The classic social contract theorized by Hobbes, Rousseau, and Locke advocates for establishing the state based on individual freedom; for them, a contractual agreement rather than nature serves as the foundation for the state. In his Philosophy of Rights, Hegel rejects this classic view of the social contract, which sees freedom and consent as the basis of the state. While the perspectives of the classic social contract and Hegel yield differing interpretations of freedom, the dissertation explores their relationship against contemporary challenges to freedom. Advanced Care Planning (ACP) is a process used to identify or update a specific patient’s preferences of care and treatment in the event that they lack the capacity to do so. ACP is especially important for the aging population and those living with chronic conditions. Currently 11% of the world’s population is above the age of 60 and this number is projected to climb to 22% by 2050. The effects of aging are even greater in rural areas, such as South Dakota, where there is a disproportionate number of older and sicker people.

Recent legislation in Illinois schools has focused on enhancing cardiac preparation in schools by establishing written emergency plans that are to be disseminated to employees within thirty days from the start of the school year. This web-based education tool was created as a centralized hub for employees to locate pertinent information regarding cardiac emergencies. The intent is to increase cardiac emergency preparedness in schools with access to evidence-based resources and content. This data analysis examined associations between cardiovascular and muscular fitness indicators and the presence of cardiovascular disease (CVD) in older adult athletes. Using cross-sectional data from 1,110 participants aged 50 and older who completed the Sustained Athlete Fitness Exam (SAFE) at state-level Senior Games events, we evaluated both objective performance metrics and self-reported exercise behaviors. Cardiovascular measures included systolic blood pressure (SBP), waist circumference (WC), waist-to-hip ratio (WHR), and body mass index (BMI). Tens of millions of Americans suffer from metabolic disorders, predisposing patients to cardiovascular disease, kidney disease, liver disease, infertility, and cancer. Unfortunately, the etiology of many of these diseases remains a mystery, limiting our ability to identify targets for pharmacologic therapies. Due to the relationship between altered lipid dynamics and metabolic dysfunction, we decided to study the effect of the posttranslational lipid modification, S-palmitoylation, on metabolism. S-palmitoylation is the only known reversible lipid modification for proteins, and this depalmitoylation process is catalyzed by acyl-protein thioesterases including APT1 and APT2. Achieving flexible and efficient communication in free space requires compact, robust optical components capable of dynamically tuning laser light without relying on bulky lenses or mechanically driven mirrors. Of the protein structures deposited in the Protein Data Bank, less than half have pockets suitable for the binding of drugs. Even when proteins contain pockets in their ground state structures (e.g., the nucleotide-binding active site in myosin motors), achieving specificity remains a central challenge in drug design as many protein families share common structural motifs. Cryptic pockets are cavities absent in ligand-free experimental structures that form due to protein fluctuations in solution. This research aimed to evaluate the effectiveness of a two-and-a-half hour training session for master's students in counselor education. The training focused on using mindfulness to enhance spiritual and religious counseling competencies. Utilizing a pre-and post-test design, the quantitative study employed mindfulness to train master's students with the aim of building and fostering spiritual and religious competencies in counseling. Biological systems must navigate dynamic and complex environments while operating under constraints such as limited molecular resources, biochemical noise, and structural trade-offs. This dissertation investigates three distinct cases in which biological systems allocate resources and process information efficiently, revealing emergent simplicity from underlying complexity. The first study develops a generalized end-product inhibition model with cross-talk and an excess of regulators, providing a proof-of-principle that simple biochemical circuits could, in principle, dynamically adjust their responsiveness to high-dimensional environmental variation. Transposable elements (TEs) contribute to nearly half of the human genome yet have been overlooked in genomic medicines due to the stereotypic impression as “junk DNA” and technical challenges. However, studies in the last two decades demonstrated that dysregulation of the epigenome in cancer cells resurrects the cis-regulatory functions of TEs, including the promoter activities. Infant leukemias arise as either B cell acute lymphoblastic leukemia (B-ALL) or acute myeloid leukemia (AML) and are primarily driven by MLL/KMT2A-rearrangements (MLLr). Prior work has shown that MLLr transform fetal/neonatal hematopoietic progenitors more efficiently than adult progenitors. However, the genetics and epidemiology of MLLr leukemias present an important paradox. MLLr arise during late gestation under conditions that would seemingly favor leukemic transformation (i.e., fetal/neonatal progenitors have high proliferation rates and self-renewal). Furthermore, MLLr infant leukemias require very few cooperating mutations for transformation. Yet congenital leukemias are 10-fold less common than infant leukemias and >100-fold less common than childhood leukemias …

