Cell-Mediated Cytotoxicity Of Jensen Tumor In Sprague-Dawley Rats, Theodore D. Masek Sep 1976

Cell-Mediated Cytotoxicity Of Jensen Tumor In Sprague-Dawley Rats, Theodore D. Masek

Loma Linda University Electronic Theses, Dissertations & Projects

An in vitro system for the study of the cell-mediated immune response to Jensen Sarcoma (JS) is described. The tumor grew unrestricted and lead to the demise of 80% of successfully challenged Sprague-Dawley rats (persistors) while 20% of challenged rats (regressors) destroyed the tumor by an unknown mechanism(s). Lymphocytes from both persistors and regressors were shown to be cytotoxic for tumor cells in vitro. Pretreatment of tumor target cells with "immune" serum from persistors or regressors had no effect on the in vitro cytotoxicity of lymphocytes from tumor-challenged rats. Pretreatment of lymphocytes with irmnune serum, however, was shown to reduce …

Concurrent Infection Of Trypanosoma Musculi Kendell, 1906, And Schistosoma Mansoni Sambon, 1907 In Mice, Berhane Andeberhan

Loma Linda University Electronic Theses, Dissertations & Projects

The possible effects of ablastin on Schistosoma mansoni were investigated by (1) establishing a concurrent infection of Trypanosoma musculi and Schistosoma mansoni in mice, and (2) by obtaining Trypanosoma lewisi immune rat serum (ablastin) and injecting it into S. mansoni infected mice. These procedures were followed on days 4, 14 and 28 of S. mansoni infection. For the groups that were given immune rat serum, controls were kept into which normal rat serum was injected.

Ablastin does not have any effect on the development or morphology of the S. mansoni worms as tested in this study. Although ablastin inhibits glycolytic …

Comparison Of Two Techniques For Enumeration Of Human B And T Lymphocytes, William F. Forsythe Jr. Jun 1976

Comparison Of Two Techniques For Enumeration Of Human B And T Lymphocytes, William F. Forsythe Jr.

Loma Linda University Electronic Theses, Dissertations & Projects

The assay of Bursa-derived and Thymus-derived lymphocytes (B and T cells) in man has become increasingly useful during the past few years in normal and in clinical conditions. There has been quite a range reported in the literature in the percent of B and T cells in normal subjects by different investigators.

The purpose of this study was to see if monocyte contamination was one cause of non-specific rosette formation. Twenty-one subjects were used to compare the technique involving trypsin and neuraminidase treatment of erythrocytes and the technique of glass wool removal of monocytes. A peroxidase stain was used to …

Fixation Of Dormant Tilletia Teliospores For Thin Sectioning, John Scott Gardner Apr 1976

Fixation Of Dormant Tilletia Teliospores For Thin Sectioning, John Scott Gardner

Theses and Dissertations

Dormant Tilletia caries teliospores in fixative solution or distilled water were frozen onto specimen chucks of an FTS Sorvall- Christensen frozen thin sectioner and cut or fractured at various temperatures (-20 to -75 C) and thickness settings (10, 15, 20, and 25 um). Cytoplasm of dormant spores was well preserved and organelles were found to differ from those of germinated spores in morphology. Irregular electron dense and electron opaque areas of lipid bodies were sometimes evident in dormant and germinating Tilletia caries teliospores. When dormant teliospores were sectioned in buffered fixative, lipids had an appearance similar to lipids in thin …

Root Rot Of Subterranean Clover In W.A, G C. Macnish, M. J. Barbetti, D. Gillespie, K. Hawley Jan 1976

Root Rot Of Subterranean Clover In W.A, G C. Macnish, M. J. Barbetti, D. Gillespie, K. Hawley

Journal of the Department of Agriculture, Western Australia, Series 4

Root rot of subterranean clover has occurred sporadically in the south west of Western Australia for a number of years.

In most seasons the disease has affected the clover paddocks of only a few farms, but in 1973 there was widespread pasture decline due to root rot in the South-West and south coastal districts.

At present the most promising approaches for minimising the effect of root rot appear to be the use of cultivation techniques and eventually the use of resistant varieties, or other pasture species.

