Poly- and perfluoroalkyl substances (PFAS) are an anthropogenic group of chemicals known for their persistence in the environment and their potential to bioaccumulate. PFAS can bioaccumulate in largemouth bass (Micropterus nigricans), yet the effects of exposure on bass larvae are unknown. While largemouth bass are commonly raised for aquaculture, techniques for rearing bass which enable controlled experiments at known densities, a necessity for toxicity studies, are poorly understood. Furthermore, the live prey typically used in existing approaches are not suitable for PFAS studies due to high background concentrations. Regulatory toxicology depends primarily on animal testing, which limits throughput, mechanistic interpretability, and applicability to human-relevant endpoints. This dissertation presents computational approaches that integrate publicly available bioactivity data with machine learning to support chemical safety evaluation. Oral carcinogenicity was modeled using curated chemicals from the US Environmental Protection Agency’s Integrated Risk Information System (IRIS) and activity data from over 1,900 PubChem assays. Quantitative structure-activity relationship (QSAR) models trained on statistically prioritized assays demonstrated strong predictive performance and were applied to rank the carcinogenic potential of more than 16,000 compounds. Diabetic nephropathy (DN) is one dominating reason for death in diabetic patients, and its incidence is high. It has been reported that Yiqi Yangyin Tongluo prescription (YYTP) can relieve inflammation, and it owns better clinical effects in the treatment of DN. However, the molecular mechanisms of YYTP in the treatment of DN still keep unclear, and deeply investigations are needed. Following the observed significant improvements in overall survival and progression-free survival in clinical trials, the combination of atezolizumab and bevacizumab has been recommended as a first-line therapy for patients with unresectable hepatocellular carcinoma. Despite its clinical benefits, the high cost associated with this treatment poses a substantial challenge in routine practice in Taiwan. This study aims to assess the cost-effectiveness of atezolizumab plus bevacizumab in comparison to sorafenib monotherapy. The Ping-Chong-Jiang-Ni Formula (PCJNF), a compound of Traditional Chinese Medicine (TCM), has demonstrated remarkable clinical effectiveness and is widely utilized in the treatment of endometriosis (EMs). Previous studies have shown that PCJNF inhibits the proliferation and induces the apoptosis of ectopic endometrial stromal cells (EESCs) by modulating the JNK signaling pathway. Synthetic cathinones are a widely abused class of new psychoactive substances that pose significant challenges in forensic toxicology owing to their complex metabolic pathways and lack of verified reference standards. This study investigated the in vitro metabolism of nine methylenedioxy-substituted synthetic cathinones, including methylone and eight structural isomers, which were categorized into three groups: Group 1 (dimethylone, ethylone, and butylone), Group 2 (dibutylone, pentylone, and eutylone), and Group 3 (N-ethylpentylone and dipentylone). Chronic atrophic gastritis (CAG) is precancerosis with limited treatment options. This study investigated the therapeutic effects of velvet antler (VA), a traditional natural medicine, on CAG, with a focus on ferroptosis. In vivo experiments demonstrated the efficacy of VA in alleviating CAG, and network pharmacology analysis revealed its involvement in related terms, including inflammation, oxidative stress, and cell proliferation. Analyses further confirmed the modulation of ferroptosis by VA, particularly via the Keap1-Nrf2/HO-1/GPX4 pathway. In vitro assays supported these findings, demonstrating the ability of VA to reduce methylnitronitrosoguanidine-induced inflammation, oxidative stress, and ferroptosis in GES-1. Jiao Sanxian (JSX) consists of three traditional Chinese medicine including charred hawthorn, malt and Liu Shen Qu, they are also from Chinese medicine homologous food. It is commonly used to relieve functional dyspepsia (FD). The present study aims to explore the mechanism of JSX (granule), with the addition of prebiotics for the treatment of FD using mice model. The network pharmacology and molecular docking were used to forecast therapeutic targets for JSX. Pesticides are ubiquitous in aquatic environments, alongside numerous contaminants and other stressors that can result in combined exposure to target and non-target organisms. Nevertheless, traditional toxicity studies quantify the effects of these compounds at an individual level, and to a narrow range of traditional test organisms. Through this dissertation, I explored the presence and effects of commonly detected pesticides of concern in the Pacific Northwest in order to realistically quantify potential effects from combined stressors. Per- and polyfluoroalkyl substances (PFAS) are synthetic chemicals that raise significant toxicological concerns due to their environmental persistence and potential health hazards. This research examines the presence and concentration of selected PFAS compounds in human hair samples from volunteers in New York, comparing these results with global trends found in peer-reviewed research. Hair was selected as the biological matrix for its non-invasive collection method, ability to reflect exposure, and long-term chemical stability. Long-term treatment of patients with medications is associated with adverse drug reactions and continues to be a challenge. Sodium valproate is commonly used to treat bipolar disorder and epilepsy. It can induce adverse