This thesis, The Space Between: Engaging and Withholding the Self, documents my journey in understanding the self through the act of oil-painted self-portraiture. Through an exploration of eye contact, body language, non-performative representations of self, and the tension between engagement and detachment, this body of work examines the complexities of visibility, agency, and self-perception. Using my own body as both subject and metaphor, I investigate the interplay between revealing and withholding, questioning the boundaries between what is seen and what remains obscured. In examining Kate Chopin’s The Awakening alongside anthropologic theories of liminality, this paper examines the becoming women traverse as they transition between various social roles: girl, woman, mother. Edna is a maternal character attempting to unweave herself from her roles as mother and wife, and she finds this desire to go backwards to the freedom she experienced as a girl is proven to be impossible. The unmooring of her psyche and her subsequent suicide is indicative of a failure, not of her own accord, but in the social preparation and support she receives within her new identity. This research explores the potential of a cloud-native multi-user computer-aided design (MUCAD) system to improve communication and efficiency during engineering design reviews and shorten the design timeline for military tools. First, a literature review is used to generate a list of thirty-five collaboration-oriented requirements that aim to ensure concurrency, annotations, heterogeneity, security, IP protection, version control, and viewing within a MUCAD system. A comparative evaluation of existing collaborative CAD system architectures, including cloud-native, cloud-enabled, and a single-user system with external file sharing, is conducted to determine how well each system architecture fulfills the generated collaboration-oriented requirements. The underrepresentation of Black women notably impacts the landscape of educational leadership as a result of a predominantly White workforce within the teaching profession (N. N. Johnson & Fournillier, 2021). This inequity not only highlights systemic inefficiencies as evidenced by worsening educational experiences for African American students following Brown v. BOE but also underscores the silence within studies exploring the experiences of Black women faculty in instructional leadership roles (Brown, 2005; Foster, 2005; N. N. Johnson, 2024; N. N. The first chapter examines the role of return migration in driving innovation within India’s tech startup ecosystem, using a novel dataset constructed from web scraping of startup profiles, LinkedIn data, and patent databases. By capturing founders’ international experience, education, and innovation outcomes, the analysis reveals that returnees significantly contribute to higher inno- vation levels, particularly through patent filings. Returnee-founded startups exhibit an 18% higher rate of patent filings compared to those led by locals. The study employs OLS, Poisson regression, Coarsened Exact Matching (CEM), and Instrumental Variables (IV) to address selection bias and endogeneity concerns. The sacral knot, a term first coined by Sir Arthur Evans, is an iconic symbol in Minoan iconography, but there is surprisingly little information about it. Various interpretations have been suggested regarding the meaning of this knot, the most popular and well-known being a religious function.There is no doubt that the so-called "sacral knot” had an important meaning in Bronze Age Greece, with a number of depictions showing the knot alongside other significant symbols, such as bulls, figure-of-eight shields, and lions. Heterotopic ossifications (HOs) are the pathologic process by which bone inappropriately forms outside of the skeletal system. Despite HOs being a persistent clinical problem in the general population, there are no definitive strategies for their prevention and treatment due to a limited understanding of the cellular and molecular mechanisms contributing to lesion development. One disease in which the development of heterotopic subcutaneous ossifications (SCOs) leads to morbidity is Albright hereditary osteodystrophy (AHO). Albright hereditary osteodystrophy is caused by heterozygous inactivation of GNAS, the gene that encodes the α-stimulatory subunit (Gαs ) of G proteins. Previously, we had shown using our …

The evolution of isocitrate dehydrogenase (IDH)-wildtype glioblastoma (GBM) after standard-of-care therapy remains poorly understood. Here we analyzed matched primary and recurrent GBMs from 59 patients using single-nucleus RNA sequencing and bulk DNA sequencing, assessing the longitudinal evolution of the GBM ecosystem across layers of cellular and molecular heterogeneity. The most consistent change was a lower malignant cell fraction at recurrence and a reciprocal increase in glial and neuronal cell types in the tumor microenvironment (TME). The most dynamic and repetitive regions of great ape genomes have traditionally been excluded from comparative studies. Consequently, our understanding of the evolution of our species is incomplete. Here we present haplotype-resolved reference genomes and comparative analyses of six ape species: chimpanzee, bonobo, gorilla, Bornean orangutan, Sumatran orangutan and siamang. We achieve chromosome-level contiguity with substantial sequence accuracy (< 1 error in 2.7 megabases) and completely sequence 215 gapless chromosomes telomere-to-telomere. We resolve challenging regions, such as the major histocompatibility complex and immunoglobulin loci, to provide in-depth evolutionary insights. Comparative analyses enabled investigations of the evolution and diversity of regions previously uncharacterized or incompletely studied without bias from mapping to the human reference genome. Such regions include newly minted gene families in lineage-specific segmental duplications, centromeric DNA, acrocentric chromosomes and subterminal heterochromatin. This resource serves as a comprehensive baseline for future evolutionary studies of humans and our closest living ape relatives.