Opioid use disorder is an urgent public health crisis in the United States. Over 800,000 Americans have died from opioid-related overdoses since 2000. Nearly 30% of people who misuse opioids begin with prescription oral opioids, such as oxycodone, and continue misusing to avoid the immediate aversive state of withdrawal and the persistent dysphoria associated with protracted abstinence from opioids. The persistency of these dysphoric symptoms increases the risk of relapse even after long periods of abstinence. Treatment for opioid use disorder currently consists of pharmacological opioid treatment to reduce withdrawal symptoms and behavioral counseling. Deciphering the “genotype to phenotype” map is a crucial goal of biology. We will not improve our ability to predict phenotypes from DNA sequences until we better incorporate epigenetic and environmental variation into our models of human health and disease. Genes are typically presumed to express both parental alleles equally, but genetic and epigenetic factors can cause one allele to function differently than the other (e.g. expressed or silenced more). These allele-specific imbalances can alter the final composition of a gene’s transcriptional product and ultimately shape phenotypes. Natural Killer (NK) cells are cytotoxic innate lymphoid cells that play an important role in the surveillance and elimination of virally-infected and malignantly-transformed cells. These primary effector functions are achieved by producing cytokines and chemokines which activate and recruit additional immune effectors, as well as direct cytotoxic killing via perforin and granzymes. NK cell function is tightly regulated by a balance of germ-line encoded activating, inhibitory, co-stimulatory, and cytokine receptors expressed at the cell surface. The therapeutic landscape in ALL has changed dramatically over the last decade. Allogeneic blood or marrow transplantation (AlloBMT) has also evolved and remains an important option for consolidation. We assessed the interplay between these factors by analyzing the outcomes of 251 adult ALL (214 B and 37 T ALL) patients undergoing alloBMT with post-transplantation cyclophosphamide (PTCy) across two eras: 2008-2014 (ERA1) and 2015-2022 (ERA2). ERA1 patients were younger (median age 45.5 vs. 50, p=0.03), less likely to have an HCT-CI ≥4 (9% vs. 21%, p=0.01), more likely to have MRD by flow cytometry (20% vs. 9%, p=0.01) and receive myeloablative …

24-hour biological rhythms are essential to maintain physiological homeostasis. Disruption of these rhythms increases the risks of multiple diseases. Biological rhythms are known to have a genetic basis formed by core clock genes, but how individual genetic variation shapes the oscillating transcriptome and contributes to human chronophysiology and disease risk is largely unknown. Here, we mapped interactions between temporal gene expression and genotype to identify quantitative trait loci (QTLs) contributing to rhythmic gene expression. These newly identified QTLs were termed as rhythmic QTLs (rhyQTLs), which determine previously unappreciated rhythmic genes in human subpopulations with specific genotypes. Background: Patients with spinal cord injury (SCI) who develop urinary tract infection (UTI) present differently than the non-SCI population. UTIs can cause loss of quality of life and even lead to life-threatening complications including urosepsis. Challenges in SCI management include distinguishing symptomatic UTI from asymptomatic bacteriuria (ASB), which occurs often in patients with SCI, and the lack of standardization in UTI diagnosis in SCI.

This three-paper dissertation explores law enforcement phlebotomy, the ability of law enforcement officers to draw blood. Phlebotomy is both a ubiquitous, invasive diagnostic tool as well as a social site with complex relational dynamics at play between phlebotomist and patient. Through law enforcement phlebotomy, the blood draw is co-opted from the medical field into the policing field, and the normative framework through which it is used changes: whereas medical edicts instruct providers to work with patients through an ethics of care--respecting consent and refusal--policing is not similarly bound, instead operating through a prioritization of security and efficiency. The central High Cascades graben in Oregon has been a center for mafic volcanism since its initiation. Three Fingered Jack is one of the larger mafic composite cones in the graben, located 20km south of Mount Jefferson. Three Fingered Jack erupted between 142.1 - 150.8ka, based on Ar40/Ar39 age dates. This study provides stratigraphic, compositional and petrographic evidence to understand the eruptive history of Three Fingered Jack and the processes involved in generating compositionally diverse basaltic andesites here.

After purchasing the building, the landlord moved for summary judgment, claiming a lawful high-rent deregulation occurred in 2009. The tenant countered that the apartment had been used commercially by a prior tenant, disqualifying it from deregulation. The motion court had granted the landlord summary judgment and ejectment. Situation selection is a form of emotion regulation where people approach or avoid certain things, places, or objects to proactively regulate their emotions. Though it is the only strategy that occurs before encountering emotion-eliciting events, situation selection remains understudied. The present research proposes a new measure of Day Recollection Situation Selection (DRSS) in which participants are prompted to recall the day's events before reporting their approach and avoidance behaviors. Even though sexist comments and behavior occur frequently in everyday life, people do not often confront the individuals who are responsible. However, interpersonal bias confrontations can be powerful tools for reducing expressions of bias and norm setting. The current studies tested an intervention designed to increase confrontations by harnessing theory and research on what holds people back from confronting. We designed a half-hour intervention consisting of engaging and evidence-based videos on confronting bias and self-reflection exercises to increase personal relevance. Recent advances in condensed matter physics have revealed a wide range of novel quantum phenomena in two-dimensional (2D) materials. The discovery of unique topological, magnetic, and optical properties in solids has greatly expanded our understanding of quantum phases in electronic structures. As more material systems are proposed theoretically and realized experimentally, our research focuses on uncovering and explaining these phenomena through effective models and first-principles calculations. First, we explore topological mosaic patterns in moiré superlattices—structures composed of periodic, spatially distinct domains, each with different topological properties.