gastrointestinal effects, and its absorption may vary depending on the excipient used. Arsenic is a prevalent toxicant found to affect around 94 to 240 million individuals in over fifty countries who are exposed to levels above the EPA and WHO standard of 10 ppb. Lung cancer is the leading cause of cancer death worldwide. Hexavalent chromium [Cr(VI)] is a human lung carcinogen with widespread occupational and environmental exposure. Despite the known health risks, how Cr(VI) causes lung cancer remains unclear. This dissertation investigates the mechanisms of Cr(VI)-induced carcinogenesis. Cr(VI) causes DNA damage, specifically DNA double strand breaks, and inhibits homologous recombination repair, a high-fidelity repair pathway. Unrepaired DNA double strand breaks or those repaired through error-prone pathways, progress to chromosomal damage, and chromosome instability. Each year cancer affects nearly 20 million people worldwide and genetic differences across populations can impact cancer onset and progression. Specifically, tumors with high levels of HSF1, the master regulator of the cytoprotective heat shock response (HSR), are correlated with poor patient outcomes in multiple cancers such as prostate, breast, and melanoma. Subsequently, the development of pharmacological inhibitors of HSF1 represents a promising strategy for anticancer therapeutics. Using a luciferase-based transcriptional reporter, two small molecule libraries were screened for inhibitors of HSF1 expression in human embryonic kidney cells, yielding ten compounds that decrease HSF1 expression. The prevalence of vaping has rapidly increased. ECs are often perceived as less harmful than tobacco cigarettes. A recent meta-analysis showed electronic cigarette (EC) aerosols adversely affected the respiratory system, similar to cigarette smoke, with some effects specific to EC use. Failure to prevent these alterations could lead to life-long comorbidities, and predisposition to respiratory diseases, including COPD and squamous cell carcinoma. Lung cancer remains the leading cause of cancer death, often linked to smoking. However, smoking alone does not account for all cases. Exposure to environmental metals like hexavalent chromium [Cr(VI)] also contributes to lung cancer. Cr(VI) causes lung cancer by inducing chromosome instability (CIN), an early hallmark of its carcinogenic mechanism. While it has been established that Cr(VI) induces chromosome instability, the precise mechanisms underlying the chromosome instability are poorly understood. Pyridostigmine bromide (PB) is an organophosphate (OP) nerve agent prophylactic used to protect warfighters against chemical warfare agents. OPs inhibit the neural enzyme acetylcholinesterase (AChE) in the brain. This leads to the buildup of acetylcholine, causing respiratory failure, seizures, and death. PB functions by inhibiting ~30% of peripheral AChE temporarily, protecting the enzymes so that the body is able to restore sufficient cholinergic function post-exposure. PB does not cross the blood-brain barrier, though it may still indirectly affect toxicity in the brain due to repeated AChE inhibition throughout treatment, or by inhibition of non-target ChE’s in the blood. Gray wolves (Canis lupus), northern sea otters (Enhydra lutris) and salmon (Oncorhynchus spp.) play vital roles in southeast Alaska, yet all are exposed to trace element (TE) bioaccumulation. This study examines dietary patterns and TE accumulation in four northern Lynn Canal wolf populations (Douglas Island, Juneau, Gustavus, and Pleasant Island) using stable isotope analysis (SIA) (δ13C and δ15N) and TE assessments. SIA revealed that wolves in Juneau and Douglas Island primarily consumed terrestrial prey, whereas wolves in Gustavus and Pleasant Island exhibited enriched isotopic signatures indicative of a marine-based diet, particularly …

Background: Ethanol (EtOH) consumption causes hepatocellular mitochondrial depolarization (mtDepo) and mitophagy. Here, the Aims were to develop techniques to characterize sublobular hepatocellular metabolism and the distribution of mtDepo and mitophagy after acute EtOH. Methods: Wildtype and GFP-LC3 transgenic mice were treated with EtOH or vehicle and administered the covalently labeling mitochondrial membrane potential (ΔΨ) reporter, MitoTracker Red (MTR), and/or the non-covalent ΔΨ reporter, rhodamine 123 (Rh123), prior to intravital imaging, liver fixation, or hepatocyte isolation. Mercury contamination in aquatic ecosystems is a pressing environmental and public health concern, particularly in urban areas where industrial runoff and wastewater contribute to heavy metal accumulation. This study investigates mercury presence in fish from Richland Creek in Nashville, TN, by analyzing histological changes in liver and muscle tissues. Mercury bioaccumulates in aquatic food chains, potentially leading to physiological damage in fish and posing health risks to humans who consume contaminated seafood. 5-Fluorouracil (5-FU) disrupts intestinal cells and causes dysbiosis in the gut microbiota. This study explores the potential of Bacillus coagulans-198 (BC198) to mitigate gut microbiota imbalance and mucositis caused by 5-fluorouracil. L-glutamine is used to alleviate mucositis, and this study found that BC198 exhibits protective effects on the gut, including maintaining a healthy microbiota and reducing intestinal inflammation, regardless of whether L-glutamine is used in combination. Therefore, it can help reduce the deterioration of the gut environment caused by 5-fluorouracil. BC198 can be provided to cancer patients to prevent severe side effects, thereby improving their treatment outcomes and nutritional …
