This qualitative study examined how special education teachers perceived their teaching effectiveness in supporting students identified with emotional and behavioral disorders (EBD). The study addressed a gap in the literature by examining the factors that influence special educators’ sense of self-efficacy when working with this population. Bandura’s theory of self-efficacy served as the conceptual framework. A generic qualitative research design was employed, and data were collected through semi-structured interviews with eight participants from two central Texas school districts. The problem that was addressed through this study is that the experiences of early childhood professionals (ECPs) working with children with selective mutism are unknown. Grounded in Bronfenbrenner’s ecological theory of child development, the purpose of this basic qualitative study was to explore the experiences of ECPs working with children with selective mutism. Data were collected through semistructured interviews with 12 ECPs who support children with selective mutism in several districts in southern California. Background: We previously developed a digital quality measure (dQM) of emergency presentations (EPs) in colorectal cancer (CRC) and found it to be associated with worse outcomes. Potentially avoidable EPs were common in this cohort, but identifying them required time-intensive chart reviews. We aimed to enhance the existing dQM to automate the detection of potentially avoidable EPs.

Materials and methods: We defined potentially avoidable EPs as those preceded by a CRC red flag (iron-deficiency anaemia or haematochezia ≥60 days prior, or positive stool-based screening test ≥180 days prior). The enhanced dQM was applied to a national cohort of incident CRC cases …

Endometriosis is a pathological condition characterized by the ectopic growth of endometrial cells, leading to chronic pelvic pain and infertility. Epidemiological studies have associated exposure to dioxin-like polychlorinated biphenyls, particularly PCB126, with an increased risk of endometriosis. However, the underlying mechanisms of this association remain poorly understood. We utilized a surgically induced endometriosis mouse model and human endometrial cell lines to assess the impact of PCB126 on endometriosis progression. Mice were exposed to environmentally relevant doses of PCB126. Long-read RNA sequencing is a powerful technology for transcriptomics, but low throughput and high cost pose challenges. Adaptive sampling, a feature of Oxford Nanopore Technologies, offers real-time enrichment by selectively ejecting non-target molecules. We evaluate adaptive sampling for human transcriptome analysis. Adaptive sampling modestly enriches target transcripts (1.3 × for cDNA sequencing, 1.9 × for direct RNA sequencing) while preserving gene expression and splicing profiles, but is significantly less effective than cDNA hybridization capture. Short read lengths and low sequencing quality limit performance. Adaptive sampling on direct RNA sequencing can boost target yield (~ 20%) within fixed run times, potentially …

Breakthrough therapy medicines are designed to address unmet medical needs and are often approved on an expedited timeline; however they are also criticized for lacking an adequate understanding of their risk profile at the time of approval. The illegal wildlife trade, valued at tens of billions annually, presents a major conservation and law-enforcement challenge; this article examines how novel forensic techniques (e.g. biometric hand-print matching, DNA analysis) are empowering authorities to trace trafficked specimens and prosecute operators. It argues that while promising, these methods require further development, broader deployment, and stronger legal frameworks to truly shift the balance against organized wildlife crime.

