The Timing Of Pubertal Onset Affects Adults Anxiety Behavior In A Sex-Specific Manner, Evan Raubenolt
Honors Projects
This study aims to look at the relationship between the timing of pubertal onset and its long-term effects on adult anxiety behavior in rats. Though the topic of pubertal onset has been frequently discussed in developmental research, its exact effects have been inconsistent. Human models have shown that early and late maturing males are associated with a notable increase in adult anxiety behavior, while in rat models, similar sex-specific effects regarding anxiety have been demonstrated, though those studies focus on a causal relationship between exposure to stress and age of pubertal onset. This study first aims to examine that relationship …

Discovery Undergraduate Interdisciplinary Research Internship
Per- and polyfluoroalkyl substances (PFAS) are highly persistent synthetic chemicals that are now ubiquitous in the environment. Due to PFAS stability leading to its environmental persistence, there is concern regarding potential long-term health effects as PFAS accumulation occurs in humans from dietary and inhalation exposure routes. Chronic exposure may lead to adverse effects such as fertility issues or liver damage. Although a wide range of PFAS have been found in human samples, specifically human serum, there remains data gaps for what PFAS are present in human blood. Recent innovation in detection technology has led to the ability to more accurately …

Biological Sciences Faculty Research & Creative Works
Previously, we reported final-instar lepidopteran larvae exposed to low doses of imidacloprid, clothianidin, and thiamethoxam had arrest in pupal ecdysis, which is a novel adverse outcome for neonicotinoid insecticides. Since neonicotinoids disrupt acetylcholine signaling, we hypothesized that the excitatory neurotransmitter acetylcholine plays a critical role in regulation of pupal ecdysis, likely by modulating the release of peptides from crustacean cardioactive peptide (CCAP) neurons. In this paper, using two lepidopteran species, we undertook studies with five additional nicotinic acetylcholine receptor (nAChR) agonists and three muscarinic acetylcholine receptor (mAChR) agonists to hypothesize the putative nAChR subunits that mediate pupal ecdysis. We also …

The demand for processed foods has led to increased use of synthetic phenolic antioxidants (SPAs), notably tert-butylhydroquinone (TBHQ), to inhibit lipid oxidation and prolong shelf life. While TBHQ is deemed safe at regulated levels, concerns arise from studies indicating potential adverse effects such as oxidative stress, genotoxicity, and impaired immune function. To investigate TBHQ's immunomodulatory effects, RAW 264.7 murine macrophages were pre-treated with TBHQ and subsequently stimulated with lipopolysaccharide (LPS) or polyinosinic-polycytidylic acid (poly I:C, PIC) to simulate bacterial or viral challenges. Results demonstrated that TBHQ at high non-cytotoxic doses reduced nitric oxide and IL-10 production while promoting phagocytosis and …

Fast & Forensic: Rapid Detection & Quantitation Of 30 + Emerging Novel Psychoactive Substances In Hair Using Lc-Ms/Ms, Seokjin Hwang
Student Theses
The continuous emergence of novel psychoactive substances (NPS) poses a significant challenge for forensic and clinical toxicologists, due to their structural diversity, rapid turnover on the illicit drug market, and limited toxicological information. Hair is a powerful matrix for retrospective drug exposure assessment and monitoring NPS worldwide. Not only does it provide a broad window of detection but also a simple sample exchange amongst laboratories. However, the extraction of multi-class NPS in a single method remains an analytical challenge. Therefore, there is an urgent need for extraction techniques and analytical methods that are both rapid and capable of multi-class quantitation …

Characterizing Hexavalent Chromium Neurotoxicity Across Laboratory Models., Samuel Thomas Vielee
Electronic Theses and Dissertations
Hexavalent chromium [Cr(VI)] is a ubiquitous environmental pollutant and potent toxicant. Exposure to Cr(VI) in drinking water is primarily associated with cancers, inflammation, and hepatotoxicity; however, recent studies have identified Cr(VI) as a neurotoxicant in rodent and cell culture models. The literature suggest chromium accumulates in the brain, induces widespread neurodegeneration, and alters rodent behaviors. Despite these data, we lack a detailed and consistent assessments of Cr(VI) neurotoxicity across models, and interactions between aging and Cr(VI) neurotoxicity are not reported in the literature. In this dissertation, we characterize Cr(VI) neurotoxicity across 3 laboratory models: Sprague-Dawley rats, Caenorhabditis elegans, and …

Exposure To Pm2.5 And Aging., Daniel Chris Gomes
Electronic Theses and Dissertations
Inhalation of fine airborne particulate matter (PM2.5) is an environmental risk factor for numerous health disorders, including cardiovascular diseases (CVD). While several studies have shown that PM2.5 exposure can induce oxidative stress, DNA damage, and inflammation, the underlying mechanisms that translate these effects into systemic pathology remain poorly understood. Some recent studies have shown that PM2.5 exposure can lead to telomere shortening in leukocytes, but the generality of this outcome and contribution to endothelial dysfunction has not yet been fully explored. We hypothesized that PM2.5inhalation accelerates cellular aging and senescence in both circulating peripheral …

Journal of Food and Drug Analysis
Plant-derived nanovesicles (PDNVs) offer a promising, biocompatible therapeutic platform. In this study, we isolatednanovesicles from protocorm-like bodies of Dendrobium officinale (PLBs-NVs) and evaluated their efficacy in dry eyedisease (DED) models. PLBs-NVs protected human corneal epithelial cells against hyperosmotic stress, supportedwound repair, and inhibited inflammation in vitro. In a desiccating stress-induced DED mouse model, topical PLBs-NVstreatment significantly restored tear secretion, reduced corneal epithelial damage, and lowered levels of proinflammatorycytokines. These results suggest that PLBs-NVs are a safe, natural, and effective therapeutic strategy for DED, sup-porting their potential for future clinical translation in ocular surface disorders.

Journal of Food and Drug Analysis
Jujube seed extract (JSE), rich in polyphenols and flavonoids, exhibits neuroprotective and sleep-enhancing properties. In vitro, JSE protected HT22 cells against oxidative stress by activating the Nrf2/HO-1 pathway and upregulatingantioxidant enzymes. In vivo, JSE increased non-REM sleep and delta wave activity in pentobarbital- and caffeine-induced sleep models. Spinosin contributed to these effects. Long-term JSE administration upregulated GABAA, GABAB, and 5-HT1A receptors in brain tissue. These findings suggest that JSE offers dual benefits in mitigating oxidativestress and promoting sleep through antioxidant defense and neurotransmitter modulation, supporting its potential as a natural therapeutic agent.

Efforts to remediate such spills have evolved over the years through the introduction of various chemical response agents that efficiently and effectively control the spread of oil. Chemical herding agents and dispersants, which can contain and control the oil spread, are promising additions to the spill response toolbox as potential alternatives to other response methods. These response agents are designed to be biodegradable and minimize potential harm to aquatic ecosystems. However, …

Inorganic arsenic (As), specifically arsenate (As(V)) and arsenite (As(III)) are well-documented toxicants to a range of marine organisms. However, their specific impacts on coral reef species remain largely unstudied. This study is the first to assess the acute and subacute toxicity of inorganic arsenic on two threatened scleractinian coral species, Acropora cervicornis and Orbicella faveolata. Corals were exposed for 96 hours to five analytically verified concentrations and a negative control of either arsenate or arsenite. Toxicity was evaluated through three endpoints: mortality (LC50), coral condition (EC50), and photosynthetic efficiency (IC50). The lethal concentrations to result in 50% mortality (LC50) …
