Thermal Responsive Release Of A Model Drug, Rhoadmine B, From Alginate Bead System, Heather F. Fairbairn Miss

Williams Honors College, Honors Research Projects

Smart drug delivery systems have garnered increased interest in the biomedical community during recent years. Investigation into thermal, pH, and light responsive delivery systems allows for targeted drug release systems to be developed. For this project, thermal responsive release behavior was investigated for the purpose of tuning release systems to achieve 60% release in a specified time by varying the content of thermal responsive polymer. To investigate this, the release profile of a model small hydrophilic drug (rhodamine B) from polymeric alginate beads that have been modified by the addition of thermal responsive poly(N-isopropylacrylamide) (PNIPAAm). It was determined that when …

Purification And Recovery Of Infectious Virus Particles Using Osmolyte Flocculation, Ashish Saksule

Dissertations, Master's Theses and Master's Reports

Viral infectious diseases cause millions of deaths and severe illness all around the world affecting public health and economics. Viral vaccines are helping to fight against viral diseases, but current vaccines are expensive and unavailable, especially in poor and developing countries. When measuring the entire manufacturing processing, the downstream processing of vaccines is the major cost of production. Our goal for this research is to develop a low-cost alternative downstream processing platform for new vaccine manufacturing infrastructures. We have developed a novel osmolyte flocculation method for viral particles. To create a platform purification for several types of viral particles, we …

A Petrophysical Evaluation For Permeability Of A Gas Reservoir In The Taranaki Basin, New Zealand, Musa Cetin 2016 Michigan Technological University

A Petrophysical Evaluation For Permeability Of A Gas Reservoir In The Taranaki Basin, New Zealand, Musa Cetin

Dissertations, Master's Theses and Master's Reports

The goal of this study was to evaluate permeability and study the controls on permeability in a gas saturated formation. Conventional well logs, mineral identification crossplots and empirical models were applied to analyze different lithologic and diagenetic features and to examine the effect that these features may have on the reservoir. An unusual feature was observed, and required detailed examination: there existed (in two wells) five zones of lower resistivity (higher water saturation) above the gas-water contact. This is unexpected, as above that contact, the water is usually at irreducible water saturation. I conclude that the lower resistivity (the higher …

Experimental And Theoretical Investigation Of Sustainable Fast Pyrolysis Biofuels From Woody Biomass, Bethany Jean Klemetsrud

Dissertations, Master's Theses and Master's Reports

Biofuels are an important advancement in alternative energy that can provide substantial environmental benefits compared to their conventional fossil fuel counterparts Said benefits are usually measured using life cycle assessments. However, it is not well understood yet how different methodological choices such as system boundaries, biomass feedstocks, conversion pathways, geographical data, etc. affect the conclusions drawn from biofuels LCA. This research shows large variability in life cycle assessment results and limits comparison across different biofuel pathways due to methodological choices set forth by policy and certification schemes. Advanced biofuels have not reached large scale production due to a limited understanding …

Theses and Dissertations

A novel two-stage pressure swing adsorption (PSA) system has been developed to remove metabolic CO2 removal from the spacecraft cabin air of the International Space Station (ISS). This PSA system enriches and recovers the CO2 to make it suitable for use in a Sabatier reactor (CO2 reduction). This two-stage PSA process utilizes Stage 1 to concentrate metabolic CO2 from about 0.2667 vol% to about 40 to 60 vol% and Stage 2 to further enrich the CO2 product from Stage 1 up to > 97 vol% CO2, while recovering at least 95% of it, which corresponds to removing 4.0 kg/day of CO2. …

A Theoretical Simulation Of The Settling Of Proppants In A Hydraulic Fracturing Process, Nisreen Alseamr 2016 Virginia Commonwealth University

A Theoretical Simulation Of The Settling Of Proppants In A Hydraulic Fracturing Process, Nisreen Alseamr

Theses and Dissertations

Hydraulic fracturing is a process for the extraction of hydrocarbons from underground formations. It involves pumping a specialized fluid into the wellbore under high pressures to form and support fractures in the rock. Fracturing stimulates the well to increase the production of oil and the natural gas which are the pillars of the energy economy. Key to this process is the use of proppants, which are solid materials used to keep the fractures open. Understanding the transport of proppant particles through a fluid is important to improve the efficiency and reduce environmental impact of fracturing. An increase of the settling …

Department of Chemical and Biomolecular Engineering: Faculty Publications

Chlorovirus Paramecium bursaria chlorella virus 1 (PBCV-1) contains a viral-encoded K+ channel imbedded in its internal membrane, which triggers host plasma membrane depolarization during virus infection. This early stage of infection was monitored at high resolution by recording the cell membrane depolarization of a single Chlorella cell during infection by a single PBCV-1 particle. The measurement was achieved by depositing the cells onto a network of one-dimensional necklaces of Au nanoparticles, which spanned two electrodes 70 μm apart. The nanoparticle necklace array has been shown to behave as a single-electron device at room temperature. The resulting electrochemical field-effect transistor …

