Dr. Dave Berkowitz was born in Urbana, Illinois. He earned his bachelor's degree in chemistry from the University of Chicago, where he did undergraduate research. Next, he earned his doctorate from Harvard University. However, his advisor, Steve Benner, moved from Harvard to ETH Zurich, also called the Swiss Federal Institute of Technology. Dave accompanied him to Zurich, where he completed his dissertation work. When Dave returned to the U.S., he did a postdoc with the legendary Samuel Danishefsky at Yale University. In 1991, he joined UNL as an assistant professor of chemistry and quickly rose through the ranks. Dave is …

Net Carbonyl Ene Reactions Of Olefins With Aldehydes, Alexandra Mock

Aldehydes and olefins react in the presence of base and TMSOTf to yield homoallylic alcohols. In this net ene reaction, TMSOTf acts as a Lewis acid to activate the aldehyde, leading to a strong oxygen-silicon bond that acts as a driving force in the reaction. In the proposed mechanism, the α-methylstyrene attacks the carbonyl carbon of the TMSOTf-activated aldehyde to produce a carbocationic intermediate. Elimination using a relatively bulky base (2,6-lutidine) provides the homoallylic alcohol. This reaction has proven robust in terms of the aldehyde and olefin scope.

Radical polarity is one of the key factors in studying nitrogen-centered radicals’ reactivity, and the functional groups attached to the nitrogen atom are of vital importance. Among different types of nitrogen-centered radicals, aminium radical and metal aminium radical are electrophilic due to the positive charge on the intermediate. The enormous importance of aromatic rings in organic molecules cannot be understated. Aromatic rings are present in the majority of pharmaceuticals, agrochemicals, and consumer products. The way in which these rings are arranged and substituted results in a molecule’s unique properties. Because of this, the current limitations of the chemical methods used to synthesize these molecules results in limited structural diversity and thus missed opportunities for the development of value-added products. This research explores the application of mechanochemistry in the bottom-up synthesis of nanographene, specifically focusing on the mechanochemical Diels-Alder (DA) reaction. The study investigates the effect of different reaction conditions and dienophiles on the mechanochemical DA reaction, aiming to optimize reaction efficiency and yield using a ball mill. The initial trials with a sterically hindered dienophile (2) and an electron-deficient diene experienced repeated failure, while reactions using dienophile (B), a terminal alkyne showed more promising results. Throughout Earth’s history, organic molecules from both abiogenic and biogenic sources have been buried in sedimentary rocks. Most of these organic molecules have been significantly altered by geologic processes through deep time. Nonetheless, the nature and distribution of those ancient fragmentary organic remains have the potential to reveal diagnostic biomolecular information after billions of years of burial. Here, we analyzed 406 fossil, modern biological, meteoritic, and synthetic samples using pyrolysis gas chromatography and mass spectrometry. We explored these analytical data via supervised machine-learning methods to discriminate samples of biogenic vs. abiogenic origin, plant vs. animal phylogenetic affinity, and photosynthetic vs. …

The carbon scaffold of many drugs and natural products contain multiple stereogenic centers bearing heteroatoms. As a result of this, chemists have long sought methods to efficiently install these multi-heteroatom functionalities. Reductive coupling reactions have been extensively studied over the past decades, and the allylation of carbonyls via the reductive coupling approach has been a key method for generating chiral allylic alcohols. Arynes hold immense potential as reactive intermediates in organic synthesis as they engage in a diverse range of mechanistically distinct chemical reactions. However, the poor functional group compatibility of generating arynes or their precursors has stymied their widespread use. Here, we show that generating arynes by deprotonation of an arene and elimination of an “onium” leaving group is mild, efficient and broad in scope. This is achieved by using aryl(TMP)iodonium salts (TMP = 2,4,6-trimethoxyphenyl) as the aryne precursor and potassium phosphate as the base, and a range of arynophiles are compatible. Spin polarized photocathodes are key to the future operation of electron accelerators such as the ones at Thomas Jefferson National Accelerator Facility and Brookhaven National Laboratory. Currently, these photocathodes come in short supply due to limited production by molecular beam epitaxy. By developing a process to implement similar structures using metal organic chemical vapor deposition, the availability of these devices can be increased. Herein, we examine data on DFW% (i.e., % of D and F grades and withdrawals) in our general chemistry courses over roughly a quarter-century period, focusing on sections taught by the authors. Keto-enol tautomerization (KET) is a fundamental process impacting a range of molecular phenomena in organic and biochemistry. However, the accurate computation of solution-phase KET energies remains a challenge, even for prototypical acetone.

