Microbial persistence in confined environments, such as spacecraft, can pose challenges to astronaut health and habitat maintenance. This project investigates Staphylococcus aureus, a Gram-positive bacterium commonly associated with human microbiota and opportunistic infections, in spaceflight-relevant environmental conditions. Further, this project will assess the influence of the antimicrobial/antioxidant compound quercetin to assess its effects on bacterial growth and virulence under standard Earth gravity and simulated International Space Station (ISS) conditions. This approach will assess whether spaceflight-relevant stressors, including microgravity and radiation, alter treatment efficacy by influencing microbial physiology and antimicrobial susceptibility. While astronauts are still preparing to travel to Mars, scientists must learn how to simulate a more realistic Martian environment. Simulated microgravity presents multiple physiological stressors for astronauts, including bone demineralization, muscle loss, and weakening of the immune system. Bacteria serve as viable model organisms to study physiological changes because of their abundance in the human digestive system and skin. Motility in bacteria plays a significant role in cell survival, but few studies have been conducted on microgravity’s effect on motility phenotypes. While motile strains like Escherichia coli K12 have been studied extensively under microgravity conditions, the effects of simulated microgravity on non-motile strains are largely unknown. Antimicrobial resistance is a growing global concern that necessitates the development of effective strategies for selecting appropriate antibiotics, especially in the context of future long-duration space missions, where infection control and treatment options are limited. This study employs the Kirby–Bauer disk diffusion assay to evaluate the antimicrobial susceptibility of Pseudomonas aeruginosa and other clinically and spaceflight-relevant bacterial pathogens such as Klebsiella pneumoniae, Staphylococcus aureus, and Streptococcus pneumoniae. From Dust to Growth: Microbial Pathways to Extraterrestrial Farming addresses the fundamental challenge of sustaining long-term human habitation on other planets. In the era of the Artemis missions, establishing a cost-effective, bioregenerative food source is critical to reducing logistical dependency on Earth, and mitigating the physiological stressors faced by spacecrew through improved nutrition. This study initially investigated the viability of cultivating Mizuna, a proven ISS model organism, in a substrate of Martian regolith simulant amended with organic compost. Biofilms are collections of bacteria that commonly grow on a variety of surfaces. They are resistant to environmental impacts as bacteria fortify themselves to a surface. Biofilms are commonly found on Earth, but more problematically are also found on spacecraft. On the ISS biofilms have developed in spaces like the workout area, food spaces, and most concerning in the Water Recovery System (WRS). Malate Dehydrogenase (MDH) is a key protein in the citric acid cycle; its dysregulation can have catastrophic impacts, such as neurological and metabolic disorders, and various cancers. MDH catalyzes the reversible oxidation-reduction reaction between malate and oxaloacetate, also reducing NAD+ to NADH in the process. The citric acid cycle is one of the central metabolic pathways, connecting to other essential pathways, such as amino acid and fatty acid biosynthesis, highlighting the importance of understanding how MDH works. Malate Dehydrogenase (MDH) is an essential enzyme that converts malate to oxaloacetate using the reduction of NAD+ to NADH. In humans, MDH plays a role in energy metabolism by catalyzing a key step of the citric acid cycle, important for energy production. Other indirect roles for MDH include gluconeogenesis and reoxidizing NADH from fatty-acid β-oxidation. An important feature of MDH is a flexible loop region that can alter substrate binding and enzyme activity, but its role is not well understood. Harmful algal blooms produced by the diatom Pseudo-nitzschia regularly expose marine mammals along the California coast to domoic acid (DA), a neurotoxin that bioaccumulates in marine food webs. In California sea lions (Zalophus californianus), domoic acid exposure is associated with seizures, hippocampal injury, and maternal-fetal transfer of the toxin that can result in abortion. Bloom events have led to large numbers of stranded sea lions along the California coast. Coral reefs thrive in primarily oligotrophic environments, depending on factors such as temperature and salinity for their stability. While moderate levels of nutrients sustain coral growth, excessive nutrient inputs promote algal blooms which can outcompete corals for space. Over the past century, Maunalua Bay (O'ahu, HI) has experienced dramatic development and increase in population, which has affected the water quality in the nearshore coral reefs of the bay. The human microbiome, a complex ecosystem of trillions of microorganisms including bacteria, fungi, and viruses, represents a critical "gut-diet axis" that significantly influences cancer progression and therapeutic efficacy. While a state of homeostasis supports systemic immunity, dysbiosis is linked to chronic inflammation and can actively promote resistance to standard cytotoxic chemotherapies and modern immune checkpoint inhibitors. Specifically, research indicates that Bacteroides fragilis (ETBF) promotes resistance to 5-Fluorouracil and Oxaliplatin in colorectal cancer by activating Notch1 receptors to inhibit apoptosis and push cancer cells into a resilient, stem-cell-like state. This project investigated the use of engineered bacterial reporter systems to detect cellular stress responses associated with antibiotic activity. While constructs were successfully introduced and tested against known antibiotics, the reporter consistently produced a blue signal across conditions, limiting the ability to distinguish specific stress responses. This suggests issues such as leaky expression, insufficient regulatory control, or lack of specificity in the reporter design. Although results were inconclusive, this work highlights key challenges in developing reliable biosensors and provides a foundation for future optimization. This study measures whether microbial communities in the shallow coastal waters of Virginia Beach are capable of consuming methane in the water column. Using methane enrichment incubations and gas chromatography, we found no measurable methane oxidation of our samples over multi-day periods. These results suggest that these coastal waters may act as net sources of atmospheric methane.

