From 1941 to 2003, the United States Navy and North Atlantic Treaty Organization (NATO) used Vieques, Puerto Rico, for military training, which led to long-term pollution from metals, organic chemicals, explosives, and per- and polyfluoroalkyl substances (PFAS). While earlier research found these contaminants in the environment and in living organisms, their combined effects on local ecosystems remain poorly understood.
This dissertation used a Genomic-Based Environmental Health Assessment (GEHA) framework to study the ecological risks from past military pollution in Vieques. Environmental pollutants can disrupt normal developmental processes, leading to neurobehavioral and developmental abnormalities. Toxicity tests are important for evaluating environmental contaminants; however, standard assays are often time- and cost-intensive and typically require large numbers of vertebrate animals. These limitations highlight the need to reduce reliance on vertebrate testing and to develop New Approach Methodologies that provide comparable results while improving efficiency and cost-effectiveness. Spiders are one of the most diverse groups of organisms on the planet with over 53,000 species described as of 2026. They provide a suite of ecosystem services including their regulation of problematic pests in agricultural systems, appearing in high biomasses across a breadth of ecological niches. While beneficial as pest regulators, they are also susceptible to the same management techniques as pest organisms and are especially vulnerable to chemical control. Micro-and Nanoplastics (MNPs) are ubiquitous biological pollutants that have been identified throughout a variety of biological samples. MNPs have been detected in several human tissues, such as the brain, kidneys, and liver. Lung tissue remains understudied for MNP accumulation despite the large surface area and clear path for exposure. Using Pyrolysis-Gas Chromatography Mass Spectrometry (Py-GC/MS), human lung tissue was evaluated for 12 different polymers, enabling quantification of total plastic per gram of tissue to be identified. Francisella tularensis is a highly infectious intracellular bacterium and the causative agent of tularemia, with pulmonary infection resulting in severe disease and high mortality if untreated. While protective immunity to F. tularensis has been largely attributed to CD4⁺ T cell–mediated IFN-γ responses, the contribution of γδ T cells to mucosal immunity and vaccine-induced protection remains poorly defined. The objective of this study was to characterize the role of γδ T cells during primary pulmonary infection with F. tularensis live vaccine strain (LVS) and following intranasal immunization with inactivated F. tularensis (iFt) prior to LVS challenge.

This thesis examines the disproportionate burden of lead exposure carried by low income, POC communities. The systemic nature of this problem is a symptom of a longstanding legacy of environmental injustice in the United States. Decades of federal neglect are reflected in the higher statistics of lead exposure and poisoning in predominantly black communities. While it is understood that lead exposure poses a serious threat to physical health and early cognitive development, there is a discouraging lack of urgency to remove the toxin from non-wealthy communities. The Environmental Influences on Child Health Outcomes Fetal Growth Studies (ECHO-FGS) birth cohort data was used to identify chemical mixtures associated with adverse outcomes. This biomonitoring study associated specific chemical mixtures in maternal serum with low birth weight or childhood obesity (Pearce et al., 2021). Mixture Profiles 1, 3, and 4, comprised of different observed concentrations of DDE, PCB153, BDE47, and PFOS were prioritized. Mixtures were tested to determine whether they perturbed mitochondrial metabolism and energy sensing pathways. This paper examines the hidden environmental, geopolitical, economic, and social costs of the cobalt supply chain. Fueled by the demand for battery technology and electric vehicles, cobalt has become an essential element to the green energy transition. With the largest share of global cobalt deposits, the Democratic Republic of the Congo bears a disproportionate share of the industry’s harms. Cobalt mining in the DRC is marked by environmentally degrading practices and inhumane working conditions, compromising Congolese health, safety, and well-being. This research was conducted to explore the possibility of constructing floating treatment wetlands (FTWs) without using plastic materials. FTWs are floating platforms that hold plants on the water’s surface, letting roots dangle within the water column, to help remove contaminants from contaminated water, especially in stormwater ponds. Many conventional FTWs are made with plastic components, such as mats and plastic cup containers, which can release microplastics into the environment over time. The US population faces simultaneous public health threats from both aging and environmental pollutants. Chief among these is hexavalent chromium [Cr(VI)], whose toxic effects in peripheral tissues are well-established but its effects on the brain are still understudied. Following previous results indicating hippocampal metals dyshomeostasis after Cr(VI) exposure, we aim to assess Cr(VI)’s impact on the blood-brain barrier (BBB), a key structure for brain protection. Pharmaceutical contaminants are increasingly prevalent in aquatic environments, yet their sublethal effects on ecologically relevant behaviors remain poorly understood, particularly in estuarine systems. Selective serotonin reuptake inhibitors (SSRIs), such as fluoxetine, are of particular concern due to the conserved role of serotonin in regulating behavior across vertebrates. Here, we investigated how acute exposure to environmentally relevant concentrations of fluoxetine influences anti-predator behavior in Gulf killifish (Fundulus grandis), a key estuarine fish commonly found in saltmarsh habitats. These sessile organisms are vulnerable to anthropogenic impacts, habitat degradation, climate change, and pollution, particularly Petroleum hydrocarbon contamination. Previous research has