The wave of next-generation sequencing data has arrived. However, many questions still remain about how to best analyze sequence data, particularly the contribution of rare genetic variants to human disease. Numerous statistical methods have been proposed to aggregate association signals across multiple rare variant sites in an effort to increase statistical power; however, the precise relation between the tests is often not well understood. Objective: The use of haplotypes to impute the genotypes of unmeasured single nucleotide variants continues to rise in popularity. Simulation results suggest that the use of the dosage as a one-dimensional summary statistic of imputation posterior probabilities may be optimal both in terms of statistical power and computational efficiency; however, little theoretical understanding is available to explain and unify these simulation results. In our analysis, we provide a theoretical foundation for the use of the dosage as a one-dimensional summary statistic of genotype posterior probabilities from any technology. Stroke is one of the leading causes of death and disability, with major global public health implications. Stroke ranks No. 4 among all causes of death, behind heart disease, cancer, and chronic lower respiratory disease (CLRD). Stroke accounts for almost 1 of every 18 deaths in the United States. Women accounted for 60.6% of stroke deaths. Death certificate data show that the mean age at stroke death was 79.6 years; males had a younger mean age (76.3) than females.

Distillers dried grains (DDGS), the coproducts of corn ethanol production, were used as a textile sizing agent on cotton, polyester and polyester/cotton blends in an effort to find inexpensive and biodegradable alternatives to sizing agents such as poly(vinyl alcohol) that are currently used. Although DDGS is an inexpensive, biodegradable and abundant co-product, it has limited industrial applications. DDGS is a mixture of carbohydrates, proteins and oil which are used as sizing agents or as size additives. The effects of DDGS extraction conditions on sizing evaluation parameters such as fiber adhesion, film properties, viscosity and fabric abrasion were studied in comparison …

Various characterizations of the Beta Weibull Geometric distribution of Bidram et al. (2011), the New Generalized Exponential distribution of Bidram et al. (2012), and the Gamma Exponentiated Weibull distribution of Pinho et al. (2012) are presented. These characterizations are based on: (i) a simple relationship between two truncated moments (ii) certain functions of the nth order statistic, and (iii) certain functions of the 1st order statistics.

This paper examines the predictive power of the Scholastic Aptitude Test (SAT) for Macalester students’ college success and academic choices. We use linear regression to study whether the SAT can predict students’ first year or four-year grades. Using Kullback-Leibler divergence and classification trees, we also examine the SAT’s predictive ability for other aspects of students’ academic experience, for example, major selection, or academic division of study. After controlling for major and course level, we find that the SAT does not explain a large proportion of the variability in Macalester students’ college success. Next-generation sequencing (NGS) technology has become a prominent tool in biological and biomedical research. However, NGS data analysis, such as de novo assembly, mapping and variants detection is far from maturity, and the high sequencing error-rate is one of the major problems. .

To minimize the impact of sequencing errors, we developed a highly robust and efficient method, MTM, to correct the errors in NGS reads. This dissertation aims to address two problems in regression
analysis. One problem is the model selection and robust parameter estimation in high dimensional linear regressions. The other is concerning developing a robust and efficient estimator in nonparametric regressions.
In Chapter 1, we introduce the robust and efficient regression analysis, discuss those two interesting problems and our motivations, and present several exciting results.

The first part of my dissertation demonstrates that a modified simulated annealing algorithm can successfully determine highly efficient D-optimal designs for second order polynomial regression for a variety of correlated error structures.

In the second part, I solved weak universal optimal block designs for the nearest neighbor correlation structure and multiple block sizes, for the hub correlation structure with any block size, and for circulant correlation with odd block size.

For over twenty years, the space physics community has studied a hypothesis of global cooling in the thermosphere, attributable to greenhouse gases. Functional data analysis (FDA) has grown into a substantial field of statistical research, with new methodology, numerous useful applications and interesting novel theoretical developments. My dissertation focuses on the empirical properties of functional regression models and their application to financial data. We start from testing the empirical properties of forecasts with the functional autoregressive models based on simulated and real data. The Negative Hypergeometric distribution represents waiting times when drawing from a finite sample without replacement. It is analogous to the negative binomial, which models the distribution of waiting times when drawing with replacement. Even though the Negative Hypergeometric has applications it is typically omitted from textbooks on probability and statistics and is not generally known. The main purpose of this thesis is to derive expressions for the mean and variance of a new application of the Negative Hypergeometric to gaming and gambling. Motivated by modern observational studies, we introduce a class of functional models that expands nested and crossed designs. These models account for the natural inheritance of correlation structure from sampling design in studies where the fundamental sampling unit is a function or image. Inference is based on functional quadratics and their relationship with the underlying covariance structure of the latent processes. A computationally fast and scalable estimation procedure is developed for ultra-high dimensional data. Methods are illustrated in three examples: high-frequency accelerometer data for daily activity, pitch linguistic data for phonetic analysis, and EEG data for studying electrical brain activity …
