This study evaluated the efficacy and safety of unengineered tumor-infiltrating lymphocytes (TILs) combined with pembrolizumab and either high (HD, Arm-1) or low (LD, Arm-2) doses of IL-2 in patients with metastatic melanoma (MM). Patients were lymphodepleted with cyclophosphamide and fludarabine, followed by TIL infusion and IL-2 (Arm-1: 720,000 IU/kg IV q 8 hrs up to 15 doses; Arm-2: 2 million IU SC for 14 days). Patients received pembrolizumab 200 mg IV starting 21 days post-TIL infusion, and every 3 weeks for up to 2 years. The primary endpoint was overall response rate (ORR) per RECIST 1.1. Background: Vertebral compression fractures (VCFs) are the most common type of vertebral body fracture. The Kiva VCF Treatment System is a relatively novel technique to manage VCFs. The aim of this study was to compare the efficacy of Kiva versus standard Balloon Kyphoplasty (BK) through evaluation of published randomized controlled trials (RCTs).
Methods: This study was performed following the guidelines for PRISMA. We performed a systematic literature search using PubMed and MEDLINE in June 2023. Aneurysmal subarachnoid hemorrhage (aSAH) is a devastating neurological disease, and one of the primary drivers of morbidity after aneurysm rupture is the phenomenon of delayed cerebral ischemia (DCI). Significant knowledge has been gained over the past two decades of the impact of neuroinflammation in DCI; and neutrophils are now believed to play a major role. There is significant human subject data showing the rise of neutrophil related inflammatory markers and neutrophil's association with poor outcome after aSAH, but as of yet no trials involving human subjects have been done specifically targeting neutrophils. SARS-CoV-2 is the betacoronavirus causing the COVID-19 pandemic. Although the SARS-CoV-2 genome and transcriptome were reported previously, the function of individual viral proteins is largely unknown. Utilizing biochemical and molecular biology methods, we identified that four SARS-CoV-2 RNA-binding proteins (RBPs) regulate the host RNA metabolism by direct interaction with mature miRNA let-7b revealed by Nuclear Magnetic Resonance spectroscopy (NMR). SARS-CoV-2 RBP Nsp9 primarily binds mature miRNA let-7b, a direct ligand of the Toll-like Receptor 7 (TLR7), one of the potential SARS-CoV-2 therapeutics. Background: Apparent Diffusion Coefficient (ADC) as measured by diffusion weighted imaging is known to negatively correlate with prostate tumor aggressiveness. Heterogeneity in system and protocol performance causes potential variability in ADC acquired across a large scanner network, prompting a need to evaluate quantitative ADC from a prostate-specific MR diffusion protocol as part of quality assurance (QA). Due to the temperature dependence of ADC, repeatability and reproducibility assessments typically require phantoms to maintain a temperature of 0°C, imposing a considerable burden when assessing large numbers of scanners.

The F. nucleatum group (sensu lato) is historically composed of four subspecies (F. nucleatum subsp. animalis, F. nucleatum subsp. nucleatum, F. nucleatum subsp. polymorphum, F. nucleatum subsp. vincentii). Although F. nucleatum sensu lato members are typically associated with oral disease, they have recently been shown to disseminate to the gastrointestinal tract and are associated with adverse health conditions such as inflammatory bowel disease and colorectal cancer (CRC). Cancerous tissue is a largely unexplored microbial niche that provides a unique environment for the colonization and growth of specific bacterial communities, and with it, the opportunity to identify novel bacterial species. Here, we report distinct features of a novel Fusobacterium species, F. sphaericum sp. nov. (Fs), isolated from primary colon adenocarcinoma tissue. We acquire the complete closed genome and associated methylome of this organism and phylogenetically confirm its classification into the Fusobacterium genus, with F. perfoetens as its closest neighbor. Fecal Microbiota Transplant (FMT) is a treatment for recurrent Clostridium difficile infections and is being explored for other clinical applications, from alleviating digestive and neurological disorders, to restoring microbiomes impacted by cancer treatment. Quantifying the extent of engraftment following an FMT is important in understanding a recipient's response to treatment. Engraftment and clinical response need to be investigated independently to evaluate an FMT's role (or lack thereof) in achieving a clinical response. Standardized bioinformatics methodologies for quantifying engraftment extent would not only improve assessment and understanding of FMT outcomes, but also facilitate comparison of FMT results and protocols across studies. …

