Aspiration pneumonia is a common mild case of pneumonia, a disease where the lungs are inflamed and filled with fluid, compromising respiration and gas exchange. One common cause of mild pneumonia is bacterial infection via a pathogen known as Mycoplasma pneumoniae. M. pneumoniae causes a variety of respiratory illness- ranging from pharyngitis, tracheobronchitis, mild pneumonia- and in rare cases encephalitis. M. pneumoniae produces the CARDS toxin, which participates in ADP-ribosylation and vacuolization (Kannan et al., 2006)1. The rapid rise of e-cigarette use among young adults has created a significant public health challenge, as the popularity of "vaping" has grown faster than our understanding of its long term cellular consequences. Current research indicates that e-cigarette vapor (ECV) induce high levels of oxidative stress and impair the lungs' ability to clear bacterial pathogens. Furthermore, ECV exposure upregulates specific receptors on the surface of airway cells, essentially making it easier for bacteria to adhere to and infect the lungs. The Effects Of Mycoplasma Pneumoniae Cards Toxin On M2 Macrophage Polarization And Formation, Ria Modi

Macrophages are key cells of the innate immune system, playing essential roles in host defense, tissue homeostasis, and inflammation. Depending on stimuli, macrophages can polarize into distinct functional phenotypes, characterized as classically activated (M1) or alternatively activated (M2) macrophages. M2 macrophages are associated specifically with anti-inflammatory responses, tissue repair, and immune regulation. They contribute to wound healing and can regulate host responses during infection. Community-acquired respiratory distress syndrome (CARDS) toxin, a key virulence factor of Mycoplasma pneumoniae, plays a role in altering host immune responses, but its impact on M2 macrophage function and polarization remains unclear.

The DePauw Nature Park Quarry Bottom is a former limestone quarry that was mined for decades and then lay dormant. Through the years, it has developed from being uninhabited to being populated by a diversity of organisms at every scale of life. All organisms in the Quarry Bottom must withstand the changing conditions of the quarry, including the proliferation of invasive plants that are well-adapted to stress. Stationary Phase Mutagenesis is a process where a bacterium can accumulate mutations in the absence of cell division. This may occur via competent DNA uptake and oxidant DNA damage and repair. Pseudomonas aeruginosa (P. aer.) is the most common cause of ventilator associated pneumonia (VAP) in ICU patients. Different combinations of exoenzymes (S, T, Y, and U) are found in P. aer. strains. ExoY is most often associated with VAP in ICU patients. ExoY infection of pulmonary microvascular endothelial cells (PMVECs) induces cell rounding but does not activate intracellular caspase-3/7 or cause cell death. ExoS and ExoT infection of epithelial cells leads to caspase-3/7 dependent cell death. Previous studies using epithelial cells show that ExoY alleviates the cytotoxic effects of ExoS and ExoT. Small RNAs (sRNAs) are noncoding RNA molecules in bacteria that function primarily to regulate gene expression, including those associated with stress. The Borchert Lab has identified 173 sRNAs and > 500 sRNAs that are responsive to desiccation and carbon starvation (c-starvation), respectively. Prokaryotic transcription involves an RNA Polymerase (RNAP) that transcribes DNA into RNA. RNAP involves sigma factors, which recognize promoters for the initial steps of RNA synthesis. The Borchert Lab has determined that there are significant overlaps between sRNAs expressed in cells subjected to both short-term durations and long-term durations of carbon starvation. Streptomyces bacteria produce antibiotics that inhibit competing microorganisms. We hypothesized that Streptomyces from the rhizosphere (around plant roots) would be more likely to produce antibiotics than those from bulk soil (further away from roots). Soil samples were collected from rhizosphere and bulk soils in transitional and long-term organic fields of corn and alfalfa. Streptomyces from these soils were cultured on glucose-asparagine agar media. Their antibiotic production was evaluated by testing whether they could inhibit the growth of two target Streptomyces strains. No significant effects of soil type (rhizosphere vs. bulk), crop, or management on antibiotic production were observed for one …

To learn the pathway by which the peripheral immune system interacts with dopaminergic neurons and triggers an addiction response, we compared cytokine and chemokine production levels before and after a 20-minute ethanol injection. Since some cytokines and chemokines can cross the blood-brain barrier, their peripheral production may influence neural activity. We had four groups: ethanol, saline, domperidone-ethanol, and domperidone-saline. As domperidone is a dopamine antagonist, we will use it as a control to verify that the change in cytokine production is due to ethanol rather than other factors.

Pacific freshwater fish are less likely than Atlantic freshwater fish to exhibit Enteric Redmouth Disease (ERM), or Yersiniosis as caused by Yersinia ruckeri. Based on the hypothesis that these fish may harbor commensal bacteria that inhibit Y. ruckeri growth, we isolated many kinds of commensal bacteria from fish intestines that we have collected from Alaska and local streams. Using co-culture experiments, we have conducted competitive assays to determine if the bacteria found in the gut biomes of these fish inhibit the growth of Y. ruckeri. These may be potential probiotic organisms that could provide resistance to ERM disease.

Pectobacterium carotovorum is a plant-pathogenic bacterium, causing soft rot diseases in vegetables such as potatoes and carrots. Mainly, this is achieved by the production of plant cell walldegrading enzymes, allowing for invasion of host cells. The general mechanisms underlying the pathogenicity of P. carotovorum are well characterized, but the phenotypic effects of some core metabolic processes are not. An important part of metabolism in bacteria is pyruvate production and utilization as a metabolic intermediate. P. carotovorum has two critical genes— pflB and aceE—that play roles in directing pyruvate metabolism. PflB encodes pyruvate formate-lyase, enabling the bacterium to convert pyruvate into …

René Dubos (1901–1982), a prominent 20th century scientist, humanist, and philosopher, was a prolific author. Affiliated for more than 50 years with The Rockefeller Institute for Medical Research/The Rockefeller University, his studies ranged from antibiotics and the role of bacteria in human diseases to formulating broad ecological and environmental questions affecting human health and disease. Salmonella can colonize plant tissue effectively. Understanding the molecular mechanisms underlying Salmonella’s interactions with plants is essential for developing effective strategies to reduce its impact on human health and agriculture. Effector proteins, known for their role in modulating host-pathogen interactions, have been implicated in the virulence and pathogenicity of Salmonella. However, their specific functions and contributions to Salmonella’s interactions with plants remain largely unknown. Here, we aim to investigate the functions of Salmonella effector proteins during the colonization of plants.

Streptomyces bacteria from soil often produce antibiotics which inhibit growth of other microbes. When comparing Streptomyces’ inhibitory ability between different soils, it may be important to match the pH of the medium used in assays to the pH of the soil they came from. We assayed Streptomyces isolates collected from a long-term agricultural experiment with nitrogen-fertilized and non-fertilized treatments to determine their ability to inhibit growth of other bacteria by producing antibiotics. Average soil pH was 6.8 in fertilized soil and 7.2 in non-fertilized soil. We dotted isolates on a glucose-asparagine agar medium adjusted to different pH values and overlaid …
