model, thermogravimetry-differential scanning calorimetry, and the use of polycyclic aromatic hydrocarbons as tracers. However, there is limited knowledge about the comparability of these methods in marine sediments and their suitability as wildfire proxies. Here, we examined a sediment core from the Congo River …

Atmospheric Concentrations Of Per- And Polyfluoroalkyl Substances In Michigan's Ambient Air Using Passive Sampling, Melissa Woodward, Jitka Becanova, Simon Vojta, Rainer Lohmann

Graduate School of Oceanography Faculty Publications

As a part of Michigan's efforts to identify and address per- and polyfluoroalkyl substances (PFAS) contamination statewide, more knowledge of atmospheric PFAS concentrations is needed to better understand their atmospheric transport and impact on other environmental media. This research aimed to measure atmospheric concentrations of PFAS in Michigan's ambient air using low-cost and easy-to-use passive samplers and identify relationships with environmental factors. Passive samplers, consisting of polyurethane foam (PUF) discs and sorbent-filled polyethylene tubes (radiello-XAD samplers), were deployed for a month at 27 sites across Michigan and analyzed for both ionic and volatile, neutral PFAS. Short chain perfluorocarboxylic acids (PFCAs), …

Graduate School of Oceanography Faculty Publications

Per- and polyfluoroalkyl substances (PFAS) are a group of compounds of high concern due to their ubiquity, persistence, and adverse health impacts. With a diversity of chemical structures and properties, detection tools are needed to capture as many PFAS as possible. In this study, a microporous polyethylene tube (MPT) passive sampler was calibrated for 25 target compounds, 8 suspect PFAS, and extractable organofluorine (EOF) during 1–2 week deployments in groundwater, freshwater river, and estuary contaminated by aqueous film-forming foam (AFFF). Targeted analysis, suspect screening, and EOF were performed on passive and grab samples to derive sampling rates, Rs. …

Characterization Of The Potential Long-Term Impact From Sedimentary Pfas At A Historically Contaminated Textile Waste Site, Jarod Snook, Jitka Becanova, Simon Vojta, Rainer Lohmann

Graduate School of Oceanography Faculty Publications

Per- and polyfluoroalkyl substances (PFAS) are pervasive pollutants at historically contaminated sites throughout the United States and beyond. Two such sites in Rhode Island, USA, are textile-mill-associated waste retention ponds known to introduce PFAS contamination to the adjacent river, estuary, and eventually the Atlantic Ocean. Here, we thoroughly investigated the retention ponds as a long-term source of PFAS via water passive sampling, sediment coring, and laboratory-derived partitioning coefficients, Kd, with field sediment and water. Additional studies were performed to assess the mobility and estimate the mass fluxes of PFAS from sediment to water. Retention pond 1 was more …

Resort Enhances Reference-Based Cell Type Deconvolution For Spatial Transcriptomics Through Regional Information Integration, Linhua Wang, Ling Wu, Guantong Qi, Chaozhong Liu, Wanli Wang, Xiang H-F Zhang, Zhandong Liu

Duncan NRI Faculty and Staff Publications

Motivation: Spatial transcriptomics (ST) captures positional gene expression within tissues but lacks single-cell resolution. Reference-based cell type deconvolution methods were developed to understand cell type distributions for ST. However, batch/platform discrepancies between references and ST impact their accuracy.

Results: We present Region-based Cell Sorting (ReSort), which utilizes ST's region-level data to lessen reliance on reference data and alleviate these technical issues. In simulation studies, ReSort enhances reference-based deconvolution methods. Applying ReSort to a mouse breast cancer model highlights macrophages M0 and M2 enrichment in the epithelial clone, revealing insights into epithelial-mesenchymal transition and immune infiltration.

Peroxisomal Biogenesis Disorders Zellweger Spectrum (PBD-ZSD) disorders are a group of autosomal recessive defects in peroxisome formation that produce a multi-systemic disease presenting at birth or in childhood. Well documented clinical biomarkers such as elevated very long chain fatty acids (VLCFA) are key biochemical diagnostic findings in these conditions. Additional, secondary biochemical alterations such as elevated very long chain lysophosphatidylcholines are allowing newborn screening for peroxisomal disease. In addition, a more widespread impact on metabolism and lipids is increasingly being documented by metabolomic and lipidomic studies. Here we utilize Drosophila models of pex2 and pex16 as well as human plasma …

Leadership—whether as a parent, healthcare provider, soldier, or business executive—requires intentional effort to master its art and science. Just as we would not expect a physician or a soldier to learn their craft entirely “on the job,” we should not expect leaders to develop their leadership skills the same way. Society demands years of preparation for physicians and soldiers before they assume their professional responsibilities. However, when it comes to leadership, we often accept that individuals take on leadership roles with neither formal leadership training nor the “education of experience,” despite the profound and life-changing consequences of good and bad …