As the global demand for renewable energy intensifies, piezoelectric energy harvesting from roadways has emerged as a promising avenue for sustainable power generation. This systematic literature review analyzes 61 peer-reviewed studies to assess the feasibility, performance, and potential of integrating piezoelectric systems into roadway infrastructure. While technology faces challenges, such as high installation costs, limited energy output, and a scarcity of thorough economic evaluations, findings suggest it holds considerable promise as a supplementary renewable energy source. The adoption of rooftop photovoltaic (PV) systems can create upward pressure on retail electricity rates as utilities are forced to spread their fixed costs of generation and transmission across a smaller customer base. Since high-income households are more likely to purchase PV systems, low-income households may be disproportionately impacted by these rate increases. Using a novel combination of agent-based computational economic modeling and a choice experiment of rooftop solar adoption, we show how this pecuniary externality between low- and high-income customers increases low-income electricity bills by 10% in an area with some of the highest poverty rates in the United …

Sinkhole formation poses a significant geohazard in karst regions, where unpredictable subsurface erosion often necessitates costly grouting for stabilization. Accurate estimation of grout volume remains a persistent challenge due to spatial variability, site-specific conditions, and the limitations of traditional empirical methods. This study introduces a novel machine learning-based regression model for grout volume prediction that integrates cone penetration test (CPT)-derived Sinkhole Resistance Ratio (SRR) values, spatial correlations between CPT and grouting points (GPs), and field-recorded grout volumes from six sinkhole sites in Florida. During a first-year engagement program, a librarian and a cohort of students worked towards a shared understanding of dominant narratives, historical silences, and creating counterstories through research to center marginalized stories and complicate existing narratives. As information literacy professionals, we should design interactive lessons that demonstrate critical search and evaluation skills through topics that are relevant to students and encourage them to work with issues that impact their communities. Incorporating critical information literacy, feminist pedagogy, and critical race theory, this two-day micro-seminar introduced first-year students to the legacy of youth community activism in the neighborhoods around their university. Objective: High-quality patient matching from several sources without a common identifier (ID) requires interactive record linkage (RL) using a hybrid human-computer system. MiNDFIRL (MInimum Necessary Disclosure For Interactive Record Linkage) is a hybrid prototype software system that facilitates maximizing linkage accuracy while minimizing information disclosure. We present and evaluate MiNDFIRL using two real-world case studies.
Materials and methods: Two user studies were conducted linking 10,000 data pairs from EHR data and 18,240 unique patient IDs from patient generated data. After automated RL, manual review was conducted by three teams of four reviewers (12 total) using MiNDFIRL to resolve potential matches …

Military voluntary kin (MVK) refer to relationships between Service members and/or Veterans that they consider to be “like family,” despite lacking blood or legal ties. The purpose of this qualitative/interpretive communication study was to explore how female Veterans, who are the largest growing population of U.S. Veterans, discursively construct MVK relationships, navigate communication, privacy management, and boundary turbulence with their MVK and biolegal families (BLF). The researcher undertook in-depth interviews with nine female U.S. Veterans who reported having at least one MVK. The researcher posed four research questions, engaging in an interpretive analysis.

