The Dynamic Nature And Biophysical Characterization Of Isu1, Fe-S Cluster Assembly Scaffold Protein In Saccharomyces Cerevisiae, And Its Significance To Human Disease, Brianne Elizabeth Lewis

Wayne State University Dissertations

Mitochondrial Fe-S cluster biosynthesis is accomplished within yeast utilizing the biophysical characteristics of the “Isu1” scaffold protein. As a member of a highly homologous protein family, Isu1 has sequence conservation with orthologs and a conserved ability to assemble [2Fe-2S] clusters. Regardless of species, scaffold orthologs can exist in both “disordered” and “structured” conformations and is directly related to conformations utilized during Fe-cofactor assembly. During assembly, the scaffold directs the delivery and the utilization of both Fe(II) and sulfide substrates in order to produce [2Fe-2S] clusters, however Zn(II) binding can alter the activity of the scaffold with stabilizing the protein in …

Photoluminescent Zirconium Complexes For Visible Light Photoredox Catalysis, Yu Zhang Jan 2019

Photoluminescent Zirconium Complexes For Visible Light Photoredox Catalysis, Yu Zhang

Graduate Theses, Dissertations, and Problem Reports (ETD)

Early transition metals like zirconium are attractive candidates for the design of luminescent molecules based on earth-abundant elements. Here, a series of zirconium complexes carrying two pyrrolyl-based ligands have been synthesized and characterized. These complexes are photoluminescent upon excitation with visible light and exhibit remarkably long emission lifetimes of hundreds of μs in solution at room temperature. Computational studies using time-dependent density functional theory (TD-DFT) were conducted to identify the nature of the luminescent excited states as mixed intra-ligand/ligand-to-metal charge-transfer (ILCT/LMCT) states. The electrochemistry studies revealed multiple reversible redox events for each zirconium complex. Furthermore, the excited state of the …

Experimental And Modeling Investigation Of The Oh-Initiated Oxidation Of Semi-Solid And Aqueous Saccharide Aerosols, Hanyu Fan

Graduate Theses, Dissertations, and Problem Reports (ETD)

My research focuses on investigating the impact of moisture-induced and oligomer-induced viscosity changes on OH-initiated oxidation of semi-solid aerosols, and the role of gas-liquid interfaces in regulating aqueous aerosol chemistry. Saccharides, which are a major constituent of aqueous atmospheric aerosols, are chosen as model molecules to form highly oxygenated organic aerosols. The experiments are performed using an atmospheric pressure flow-tube reactor with both online VUV-AMS (Vacuum-Ultraviolet Aerosol Mass Spectrometer) and offline GC-MS analysis techniques. The decay rates of saccharide are determined by measuring the loss signal of saccharide in the particle phase as a function of OH exposure (time-integrated total concentration …

The functionalization of boron-based bioconjugates has long been studied for their potential use as a drug candidate for boron neutron capture therapy (BNCT). Also, magnetic nanoparticles have received greater attention due to their interesting magnetic properties and behaviors for selectively guiding them with an external magnetic field through the blood. A combination of the two ideas gives the framework for nanotechnology that has the potential to revolutionize drug delivery systems. This thesis will focus systematically on individual elements required for the many moving pieces required for magnetic drug targeting.

The research discussed in this thesis is divided into three parts. …

Thermostability And Excision Activity Of Polymorphic Forms Of Hogg1, Kathryn Mouzakis Jan 2019

Thermostability And Excision Activity Of Polymorphic Forms Of Hogg1, Kathryn Mouzakis

Chemistry and Biochemistry Faculty Works

Objectives: Reactive oxygen species (ROS) oxidize guanine residues in DNA to form 7,8-dihydro-oxo-2′-deoxyguanosine (8oxoG) lesions in the genome. Human 8-oxoguanine glycosylase-1 (hOGG1) recognizes and excises this highly mutagenic species when it is base-paired opposite a cytosine. We sought to characterize biochemically several hOGG1 variants that have been found in cancer tissues and cell lines, reasoning that if these variants have reduced repair capabilities, they could lead to an increased chance of mutagenesis and carcinogenesis. Results: We have over-expressed and purified the R46Q, A85S, R154H, and S232T hOGG1 variants and have investigated their repair efficiency and thermostability. The hOGG1 variants showed …

