Osteoarthritis is a prevalent joint disease in the aging population. The hallmark of osteoarthritis is the degeneration of the joint cartilage, characterized by changes in chondrocytes including mitochondrial dysfunction. However, the precise mechanisms of how this affects chondrocyte homeostasis and whether such processes can be explored as therapeutic targets for osteoarthritis remain unclear. Here, we show that impaired mitochondrial function and disrupted cartilage matrix metabolism due to loss of mitofusin-2 (MFN2) expression in chondrocytes leads to the development of osteoarthritis. A growing number of archaeologists are now demanding systemic disciplinary transformations that directly address how white supremacy and settler colonialism enact Indigenous dispossession and erasure as well as anti-Blackness, gender discrimination, and ableism. Follicular lymphoma (FL) has a clinical course that is often characterized by high response rates to first-line therapy, followed by multiple relapses over a prolonged natural history. Currently, there are multiple possible approaches to frontline therapy for untreated advanced-stage FL, but there is an ongoing debate around what is the preferred approach. In a labor context, the parties may disagree on the hourly wage, number of paid holidays, and fringe benefits as well as other conditions of employment. Background: In the phase 3 FRESCO (NCT02314819) and FRESCO-2 (NCT04322539) studies, fruquintinib vs placebo, plus best supportive care, significantly improved overall survival (OS) in patients with metastatic colorectal cancer (mCRC). These studies were conducted in temporally and geographically diverse populations that received distinct prior therapies; FRESCO patients were less pretreated than FRESCO-2 patients. This analysis assessed the efficacy and safety of fruquintinib in a less pretreated global population than the FRESCO-2 intention-to-treat (ITT) population.

In this nonpayment proceeding for a 3-unit building, the Civil Court of the City of New York granted the tenant's motion for a directed verdict, dismissing the petition. The landlord failed to establish a prima facie case by not providing a complete lease with rental amounts or a proper rent breakdown. Although the tenant testified to a $1,650 monthly rent, the rent demand sought $2,900, rendering it defective and non-amendable. The court also noted the landlord's violation of a discovery order to produce all necessary documents. The tenant's counterclaims were severed without prejudice.

Objective

This study aimed to explore associations between self-reported first-trimester vaginal bleeding status and fetal body composition and organ volumes measured by three-dimensional (3D) ultrasound across pregnancy.

Study Design

The NICHD Fetal 3D Study (2015–2019) included individuals with singleton pregnancies at low risk for fetal growth abnormalities (n = 2,634). Fetal body composition measures of arm, abdomen, and thigh, and organ volumes including cerebellum, lung, kidney, and liver, were measured up to five times between 15 and 40 weeks using 3D ultrasound. Methylmalonic acidemia (MMA) is a severe metabolic disorder affecting multiple organs because of a distal block in branched-chain amino acid (BCAA) catabolism. Standard of care is limited to protein restriction and supportive care during metabolic decompensation. Severe cases require liver/kidney transplantation, and there is a clear need for better therapy. Here, we investigated the effects of a small molecule branched-chain amino acid transaminase (BCAT) inhibitor in human MMA hepatocytes and an MMA mouse model. In mammals, sperm formation is completed in the seminiferous tubules within the testis, and sperm maturation occurs during the epididymal transit of the spermatozoa. Sperm morphology drastically changes when abnormal spermatozoa migrate from the testis to the epididymis. Detailed molecular mechanisms for sperm survival in the epididymis have not been determined yet. Globozoospermia is a cause of male infertility and is characterized by round-headed spermatozoa without acrosomes, an abnormal sperm nuclear membrane, and sperm midpiece defects. Several prediction scores have been developed to guide success during spontaneous breathing trials to help with weaning decisions. Despite widespread concern about teacher shortages, there is little evidence on when teaching positions are posted and how many people apply for them. Using detailed job-posting and application data on nearly 57,000 applicants from 19 traditional school districts and 24 charter organizations, we provide descriptive evidence on the timing of job postings, the supply of applicants, and the characteristics of applicants, emphasizing how each varies across organizations and between the district and charter sectors. We find that late hiring is less prevalent than prior single-district studies suggest—about 11% of openings are posted late—but varies widely across organizations. Applicant supply differs …

Peripheral sensory neurons regenerate their axons after injury to regain function, but this ability declines with age. The mechanisms behind this decline are not fully understood. While excessive production of endothelin 1 (ET-1), a potent vasoconstrictor, is linked to many diseases that increase with age, the role of ET-1 and its receptors in axon regeneration is unknown. Using single-cell RNA sequencing, we show that satellite glial cells (SGCs), which completely envelop the sensory neuron soma residing in the dorsal root ganglia (DRG), express the endothelin B receptor (ETBR), while ET-1 is expressed by endothelial cells. Background and study aims: Data on colorectal endoscopic mucosal resection (C-EMR) training during advanced endoscopy fellowship remain limited. We aimed to determine the number of procedures required by an "average" advanced endoscopy trainee (AET) to achieve competence in cognitive and technical C-EMR skills.

Methods: AETs from advanced endoscopy training programs (AETPs) were graded on every C-EMR using a standardized assessment tool. Though there has been substantial progress in the development of anti-human epidermal growth factor receptor 2 (HER2) therapies to treat HER2-positive metastatic breast cancer (MBC) within the past two decades, most patients still experience disease progression and cancer-related death. HER2-directed tyrosine kinase inhibitors can be highly effective therapies for patients with HER2-positive MBC; however, an understanding of resistance mechanisms is needed to better inform treatment approaches. We performed whole-exome sequencing on 111 patients with 73 tumor biopsies and 120 cell-free DNA samples to assess mechanisms of resistance. The development of a region is frequently followed by the emergence of slum settlements which further obstruct the growth of productive activities. This study evaluates the effects of Slum Upgrading Program in Indonesia to the growth of small-micro industries, cooperatives, and hotels in slum area. By using difference in difference (DID) method, this study analysed 3427 villages in 33 provinces during 2016-2018. It was found that program increased the number of micro and small industries and hotels, and reduced the number of cooperatives. Over the last decade, the manufacturing sector's contribution to Indonesia's economy has declined. In 2010, it was 29.10% of GDP, but by 2022, it had dropped to 19.14%. Employment in the sector also fell, from 14.91% of the workforce in 2010 to 13.80% in 2022. To reverse this trend, the government introduced a policy to boost the Domestic Component Level (DCL) in manufacturing. By offering fiscal incentives to industries meeting certain DCL targets, the government aims to drive economic growth and job creation. A study using BPS data from 2008-2019 shows that this policy raised DCL by 23.5%.

This study aims to inquire the Political Budget Cycle (PBC) by examining the relationship between the presence of an incumbent during the simultaneous district/city level elections in 2015, 2017, 2018, 2020 and village financial assistant. Using the village financial assistant data in 2013-2021, this study conducts a random effect model. The results showed that the existence of an incumbent is significantly correlated with an increase in village financial assistant in one or two years prior to election. Meanwhile, in the election year, there is no significant relationship between variables. The PBC took place in the years prior to election.

Background: Delirium is a common, underdiagnosed symptom of acute illness that can be prevented through early recognition and intervention. There is limited understanding of effective delirium education for pre-registration health students, and no evidence-based interdisciplinary digital resources exist. Using mixed methods, this study evaluates a co-designed digital tool for health students in Ireland to enhance their ability to prevent, identify, and manage delirium collaboratively [1-3].