Recently, a recommendation has emerged for law enforcement to record the full eyewitness identification procedure, recognizing the critical role eyewitness testimony plays in the justice system (Wells et al., 2020). However, in some states, such as New York, law enforcement is required to seek consent from witnesses before recording (Municipal Police Training Council & Division of Criminal Justice Services, 2017). Since this practice is relatively new, there has been limited research on how obtaining consent may affect eyewitness accuracy. Fenestrated and branched technology for endovascular aortic repair of the thoracoabdominal aorta and aortic arch has evolved significantly over the last decade. The spectrum of these devices is largely available outside the United States (US), while patient-specific company manufactured devices are limited to ten sites in the US, with ongoing expanding use of market-approved off-the-shelf devices. Halophila stipulacea (Forsskål & Niebuhr) is a tropical seagrass native to the Red Sea, Persian Gulf, and Indian Ocean. In 2002, invasive populations of H. stipulacea were identified in the Caribbean Sea where it now forms large meadows, displaces native seagrasses, alters invertebrate and fish communities, and potentially affects organic matter dynamics in invaded meadows. This study investigated differences in organic matter dynamics between meadows of H. stipulacea and native Thalassia testudinum within the Jobos Bay National Estuarine Research Reserve (JBNERR) on the southern coast of Puerto Rico. Urbanization alters landscapes in ways that can reshape wildlife populations by changing habitat connectivity, resource availability, and selective pressures. To investigate how urbanization influences both genetic differentiation and morphology in small mammals, I examined Peromyscus leucopus (PELE) across an urban–rural gradient in northern Georgia. Using RADseq SNP data from PELE individuals across 13 sites, I quantified pairwise genetic differentiation (FST) and tested how various environmental variables, including impervious surface cover, human population density, and tree canopy cover, predict genetic differentiation after accounting for geographic distance. This qualitative case study examined how peer mentoring influences teacher self-efficacy at a rural elementary school in southeastern Georgia. Using Bandura's self-efficacy theory and Wenger's communities of practice framework, the research investigated how teachers engage in mentoring relationships and how these practices contribute to professional growth.
Semi-structured interviews and reflection journals were conducted with ten elementary teachers representing diverse experience levels (1-21+ years) and grade levels. Findings revealed that meaningful peer mentoring occurred primarily through informal, trust-based relationships within grade-level teams rather than formal programs. Teachers engaged collaboratively in lesson planning, data analysis, and problem-solving.

This single qualitative case study examined how detracking Algebra 1 at Mountainview High School (pseudonym) restructured the course’s rules and resources and how teachers exercised agency in response. Guided by Giddens’ structuration theory and Weis and Fine’s critical bifocality, the study explored how institutional structures both shape and are reshaped by teacher action within broader systems of inequality. Participants were seven Algebra 1 teachers involved in the first two years of detracking. Peacebuilders have utilized positive intergroup contact in various settings to help groups encounter each other. I implemented three different intimate imagined contact scenarios in Peja, Kosovo with Albanian and Serb participants. I conducted this research to understand how multiple intimate social settings could impact individuals’ encounters with the outgroup during each imagined scenario, thus gathering better operationalization efforts regarding intimacy and imagined contact. Using written responses, interviews, and one-year follow-up interviews, the café and workplace scenarios created a positive experience for most participants, and while half of participants had a positive experience with the fiancé scenario, many more had negative …

Peripheral Artery Disease (PAD) affects an estimated 200 million people worldwide and is the leading cause of amputation in the United States. PAD is characterized by atherosclerotic narrowing of arteries, resulting in ischemia from reduced blood flow to distal tissues. Although surgical and percutaneous interventions can restore perfusion, fewer than 50% of patients experience sustained benefit, highlighting the need for alternative therapeutic strategies. One promising approach is promoting arteriogenesis, the growth and outward remodeling of pre-existing collateral vessels that improve blood flow around the occluded arteries. Arteriogenesis is regulated by regenerative, M2-polarized macrophages, making macrophage polarization a key therapeutic target. …

Background: Despite significant progress in patient safety, human error continues to occur at high rates in procedural settings. Traditional safety models tend to focus on retrospective incident analysis, often overlooking the systemic conditions that allow errors to occur.