Regulation Of Expression Of The Bacteriophage T4 Genes 45, 44, And 62, Margaret G. Bowles Jan 1976

Regulation Of Expression Of The Bacteriophage T4 Genes 45, 44, And 62, Margaret G. Bowles

MUSC Theses and Dissertations

The functions of T4 genes 45, 44, and 62 are required for phage DNA replication. Some evidence suggests that in T4-infected E. coli these genes are cotranscribed in the 45 to 44 to 62 direction. In this study regulation of expression of genes 45, 44, and 62 has been examined by using SDS-polyacrylamide gel electrophoretic assays to analyze the protein products synthesized in T4-infected E. coli at various times after infection under different conditions. Genes 45, 44, and 62 were shown to be cotranscribed by the criterion of translational polarity, but the degree of polarity shown by gene 44 nonsense …

The Effect Of Urea On The Sedimentation Coefficient Of The Curly Top Virus Dimer, Allen H. Smith Jan 1976

The Effect Of Urea On The Sedimentation Coefficient Of The Curly Top Virus Dimer, Allen H. Smith

Undergraduate Honors Capstone Projects

The purpose of this project is to investigate the effect of increasing concentrations of urea on the sedimentation rate of the curly top virus dimer as measured by ultracentrifugation techniques. In general, urea causes the breaking of hydrogen bonds in macromolecules--in the case of the virus, increased concentrations of urea should cause changes in the configuration of the virus and may possibly cause a separation of the dimer into monomers. From previous centrifugation studies, we know that the dimer has a characteristic sedimentation coefficient(S) of about 80 Svedbergs; the monomer has an S value of about 55 Svedberg units. Increased …

Lipid Metabolism During Bacterial Growth, Sporulation, And Germination: Differential Synthesis Of Individual Branched And Normal-Chain Fatty Acids During Spore Germination And Outgrowth Of Bacillus Thuringiensis, Kenneth W. Nickerson, Lee A. Bulla Jr., Timothy L. Mounts Dec 1975

Mounts

Papers in Microbiology

The biosynthesis of individual branched- and normal-chain fatty acids during Bacillus thuringiensis spore germination and outgrowth was studied by comparing pulsed and continuous labeling of these fatty acids with [U- 14C ]acetate. The relative specific activity of each fatty acid varies with time as the cell progresses through outgrowth. However, fatty acid synthesis does occur in two distinct phases. Upon germination, acetate is incorporated only into the iso-isomers i-C13, i-C14, and i-C16; no normal or anteiso synthesis occurs. Subsequent to T30, the full complement of branched- and normal-chain homologues is formed …

Lipid Metabolism During Bacterial Growth, Sporulation, And Germination: An Obligate Nutritional Requirement In Bacillus Thuringiensis For Compounds That Stimulate Fatty Acid Synthesis, Kenneth W. Nickerson, Lee A. Bulla Jr. Aug 1975

Bulla Jr.

Papers in Microbiology

The regulation of fatty acid biosynthesis by compounds that are required for growth of Bacillus thuringiensis was investigated using an in vivo assay developed to measure fatty acid synthesis in germinating spores. A minimal glucoseammonium- salts medium does not support growth even though previous radiorespirometric studies have shown B. thuringiensis to possess intact tricarboxylic acid and Embden-Meyerhof-Parnas pathways. Abundant growth does occur, however, when this medium is supplemented with either glutamate, aspartate, citrate, thiosulfate, cystine, or ethylenediaminetetraacetic acid. Cells held under non-growing conditions incorporate acetate into fatty acids; fatty acid synthesis is stimulated by the compounds that permit growth. These …

B And T Lymphocytes In Human Pregnancy, Ted Fong Jun 1975

B And T Lymphocytes In Human Pregnancy, Ted Fong

Loma Linda University Electronic Theses, Dissertations & Projects

The success of the fetus as a transplant has been attributed to an immune depression in the mother. It has not been shown whether this purported depression was due to fewer T-lymphocytes or a dysfunction of these cells. T cells were measured by the rosette technique using neuraminidase treated sheep red blood cells (SRBC). B cells were measured using a SRBC-hemolysin-guinea pig complement indicator system. The total leucocyte count in the pregnant group was significantly higher than the control group but this was not reflected in the absolute number of lymphocytes. Compared to non-pregnant women, the pregnant group did not …

Detection Of Neonatal Calf Diarrhea Virus (Ncdv) And Human Infant Reolike Diarrhea Virus, Martin Wyatt Peterson May 1975

Detection Of Neonatal Calf Diarrhea Virus (Ncdv) And Human Infant Reolike Diarrhea Virus, Martin Wyatt Peterson

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

The purpose of this study was to develop a diagnostic test and conduct a survey for the neonatal calf diarrhea virus (NCDV) and human infant reolike diarrhea virus. Two immunologic methods were developed in this investigation.