Haematopoietic stem cells maintain blood production throughout life1 . Although extensively characterized using the laboratory mouse, little is known about clonal selection and population dynamics of the haematopoietic stem cell pool during murine ageing. We isolated stem cells and progenitors from young and old mice, identifying 221,890 somatic mutations genome-wide in 1,845 single-cell-derived colonies. Mouse stem cells and progenitors accrue approximately 45 somatic mutations per year, a rate only approximately threefold greater than human progenitors despite the vastly different organismal sizes and lifespans. PURPOSE: This study investigates genes contributing to late-adult corneal dystrophies (LACDs) in aged mice, with potential implications for late-onset corneal dystrophies (CDs) in humans.
METHODS: The International Mouse Phenotyping Consortium (IMPC) database, containing data from 8901 knockout mouse lines, was filtered to include late-adult mice (49+ weeks) with significant (P < 0.0001) CD phenotypes. Candidate genes were mapped to human orthologs using the Mouse Genome Informatics group, with expression analyzed via PLAE and a literature review for prior CD associations. Comparative analyses of LACD genes from IMPC and established human CD genes from IC3D included protein interactions (STRING), biological processes (PANTHER), and molecular pathways (KEGG).
RESULTS: Analysis identified 14 genes linked to late-adult abnormal corneal phenotypes. Of these, 2 genes were previously associated with CDs in humans, while 12 were novel. Seven of the 14 genes (50%) were expressed in the human cornea based on single-cell transcriptomics. Protein-protein interactions via STRING showed several significant interactions …

A milestone in sickle cell disease (SCD) therapeutics was achieved in December 2023 with the FDA-approved gene therapy for patients aged 12 years and older. However, these therapies may only suit a fraction of patients because of cost or health risks. A better understanding of SCD outcome heterogeneity is needed to propose patient-specific pharmacological interventions. To achieve this, humanized and genetically diverse mouse models are essential for associating candidate genotypes with specific hematological traits, organ function, and disease resilience. Although vaccines are usually given intramuscularly, the intranasal delivery route may lead to better mucosal protection and limit the spread of respiratory virus while easing administration and improving vaccine acceptance. The challenge, however, is to achieve delivery across the selective epithelial cell barrier. Here we report on a subunit vaccine platform, in which the antigen is genetically fused to albumin to facilitate FcRn-mediated transport across the mucosal barrier in the presence of adjuvant. Intranasal delivery in conventional and transgenic mouse models induces both systemic and mucosal antigen-specific antibody responses that protect against challenge with SARS-CoV-2 or influenza A. Mitochondrial translation release factor in rescue (MTRFR) catalyzes a termination step in protein synthesis, facilitating release of the nascent chain from mitoribosomes. Pathogenic variants in MTRFR cause MTRFR deficiency and are loss-of-function variants. Here, we tested gene replacement as a possible therapeutic strategy. A truncating mutation (K155*) was generated in mice; however, homozygotes die embryonically whereas mice heterozygous for this K155* allele are normal. We also generated transgenic strains expressing either wild-type human MTRFR or a partially functional MTRFR. Despite dose-dependent phenotypes from overexpression in vitro, neither transgene caused adverse effects in vivo. In K155* homozygous mice, the wild-type MTRFR …

Social media, particularly TikTok, has become a major influence on pre-service teachers (PSTs), shaping their professional identity, pedagogical expectations, and instructional decision-making. While TikTok offers networking and inspiration, its algorithm-driven content prioritizes entertainment over evidence-based pedagogy, reinforcing idealized, oversimplified, and commercialized portrayals of teaching. Many PSTs consume and apply TikTok-derived strategies without critical evaluation, contributing to the casualization and deprofessionalization of the profession. The research focused on addressing equity and communication challenges in co-teaching through the implementation of a Professional Learning Community (PLC) and peer mentoring within a suburban middle school. The study took place in a predominantly white, lower-middle-class suburban school that utilized co-teaching across various subjects to provide students with disabilities greater access to the general education curriculum in the least restrictive environment. Some current building codes for structural concrete allow the use of alternative cement concrete if their structural, durability, and fire performance can be shown to be comparable to portland cement (PC) concrete. One such alternative cement, belitic calcium sulfoaluminate (BCSA) cement, is a promising standalone replacement for portland cement given its lower global warming potential (GWP), fast setting time, and high early age compressive strength. Much research has allowed for in-depth analyses of student engagement, a growing issue in education, without uncovering a solution. The purpose of this study was to analyze the relationship between school culture and student engagement, a connection not yet studied, by using two valid and reliable instruments, the School Culture Survey and Instructional Practices Inventory among Indiana Principal Leadership Institute elementary schools. Data were gathered quantitatively, as well as qualitatively, to determine that there is no statistically significant relationship between culture and engagement. Optic disc edema, which may arise from a range of inflammatory, ischemic, compressive, infiltrative, or hereditary causes, can also indicate elevated intracranial pressure in the form of papilledema. Given its potential association with life-threatening conditions such as mass lesions or venous sinus thrombosis, timely identification and thorough evaluation are critical. In this case, a 61-year-old female presented with a complaint of a new-onset floater in her left eye. Despite minimal symptoms and an unremarkable systemic review, dilated fundus examination revealed bilateral optic disc edema with hyperemia and hemorrhages, prompting urgent neuroimaging.
