The effect of ethyl alcohol (10%) on the postnatal development of cerebellar cortex was studied using histological and morphometric methods. The external granular layer was much increased in thickness in alcohol fed rat than those of the control while the molecular and granular layer were reduced. The cells of themolecular layer showed neuronal degeneration and many spaces were present in this layer esp < /strong>ecially late postnatal. The Purkinje cells failed to reach their final position up the tenth day and still arranged in more than one row. The degenerative changes appeared at five days and increased with age. The extent …

Trimethyltin (TMT) is a neurotoxin occurring in the environment. Exposure to (TMT) is known to destroy specific neuronal components of the hippocampus in the rat and to cause clinical symptoms in exposed humans, including mnemonic deficits, that indicate hippocampal involvement. In addition to hippocampal cell loss TMT causes significant increases in cholinergic markers such as acetylcholinesterase (AChE) stain density and choline acetyltransferase (ChAT) activity in the hippocampus of rats. However, despite these observations the effect of TMT on hippocampal cholinergic system has not been investigated in detail. Several industrial and environmental chemicals cause distal and/or central neuropathy among other diverse toxic effects. Spague-Dawley derived rats were fed doses of 2,5-hexanedione, acrylamide, tri-o-tolyl phosphate, Leptophos and methylmercury via gavage. The dose levels and administration periods were established in previous experiments designed to assess clinical neuropathy using rats trained to walk on a rotorod apparatus fitted with an electrode floor. After intravenous injections of 3H-Tryptophan, whole rat brain homogenates were analyzed using liquid scintillation and spectrofluorometric techniques for levels of tryptophan, serotonin and 5-hydroxyindoleacetic acid. The eggs of the Japanese medaka (Oryzias latipes) were employed in a number of studies to develop a routine, standardized assay which can assess the acute and sublethal impacts of individual toxicants and complex mixtures. The eggs of this Cyprinodont minnow were topically treated with each toxicant or mixture dissolved in membrane permeable dimethylsulfoxide (DMSO) solvent. Nine chemicals were initially evaluated with two, pentachlorophenol (PCP) and tributyltin chloride (TBTCl), subsequently selected for extensive evaluations of salinity tolerance and the accuracy of the topical exposure method for representing a typical immersion exposure. This study describes morphological changes in the lung at several time intervals following a single injection of 1-Nitronaphthalene (100 mg/kg intraperitoneal) in male Sprague-Dawley rats using transmission and scanning electron microscopy. Dilation of the smooth endoplasmic reticulum and mitochondrial distension in the nonciliated bronchiolar (Clara) cells was apparent as early as 1 hour after injection, while adjacent ciliated cell showed minimal changes. At 6 hours, when respiratory distress was apparent in the animals, Clara and ciliated cells exhibited chromatin clumping, loss of mitochondrial cristae and cytoplasmic vacuolization. Nitrogen dioxide (NO$\sb2$), an air pollutant produced by burning fossil fuels and a component of cigarette smoke, is thought to contribute to the pathogenesis of pulmonary diseases, such as emphysema. To gain information on the mechanism by which NO$\sb2$ damages the lung, in vitro exposures of $\alpha\sb1$-proteinase inhibitor ($\alpha\sb1$-PI), elastin, bovine serum albumin (BSA), human serum albumin (HSA) and synthetic poly-L-lysine were performed. A genetic deficiency of $\alpha\sb1$-PI predisposes humans to emphysema and NO$\sb2$ has been hypothesized to damage $\alpha\sb1$-PI, which would leave proteases such as human neutrophil elastase, (HNE) free to attack lung structural proteins. This study examined biological mercury removal from soil using mercury-resistant bacteria in soil microcosms. Mercuric chloride was used to artificially contaminate Kidman soil to mercury concentrations of 5 ppm and 10 ppm. Soil moisture content was maintained at three levels, 20%, 30% and 50%. Mercury resistant-bacteria were added to soil samples and the mercury removal rate was compared to control samples without added bacteria. Mercury removal rate was initially enhanced by the addition of bacteria. After 30 days, no difference was observed between samples and controls with initial mercury concentration of 5 ppm when soil moisture content was 20%. At …

In this study, 28 volatile organic compounds (VOCs) emitted from the Linden Roselle Sewerage Authority Plant (LRSA) were monitered. The determination by gas chromatography (GC) is presented. This work emphasizes the collection of whole air samples, using a canister-based sampling system with cryogenic concentration. An appropriate temperature program was selected to obtain the best sample separation. The relative merits of eight different coal fly ashes for use in the stabilization/Solidification of a waste containing both heavy metals and pentachlorophenol were investigated. Also studied, was the possible use of either untreated or treated (with hexade-cyl ammonium bromide) clay to enhance the stabilization of the organic. The sample composition was : 62% foundry baghouse dust (or 60% if 2% clay as added to the sample), 4.5% cement, 14% coal fly ash, 19.5% water. Aflatoxin B1 (AFB1), a secondary metabolite of Aspergillus flavus and Aspergillus parasiticus, is known for its potent carcinogenicity and immunosuppressive effects. It is also known that AFB1 toxicity appears in different degrees in different animal species and strains.
The present study was performed to reveal the involvement of the hypothalamus-pituitary-adrenal gland (HPA) axis in the immunosuppressive effects of AFB1 on C57BL/6 mice. Splenic lymphocytes were assayed to investigate their phenotyping using flow cytometry, proliferative response against mitogen and allogenic lymphocytes, cytolytic cell activity, and IL-2 production. The objective of this research is to develop a method to degrade refractory Tar materials, higher molecular weight 3-5 ring Poly Aromatic Hydrocarbons (PAH's) or degrading these highly refractory PAH structures so that they are amenable to further treatment by biological or chemical methods. The process utilized to achieve this is to treat contaminated soils or tar materials with hydrogen peroxide or ozone plus ultraviolet light. Analysis of 27 specific PAH's was done by gas chromatograph (Flame Ionization Detection) for the reactants and products. The contaminated soil showed significantly lower levels of these PAH's after treatment.

