There is an urgent need for high-throughput screening (HTS) models to replace, refine, and/or reduce (“3Rs”) vertebrate toxicity testing. Replacing in vivo animal studies is challenging for neurotoxicity and developmental neurotoxicity (DNT), where the functional relevancy of adverse outcomes needs to be assessed on the whole organism. We previously screened the NTP 87-compound library (NTP87), consisting of known and suspected developmental neurotoxicants, and showed that planarian HTS can identify known (developmental) neurotoxicants. BACKGROUND AND AIMS: The Doublecortin-like kinase-1 (DCLK1) plays a chemosensory role in the gut. It's role in the context of inflammatory diseases including inflammatory bowel disease (IBD), has not been thoroughly investigated. This study explored the role of the DCLK1 isoform (DCLK1-S) in promoting infectious/chemical colitis by utilizing high-throughput imaging mass cytometry (IMC).

METHODS: Transgenic mice were either infected with Citrobacter rodentium (CR) or received DSS and tissues/cells were processed via standard techniques. Abstract

This thesis traces the evolving symbolism of the Naga motif (a mythic water serpent) in Northeast Thailand through a framework I call meta-motion, which maps symbolic persistence of this cultural signifier and boundary marker across four cultural horizons—animist (Z), Hindu–Khmer (B), Buddhist (A), and modern/global (C). Like electrons in quantum super position, symbolic motifs circulate as overlapping “waves” of potential meaning and are concretized into cultural “particles” only when recruited for specific ritual, commercial, or political purposes. Objective: The real-world evidence on the association between glucagon-like peptide-1 receptor agonists (GLP-1RAs) and cancer risk remains limited and mixed.

Methods: In 2013-2020 national Medicare claims data, we included cancer-naïve patients with type 2 diabetes (T2D). We identified those who initiated GLP-1 RA, sodium-glucose cotransporter 2 inhibitor (SGLT2i), or dipeptidyl peptidase 4 inhibitor (DPP4i) and conducted 1:1 propensity score matching for confounding adjustment. Cox proportional hazards models were used to estimate hazard ratios (HR) of nine obesity-associated cancers (thyroid, pancreatic, bladder, colorectal, lung, kidney, breast, endometrial, and prostate cancer).

Tort liability for failure to “age gate” is a promising legal response to the public health hazards of AI, social media, sports gambling, and other digital spaces. Tort liability for failure to “age gate” hinges liability for harms to minors on an app’s failure to take reasonable steps to prevent minors from gaining access or otherwise to apply appropriate governance rules, such as privacy-protective default settings or ensuring genuine parental consent. Leveraging Sequences Missing From The Human Genome To Diagnose Cancer, Ilias Georgakopoulos-Soares, Christopher A Maher, Et Al.

BACKGROUND: Cancer diagnosis using cell-free DNA (cfDNA) has the potential to improve treatment and survival but has several technical limitations.

METHODS: In this study, we developed a prediction model based on neomers, DNA sequences 13-17 nucleotides in length that are predominantly absent from the genomes of healthy individuals and are created by tumor-associated mutations.

RESULTS: We show that neomer-based classifiers can accurately detect cancer, including early stages, and distinguish subtypes and features. Analysis of 2577 cancer genomes from 21 cancer types shows that neomers can distinguish tumor types with higher accuracy than state-of-the-art methods. Generation and analysis of 465 cfDNA …

Background: Bystanders have the potential to be a critical component of the Road Injury Chain of Survival, particularly in the early phases of injury recognition, scene assessment, and emergency service activation. Their potential role in trauma triage remains underexplored. This scoping review aims to examine the existing evidence of the use of bystanders in triage—both laypersons and non-clinical professionals—during the prehospital phase of road injury and other trauma scenarios. Synthetic Aperture Radar (SAR) technology offers unparalleled advantages by delivering high-quality images under all-weather conditions, enabling effective flood monitoring. This capability provides massive remote sensing data for flood mapping, while recent rapid advances in deep learning (DL) offer methodologies for large-scale flood mapping. Quantum computing is a promising tool for solving computational problems in several areas, including the simulation of physical and chemical systems. The design of practical quantum computing systems for these applications is a daunting task, requiring collaborative work by interdisciplinary teams considering different abstractions of the same systems. Background: Lymph node metastasis is a key driver of poor outcomes in cervical cancer. However, the molecular mechanisms of circular RNAs (circRNAs) driving cervical cancer lymph node metastasis remain unclear.