Enabling Method To Design Versatile Biomaterial Systems From Colloidal Building Blocks, Shalini Saxena, L. Andrew Lyon

Biology, Chemistry, and Environmental Sciences Faculty Articles and Research

Development of materials with fine spatial control over topographical, mechanical, or chemical features has been investigated for a variety of applications. Here we present a method to fabricate an array of polyelectrolyte constructs including two-dimensionally and three-dimensionally patterned assemblies using both compressible and incompressible colloidal building blocks. This method eliminates prior constraints associated with specific chemistries, and can be used to develop modular, multi-component, patterned assemblies. In particular, development of constructs were investigated using microgels, which are colloidally stable hydrogel microparticles, polystyrene (PS) beads, and PS-microgel core-shell building blocks in conjunction with the polycation poly(ethyleneimine) (PEI). The topography, mechanical properties, …

Discrete Model For Cancer Tumor Growth And Effect Of Chemotherapy, Karim Virani 2016 CUNY City College

Discrete Model For Cancer Tumor Growth And Effect Of Chemotherapy, Karim Virani

Dissertations and Theses

Many mathematical models have focused on growth of a single tumor with their spreading rate with some have included its metastasis. Some of the earlier models focus on exponential growth and its shift with time on to a linear model. The Gompertzian model involves a quadratic terms and that limit growth. We looked into Iwata et al paper it discusses a different model and takes into account the growth and metastasis of a tumor population but does not look into tumor shrinking or chemotherapy. Our new model focuses on the extending Iwata et al model to take into account tumor …

Bioactive Poly(Beta-Amino Ester) Biomaterials For Treatment Of Infection And Oxidative Stress, Andrew L. Lakes

Theses and Dissertations--Chemical and Materials Engineering

Polymers have deep roots as drug delivery tools, and are widely used in clinical to private settings. Currently, however, numerous traditional therapies exist which may be improved through use of polymeric biomaterials. Through our work with infectious and oxidative stress disease prevention and treatment, we aimed to develop application driven, enhanced therapies utilizing new classes of polymers synthesized in-house. Applying biodegradable poly(β-amino ester) (PBAE) polymers, covalent-addition of bioactive substrates to these PBAEs avoided certain pitfalls of free-loaded and non-degradable drug delivery systems. Further, through variation of polymer ingredients and conditions, we were able to tune degradation rates, release profiles, cellular …

Synthesis And Characterization Of Antioxidant Conjugated Poly(Βeta-Amino Ester) Micro/Nanogels For The Suppression Of Oxidative Stress, Prachi Gupta 2016 University of Kentucky

Synthesis And Characterization Of Antioxidant Conjugated Poly(Βeta-Amino Ester) Micro/Nanogels For The Suppression Of Oxidative Stress, Prachi Gupta

Theses and Dissertations--Chemical and Materials Engineering

Oxidative stress is a pathophysiological condition defined by an increased production of reactive oxygen species (ROS), which can result in the growth arrest of cells followed by cell disintegration or necrosis. A number of small molecule antioxidants (e.g. curcumin, quercetin and resveratrol) are capable of directly scavenging ROS, thereby short-circuiting the self-propagating oxidative stress state. However, poor solubility and rapid 1st pass metabolism results in overall low bioavailability and acts as a barrier for its use as a drug to suppress oxidative stress efficiently.

To overcome this limitation, these small molecule antioxidants were covalently conjugated into poly(β-amino ester) (PβAE) …

Theses and Dissertations--Chemical and Materials Engineering

Protein adsorption on solid surfaces is a common yet complicated phenomenon that is not fully understood. Self-assembled monolayers have been utilized in many studies, as well-defined model systems for studying protein-surface interactions in the atomic level. Various strategies, including the use of single component SAMs[1, 2], combinations of long and short alkanethiolates with methyl- and hydroxyl- terminal groups[3, 4], and using mixtures of alkanethiolates with similar chain length and varying terminal functional group [5] have been used to effectively control the surface wettability and determine the effect of surface composition and wettability on protein adsorption. In this dissertation we report …

Synthesis Of Titania Thin Films With Controlled Mesopore Orientation: Nanostructure For Energy Conversion And Storage, Suraj R. Nagpure 2016 University of Kentucky

Synthesis Of Titania Thin Films With Controlled Mesopore Orientation: Nanostructure For Energy Conversion And Storage, Suraj R. Nagpure