Moreover, B(a)P has often been used as an indi¬cator for the carcinogenic potency of environmental matter. Formaldehyde has been shown in the laboratory to play a critical role in the chemistry of polluted air. The method which we discuss in this paper was evaluated under practical field conditions in the Northern New Jersey Staten Island Urban Air Toxics Project. Formaldehyde in ambient air is concentrated and derived by passing a known quantity of air through a silica column impregnated with 2,4-dinitrophenylhydrazine (DNPH). The silica is eluted with acetonitrile and the concentration of formaldehyde is determined by High Performance Liquid Chromatograph (HPLC) with UV detection. In mid 1987, the organochlorines (O/Cs) DDT, dieldrin, heptachlor and chlordane were deregistered for agricultural and horticultural use in Australia. These chemicals had been used widely, and land on several hundred Western Australian farms, particularly where potatoes had been grown, was contaminated with chemical residues. Livestock grazing pastures on such land was also contaminated, and our beef exports were threatened. In 1988, the Western Australian Department of Agriculture started a two-year project to collect information from sheep which were grazed on land that had previously been treated with O/Cs, particularly dieldrin. Processes affecting compartmentalization, biogeochemical cycling, and transport of trace metals in ecological systems must be elucidated if the impact of trace metal releases on biota are to be understood. Because elemental cycles are often closely linked to hydrologic processes, freshwater and marine systems serve as both vectors and sinks for trace metals in the environment. Three phenol degraders were isolated from the mixed liquor of the Livingston, NJ municipal treatment plant. Shaker flask experiments using one of the degraders (a Pseudomonas sp.) resulted in the determination of its kinetic rate constants. The second phenol degrader (a Xanthamonas) lost its phenol degrading ability while being stored, and its kinetic rate constants could not be determined. The third phenol degrader (a Pseudomonas cepacia) formed flocs which interfered with the spectrophotometric determination of biomass and therefore its kinetic rate constants also could not be determined.

The purpose of this thesis is to study the thermal desorption behavior of toxic organic compounds, specifically o-xylene, p-xylene, ethyl benzene, p-chlorotoluene, and anthracene, from soil with respect to temperature and time, and to develop a mathematical model that will describe this desorption behavior. A method developed by Becher and Bjorseth for analysis of PAH metabolites in urine was employed to investigate human exposure to benzo(a)pyrene (BaP) - an ubiquitous environmental carcinogen. Preliminary results are presented showing the relationship between exposure to BaP and urinary elimination. Although the correlation between the two variables is not statistically significant, there appears to be a positive association with selected exposure varibles such as smoking. The identification of an association may establish urinary BaP as a marker of exposure.
However, detailed study of the analytical procedure indicated that recoveries with this method were as low as 3-8% for …

X-ray flourescence (XRF) and atomic absorption spectrophotometry (AAS) measurements for manganese, iron, copper, and zinc were compared for 96 samples of 21 foods from different sources. Correlation coefficients were 0.94 for manganese, 0.99 for iron, 0.93 for copper, and 0.91 for zinc for XRF vs. AAS determinations.

Nitrous oxide (N2O) oxidizes vitamin B12. disrupting deoxyribonucleic acid (DNA) synthesis. Occupational exposures to subanesthetic levels of the gas have been documented that may result in suppressed proliferative cell activities. Male CD-I mice were exposed to 0, 50, 500, and 5000 parts of N2O per million parts of air (ppm) for 6 hr/day, 5 days/week for 2 and 13 weeks. Splenic lymphocytes were assayed for responsiveness to mitogens and for the ability to produce interleukin-2 (lL-2) . Effects of Fe and Se status on GSHPx activity and lipid peroxidation in liver and intestinal mucosa were studied. Rats were Se and Fe supplemented (+Se+Fe), Se-deficient and Fe-supplemented (-Se+Fe), Se supplemented and Fe overloaded (+Se++Fe) by intramuscular injection, or Se deficient and Fe overloaded (-Se++Fe) for 20d. Fe overloaded tissues had more Fe, but hemoglobin was unaffected. Liver and mucosal GSHPx activity was low in Se-deficient rats. Thiobarbituric acid reactive substances (TBARS) were higher in Fe overloaded and -Se++Fe vs +Se++Fe tissues. Mucosal TBARS was higher in -Se++Fe rats gavaged with CBrCl3. This study is undertaken from the standpoint of water pollution evaluation in bioethanol technology, using immobilized yeast cells. Evaluation of water pollution caused by immobilized yeast fermentation is the main objective of this research.
The pollution in water remaining at the end of ethanol production, which is normally discharged in the wastewater stream, has been analyzed by the measurement of biological oxygen demand (BOD), since BOD measurement is one of the widely used parameters for water pollution evaluation.