Obesity is of critical concern as excess body fat storage is a comorbidity to various diseases including type-2 diabetes, cardiovascular disease, and cancer. There is growing interest in the role of soil microorganisms in prairie ecosystems, which may be crucial for restoration of the plant community through interspecies interactions within the soil near plant roots. Background: Respiratory viruses are a significant cause of pediatric illness and healthcare utilization. Diagnostic testing for respiratory viruses is generally performed using nasopharyngeal (NP) specimens obtained by trained staff, limiting testing to medical settings. Evaluating additional sampling methods in children that are easier to collect and can be performed outside of medical settings, such as anterior nasal swab (NS) specimens may expand access to diagnostic testing and aid public health surveillance.

Biofilms are structured communities of bacterial cells encased in a self-produced extracellular matrix, composed of polysaccharides, extracellular DNA, and proteins. Bacteria in a biofilm pose significant challenges in healthcare due to their resistance to disinfectants and tolerance to antibiotics. In some species, histone-like proteins were found to be important structural components of the biofilm. Nε-Lysine acetylation is a widespread post-translational modification (PTM) in bacteria, which can alter a protein’s function. Canada Geese (Branta canadensis) are migrating birds. They can transmit antibiotic resistant bacteria when they are traveling. They do this by ingesting antibiotic resistant bacteria that is on the food they eat in one area, and then excrete some of it at the next place they travel to. Retention ponds have increased amounts of antibiotic-resistant bacteria that Canada geese can pick up if they stop there while migrating. Plastics, including microplastics, pose significant environmental threats due to their widespread presence and harmful ecological effects. This dissertation present three distinct, yet interconnected studies. First, a study on microplastic contamination in urban recreational parks was conducted in Al Ain, UAE. Results showed that low-density microplastics were present in 87% of soil samples, with FTIR analysis identifying synthetic polymers such as PE and PP. Additionally, these microplastics were found to negatively affect soil pH and moisture levels, emphasizing the need for enhanced monitoring and management strategies. The Aeromonas hydrophila are gram-negative, opportunistic bacteria that can infect many groups of organisms, mainly fish and amphibians. In channel catfish, A. hydrophila causes fatal bacterial septicemia, leading to massive economic losses for catfish farmers. Therefore, an understanding of its genome will aid in identifying its virulence factors, metabolic pathways and survival mechanisms within the host. This study details the draft genome sequence of A. hydrophila strain S14-452, isolated from channel catfish exhibiting bacterial septicemia during a disease outbreak at a catfish farm.

Bacteriophages are an abundant type of virus with the ability to infect and kill bacteria while using them to replicate through lytic or lysogenic processes. Bacteriophages play a key role in the regulation of bacterial populations in various environments and are being explored as potential tools in phage therapy. The aim of this study was to isolate, characterize, and manually annotate the genome of the bacteriophage MUWow collected from the campus of the Mississippi University for Women in Columbus, MS at GPS coordinates 33.493021 N, 88.419611 W. Widespread antibiotic use has led to antibiotic resistance of infectious bacteria. (Young, 2017) As part of the TinyEarth Program, the lab group collected and tested local soil samples for bacteria with hopes of finding novel antibiotic properties. From the soil samples, a bacteria was isolated, subsequently sequenced, and determined to be Pseudomonas parafulva. P. parafulva is a Gram-negative bacterium that has been known to frequent rice paddies of eastern Asian countries and has been collected in select regions of North America, including that of Cache Valley.
