Contamination of public recreational water and sand is a human health concern, creating a management challenge for public health officials. Past research has shown that Escherichia coli can persist in sediment and can be washed into recreational water. Currently, beach monitoring practices only utilize E. coli concentrations in water samples to determine beach safety and do not consider potential exposure to sediment/sand concentrations. "This study investigated the impact of climate change, particularly drought, on leaf decomposition and associated fungal and macroinvertebrate communities in freshwater ecosystems. With climate change, streams may experience altered hydrology, leading to shifts in community composition and biogeochemical processes. Fungi decompose organic matter, facilitate nutrient cycling, and support food webs. However, drought can disrupt these processes by reducing water availability, causing declines in fungal diversity and activity. I examined leaf breakdown and associated biota in 13 stream sites across varying hydrologic conditions, from perennial to ephemeral flow patterns. Flow duration varied during the 8-week study. This thesis describes the isolation and characterization of a halophilic iron- oxidizing bacterium from sediments obtained from Lake Magic; an extremely acidic, saline lake located in Western Australia. The purpose for the isolation of novel, halotolerant/halophilic iron-oxidizing bacteria is to aid in metal-mining bioleaching operations. Currently used iron- and sulfur-oxidizing bacteria are extremely sensitive to chloride, which restricts their use in these industries.

Temperature changes affect these organisms by altering their metabolic rate and energy balance. Domestic hot-water systems quietly grow biofilms under elevated temperatures that can serve as shelter to Opportunistic Premise Plumbing Pathogens. Although focus has been placed on microbes in domestic systems, there has been no clear conclusion on the mechanism of survival and persistence of opportunistic pathogens despite various disinfection strategies. In this study, we determined the distinction between showerhead and drip-flow biofilms at the whole-community level and analyzed the coexistence of thermotolerant organisms with opportunistic genera, such as Legionella. "The highly contaminated subsurface matrix of the Baird and McGuire Superfund site is currently threatening the health and safety of the surrounding environment of Holbrook, MA. Contaminants of significant concern due to high concentration are inorganic arsenite and petroleum hydrocarbons, such as naphthalene. Parsons Corporation and the Massachusetts Department of Environmental Protection have implemented a bioremediation pilot to attempt to degrade the hydrocarbons and arsenic with the application of nitrate. Tallgrass prairies have virtually disappeared in many parts of their former range due to the conversion of this ecosystem to farmland. In more recent years there have been efforts to restore these prairies on reclaimed agricultural land. Soil microbes have profound impacts on plant growth and survival and can either promote or inhibit plant dominance. Exotic plants are often strongly invasive because they have escaped their natural enemies, potentially including antagonistic soil microbes. I examined how the invasive shrub Lonicera maackii and a common native tree, Acer negundo, responded to soil microbial communities to determine the role of soil microbes in regulating invasion success. Sinorhizobium meliloti, which is the nitrogen fixing symbiont of Medicago sativa (alfalfa), was used to test the hypothesis that D-chiro-inositol is an intermediate of pinitol catabolism and that pinitol catabolism plays a role in S. melilot s ability to compete for nodule occupancy. An analytical method, involving gas chromatography and mass spectrometry (GCMS), was developed to detect and separate pinitol and D-chiro-inositol. Bile acids are one group of steroids that can be metabolically transformed by human gut anaerobes. One of these transformations is the conversion of primary bile acids to secondary bile acids by 7α-dehydroxylating gut bacteria such as Clostridium scindens, Clostridium hylemonae, and Clostridium hiranonis. These anaerobes metabolize bile acid derivatives via 3α- and 7α-hydroxysteroid dehydrogenases (HSDH). Anastrepha suspensa (Diptera; Tephritidae) is an agricultural pest species causing severe economic damage and is controlled, in part, by applying knowledge of this species' reproduction (e.g., disrupting fertile copulations by SIT). During copulation, males transfer sperm as well as protein rich fluids through an aedeagus and distiphallus, which females then store in three spermathecae and one ventral receptacle. Within the female reproductive tract, the ventral receptacle and the three spermathecae are strategically separated from each other. Bile acids (cholate and chenodeoxycholate) are synthesized from cholesterol in the human liver, and secreted in small intestine for food digestion. However, a portion of these primary bile acids is converted in the colon to toxic secondary bile acids, deoxycholate and lithocholate. Proper treatment of municipal wastewater is important not only for prevention of eutrophication but also for public health concerns. One consideration for wastewater treatment is the presence of organisms carrying antibiotic resistance genes. If these organisms survive the wastewater treatment process and are released into the streams, they can pose significant risks to public health. In addition, their presence also raises concerns for lateral gene transfer events at the wastewater plant. "Insertion sequences are the smallest prokaryotic transposable elements. These genes play a significant evolutionary role by promoting genome plasticity. Insertion sequences are highly diverse elements that have largely been uncharacterized. As such, the ability to accurately identify, annotate, and infer genomic impact of insertion sequences is lacking. The study of new insertion sequences contributes knowledge to their annotation and evolution. Halanaerobium hydrogeniformans is a unique organism with an abnormally high number of insertion sequences.