With diabetes prevalence around the world nearing 10%, and half of the US population having either diabetes or pre-diabetes, many epidemiological studies show diabetic patients have both increased risk for developing major cardiovascular disease (CVD), and increased mortality rates due to CVD. Additionally, within this field, studies have shown that indi-viduals with previously higher HbA1C levels have higher long-lasting risk for major ad-verse cardiac events compared to those with previously lower HbA1C levels—establishing the concept of “metabolic memory”. Within the diabetic milieu, cardiac protein O-GlcNAcylation (O-linked B-N-acetyl-glucosamine) is important for cardiac pathology de-velopment. The toxicological effects of polycyclic aromatic hydrocarbons (PAHs) and polycyclic aromatic compounds (PACs) have attracted considerable research interest due to their known bioaccumulation. However, time-resolved and longitudinal studies that examine in vivo bioaccumulation and clearance remain limited. Here we use Surface-Enhanced Raman Scattering (SERS) and Surface-Enhanced Infrared Absorption (SEIRA) spectroscopies to detect and track the presence of two PAHs: pyrene, benzo[a]pyrene, and one PAC: 5,12-naphthacenequinone, in murine organ tissues. Previous studies show that orphan nuclear receptor 4A1 (NR4A1) regulates endometriotic cell growth, survival, estrogen receptor β (ERβ), mechanistic target of rapamycin signaling and fibrosis. NR4A2 is also expressed in epithelial and stromal derived endometriotic cells, and in this study the effects of 1,1-bis(3'-indolyl)-(3,5-disubstitutedphenyl)methane (DIM-3,5) dual NR4A1/nuclear receptor 4A2 (NR4A2) ligands and knockdown of NR4A1 and NR4A2 were investigated. The dual NR4A1/2 DIM-3,5 analogs inhibited previously identified proendometriotic pathways and gene products, and they also inhibited TWIST1 and multiple markers associated with epithelial-to-mesenchymal transition (EMT). Purpose: Intimate partner violence (IPV) victimization and traumatic brain injury (TBI) are common correlates of mental health problems in adolescence. However, research has yet to investigate whether IPV victimization is related to developmental changes in mental health, as well as the contributing role that TBI may play in conditioning this association. We address this gap by analyzing longitudinal data with well-validated measures of mental health from an ethnically diverse sample of adolescents.

Background: Recent data have suggested that germline genetic aberrations can affect outcomes in patients with large B-cell lymphoma (LBCL) treated with chimeric antigen receptor T-cell therapy (CART). However, a comprehensive analysis of germline determinants of response and toxicity after CART has not yet been described.

Methods: Genome-wide genotyping was performed in 170 patients with LBCL treated with standard of care axicabtagene ciloleucel. Polygenic risk score instruments for blood cell traits and inflammatory markers were obtained from the PGS Catalog and analyzed using PRSice-2. The success of nanoparticle-based cancer therapeutics relies on their efficient tumor uptake and retention. Given this, improving nanoparticle localization in tumors is paramount to maximize their therapeutic potential. A common approach to achieve this is to functionalize nanoparticles with active targeting moieties that bind to specific tumor-associated receptors. Among these, arginine-glycine-aspartic acid (RGD) peptides have shown a potential to promote tumor accumulation by targeting the ανβ3 integrin receptor, a receptor commonly overexpressed by tumors owing to its role in promoting angiogenesis, metastasis and proliferation. Intratumoral B Cell And Interferon Signatures In Newly Diagnosed Glioblastoma Are Associated With Longer Survival In Patients Treated With Survaxm, Manmeet Ahluwalia

Glioblastoma (GBM) has proved difficult to treat, and there is dire need for more effective therapies. In a single arm phase IIa trial (NCT02455557), treatment of newly diagnosed GBM patients with the peptide vaccine SurVaxM resulted in promising median progression-free and overall survival. To investigate molecular features that associate with GBM responsiveness to SurVaxM, retrospective whole exome and RNA sequencing was performed on patient tumors (n = 34) collected prior to standard of care treatment plus SurVaxM. Differential gene expression and mutational profiles were characterized between patients with short-term (OS < 18 months) or long-term (OS ≥ 18 months) overall survival. Greater expression of interferon, complement, and humoral immunity signatures were associated with long-term survival. Deconvolution of transcriptomes identified enrichment of intratumoral memory B cell populations in long-term survivors that were validated by CD20 staining in matched samples. A five-gene expression signature and a B cell specific signature predicted survival within the SurVaxM-treated cohort, however, these signatures were not associated with improved outcomes in a similarly treated population obtained from The Cancer Genome Atlas (TCGA) that did not receive immunotherapeutic intervention. Although prospective validation is ongoing, the findings in this discovery cohort specify molecular features of GBM associated with better overall survival and potential responsiveness to immunotherapy with SurVaxM.

In recent years, there has been an increase in the frequency of severe weather events (like hurricanes). These events are responsible for most power outages in power distribution systems (PDSs). Particularly susceptible to storms are overhead PDSs. In this study, the dynamic Bayesian network (DBN)-based failure model was developed for different hurricane scenarios to predict the line failure of overhead lines.
