More than 57 million people globally suffer from neurodegenerative diseases, a figure expected to double every 20 years. Despite this growing burden, there are currently no cures, and treatment options remain limited due to disease heterogeneity, prolonged preclinical and prodromal phases, poor understanding of disease mechanisms, and diagnostic challenges. Identifying novel biomarkers is crucial for improving early detection, prognosis, staging and subtyping of these conditions. High-dimensional molecular studies in biofluids ('omics') offer promise for scalable biomarker discovery, but challenges in assembling large, diverse datasets hinder progress. Bifidobacteria are beneficial saccharolytic microbes that are widely used as probiotics or in synbiotic formulations, yet individual responses to supplementation can vary with strain type, microbiota composition, diet and lifestyle, underscoring the need for strain-level insights into glycan metabolism. Here we reconstructed 68 pathways for the utilization of mono-, di-, oligo- and polysaccharides by analysing the distribution of 589 curated metabolic gene functions (catabolic enzymes, transporters and transcriptional regulators) across 3,083 non-redundant Bifidobacterium genomes of human origin. Thirty-eight predicted phenotypes were validated in vitro for 30 geographically diverse strains, supporting genomics-based predictions. This project addresses alternatives to fixed schedule local transportation service offerings to members of retirement communities. The project design measures the effects of the contrast between fixed schedule (Fixed) service and demand-responsive services (DRT) in retirement communities in Santa Clara County that are closely matched in demographic profiles and geographic location. The research team investigates the effects of scheduling alternatives in a diary format with a within-person variable of four recent trips. Mobile LiDAR systems are powerful tools that help us map roads and their surroundings in 3D with great speed and precision. The data provided by these systems support urban planning efforts, digital mapping, transportation infrastructure maintenance, and more. This report presents a comprehensive workflow for roadside asset extraction using Mobile Terrestrial Laser Scanning (MTLS) data, focusing on road lane detection, cross-section slope analysis, and point cloud classification. Roadside asset extraction is the identification and classification of roadside features like signs and poles. Considerable research addresses the dynamics between land use and public transportation, including the impacts of transit systems on urban development and vice versa. However, this research has underestimated the heterogeneity of contributing factors to urban developments near transit by property type: vacant lot and occupied property. That is, what drives development in vacant lots differs from what drives development in already-occupied property. To improve understanding of the impact of natural hazards on the U.S. railway system, this project developed a comprehensive, geospatially precise dataset that maps historical hazard events to specific rail segments. The project provides insight into when, where, and how much damage natural hazards have inflicted on rail infrastructure across the country. While a rail-specific hazard database did not previously exist, this research combines data from the Geological Survey (USGS) Flood Events Database, Hydrologic Unit Codes (HUC), the National Centers for Environmental Information (NCEI) from the National Oceanic and Atmospheric Administration (NOAA), and the National Weather Service (NWS) forecast zones. …

With age, clonal expansions occur pervasively across normal tissues yet only in rare instances lead to cancer, despite being driven by well-established cancer drivers. Characterization of the factors that influence clonal progression is needed to inform interventional approaches. Germline genetic variation influences cancer risk and shapes tumor mutational profile, but its influence on the mutational landscape of normal tissues is not well known. Here we studied the impact of germline genetic variation on clonal hematopoiesis (CH) in 731,835 individuals. BACKGROUND: Predicting mortality risk in neonatal intensive care units (NICUs) is challenging due to complex, variable clinical and physiological data. Machine learning (ML) offers potential for more accurate risk stratification.

OBJECTIVE: To compare the performance of various ML models in predicting NICU mortality using a team-based modeling competition.

METHODS: We conducted a modeling competition with five neonatologist-led teams applying ML techniques-logistic regression, CatBoost, neural networks, random forest, and XGBoost-to a shared dataset from over 6,000 NICU admissions. In the emergency department-initiated buprenorphine for opioid use disorder (EMBED) trial, a clinical decision support (CDS) tool had no effect on rates of buprenorphine initiation in emergency department (ED) patients with opioid use disorder. The Agency for Healthcare Research and Quality (AHRQ) recently released a CDS Performance Measure Inventory to guide data-driven CDS development and evaluation. OBJECTIVE: Use of the Heart Team has been the standard of care for the treatment of aortic valve disease; however, its efficacy has not been evaluated. We sought to analyze its impact using the transcatheter aortic valve replacement (TAVR) use ratio (number of TAVR/total aortic valve replacement [AVR] volume) on TAVR, surgical aortic valve replacement (SAVR), and overall AVR outcomes.

OBJECTIVE AND METHODS: This study aimed to evaluate Medicaid accessibility for ophthalmologists and optometrists in Florida. A cross-sectional telephone survey was conducted between February and March 2018 using a geographically uniform sample of 22 ophthalmologists and 80 optometrists registered with Medicaid. Providers were asked whether they accepted new Medicaid patients (adult and/or pediatric) and for the earliest available appointment. Categorical variables were compared using Fisher's exact test, and time to next appointment was analyzed using the Wilcoxon rank-sum test (α = 0.05).

AIMS: The PANORAMA-HF trial demonstrated significant N-terminal pro-B-type natriuretic peptide (NT-proBNP) reductions in paediatric patients with left ventricular systolic dysfunction with sacubitril/valsartan or enalapril treatment over 52 weeks. This post hoc analysis aims to correlate changes in NT-proBNP levels with clinical outcomes in PANORAMA-HF patients receiving either sacubitril/valsartan or enalapril. Additionally, NT-proBNP reductions in the paediatric population were compared with a subset of adult heart failure with reduced ejection fraction (HFrEF) patients from the PARADIGM-HF trial.

Background: Social prescribing link workers formed part of the Additional Roles Reimbursement Scheme introduced into primary care in England from 2019. Link workers assist patients experiencing issues affecting their health and well-being that are 'non-medical' (e.g. lack of social connections, financial difficulties and housing problems). They give patients space to consider these non-medical issues and, when relevant, connect them to support, often within the voluntary-community-social-enterprise sector. Schools commonly teach a history of mathematics and science that is Eurocentric. This selective version of the history of science, called Classical Narrative by some, is rooted in colonial times and mindset, and has reinforced certain negative opinions about other cultures. It ignores or downplays contributions to mathematics and sciences from non-western civilizations, and falsely attributes many scientific discoveries to European scholars. Medieval Islamic Civilization, which had strong connections to Renaissance Europe, serves as a clear example of how this narrative is distorted.
