Intercellular communication is vital for quick adjustments and maintenance for cell function and development. Gap junctions are membranes channel proteins that enable this direct communication between adjacent cells throughout the body. The compatibility of connexins (Cx), which make up a gap junction, determines whether a gap junction can form. Though many studies show which connexins are compatible, the molecular basis is not known (Bai & Wang, 2014). Through computational modeling, we identify the residues that energetically contribute most favorably at the docking interface of homotypic and heterotypic combinations of Cx43, Cx46, and Cx50 gap junctions. The Anterior Cruciate Ligament (ACL) is the primary source of rotational stability in the knee by preventing the tibia from sliding in front of the femur. When the ACL is torn, it typically must be repaired through reconstructive surgery which results in proprioceptive deficiencies in the knee. Proprioception plays an important role in understanding where one’s knee is in space, sensing movement and reacting accordingly. This study examines an alternative method of measuring proprioceptive responses to a stimulus (motion) by using electromyogram (EMG) and electroencephalogram (EEG) signals to observe muscle and brain activity. Wireless body sensor networks (WBSNs) mostly consist of low-cost sensor nodes and implanted devices which generally have extremely limited capability of computations and energy capabilities. With exponential growth on a daily basis, there is potentially valuable information hidden in complex electronic medical records (EMR) systems. In this thesis, several efficient data mining algorithms were explored to discover hidden knowledge in insurance claims data. The first aim was to cluster three levels of information overload(IO) groups among chronic rheumatic disease (CRD) patient groups based on their clinical events extracted from insurance claims data. Increased expression of cytosolic DNA sensors, a category of pattern recognition receptor, after control plasmid DNA electrotransfer was observed in our previous studies on B16.F10 murine melanoma cells. This expression was correlated with the upregulation of proinflammatory cytokines and chemokines and was associated with cell death. Here, we expanded our research to include the influence of features of cells in a 3-dimensional environment, which better represents the tumors’ organization in vivo. Antibiotic-resistant strains of bacteria may result in serious infections which are difficult to treat. In addition, the poor antibiotic pipeline has also contributed to the crisis. Recently, the complex of furosemide and silver (Ag-FSE) has been reported as a potential antibacterial agent. However, its poor aqueous solubility is limiting its activity as an effective antibacterial agent. The purpose of this study was to encapsulate Ag-FSE into chitosan nanoparticles (CSNPs) and evaluate antibacterial efficacy. Ag-FSE CSNPs were prepared using ionic gelation technique. Intense electric shocks of nanosecond (ns) duration can become a new modality for more efficient but safer defibrillation. We extended strength-duration curves for excitation of cardiomyocytes down to 200 ns, and compared electroporative damage by proportionally more intense shocks of different duration. Enzymatically isolated murine, rabbit, and swine adult ventricular cardiomyocytes (VCM) were loaded with a Ca2+ indicator Fluo-4 or Fluo-5N and subjected to shocks of increasing amplitude until a Ca2+ transient was optically detected. Deep wounds in the gingiva caused bytrauma or surgery require a rapid and robust healing of connective tissues. Wepropose utilizing gas-brushed nanofibers coated with collagen and fibrin for that purpose. Our hypotheses are that protein-coated nanofibers will: (i) attract and mobilize cells in various spatial orientations, and (ii) regulate the expression levels ofspecific extracellular matrix (ECM)-associated proteins, determining the initial conformational nature ofdense and soft connective tissues. Research on the discovery, classification and validation of biological markers, or biomarkers, have grown extensively in the last decades. Newfound and correctly validated biomarkers have great potential as prognostic and diagnostic indicators, but present a complex relationship with pertinent endpoints such as survival or other diseases manifestations. This research proposes the use of computational argumentation theory as a starting point for the resolution of this problem for cases in which a large amount of data is unavailable. A novel crosslinker [4,4′-dihydroxybiphenyl diacrylate (44BDA)] was developed, and a series of temperature-responsive hydrogels were synthesized through free radical polymerization of N-isopropylacrylamide (NIPAAm) with 44BDA. The temperature-responsive behavior of the resulting gels was characterized by swelling studies, and the lower critical solution temperature (LCST) of the hydrogels was characterized through differential scanning calorimetry. Increased content of 44BDA led to a decreased swelling ratio and shifted the LCST to lower temperatures. These novel hydrogels also displayed resiliency through multiple swelling–deswelling cycles, and their temperature responsiveness was reversible. 3D printing was used to fabricate porous artificial knee meniscus material from biocompatible polymeric blends of polycarbonate-urethane (PCU) and ultra-high-molecular-weight polyethylene (UHMWPE) to enable “weep” lubrication that mimics the native meniscus. 3D printed and molded pure PCU, as well as molded PCU and UHMWPE, were used for comparison. Preliminary printing was done to evaluate the impact of process parameters on the results. The samples were subject to a variety of rotational oscillating friction and wear tests under simulated body fluid and loading conditions to replicate the natural motion of the knee.
