Developing An Optomechanical Approach For Characterizing Mechanical Properties Of Single Adherent Cells, Ali Mehrnezhad

Mechanical properties of a cell reflect its biological and pathological conditions including cellular disorders and fundamental cellular processes such as cell division and differentiation. There have been active research efforts to develop high-throughput platforms to mechanically characterize single cells. Yet, many of these research efforts are focused on suspended cells and use a flow-through configuration. Therefore, adherent cells are detached prior to the characterization, which seriously perturbs the cellular conditions. Also, methods for adherent cells are limited in their throughput.

Background: Reconstruction of bone fractures and defects remains a big challenge in orthopedic surgery. While regenerative engineering has advanced the field greatly using a combination of biomaterial scaffolds and stem cells, one matter of difficulty is inducing osteogenesis in these cells. Recent works have shown electricity’s ability to promote osteogenesis in stem cell lines when seeded in bone scaffolds; however, typical electrical stimulators are either (a) externally housed and require overcomplex percutaneous wires be connected to the implanted scaffold or (b) implanted non-degradable devices which contain toxic batteries and require invasive removal surgeries.

We describe Bayesian Model Optimized Reference Correction (BaMORC), a software package that performs 13C chemical shifts reference correction for either assigned or unassigned peak lists derived from protein NMR spectra. BaMORC provides an intuitive command line interface that allows non-nuclear magnetic resonance (NMR) experts to detect and correct 13C chemical shift referencing errors of unassigned peak lists at the very beginning of NMR data analysis, further lowering the bar of expertise required for effective protein NMR analysis. This model will be used to test mapping probe catheters used for locating cardiac arrhythmias against current technology used in practice. In this dissertation, we have developed advanced engineering tools to address three distinct problems that researchers, clinicians and patients are facing in SCI research. Methods for prolonged compassionate care for persons with Profound Intellectual and Multiple Disabilities (PIMD) require a rotating cast of import people in the subjects life in order to facilitate interaction with the external environment. As subjects continue to age, dependency on these people increases with complexity of communications while the quality of communication decreases. It is theorized that a machine learning (ML) system could replicate the attuning process and replace these people to promote independence. Multiple myeloma (MM) is an incurable malignancy characterized by the uncontrolled proliferation of long-lived plasma cells (PCs) in the bone marrow (BM), which constitute at least 10% of BM cellularity. Normally, long-lived plasma cells make up less than 1% of BM cells. Plasma cells become neoplastic when a clonal PC population produces a monoclonal immunoglobulin protein. A diagnosis of monoclonal gammopathy of undetermined significance (MGUS) is made when there is an increase in monoclonal PCs within the BM, but less than 10%, and the patient does not present with end-organ damage, which is associated with active MM. Left ventricular (LV) remodeling is nominally an adaptive process that restores biomechanical function following myocardial injury and/or sustained alterations in loading conditions. This remodeling can materialize as changes in myocardial geometry, composition, and mechanical properties. When these changes fail to restore LV biomechanical function, remodeling is termed maladaptive. It is generally accepted that maladaptive LV remodeling underlies the progression to heart failure in various forms of heart disease.