Theses and Dissertations--Chemical and Materials Engineering

This dissertation addresses the synthesis mechanism of mesoporous titania thin films with 2D Hexagonal Close Packed (HCP) cylindrical nanopores by an evaporation-induced self-assembly (EISA) method with Pluronic surfactants P123 and F127 as structure directing agents, and their applications in photovoltaics and lithium ion batteries. To provide orthogonal alignment of the pores, surface modification of substrates with crosslinked surfactant has been used to provide a chemically neutral surface. GISAXS studies show not only that aging at 4°C facilitates ordered mesostructure development, but also that aging at this temperature helps to provide orthogonal orientation of the cylindrical micelles which assemble into an …

Smart Fabrics For Colorimetric Detection, Breland Edwards 2016 Virginia Commonwealth University

Smart Fabrics For Colorimetric Detection, Breland Edwards

UROP Posters

Due to the prevalence of chemical warfare, soldiers often carry chemical sensing devices, to warn of oncoming nerve agents. However, these devices are cumbersome. A lightweight, wearable chemical sensing fabric that doubles as a protectant against toxins would be beneficial. This fabric can be created by incorporating (1) organophosphate hydrolase, an enzyme capable of degrading organophosphates and releasing an acidic by product, and (2) polyaniline, a color changing polymer that changes from purple to green in the presence of acid, into nylon or polyvinyl alcohol nanofibers. The first step is to produce fabrics containing polyaniline and demonstrate that the fabrics …

Hepatitis C is a liver disease caused by the hepatitis C virus (HCV). According to the World Health Organization, hepatitis C infections affect between 130-150 million people worldwide. It is a major cause of liver cirrhosis and liver cancer, and approximately 500,000 people die from hepatitis C infections or related complications each year. Although new treatments are 90% effective at curing chronic hepatitis C infections, they cost tens of thousands of dollars. Available treatments are incredibly cost prohibitive, especially considering that the regions most affected by hepatitis C are Africa and central and eastern Asia (Hepatitis C). This creates a …

G-Quadruplex Mediated Splicing Of P53 Mrna, Melissa Holy 2016 University of Mississippi. Sally McDonnell Barksdale Honors College

G-Quadruplex Mediated Splicing Of P53 Mrna, Melissa Holy

Honors Theses

The protein p53 is a master regulator of the cell cycle and its mutation/inactivation is associated with over 50% of cancer cases. Mutations in p53 are associated with worse survival rates, resistance to therapies, and other risk factors in breast cancer specifically. p53 is encoded by the TP53 gene and is expressed as several transcripts. In the G-rich portion of intron 3, secondary DNA structures known as G-quadruplexes are able to form. The stabilization of these G-quadruplexes leads to correct splicing of intron 2, thus making a functional p53 protein. A known quadruplex-stabilizing compound (TMPyP4) was tested to determine its …

The Effects Of Genistein, Thymoquinone, Epigallocatechin-3-Gallate,5-Fluorouracil, And Low Level Laser Therapy On Fadu Hypopharyngeal And Laryngeal Carcinoma Cells, Osasu Adah 2016 University of Mississippi. Sally McDonnell Barksdale Honors College

The Effects Of Genistein, Thymoquinone, Epigallocatechin-3-Gallate,5-Fluorouracil, And Low Level Laser Therapy On Fadu Hypopharyngeal And Laryngeal Carcinoma Cells, Osasu Adah

Honors Theses

Laryngeal and hypopharyngeal carcinoma are two types of head and neck squamous cell carcinomas. The location of the tumor makes it hard to detect without complex tests. Overall, these types of cancer are not common, and there are no simple screening tests for such cancers. Due to this issue, laryngeal and hypopharyngeal carcinoma are usually diagnosed when signs and symptoms occur, such as sore throat, trouble swallowing, and constant coughing. With the use of FaDu, a head and neck cancer cell line of the hypopharyngeal origin, experimentation can be performed in order to initiate possible treatments. The standard treatments for …

Design And Synthesis Of Dual Opioids/Npff Ligands As Novel Analgesics With Reduced Tolerance Potential, John N. Yi 2016 University of Mississippi. Sally McDonnell Barksdale Honors College

Design And Synthesis Of Dual Opioids/Npff Ligands As Novel Analgesics With Reduced Tolerance Potential, John N. Yi

Honors Theses

Opioid use has become prevalent in today's culture. As a treatment for acute and chronic pain, opioids permeate in the public sphere as a seemingly easy solution. However, problems that come from the increased use of opiates, especially in the last fifteen years, have shown detrimental effects in society. Not only are all opioid-based analgesics considered addictive, but the nature of such painkillers also enables a phenomenon known as tolerance, where the body adapts to the use of exogenous opioids. This creates a situation where a more potent dose of painkiller is required for the same analgesic effect. The focus …