Immune electron microscopy (IEM) and the fluorescent viral precipitin test (FVPT) are the methods used to detect NCDV and the human virus. Both methods are based upon the principle that viral aggregates form when the virus is reacted with anti-NCDV antibody. Aggregates in the IEM method are negatively stained and observed with the use of an electron microscope. Fluorescein labeled antibody is used in …

Phomopsis : The Causal Fungus Of Lupinosis, P Mcr Wood, A. G. P. Brown Jan 1975

Phomopsis : The Causal Fungus Of Lupinosis, P Mcr Wood, A. G. P. Brown

Journal of the Department of Agriculture, Western Australia, Series 4

It is now well known that lupinosis of sheep and cattle follows ingestion of lupin stubble colonised by the fungus Phomopsis rossiana. The fungus infects green lupins but persists on the stubble and under certain conditions apparently produces a toxin which causes the disease.

As Phomopsis is now widespread on lupins in Western Australia its ecology has been studied in detail as a basis for possible control measures.

This article outlines what is known so far of the disease and its control.

Lupinosis research is co-ordinated by the Department of Agriculture's Lupinosis Committee, on which are represented all groups working on the disease. These include veterinary surgeons and pathologists, chemists, plant pathologists and animal husbandry specialists.

Chairman of the Committee is Dr M. R. Gardiner, Chief of the Department's Animal Division and a veteran of lupinosis research.

In this article Dr Gardiner summarises the current state …

Nonpathogenic Free-Living Amoebae In Arkansas Recreational Waters, Leon W. Bone, David A. Becker Jan 1975

Nonpathogenic Free-Living Amoebae In Arkansas Recreational Waters, Leon W. Bone, David A. Becker

Journal of the Arkansas Academy of Science

Selected recreational waters of Arkansas were sampled for pathogenic free-living limax amoebae. Water quality parameters were determined for correlation with amoebic population densities and species diversity. Cultural criteria and animal inoculation revealed no pathogenic strains. The possibility of introduction and/or induction of pathogenic amoebic strains by environmental factors requires further ecological investigations.

A Statistical Approach To The Study Of The Ecosystems Of Seven Ponds In East-Central Illinois, Gregory Lee Orr Jan 1975

A Statistical Approach To The Study Of The Ecosystems Of Seven Ponds In East-Central Illinois, Gregory Lee Orr

Masters Theses

Gross primary productivity, heterotrophic bacterial numbers, and net phytoplankton densities of seven ponds in Coles County, Illinois, were studied in relation to physical, chemical, and biological habitat variables (light intensity and duration, turbidity, water temperature, pH, dissolved oxygen, sulfur, nitrogen, phosphorus, production, bacteria, and phytoplankton). Ten observations were made for each pond (except where otherwise noted) from 17 June through 25 August 1974. Stepwise multiple linear regression analyses of the data were used in order to determine those environmental factors which were important in predicting (i.e., significantly correlated with) bacterial and phytoplankton densities, and production. A multiple linear regression equation …

The Mechanism Of Action Of 2,4-Diamino-6-(3,4-Dichlorophenylacetamido) Quinazoline On Cryptococcus Neoformans, Harriett Camilla Emanuel Jan 1975

The Mechanism Of Action Of 2,4-Diamino-6-(3,4-Dichlorophenylacetamido) Quinazoline On Cryptococcus Neoformans, Harriett Camilla Emanuel

MUSC Theses and Dissertations

The effect of varying concentrations of 2,4-Diamino-6-(3,4-Dichlorophenylacetamido) quinazoline (DDPQ) on viable cells of cryptococcus neoformans at 37 C reveals that the compound is fungistatic at concentrations in the range of 1 to 6 ug/ml and fungicidal at concentrations above 6 ug/ml. The effect of DDPQ on C. neoformans can be measured within 30 minutes after incubation with DDPQ. Manometric studies show a decrease in oxygen uptake in both exogenous and endogenous flasks after 120 minutes. Flame spectrophotometry shows no ion leakage of sodium or potassium which suggests that no cell membrane damage is involved. Incorporation studies show an almost immediate …

Lipid Metabolism During Bacterial Growth, Sporulation, And Germination: Kinetics Of Fatty Acid And Macromolecular Synthesis During Spore Germination And Outgrowth Of Bacillus Thuringiensis, Kenneth W. Nickerson, John De Pinto, Lee A. Bulla Jr. Jan 1975

Bulla Jr.