Methods: We identified circZFR, fatty acid synthase (FASN) and YTH N6-methyladenosine RNA binding protein F3 (YTHDF3) protein expression in the cervical cancer patients with long and short disease-free survival (DFS). Functional experiments were performed to investigate the function of circZFR, FASN and YTHDF3 on cell migration and invasion. The BRAF inhibitor encorafenib and anti-epidermal growth factor receptor (EGFR) antibody cetuximab modestly improve survival for patients with microsatellite stable (MSS) BRAFV600E metastatic colorectal cancer (mCRC), characterized by higher immune activation than MSS BRAFwild-type colorectal cancer (CRC). In this phase 1/2 study (NCT04017650) of 26 participants with MSS BRAFV600E mCRC who received encorafenib, cetuximab, and anti-PD-1 antibody nivolumab, we report an overall response rate of 50% (95% confidence interval [CI] 29-71) and median progression-free survival of 7.4 months (95% CI, 5.6-9.6). Cells interact as dynamically evolving ecosystems. While recent single-cell and spatial multi-omics technologies quantify individual cell characteristics, predicting their evolution requires mathematical modeling. We propose a conceptual framework-a cell behavior hypothesis grammar-that uses natural language statements (cell rules) to create mathematical models. This enables systematic integration of biological knowledge and multi-omics data to generate in silico models, enabling virtual "thought experiments" that test and expand our understanding of multicellular systems and generate new testable hypotheses. BAF (SWI/SNF) chromatin remodelers engage binding partners to generate site-specific DNA accessibility. However, the basis for interaction between BAF and divergent binding partners has remained unclear. Here, we tested the hypothesis that scaffold proteins augment BAF's binding repertoire by examining β-catenin (CTNNB1) and steroidogenic factor 1 (SF-1, NR5A1), a transcription factor central to steroid production in human cells. BAF inhibition rapidly opposed SF-1/β-catenin enhancer occupancy, impairing SF-1 target activation and SF-1/β-catenin autoregulation. These effects arise due to β-catenin's role as a molecular adapter between SF-1 and an intrinsically disordered region (IDR) of the canonical BAF (cBAF) subunit ARID1A. In contrast …

OBJECTIVES: We aimed to examine the association between patient age and procedural intervention, especially nephrectomy, in patients with renal trauma in the National Trauma Data Bank (NTDB).

MATERIALS AND METHODS: We queried the 2013-2020 NTDB for adult renal trauma patients with an American Association for the Surgery of Trauma (AAST) grade. Patients without AAST grade or with no sign of life were excluded. We constructed a multinomial logistic regression model to demonstrate the association between age and procedural interventions (renal angioembolization, renorrhaphy and nephrectomy). Models were adjusted for patient, hospital, and clinical factors.

Water conservation is a critical issue in Utah, and several actions are already underway to help lower residential landscape water use. Strategies include smart irrigation controllers, efficient sprinkler bodies and nozzles, drip irrigation systems, soil amendments that increase water-holding capacity, and drought-tolerant grass cultivars for lawns. While additional conservation efforts will be necessary to reach sustainable water management, statewide adoption of current programs can help reduce water use and loss.

Investigation Of A Stable And Saturated Reference Electrode Using Metal Chlorides And Oxides In Eutectic Licl-Kcl Molten Salts, Carlos Mejia, Devin Rappleye

Stable and reproducible reference electrodes (REs) are essential for electrochemical systems operating in high-temperature molten salts, where consistent control of potential is required. This study uses three different solutes to investigate the development and evaluation of REs in eutectic LiCl-KCl at 773 K. The solutes tested were NiCl2, AgCl, and NiO, each prepared at multiple concentrations to assess stability, reproducibility, and saturation. Electrochemical stability was measured over three days using chronopotentiometry (CP), followed by open circuit potential (OCP), with cyclic voltammetry (CV) also integrated to monitor changes in electrochemical behavior over time. Nanometer-scale titanomagnetite crystals have been detected in nominally aphyric rhyolite pumice, but whether they are numerous enough to impact bubble nucleation in explosive silicic volcanism was unresolved. This study examines sub-micron crystals using rock magnetic techniques, Rhyolite-MELTS modeling, and physical characterization. We analyzed pumice from four eruptions spanning wide ranges in intensity, storage depth, and bubble number density (1016 to 1013 m−3 liquid): 1060 CE Glass Mountain, 1912 CE Novarupta, 232 CE Taupo, and 0.45 Ma Pudahuel. Calculations assuming monospecific assemblages of 10 and 1,000 nm cubic particles yield titanomagnetite number densities of 1021 to 1013 m−3 dense rock equivalent, …
