Endophytic actinobacteria are a diverse group of Gram-positive bacteria with high G+C content, which inhabit plants’ internal tissues, and represent a significant yet underexplored reservoir of bioactive secondary metabolites. In host plants, a remarkable repertoire of metabolites, including broad-spectrum antibiotics, anticancer, antiviral, antifungal, antioxidant, and immunomodulatory compounds, is synthesized by these microbial allies, carrying significant medicinal and agricultural potential. These endophytes, in contrast to their soil relatives, have developed unique biosynthetic gene clusters (BGCs) strongly influenced by plant-associated factors, leading to the production of structurally novel metabolites. Seroprevalence And Risk Determinants Of Toxoplasma Gondii Infection In Pregnant Women In Pakistan: A Cross-Sectional Study, Tajul Haram, Kamran Khan

Toxoplasma gondii is a globally important zoonotic parasite, and maternal infection during pregnancy may cause miscarriage, congenital anomalies, or severe neonatal complications. This cross-sectional study was conducted among 367 pregnant women attending hospitals in District Dir Upper, Khyber Pakhtunkhwa, Pakistan, to evaluate the seroprevalence of T. gondii and associated risk factors. Blood samples were analyzed using Immunochromatographic Test (ICT) for initial screening, and positive cases were confirmed by Enzyme-Linked Immunosorbent Assay (ELISA). Overall, 34.6 % (127/367) of women tested positive. HIV/AIDS remains a significant public health problem internationally, with young adults being especially at risk because of behavioral and knowledge deficits. In Pakistan, cultural inhibitions and suboptimal health education are the main reasons for limited awareness, particularly among university students. This study had the objectives of determining the knowledge level about HIV transmission and prevention among undergraduate students in Kohsar University Murree (KUM) and to investigate correlations with salient demographic variables. Polyhydroxyalkanoate (PHA) aliphatic polymers however efficiently biodegradable, still pose a contradiction with the principles of green chemistry. Use of bacterial strains capable of degrading synthetic plastics for the production of biopolymers i.e., PHAs may aid in reduction of plastic pollution. Bacterial strains PWA; Bacillus subtilis (MH142143), PWF; Bacillus tequilensis (MH142145), and PWG; Bacillus safensis (MH142146) were grown on waste shredded PET and PET flakes. The study aimed to evaluate antibiotic susceptibility patterns and extended-spectrum beta-lactamase prevalence in Escherichia coli isolated from urinary tract infections. Total fifty (50) samples of E. coli were obtained from urinary tract infection specimens in Hayatabad Medical Complex and private clinics in Peshawar, Pakistan. The present study obtained 50 E. coli isolates from 100 UTI specimens. The plates were incubated at 35 to 37°C for 24 to 48 hours. The interleukin 2 receptor alpha (IL-2Rα), which is integral to the functioning of the cytokine interleukin 2, has recently been found to be present in different cell types besides just lymphocytes. Previous studies done by our laboratory showed that several other non-lymphoid cells including smooth muscle cells express IL-2Rα. Vascular smooth muscle cell (VSMC) deficient in IL-2Rα exhibited increased proliferation, decreased size, and hypodiploid DNA content when compared to wild type (WT) cells. Previous studies also suggested that VSMC proliferate in response to IL-2 and that IL-2 increases following interventions to stimulate intimal hyperplasia in vivo. These findings, in total, …

Here, I have investigated the dynamics of calcium entry pathways, namely, store-operated calcium entry and store-independent calcium entry using a variety of cell types such as Jurkat T cells, HEK-293 cells and murine macrophages. Using fluorescence calcium imaging and selective inhibitor compounds, I have investigated the contributions of Orai and related calcium channels to calcium entry in both immune and non-immune cell types. Feathers are a waste product of the poultry industry and are considered a major pollutant of the environment. Soil associated with poultry farms is a rich source of indigenous bacteria that are involved in biodegradation of keratin of the feathers of Gallus gallus domesticus. The present study was designed to understand the role of microbes in the degradation of a feather’s keratin and its possible utilization as a bio-protein for plants. Hot springs situated at high altitudes is a kind of remarkable ecosystem for the exploration of microbial flora. It was hypothesized that hot springs can harbor bacteria with plant growth-promoting and exopolysaccharides (EPS) producing ability that can favour the growth of plants. For the investigation of this hypothesis, seven EPS producing bacterial isolates were isolated from a water sample of hot water spring of Tattapani, Azad Kashmir and characterized morphologically and biochemically. Peganum harmala L. is a wild herbal plant rich in active compounds, and is among the most used plants in local medicine. The