In Part I, keto-enol tautomers of acetone were incorporated into solvent clusters that interact via uninterrupted, cyclic hydrogen-bonding (UCHB) networks. An empirical model was created to predict accurate KET energies, Etaut, of simple carbonyl compounds. Based on the availability of experimental data and structural simplicity, acetone was selected as a prototype. Arynes are ephemeral molecules that are yet to be fully exploited in synthetic strategy because they are often challenging to access. These reactive intermediates participate in a myriad of reactions that conventional chemistry is incapable of, making them extremely useful for the synthesis of important molecules like pharmaceuticals and agrochemicals. While classical approaches for synthesizing arynes employ cheap starting material, extremely harsh reagents are necessary to generate the aryne. This research visualized the effect of different substituents, which could be either activators or deactivators on an aromatic ring for alcohol oxidants using Chromium(VI). The color of chromic acid changes from orange to blue gradually, providing a method to visualize the rate. Rate constants were used as a quantitative method to evaluate the rate.

In the presence of trimethylsilyl trifluoromethanesulfonate (TMSOTf) and 2,6-lutidine, α,α-disubstituted aldehydes condense with electron-rich aromatic aldehydes to yield β, β- disubstituted styrenes. More electron-rich aromatic aldehydes react more rapidly and in higher yield. Preliminary results suggest that the reaction may proceed via the ionization and formal deformylation of an aldol intermediate.

Due to their well-defined crystalline structures and excellent chemical and thermal stabilities, COF materials are considered promising candidates in applications such as gas adsorption, catalysis, and energy storage.Chapter 2: A new covalent organic framework (COF) based on imine bonds was assembled from 2-(4-formylphenyl)-5-formylpyridine and 1,3,6,8-tetrakis(4-aminophenyl)pyrene, which showed an interesting dual-pore structure with high crystallinity. This project investigates the synthesis of a DD-π-AA (dual donor/dual acceptor) organic dye as a potential sensitizer for dye-sensitized solar cells (DSCs). The design of this dye was based off previous research that found dual donor/dual acceptor dyes exhibited promising results when used in a DSC. The donor groups, acceptor groups, and π-bridge were chosen for their stability, ability to absorb in the near-infrared (NIR) region, and intramolecular charge transfer (ICT) abilities.

Natural products remain the most common source for drug leads, although diversity-oriented synthesis of unnatural complex molecules is gaining momentum. Our lab has been developing experimentally simple and straightforward approaches to complex molecular architectures via photochemical reactions involving a minimal number of experimental steps. In order to achieve this, photoprecursors were assembled via high yielding reactions. Next, we deployed photoinduced cycloadditions to create complex 3-dimentional structures. Lastly, with this obtained primary photoproduct, post-photochemical transformations were used to further grow its scaffold complexity.

The data obtained from an NMR spectrometer are typically shown in the form of a spectrum, which is then analyzed by an analytical chemist. Natural products produced by plants, animals, and microorganisms are of significant importance in chemistry for being major sources of molecular scaffolds essential for new drug discovery. Mansouramycins are cytotoxic bioactive compounds containing isoquinoline skeletons with substituents on the quinone and pyridine rings that may be difficult to synthesize. Synthetic processes exist for compounds B and D, however, there exists an absence of simple synthetic approaches to all four alkaloids.

The benzenoid core is a frequent and irreplaceable component of biologically active molecules. Structure function relationship of these compounds has been broadly analyzed and presented in numerous literature reports. Herein, we augmented existing knowledge by a broad analysis of the benzenoid substitution patterns in small molecule active pharmaceutical ingredients approved by the FDA. Our findings reveal a strong correlation that the most represented substitution patterns are those achieved via SEAr. Despite the rapid development of methods to functionalize benzenoid rings, the lack of synthetic methodologies to access sterically and electronically unfavorable positions is highlighted. This thesis describes the isolation, structure elucidation, and synthesis of natural products from marine cyanobacteria. A crude extract from a cyanobacterium collected in Curacao showed selective affinity for the dopamine D5 receptor in a screen against a panel of CNS receptors. Due to the high similarity of the D5 and D1 receptor, to date there are no known ligands that differentiate them. Attempts to purify the compound responsible for this affinity led to the isolation of the known compound caylobolide A.