characterized coral species' sensitivity to hydrocarbons, using a continuous passive dosing system, but the specific histological impacts of petroleum hydrocarbons remain underexplored. Amphibians are experiencing population declines due to habitat loss, climate change, and diseases, particularly chytridiomycosis caused by the fungus Batrachochytrium dendrobatidis. Protection from this fungal infection stems from the cutaneous microbiome. Pesticides have been shown to affect amphibians in a variety of ways, including reducing survival, locomotor performance, and accumulation in body tissues. Mercury is a persistent global contaminant that biomagnifies through aquatic food webs, with dietary and environmental factors serving as the primary drivers of accumulation in fishes. Trophic structure and methylmercury dynamics in Walleye (ogaa; Sander vitreus) and Yellow Perch (asaawens; Perca flavescens) were investigated in the Upper and Lower Red Lake basins in Red Lake, Minnesota, during 2024-2025. Mosquitoes are a primary transmitter of vector-borne disease, and their larvae can thrive within shallow pools of stagnant water. Rainwater drainage ditches are a common sight in many communities, and in Louisiana, they are dually used as outlets for onsite residential wastewater discharge in neighborhoods disassociated from municipal sewage treatment. However, when discharge systems malfunction, the water becomes uninhabitable to mosquito predators, allowing mosquitoes to thrive. In the present study, the pulmonary toxicity of Er2O3 NPs was assessed by examining tissues and bronchoalveolar lavage fluid (BALF) from the lungs of Golden Syrian Hamsters following inhalation exposure. Neurological function is closely linked to both longevity and quality of life. While some degree of neurodegeneration occurs naturally with aging, pathological neurodegeneration leads to devastating functional decline. The mechanisms underlying these processes remain incompletely understood; however, neuroinflammation is recognized as a key driver of neuronal dysfunction and death. Emerging evidence indicates that neuroinflammatory cascades can precede and potentiate hallmark features of neurodegenerative diseases, including synaptic loss and cognitive decline observed in Alzheimer’s disease (AD). Perfluorooctane sulfonate (PFOS) is a persistent environmental pollutant associated with potential hepatoxic effects and other health risks. Despite its widespread distribution, the mechanisms underlying its toxicities remain to be fully understood. To investigate PFOS toxicology, my study utilized HepG2 and THLE-2 human hepatic cell models to replicate conditions reflecting PFOS accumulation in the liver. Cell viability, cell stress, and cell death assays were conducted to assess the toxicological influence of the chemical on both cell lines. Heavy metal contaminated mine tailings (“stamp sands”) from historic copper mining inhibit plant establishment and contribute to ongoing metal contamination. Biochar amendments may enhance plant growth and reduce metal mobility in highly permeable mine waste soils, but the efficacies of different application methods or biochar sources and composition are uncertain. For Carcharhinid species such as the bull shark (Carcharhinus leucas), the maternal offloading of nutrients is essential for pup development and growth, however, significant amounts of OCs are also transferred. As a top predator in aquatic food webs, bull sharks can potentially bioaccumulate high concentrations of pollutants such as dichloro-diphenyl-trichloroethane (DDT) that biomagnify through aquatic food webs. Pesticides and other agrochemicals have varying effects on spiders depending on their mechanism of action and dosage, ranging from behavioral changes and ataxia, to reproductive changes and physiological stress, to lethal non-target effects. Synthetic cannabinoids represent one of the most prevalent and high-risk classes of New Psychoactive Substances (NPS), continuously evolving in structure and potency. This study aims to look at the relationship between the timing of pubertal onset and its long-term effects on adult anxiety behavior in rats. Though the topic of pubertal onset has been frequently discussed in developmental research, its exact effects have been inconsistent. Human models have shown that early and late maturing males are associated with a notable increase in adult anxiety behavior, while in rat models, similar sex-specific effects regarding anxiety have been demonstrated, though those studies focus on a causal relationship between exposure to stress and age of pubertal onset. The demand for processed foods has led to increased use of synthetic phenolic antioxidants (SPAs), notably tert-butylhydroquinone (TBHQ), to inhibit lipid oxidation and prolong shelf life. While TBHQ is deemed safe at regulated levels, concerns arise from studies indicating potential adverse effects such as oxidative stress, genotoxicity, and impaired immune function. To investigate TBHQ's immunomodulatory effects, RAW 264.7 murine macrophages were pre-treated with TBHQ and subsequently stimulated with lipopolysaccharide (LPS) or polyinosinic-polycytidylic acid (poly I:C, PIC) to simulate bacterial or viral challenges. The continuous emergence of novel psychoactive substances (NPS) poses a significant challenge for forensic and clinical toxicologists, due to their structural diversity, rapid turnover on the illicit drug market, and limited toxicological information. Hair is a powerful matrix for retrospective drug exposure assessment and monitoring NPS worldwide. Hexavalent chromium [Cr(VI)] is a ubiquitous environmental pollutant and potent toxicant. Exposure to Cr(VI) in drinking water is primarily associated with cancers, inflammation, and hepatotoxicity; however, recent studies have identified Cr(VI) as a neurotoxicant in rodent and cell culture models. The literature suggest chromium accumulates in the brain, induces widespread neurodegeneration, and alters rodent behaviors. Despite these data, we lack a detailed and consistent assessments of Cr(VI) neurotoxicity across models, and interactions between aging and Cr(VI) neurotoxicity are not reported in the literature.