Papers in Microbiology

The timing and kinetics of fatty acid synthesis are delineated for Bacillus thuringiensis spore germination and outgrowth by analyzing [U-14C ]acetate and [2-3H ]glycerol incorporation into chloroform-methanol-extractable and trichloroacetic acid-precipitable lipids. In addition to measurement of pulsed and continuous labeling of fatty acids, monitoring the incorporation of radioactive phenylalanine, thymidine, and uridine from the onset of germination through first cell division provides a profile of biochemical activities related to membrane differentiation and cellular development. Upon germination, ribonucleic acid synthesis is initiated, immediately followed by rapid and extensive fatty acid synthesis that in turn precedes protein, deoxyribonucleic …

A Photomicrographic Guide To The Saprolegniaceae Of Illinois, Ronald L. Butler Jan 1975

A Photomicrographic Guide To The Saprolegniaceae Of Illinois, Ronald L. Butler

Masters Theses

There is a paucity of information concerning the occurrence and distribution of the Saprolegniaceae of Illinois. These fungi, commonly called "water molds", are of particular interest as many are potential parasites of fish, crustaceans, and other aquatic plant and animal life. It is of considerable interest to students of the fungi and of practical value to the fisheries biologists and environmentalists to have available a means of identification for these organisms.

This paper provides a descriptive and photomicrographic account of the Saprolegniaceae of Illinois. Included herein are dichotomous keys to the genera of this family as well as generic descriptions …

Metabolic Capabilities Of Sulfur Oxidizing Bacteria And Their Role In Water Pollution, M. I. H. Aleem

KWRRI Research Reports

This report describes investigations into the physiology of microorganisms that are commonly involved in the oxidation of inorganic sulfur compounds. The metabolic activities of these bacteria play a potent role in several fields of economic importance such as strip mining operations, water pollution, corrosion, metallurgy, petroleum technology and soil fertility processes.

The oxidation of inorganic sulfur compounds was studied in the chemolithotrophs Thiobacillus denitrificans, Thiobacillus A2, Thiobacillus neapolitanus, and a photolithotroph Rhodopseudomonas palustris. Cell suspensions from all of the thiobacilli catalyzed completely the oxidation of sulfide, thiosulfate, and sulfite to sulfate. The oxidation of thiosulfate in R. palustris …

Accumulative Coliform Population In A Five-Mile Section Of The Cibolo Creek, Norman Merrill Brown Aug 1974

Accumulative Coliform Population In A Five-Mile Section Of The Cibolo Creek, Norman Merrill Brown

Theses & Dissertations

A study of the accumulative coliform population in a five-mile section of Cibolo Creek near Schertz, Texas.

The greatest danger associated with water is that it may recently have been contaminated by sewage or by human excrement. Therefore, the protection of public water supplies from intestinal contamination is a necessary obligation of the public health authorities. If there happens to be a break in the protective chain (adequate treatment, disinfection, and protection of water supply), a chance exposure to fecal contamination could trigger an explosive outbreak of disease within a community. Modern technology and more sophisticated bacteriological methods have made …

Concurrent Infections Of Trypanosoma Equiperdum And Trichinella Spiralis In Mice, Pauline A. Nembhard Aug 1974

Concurrent Infections Of Trypanosoma Equiperdum And Trichinella Spiralis In Mice, Pauline A. Nembhard

Loma Linda University Electronic Theses, Dissertations & Projects

In concurrent infections of Trichinella spiralis and Trypanosoma equiperdum, the survival times vary in the early stages of the Trichinella infection. On Days 1 and 4, the mean survival times are significantly increased, and on Days 3 and 7 they are markedly decreased. This decrease is probably due to intestinal tissue invasion. Mice infected with trypanosomes after the 7th day of the Trichinella infection have survival time increases to a significant point and a steady leveling off to Day 21. It is suggested that the antibodies produced against the Trichinella worms slow down multiplication of trypanosomes and increase the …