Green Chemistry In United States Science Policy, Emily Jarvis

Chemistry and Biochemistry Faculty Works

Although optimizing each of the Twelve Principles of Green Chemistry is necessary for achieving the greenest possible chemistry in the lab, there are additional creative ways to promote green chemistry not encompassed by these. Communicating to society the importance of green chemistry beyond the laboratory is essential. Of course, doing this effectively presents distinct challenges to those encountered when designing chemical syntheses. Here, I discuss several observations from my time working in federal science policy as an American Association for the Advancement of Science/American Chemical Society Congressional Science Fellow in the United States Senate. I suggest a practical science policy …

A fourth of the world’s population lack access to safe water, thus the need for a more effective water treatment is imperative. Interest in silver nanoparticles (AgNPs) has grown in the last decade. Unlike chlorine, AgNPs do not form disinfection by products (DBPs), making them a prime candidate for drinking water treatment. The main aim of this study was to compare the antibacterial activity of electrochemical silver nanoparticles (eAgNPs-f) of ~5 nm in diameter against well-established pathogens: Escherichia coli (E. coli), Klebsiella variicola (K. variicola), and Pseudomonas aeruginosa (P. aeruginosa) to chlorine and Ag+ for drinking water. This was achieved …

Nanostructured Materials For Photocatalysis, Water Treatment And Solar Desalination, Hiran D. Kiriarachchi

Theses and Dissertations

Maintaining a constant supply of clean drinking water is among the most pressing global challenges in our time. About one-third of the population is affected by the water scarcity and it can only get worse with climate change, rapid industrialization, and the population growth. Even though nearly 70 percent of the planet is covered by water, the consumable freshwater content is only 2.5 percent of it. Unfortunately, the accessible portion of it is only 1 percent. Even so, most of the freshwater bodies are choked with pollution. Considering the vast availability of saline water on the planet and the increasing …

Explosive Residue Transfer From Various Explosive Ordinance Disposal (Eod) Render Safe Procedures (Rsp), Joseph A. Stein Jan 2019

Explosive Residue Transfer From Various Explosive Ordinance Disposal (Eod) Render Safe Procedures (Rsp), Joseph A. Stein

Master of Science in Forensic Science Directed Research Projects

Before an IED is sent to a laboratory for analysis, it needs to be rendered safe if it did not already initiate. Render safe procedures (RSPs) include utilizing a percussion-actuated non-electric (PAN) disrupter or a mineral water bottle disrupter. Each disrupter utilizes explosives to render the device safe by breaking open the container or disrupting the fuzing system. However, the same explosives used in the RSP are also used by criminals in IED construction. As such, the explosives used in the RSP can cause problems with the interpretation of the results from forensic analysis of the IED fragments. Compounds of …

Method Considerations For Compound Identification In Complex Mixtures Using Electrospray Ionization Ultrahigh Resolution Mass Spectrometry, Tyler Leverton

Dissertations, Master's Theses and Master's Reports

Biomass burning aerosols are highly complex organic mixtures of thousands of components with consequences for global climate. Complex mixture component identification requires accurate mass measurement capability to separate components on a milli-Dalton scale, frequently using ultrahigh resolution mass spectrometry with electrospray ionization. Certain sample preparations and the ionization process may introduce artifacts that obscure the composition of the sample. Two method considerations were explored using isotopically labeled 15NH4OH and MeOH-d3 to track artifact formation in biomass burning samples. Informatics techniques and a custom molecular formula assignment software were used to identify the isotopic atoms in artifact …

Synthesis Of Oligodeoxynucleotides Containing Sensitive Electrophiles, Shahien Shahsavari Jan 2019

Synthesis Of Oligodeoxynucleotides Containing Sensitive Electrophiles, Shahien Shahsavari

Dissertations, Master's Theses and Master's Reports

Oligodeoxynucleotides (ODNs) containing electrophilic groups are useful in many studies including antisense drug development and DNA/protein interaction. Due to the use of strong nucleophiles for cleavage and deprotection, traditional ODN synthesis methods are not suitable for their preparation. To solve this problem, a new ODN synthesis technology using the 1,3- dithiane-2-yl-methoxycarbonyl (Dmoc) function as protecting groups and linker has been developed. Furthermore, Dmoc-derivatives were developed to demonstrate the feasibility of the technology. The Dmoc and Dmoc derivative functions are stable under all ODN synthesis conditions using the phosphoramidite chemistry. Upon oxidation of the sulfides in them, because of the drastically …