purpose of this study is to contribute to the evaluation of some biological activities of alkaloids extract of P. harmala roots. Using the classical method of extraction of β-carboline alkaloids, the yield of alkaloids in the roots was estimated at 2.34 %. Adenovirus is a human pathogen that causes cold like symptoms in healthy individuals, which could be lethal in pediatrics and the immunocompromised. In polarized epithelia, adenovirus uses the eight-exon isoform of the Coxsackievirus and Adenovirus Receptor (CAREx8) on the apical surface, to facilitate infection. CAREx8 protein expression level is regulated by the scaffolding protein MAGI-1, using two of its PSD95/Dlg-1/ZO-1 (PDZ) domains: PDZ2 and PDZ4. We previously showed that CAREx8 is degraded after interaction with PDZ4 and rescued following interaction with PDZ2. The human immunoglobulin heavy chain gene locus (IGH) has two 3 prime regulatory regions (3’IGHRR), each containing three enhancers (hs3, hs1.2, hs4). In animal models, the 3’IghRR regulates IgH expression and class switch recombination (CSR) to different Ig isotypes. The 3’IGHRR hs1.2 enhancer in humans is polymorphic in that an invariant sequence (IS) can be repeated one to four times in tandem. The hs1.2 polymorphism is of interest due to its association with several human autoimmune disorders and its potential sensitivity to exogenous substances such as 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD or dioxin). To ensure universal health care, the World Health Organization recognized the significance of complementary and alternative medicines (CAM) and recommended the use of natural herbs and plants bearing therapeutic potential and fewer adverse effects. Therefore, Artemisia annua herb was evaluated for its antibacterial potential and therapeutic efficacy against Staphylococcus aureus, Streptococcus and Escherichia coli both in vitro and in vivo. Artemisinin was extracted from Artemisia annua by chemical treatment. Pseudomonas aeruginosa spp are the most prevalent bacteria that cause nosocomial infections in hospitals. Most antibiotics, including novel new β-lactams, are already resistant to them, and they can become resistant during treatment, which can make the treatment fail. P. aeruginosa isolates from ICU patients who had Per-1 and VEB-1 were the main focus of this study. These two ESBLs are the two most common in ICU patients who had them. 50 isolates were gathered from Peshawar's LRH ICU facilities in the year 2021. Total thirty-two strains were isolated from both water and soil samples collected from Naran Valley which is surrounded by the rugged mountains, located in the province of Khyber Pakhtunkhwa, Pakistan. Streak plate method was used to screen the bacterial strains for resistance against selected heavy metals like chromium (Cr), cobalt (Co), copper (Cu), nickel (Ni) and zinc (Zn). Resistance against multiple heavy metals was shown by all the strains, but S15 and W15 were significant as they exhibited a maximum resistance to chromium and copper (1200 µg/ml). This study was performed to evaluate the potential contamination of the Okponha river situated near a dumpsite in Okada, Edo State, Nigeria. Water samples were collected and analyzed for bacteriological and parasitological quality using standard procedures. Isolation and enumeration of bacterial colonies were performed by pour plate technique and the isolated bacteria were identified by standard phenotypic tests. With a longitudinal (2020–2021) completion, a set of 300 urine samples of individuals suspected of UTI was characterized based on the biochemical analysis and Kirby Bauer's disc diffusion method. Throughout, a total of 167 samples revealed UTI positivity by obtaining bacterial growth. Reporting that E. coli (69.4 %) was the most dominant when compared with Klebsiella (14.3 %), Proteus (5.9 %), S. aureus (4.7 %), P. aeruginosa (3.5 %), and Enterobacter (1.7 %), respectively. Respiratory epithelial cells are the initial target for SARS-CoV-2 infection. IFN-γ is known to increase the expression of ACE-2, an initial receptor for SARS-CoV-2, on epithelial cells. This study focuses on examining the effect of IFN-γ for ACE-2, TMPRSS2, and neuropilin-1 on Vero E6 cells using two immunofluorescence methods, namely, direct (membrane) fluorescence method and Cytation5 method. Direct fluorescence was determined using an Accu-Scope and ImageJ analysis. Using this method, significance (p<0.023) was observed only for ACE-2 when Vero E6 cells were treated with IFN-γ. Cytation5 fluorescence was determined using a Bio-tek Cytation5 plate reader. The results showed that IFN-γ significantly increased (p<0.001) the expression of ACE-2, neuropilin-1, and TMPRSS2. These results indicate Cytation5 is a more sensitive method for determining the expression of receptors on Vero E6 cells. The elevated levels of SARS-CoV-2 receptors expression resulting from IFN-γ treatment makes the epithelial cells more susceptible targets of SARS-CoV-2 infection. IFN-γ is most likely provided by innate immune cells in the initial COVID-19 infection, consequently contributing to the severity of disease.
