A contentious element in the traditional method of calculating accumulated oxygen deficit (AOD) is the assumption that the oxygen demand remains constant throughout a bout of exercise. The purpose of this study was to investigate the appropriateness of this assumption. Twelve women and eight men volunteered for the study and completed cycle ergometer tests that resulted in exhaustion after ~4 min and ~8 min. The objective of this study was to compare the effects of very high supervision (VHS-RT) versus high supervision (HS-RT) ratio resistance training (RT) on irisin, brain-derived neurotrophic factor (BNDF), muscle strength, functional capacity, and body composition in elderly women. Participants performed daily undulating periodized RT over 16 weeks with two different supervision ratios: VHS-RT at 1:2 (supervisor/subject) or HS-RT at 1:5. Although synthetic testosterone-boosting pharmaceuticals have gained wide use, recent data suggests that vitamin D, ingested orally, may positively impact serum testosterone. Background: This study examined the motor unit (MU) control strategies for non-fatiguing isometric elbow flexion tasks at 40% and 70% maximal voluntary isometric contraction. Methods: Nineteen healthy individuals performed two submaximal tasks with similar torque levels: contracting against an immovable object (force task), and maintaining the elbow joint angle against an external load (position task). Recent studies have shown relationships between specific salivary microRNA levels and concussions, but it is unknown if this is due to concussive forces or physical exertion. Skeletal muscle contributes to ~40% of total body mass and has numerous important mechanical and metabolic roles in the body. Skeletal muscle is a major site for glucose disposal following a meal. Consequently, skeletal muscle plays an important role in postprandial blood glucose homeostasis. Sleep loss is well known to impair cognitive function, immunological responses, and general well-being in humans. However, sleep requirements in mammals and birds may vary dramatically, especially with changes in environment. In circumpolar regions with continuous light, sleep requirements may be little, particularly in breeding birds. The effects of sleep loss on several fitness parameters were examined in two species of Arctic-breeding passerine birds: Lapland longspurs (Calcarius lapponicus) and snow buntings (Plectrophenax nivalis). Though there are several to choose from, the Bruce protocol is the most commonly utilized treadmill protocol when assessing maximal oxygen consumption (VO2max). Originally developed for cardiac patients, the Bruce protocol may not be an appropriate treadmill protocol for young or trained individuals with higher levels of cardiorespiratory fitness. The steep grade used often leads localized muscular fatigue, which may cause participants to end the test prior to reaching their true VO2max. The aim of this thesis is to investigate the effect of physical activity and the administration of s-equol on rates of neurogenesis in HIV-1 transgenic rats. HIV-1 is a neurodegenerative disease that leads to the wide range of cognitive impairment described as HAND. Neurogenesis is a newly discovered process that involves the birth of new neurons during our adult life within the dentate gyrus of the hippocampus. Therefore, inducing neurogenesis may be a proper therapeutic that can mitigate some of the cognitive destruction seen as a result of HIV-1 infection. Prediction of energy expenditure allows for calculation of appropriate energy requirements, which is especially important for athletes. Resting metabolic rate (RMR) is the greatest contributor to total daily energy expenditure (TDEE) and is typically measured via indirect calorimetry. Indirect calorimetry is not always available, which results in the need for predictive equations. Most predictive equations have been developed with participants resembling the general population and have not been found to be appropriate for athletes, as they may incorrectly predict RMR due to the unique differences of body composition between athletes and the general population. Adverse pregnancy outcomes (APOs) are believed to be caused by poor placental formation. APOs can be characterized by elevated levels of a circulating Fms-like tyrosine kinase biomarker called Flt-1, produced by the placenta when its blood vessels are shallow or inadequately formed. Flt-1 acts directly on the maternal endothelium to impair vascular function during pregnancy and contributes to maternal features of APOs. I will be exploring the impact of the large protein Titin on heart failure with a preserved ejection fraction (HFpEF). It was hypothesized that myocardial Titin plays a significant role in the progression of HFpEF through isoform, N2BA and N2B, changes. It was hypothesized that an increase in the quantity of N2B Titin, less compliant, and a decrease in N2BA Titin, more compliant, will be observed as HF progresses, contributing to an increase in passive stiffness involved in heart contraction. I will cover a detailed report on the role of Titin in HFpEF and propose research methods to explore further.

Lactic acid is a vital indicator for shock, trauma, stress, and exercise intolerance. It is a key biomarker for increases in stress levels and is the primary metabolically produced acid responsible for tissue acidosis that can lead to muscle fatigue and weakness. During intensive exercise, the muscles go through anerobic metabolism to produce energy. This leads to decreases in the blood flow of nutrients and oxygen to the muscles and increases in lactate production, which in turn cause lactic acidosis. Women are often excluded from research studies interested in changes in muscle size and strength due to confounding influence that the menstrual cycle may have on these variables. Although there is considerable data showing that strength may fluctuate throughout the menstrual cycle, the thesis that muscle size changes throughout the cycle is based on the premise that changes in body water throughout the menstrual cycle may influence the size of the muscle. Glucose-stimulated insulin secretion (GSIS) from the pancreatic β-cell in response to elevated levels of glucose is controlled by a variety of signals including intracellular calcium and nucleotides such as cAMP, ATP and GTP. These cellular signals are responsible for activation of specific kinases that mediate phosphorylation of key exocytotic proteins that lead to GSIS. In the context of protein kinases, mixed-lineage kinases (MLKs) have been implicated in an assortment of cellular functions, including cell proliferation and apoptosis.
